This study concerns the effect of plasma from patients with insulin-dependent (type 1) diabetes mellitus (IDDM) on the capacity of normal donor lymphocytes to form rosettes with sheep erythrocytes. Parallel incubations were made of normal allogeneic peripheral lymphocytes with plasma from patients with IDDM and from normal donors. Isolated normal human peripheral lymphocytes were analyzed for their ability to bind IgE as shown by rosette formation with aldehydefixed ox red cells coated with an IgE myeloma protein (Eo'-IgE) as indicator cells. An average of 4.3% of the cells in the lymphocyte preparations of 12 donors formed Eo'-IgE rosettes. Using the method of spontaneous rosette formation the authors studied the effects of morphine and naloxone (a specific morphine antagonist) on T-lymphocytes of man peripheral blood. Treatment of the lymphocytes with morphine led to a diminution of the number of rosette-forming cells. Naloxone did not influence the rosette formation process, but eliminated the specific effect of morphine. The opiate receptors on the thymus-dependent lymphocytes may be an instrument of hormonal regulation of cell interaction processes in individual lymphocyte populations.

The immunological findings of a B-cell prolymphocytic leukaemia forming spontaneous rosettes with sheep (SRBC) and guinea-pig red blood cells (GPRBC) are reported. The B-cell lineage was revealed by Ia-like antigen, surface IgM kappa and lack of cytotoxicity of anti-human T-cell serum. Aroclor 1254 and twelve pesticides of different chemical classes were tested for their in vitro effects on lymphocyte mitogenic responses to PHA, rosette formation with sheep red blood cells (E-rosette) and neutrophil chemotaxis. By using a diluted whole blood culture, butoxide at 10 micrometers inhibit lymphocyte mitogenic response to PHA by 26%. Inhibition by other pesticides were less than 20% (5--20(). Rosettes are widely used in epithelial morphogenesis during embryonic development and organogenesis. However, their role in postnatal development and adult tissue maintenance remains largely unknown.
