Systemic Besnoitiosis in a Juvenile Roe Deer (Capreolus capreolus).

Herein, we report the first incidence of systemic besnoitiosis in a male juvenile roe deer Capreolus capreolus. The animal was found dead in an area where bovine besnoitiosis is endemic and showed cachexia and multiple skin erosions in the metacarpal and metatarsal areas. Moreover, round and elevated white structures suggestive of Besnoitia spp. tissue cysts were also present. Twenty-eight tissue samples from different anatomical locations were collected for microscopic lesion and parasite detection through histopathology and PCR. Immunohistochemistry was performed to confirm Besnoitia-positiv

Vector-borne transmission of Besnoitia besnoiti by blood-sucking and secretophagous flies: epidemiological and clinicopathological implications.

BACKGROUND: Bovine besnoitiosis has been recently diagnosed in a three-parted herd of 796 Aubrac and Charolais beef cattle in Hungary. A large scale serological, histological and molecular survey was initiated in order to uncover important factors in the local epidemiology of the disease. FINDINGS: Blood samples were collected (three times from the whole herd, and repeatedly from selected animals) for serological screening by ELISA. In addition, various organs from aborted fetuses and newborn calves, skin and colostrum samples of seropositive heifers/cows, and ticks collected from the cattle

Structural definition of babesial RAP-1 proteins identifies a novel protein superfamily across Apicomplexa.

Apicomplexan protozoa are intracellular parasites of medical and economic importance. These parasites contain specialized apical complex organelles, including rhoptries, that participate in the process of host cell invasion. Conserved antigens expressed in the rhoptries are rational vaccine targets, but whether conservation of protein structure is a functional requirement for invasion remains unknown. Novel protein structural modeling enables identification of structurally conserved protein families that are not evident by sequence analysis alone. Here we show by AlphaFold2 structural modeling

Molecular survey for cyst-forming coccidia (Toxoplasma gondii, Neospora caninum, Sarcocystis spp.) in Mediterranean periurban micromammals.

Rodents and other micromammals constitute important reservoirs of infectious diseases; their role in the life cycle of apicomplexan parasites such as Toxoplasma gondii, Neospora caninum, and Sarcocystis spp. still needs clarification. In the present study, we analyzed by PCR and Sanger sequencing methods the presence of specific parasite DNA within brain and heart tissues of 313 individuals of five synanthropic small mammal species (Apodemus sylvaticus, Mus spretus, M. musculus, Rattus rattus, and Crocidura russula) collected in Barcelona metropolitan area (NE Spain). In addition, PCR-RFLP and

Habitat-associated detection of Toxoplasma gondii and Sarcocystis spp. in Cetaceans from the Brazilian coast.

Cetaceans are sentinels of marine ecosystem health and are increasingly exposed to protozoal pathogens. This study investigated the occurrence and molecular identity of Sarcocystidae protozoa in 159 stranded cetaceans representing 21 species along the Brazilian coast. Molecular analysis based on PCR amplification and sequencing of the sarcocystid internal transcribed spacer 1 (ITS1) region revealed infections in 14 individuals, showing a clear habitat-associated distribution. Toxoplasma gondii and Sarcocystis neurona were detected in nine coastal cetaceans, consistent with transmission from te

Molecular Survey on Toxoplasma gondii and Neospora caninum Infection in Wild Birds of Prey Admitted to Recovery Centers in Northern Italy.

Toxoplasma gondii and Neospora caninum (Apicomplexa, Sarcocystidae) are protozoan parasites infecting a wide range of intermediate hosts worldwide, including birds. Raptors acquire the infections through the ingestion of both infected preys and oocysts in the environment suggesting they might be used as indicators of the spread of these pathogens. Here, we report an epidemiological survey with the aim of determining the prevalence of T. gondii and N. caninum infections in wild birds of prey, hospitalized in two Wildlife Recovery Centres (WRCs) in Northern Italy. Genomic DNA extracted from brai

Analytical validation of an immunoassay for the detection of Cystoisospora spp. coproantigen in canine and feline fecal samples.

Cystoisospora species that infect dogs and cats are ubiquitous, and oocysts can be found in the feces of both subclinically infected and sick animals. Most clinical cases are diagnosed in puppies and kittens <4-mo-old. We validated a high-throughput immunoassay (Fecal Dx immunoassay; Idexx), which uses 2 monoclonal antibodies to detect Cystoisospora spp. coproantigen, using zinc sulfate centrifugal flotation (ZCF) as the reference method and samples from experimentally infected animals. This new genus-level coproantigen assay had equivalent sensitivity for at least 4 relevant Cystoisospora spp

Bovine PMN responses to extracellular vesicles released by Besnoitia besnoiti tachyzoites and B. besnoiti-infected host cells.

Bovine besnoitiosis is a re-emerging cattle disease caused by the apicomplexan parasite Besnoitia besnoiti, which severely affects individual animal welfare and profitability in cattle industry. We recently showed that B. besnoiti tachyzoite exposure to bovine polymorphonuclear neutrophils (PMN) effectively triggers neutrophil extracellular trap (NET) formation, leading to parasite immobilization hampering host cell infection. So far, the triggers of this defense mechanism remain unclear. Emerging evidence indicates that extracellular vesicles (EVs) modulate PMN effector functions, such as ROS

Pancreatic and biliary duct Hammondia heydorni infection in a dog fed a raw elk-meat diet.

