In vitro and in vivo antiproliferative and trypanocidal activities of ruthenium NO donors.

BACKGROUND AND PURPOSE: Many compounds liberating NO (NO donors) have been used as therapeutic agents. Here we test two ruthenium nitrosyls, which release NO when activated by biological reducing agents, for their effects in vitro and in vivo against Trypanosoma cruzi, the agent responsible for the American trypanosomiasis (Chagas' disease). EXPERIMENTAL APPROACH: Ruthenium NO donors were incubated with a partially drug-resistant strain of T. cruzi and the anti-proliferative and trypanocidal activities evaluated. In a mouse model of acute Chagas' disease, trypanocidal activity was evaluated b

Ruthenium complexes with dendritic ferrocenyl phosphanes: synthesis, characterization, and application in the catalytic redox isomerization of allylic alcohols.

An efficient system for the catalytic redox isomerization of the allylic alcohol 1-octen-3-ol to 3-octanone is presented. The homogeneous ruthenium(II) catalyst contains a monodentate phosphane ligand with a ferrocene moiety in the backbone and provides 3-octanone in quantitative yields. The activity is increased by nearly 90 % with respect to the corresponding triphenyl phosphane ruthenium(II) complex. By grafting the catalyst at the surface of a dendrimer, the catalytic activity is further increased. By introducing different spacers between ferrocene and phosphorus, the influence on the elec

New nitrosyl ruthenium complexes with combined activities for multiple cardiovascular disorders.

Nitrosyl ruthenium complexes are promising platforms for nitric oxide (NO) and nitroxyl (HNO) release, which exert their therapeutic application. In this context, we developed two polypyridinic compounds with the general formula cis-[Ru(NO)(bpy)2(L)]n+, where L is an imidazole derivative. These species were characterized by spectroscopic and electrochemical techniques, including XANES/EXAFS experiments, and further supported by DFT calculations. Interestingly, assays using selective probes evidenced that both complexes can release HNO on reaction with thiols. This finding was biologically vali

Ruthenium mediated halogenation of white phosphorus: synthesis and reactivity of the unprecedented P4Cl2 moiety.

The transition metal mediated functionalization of P4 is an approach to develop a more sustainable production of organophosphorus compounds. In this paper a ruthenium complex is presented, in which the P4 unit can be selectively converted into new P4R2 molecules in a two-step synthesis. The unsaturated 16 electron species [RuX(Cp*)(PCy3)] (Cp* = C5Me5, X = Cl, Br) reacts with a half equivalent of P4 affording a bimetallic complex bearing a planar P4X2 moiety as a ligand. The latter eliminates chloride anions under reduction with magnesium. In this process the butadiene-like P4Cl2 ligand is con

The preventive effect of Daikenchuto on postoperative adhesion-induced intestinal obstruction in rats.

The present study investigated the effect of Daikenchuto (DKT) on postoperative intestinal adhesion in rats. We evaluated the effects of DKT, constituent medical herbs and active compounds on talc-induced intestinal adhesion in rats and DKT-induced contractions using isolated guinea pig ileum. DKT significantly prevented adhesion formation, and this action was inhibited by pretreatment with atropine or ruthenium red. The constituent medical herbs, Zanthoxylum Fruit and Maltose Syrup Powder significantly prevented adhesion formation. Moreover, hydroxy sanshool (HS) prevented adhesion formation,

A novel ruthenium(III) complex with a tridentate dianionic P,O,O-ligand showing high cytotoxic activity.

Tris(o-anisyl)phosphane was reacted with ruthenium(III) chloride forming a novel Ru(III) complex of formula [Ru(PAn(3))(P(An)(phenolate)(2))Cl] containing a tridentate dianionic P,O,O-ligand and a bidentate neutral P,O ligand. The tridentate ligand was formed by elimination of two methyl groups from the starting ligand in the reaction with the ruthenium salt. The molecular structure was determined by single crystal X-ray diffraction analysis. The compound shows a high cytotoxic activity in ovarian cancer cell lines comparable with cisplatin and overcoming cisplatin-resistance.

Ruthenium derivatives attenuate LPS-induced inflammatory responses and liver injury via suppressing NF-κB signaling and free radical production.

