Rumen and caecal contents, obtained from slaughterhouse cattle and rumen contents obtained from a fistulated wether were incubated in vitro with ground hay in the presence and absence of, respectively, casein hydrolysate and mucin. Differences in stoichiometry of rumen and caecal fermentations, indicative of reductive acetogenesis in the caecum, were confirmed, except for incubations with free amino acids. Net fermentation end product production was determined after correction for amounts formed in incubations without the substrate. These determined amounts of hay fermentation end products wer

The effect on ruminal motility of NH4- or Na-acetate and Na- or K-lactate of various pH and doses infused into the jugular vein (i.v.) or carotid artery (i.a.) was studied in sheep with ruminal fistula I.v. infusion of NH4-acetate whenever it was accompanied by a considerable increase of the blood NH4 level potently inhibited the amplitude and frequency of ruminal contractions. I. a. administration produced more rapid and more apparent responses and a slightly elevated venous ammonia concentration than did the i.v. infusion. Thus the augmented blood ammonia level not only affected the smooth m

The object of the study was to clarify whether rumination has a bearing on the decomposition of zearalenone in the metabolism of the toxin or not. The experiment was performed using rumen fluid in vitro. Rumen fluid samples containing various amounts of zearalenone were incubated in a carbon dioxide atmosphere at 38 degrees C for two days. The zearalenone content was determined by liquid chromatography after 0, 4, 24 and 48 hours incubation periods. The incubation of 4 hours decreased the toxin content by 11%, that of 24 h by 29% and that of 48 h by 37.5% on average. These results indicate tha

BACKGROUND: Diurnal oscillations have been reported on ruminal prokaryotes, but the daily rhythmicity of eukaryotes remains unknown. This study investigated diurnal oscillations of ruminal prokaryotes and eukaryotes under three different feeding managements and rumen fluid transplantation conditions, aiming to elucidate the regulatory mechanisms influencing the dynamic shifts of rumen microbiome through the daily feeding cycle.
RESULTS: Quantification and profiling of the microbiota of 288 rumen samples collected from lactating dairy cows (n = 12) every 6-h over 48-h feeding cycles under ad l

Rumen fermentation can produce hydrogen sulfide (H2 S), and H2 S can be rapidly absorbed by the intestinal wall in nature. If excessive H2 S was produced in rumen, it might be toxic to ruminants. This article reviews the research progress of toxicity of H2 S, rumen H2 S production pathway and its influential factors to lay a foundation for further research and application of rumen H2 S-producing regulation in ruminant.

A new rumen escapable tool is presented for cattle in prospect of developing medical treatment or supplementing trace elements for disease prevention. This tool consists of a three-layered capsule that dissolves in the lower digestive tract, but not in the rumen. The capsule was manufactured by capsule-forming techniques through the use of liquid surface tension. This method does not involve high-temperature treatment, so the capsule can contain not only lipophilic substances but also hydrophilic or heat-sensitive substances. Furthermore, the capsule has a specific gravity of 1.3 and diameter

Post-operative complications of trocarisation and rumenotomy are the most common causes of peritonitis associated with a rumen disorder. Since horn injury leading to rumen perforation has not previously been reported in the literature, two cows with this condition are reported. Small superficial skin lesions were observed in one of the cows and the other had a perforating skin lesion in the left abdomen. Both cows had signs of hypovolaemic shock. Ultrasonography revealed hypoechoic fluid, echoic lesions and occasional fibrinous septa caudoventral to the reticulum. Caudally the fluid extended t

Our objective was to review the factors that affect fiber passage from the rumen. Rumen residence time and passage from the rumen are important in control of intake, digestibility, protein metabolism, and protein escape. Physical factors associated with particle size and particle specific gravity affect passage from the rumen. Although particles of 5 cm may pass through the reticulo-omasal orifice, most particles leaving the rumen are smaller than 1 mm. Polypropylene ribbon cut into 7-cm lengths, introduced into the rumen, markedly reduced intake and were ruminated to fine particle size before

Phenol-water extraction of Megasphaera elsdenii, a predominant gram-negative coccus in rumens of cattle fed high-grain diets, yielded material that exhibited typical characteristics of endotoxin. The extract was lethal to mice and to chicken embryos, caused biphasic fever in rabbits, leukopenia in mice, and local and generalized Shwartzman reactions; and induced tolerance to the lethal effect of the endotoxin in mice. The material contained carbohydrate, protein, lipid, phosphorus, and 2-keto-3-deoxyoctonate, but no nucleic acid. The beta-hydroxymyristic acid was absent. Results imply that M e

This study evaluated the effects of dietary sea buckthorn flavonoid (SF) supplementation on growth performance, serum biochemical, antioxidant, and immune indices, rumen fermentation, and ruminal bacterial composition in Hu sheep fed a high-concentrate diet. Thirty 6-month-old fattening Hu sheep were randomly assigned to a high-concentrate control diet (HC) or the same diet supplemented with 0.30% SF (HT; n = 15 per group) for 45 days. Compared with the HC group, SF supplementation increased average daily gain (p = 0.04) and reduced the feed conversion ratio (p = 0.01) without affecting averag