Ruthenium metal complex has been shown to exert several chemical and biological activities. A series of three novel ruthenium derivatives (TQ 1, 2 and 4) were synthesized to evaluate the anti-inflammatory and hepatoprotective activities in lipopolysaccharide (LPS)-stimulated macrophages and mice liver injury. The hydroxyl radical (OH°) scavenging activity of these derivatives has also been evaluated. The results revealed that among the tested compounds, TQ-4 effectively attenuated LPS-induced abnormal alteration in liver histoarchistructure via reducing alanine transaminase (ALT) and aspartate

Diborane and Triborane Species in the Coordination Sphere of Group-8 Transition Metals.

The synthesis and structural elucidation of a series of ruthenium diborane and triborane compounds are described. Treatment of [Cp*Ru(PPh3)2Cl] (1) (Cp* = 1,2,3,4,5-pentamethylcyclopentadienyl; PPh3 = triphenylphosphine) with [BH3·THF] (THF = tetrahydrofuran) at 60 °C led to the formation of the hydrogen-rich ruthena-octahydrotetraborane arachno-[2-{Cp*Ru(PPh3)B3H8}] (2). The chemistry of 2 is explored with [Fe2(CO)9] at room temperature, which resulted in the formation of a metal-stabilized triborane species, [{Cp*Ru(PPh3)}(μ3-η1:η2:η2-B3H6){Fe2(CO)7}] (3). Compound 3 can be considered as a t

Influence of Binders and Solvents on Stability of Ru/RuOx Nanoparticles on ITO Nanocrystals as Li-O2 Battery Cathodes.

Fundamental research on Li-O2 batteries remains critical, and the nature of the reactions and stability are paramount for realising the promise of the Li-O2 system. We report that indium tin oxide (ITO) nanocrystals with supported 1-2 nm oxygen evolution reaction (OER) catalyst Ru/RuOx nanoparticles (NPs) demonstrate efficient OER processes, reduce the recharge overpotential of the cell significantly and maintain catalytic activity to promote a consistent cycling discharge potential in Li-O2 cells even when the ITO support nanocrystals deteriorate from the very first cycle. How Computations Can Assist the Rational Design of Drugs for Photodynamic Therapy: Photosensitizing Activity Assessment of a Ru(II)-BODIPY Assembly.

Ruthenium-based complexes represent a new frontier in light-mediated therapeutic strategies against cancer. Here, a density functional-theory-based computational investigation, of the photophysical properties of a conjugate BODIPY-Ru(II) complex, is presented. Such a complex was reported to be a good photosensitizer for photodynamic therapy (PDT), successfully integrating the qualities of a NIR-absorbing distyryl-BODIPY dye and a PDT-active [Ru(bpy)3]2+ moiety. Therefore, the behaviour of the conjugate BODIPY-Ru(II) complex was compared with those of the metal-free BODIPY chromophore and the R

Iridium(III) polypyridyl and cyclometalated complexes for DNA targeting, bioimaging, and light-activated therapeutics: recent progress, mechanistic insights, and emerging design strategies-a mini review.

Iridium(III) polypyridyl and cyclometalated complexes have emerged as highly versatile coordination compounds with applications spanning photocatalysis, optoelectronics, bioimaging, and photodynamic therapy. Whilst ruthenium(II) polypyridyl complexes have historically dominated nucleic acid recognition studies, iridium(III) systems possess distinct physicochemical advantages arising from strong spin-orbit coupling, tuneable metal-to-ligand charge-transfer (MLCT) states, long-lived triplet emission, and exceptional photostability. Despite these favourable features, their development as DNA-targ

(Pyridyl)benzoazole ruthenium(II) and ruthenium(III) complexes: role of heteroatom and ancillary phosphine ligand in the transfer hydrogenation of ketones.

The synthesis and structural characterization of ruthenium complexes supported by 2-(2-pyridyl)benzoazole ligands and their evaluation as catalysts in the transfer hydrogenation of ketones are reported. Reactions of 2-(2-pyridyl)benzoimidazole (L1), 2-(2-pyridyl)benzothiazole (L2) and 2-(2-pyridyl)benzoxazole (L3) with RuCl3·3H2O produced the corresponding complexes [RuCl3(L1)] (1), [RuCl3(L2)] (2) and [RuCl3(L3)] (3), respectively. Similarly, treatment of L1-L3 with RuCl2(PPh3)2 afforded the corresponding Ru(II) complexes [RuCl2(PPh3)2(L1)] (4), [RuCl2(PPh3)2(L2)] (5) and [RuCl2(PPh3)2(L3)] (

Misidentified LO2 and SGC-7901 cell lines challenge the claimed selectivity of ruthenium(II) polypyridyl complexes.