Eating and rumination activities influence rumen environmental characteristics, such as volume, digesta particle size, microbial activity, and the acidity of rumen fluid. The aim of this study was to update an existing mechanistic, steady-state rumen model to simulate diurnal patterns of rumen functioning, contributing to a better understanding of rumen metabolism. Daily patterns of eating behavior were defined as model inputs based on meal characteristics and feeding regimens. Mechanisms controlling the dynamics of rumen content and fluid volume, particle diminution of roughage, rumination be

Grazing sheep and goats are constantly exposed to helminth infections in many parts of the world, including several trematode species that causes a range of clinical diseases. The clinical picture of flukes is dependent upon the organs in which they develop and the tissues they damage within the respective organs. Accordingly, infections with the common liver fluke Fasciola hepatica, which, as juvenile worm migrates through the liver parenchyma for several weeks, may be associated with hepatic disorders such as impairment of carbohydrate, protein and fat metabolism, followed by chronic wasting

Stimulation of Ca2+ absorption can counteract hypocalcemia at the onset of lactation. The plant bioactive lipid compound (PBLC) menthol is an agonist for nonselective cation channels of the transient receptor potential (TRP) family. It acutely stimulated Ca2+ absorption in ruminal epithelia of nonadapted animals ex vivo and caused higher plasma Ca2+ concentrations in cows and sheep in vivo. To elucidate the pathway by which menthol feeding increases plasma Ca2+ level, the present study aimed to investigate the long-term dose-dependent effects of dietary menthol-rich PBLC on Ca2+ absorption and

The rumen is the hallmark organ of ruminants and hosts a diverse ecosystem of microorganisms that facilitates efficient digestion of plant fibers. We analyzed 897 transcriptomes from three Cetartiodactyla lineages: ruminants, camels and cetaceans, as well as data from ruminant comparative genomics and functional assays to explore the genetic basis of rumen functional innovations. We identified genes with relatively high expression in the rumen, of which many appeared to be recruited from other tissues. These genes show functional enrichment in ketone body metabolism, regulation of microbial co

Infections caused by multidrug resistant bacteria represent a therapeutic challenge both in clinical settings and in livestock production, but the prevalence of antibiotic resistance genes among the species of bacteria that colonize the gastrointestinal tract of ruminants is not well characterized. Here, we investigate the resistome of 435 ruminal microbial genomes in silico and confirm representative phenotypes in vitro. We find a high abundance of genes encoding tetracycline resistance and evidence that the tet(W) gene is under positive selective pressure. Our findings reveal that tet(W) is

Information available on the role of Mg for growth and cellulose degradation by rumen bacteria is both limited and inconsistent. In this study, the Mg requirements for two strains each of the cellulolytic rumen species Fibrobacter succinogenes (A3c and S85), Ruminococcus albus (7 and 8) and Ruminococcus flavefaciens (B34b and C94) were investigated. Maximum growth, rate of growth and lag time were all measured using a complete factorial design, 2(3)×6; factors were: strains (2), within species (3) and Mg concentrations (6). R. flavefaciens was the only species that did not grow when Mg was sin

In incubations in vitro with rumen fluid, the effect of two methane inhibitors, linseed oil hydrolysate (LOH) and chloral hydrate (CH) on the efficiency of microbiol growth was investigated. Total and net microbial growth were determined from 32PO43- and NH3--N incorporation respectively and expressed as g N incorporated per kg organic matter fermented (gN/kgOMf). In a first series on incubations, it was found that LOH had no influence on overall microbial growth efficiency, while with CH, a small but significant decrease of total and net growth efficiency was measured. Further experiments sho

Intra-abomasally administered 1,2-propanediol, 150 to 160 g per Japanese meat-type kid in 3 to 4 wk, increased the ratio of rumen to body weight, of rumen to total stomach, and of rumen mucosa to muscle over the control which received only water. Intra-abomasal administration of glycerol, glucose, fructose, 1.3-butanediol, xylitol, lactic acid, sodium propionate, or sodium butyrate showed no increases over the control. Only propanediol chemically stimulated rumen mucosal growth. Intraruminal administration of propanediol showed the same stimulation as intra-abomasal. Intraruminal administratio

The cell-free rumen liquor of a steer on a diet of spear grass has been shown to contain macromolecular substances in which carbohydrates and lignin-derived compounds are covalently bound to each other. The lignin-carbohydrate complexes are soluble at pH 7 or higher, but precipitate at pH 3. At the latter pH, small amounts of a polymer, assumed to be glycoprotein, remain in solution. Some of the lignin-carbohydrate linkages are broken by treatment with alkali. Treatment with 50mM sulphuric acid for a few minutes at room temperature converts part of the complex into an acetone-soluble product,

Methane production from pyruvate by mixed rumen bacteria in vitro was nearly totally inhibited by chloral hydrate (0.1 mumole/ml of incubation fluid). This effect was accompanied by an accumulation of gaseous hydrogen and an increase in propionic acid production. Infusion of chloral hydrate (4 g/day) into the rumen of a sheep produced the same effects. Evidence is presented for a direct toxic effect of chloral hydrate upon methane bacteria. Results are discussed in terms of fermentation balances.