In this commentary, I express concerns about the misidentification of the LO2 and SGC-7901 cell lines used by Tian and his colleagues in their study of ruthenium(II) polypyridyl complexes with alleged anti-cancer properties. While these compounds appear to inhibit tumor growth by inducing cell cycle arrest, mitochondrial damage, apoptosis, ferroptosis, autophagy, and immunogenic cell death, the authors' claims of selectivity are undermined by their reliance on the LO2 line as a "normal liver cell". It is well established that the LO2 line is a HeLa derivative, rather than a benign hepatocyte c

Chiral-auxiliary-mediated asymmetric synthesis of ruthenium polypyridyl complexes.

An octahedral metal complex with 6 different monodentate ligands can form 15 diastereomers as pairs of enantiomers. As a result, the elaborate stereochemistry of octahedral coordination geometries provides tremendous opportunities in the fields of catalysis, the materials sciences, and the life sciences. The demand for enantiomerically pure coordination complexes for tasks related to the selective molecular recognition of biomacromolecules led us to develop synthetic methods to control the absolute stereochemistry at octahedral metal centers. A few years ago our laboratory therefore embarked o

Recent Investigations on the Use of Copper Complexes as Molecular Materials for Dye-Sensitized Solar Cells.

Three decades ago, dye-sensitized solar cells (DSSCs) emerged as a route for harnessing the sun's energy and converting it into electricity. Since then, an impressive amount of work has been devoted to improving the global photovoltaic efficiency of DSSCs, trying to optimize all components of the device. Up to now, the best efficiencies have usually been reached with ruthenium(II) photosensitizers, even if in the last few years many classes of organic compounds have shown record efficiencies. However, the future of DSSCs is stringently connected to the research and development of cheaper mater

Ruthenium Oxide Nanozyme for Tendinopathy Treatment via Countering Ferroptosis.

The global prevalence of tendinopathy (TP) is steadily increasing, resulting in functional impairments in tendons across individuals of all ages. Excessive accumulation of reactive oxygen species (ROS) plays a pivotal role in the development of TP, which compromises tendon repair and integrity through oxidative stress. This process is often accompanied by ferroptosis-a newly recognized form of iron-dependent programmed cell death. This study explores ruthenium oxide (RuO2) hollow nanospheres as an effective nanozyme to address oxidative damage and ferroptosis in type I collagenase-induced TP.

Highly efficient and enantioselective synthesis of β-heteroaryl amino alcohols via Ru-catalyzed asymmetric hydrogenation.

Chiral β-heteroaryl amino alcohols are key fragments of many bioactive compounds and antibiotics, and the development of efficient synthetic methods for these compounds is of great value. The highly enantioselective hydrogenation of α-N-heteroaryl ketones was realized with a ruthenium-diphosphine-diamine catalyst, providing the corresponding chiral β-heteroaryl amino alcohols with up to 99% yield and up to >99% ee. The synthetic utilities of the current reaction were demonstrated by gram-scale synthesis of key intermediates of Sertaconazole and Cenobamate.

NO and NOx interactions with group 8 metalloporphyrins.

There has been an ongoing interest in the reactions of nitric oxide (NO) with heme model compounds, with the goal of interpreting related reactions occurring in biology. With recent evidence that higher oxides (NO2-, *NO2, N2O3, etc.) may also be formed under bioregulatory conditions, there is a need to understand the reactivities of these compounds with such models. This review discusses the mechanistic studies of the reactions of iron, ruthenium, and osmium metalloporphyrin complexes with NO and the higher nitrogen oxides.

Selective Ruthenium-Catalyzed Reductive Alkoxylation and Amination of Cyclic Imides.

Reported herein, for the first time, is the selective ruthenium-catalyzed reductive alkoxylation and amination of phthalimides/succinimides. Notably, this novel methodology avoids hydrogenation of the aromatic ring and allows methoxylation of substituted imides with good to excellent selectivity for one of the carbonyl groups. The reported method opens the door to the development of new processes for the selective synthesis of various functionalized N-heterocyclic compounds. As an example, intramolecular reductive couplings to afford tricyclic compounds are presented for the first time.