We describe a 3-y-old Standard Poodle that was fed a raw elk-meat diet and developed anorexia, icterus, and markedly elevated liver enzyme activities. Histopathology of the pancreatic and liver biopsy specimens revealed lymphohistiocytic pancreatitis and hepatitis with intraepithelial protozoa. Histologically, Hammondia spp. are indistinguishable from Neospora caninum. Therefore, we employed PCR and amplicon sequencing to further investigate our case. Amplification and sequencing of the ITS1 marker matched 100% with Hammondia heydorni. Intrapancreatic involvement and the histopathologic featur

First report of natural Besnoitia akodoni infection in synanthropic (Muridae) and wild (Cricetidae) rodents from Argentina.

Besnoitia spp. are cyst-forming Apicomplexa parasites within the Sarcocystidae family. The objective of this study was to identify Besnoitia spp. in synanthropic (Muridae) and wild (Cricetidae) rodents from Argentina. Samples were collected from different organs and tissues of 356 rodents and examined by histopathological methods. Besnoitia cysts were detected by histopathology in 2 % (7/356) of the animals (i.e., two Mus musculus, two Necromys spp., two Oxymycterus rufus, and one Akodon azarae). The morphological diagnosis was molecularly confirmed by PCR and bidirectional sequencing of the I

Neospora caninum-induced NETosis in canine colostral polymorphonuclear neutrophils.

Neospora caninum represents an obligate intracellular apicomplexan parasite of the family Sarcocystidae causing severe reproductive disorders in cattle, small ruminants, wild animals and canids worldwide. Neutrophil extracellular traps (NETs) were recently described as effective host defense mechanism of polymorphonuclear neutrophils (PMN) derived from cattle, dogs, goats and dolphins against N. caninum tachyzoites. Nonetheless, nothing is known so far on canine colostral PMN immune reactions against N. caninum although breeding bitches represent a susceptible dog cohort and infected bitches m

Besnoitosis in Iran: An emerging parasitic threat to livestock and wildlife.

Besnoitiosis is a chronic and emerging parasitic disease caused by protozoa belonging to the genus Besnoitia. The disease is economically and ecologically threatening livestock and wildlife in Iran. Besnoitia caprae infecting goats can lead to cyst formation in host tissues and cause reproductive symptoms, resulting in economic losses due to abortions and/or reduced productive efficiency. Its significance is particularly pronounced in provinces such as Fars and Kermanو attributed to high goat population density, climatic conditions favoring transmission, and specific animal husbandry practices

Newly Described Coccidia Goussia bayae from White Perch Morone americana: Morphology and Phylogenetics Support Emerging Taxonomy of Goussia within Piscine Hosts.

In March and April 2016, 150 white perch ( Morone americana) were collected from various localities in Chesapeake Bay and examined for coccidia. A previously undescribed species of coccidia was observed in the hepatic bile ducts and gallbladder of all white perch (100%) examined. We describe this species using morphological characteristics, histology, and gene sequences of the small-subunit ribosomal DNA ( rDNA), large-subunit rDNA, and mitochondrial genes cytochrome oxidase 1 ( COI), cytochrome oxidase b ( Cytb), and cytochrome oxidase 3 ( COIII). Oocysts of Goussia bayae n. sp. were subspher

Renal infection by a new coccidian genus in big brown bats (Eptesicus fuscus).

A novel coccidian parasite from the kidney of big brown bats (Eptesicus fuscus) is described. This coccidian (Nephroisospora eptesici nov. gen., n. sp.) was associated with a generally mild, focal or multifocal, well-demarcated cortical renal lesion less than 1 mm in diameter. The lesion represented cystic, dilated tubules with hypertrophied tubular epithelial cells and was present in the kidneys of 29 of 590 bats. Numerous coccidian parasites in various stages of development were present within the tubular epithelial cells and within the cyst lumina. Oocysts were collected from cystic dilated

Detection of Sarcocystis halieti DNA in the Blood of Western House Martin (Delichon urbicum) and Barn Swallow (Hirundo rustica) from Lithuania, and in Eurasian Griffon Vulture (Gyps fulvus) from Greece.

PURPOSE: Sarcocystis (Apicomplexa: Sarcocystidae) is a cyst-forming coccidian parasite that infects mammals, reptiles and birds. Despite the emergence of studies employing less invasive or lethal methods to study these parasites, Sarcocystis species have yet to be detected in avian blood. The objective of this study was to molecularly identify Sarcocystis species in the blood DNA samples of three avian host species. METHODS: A total of 93 DNA samples from avian blood were subjected to a screening procedure for Sarcocystis. Samples from three bird species, 30 western house martins (Delichon ur

Detection of Toxoplasma gondii DNA in milk of dairy cows from southern Brazil.

Consumption of unpasteurized cow's milk may be a transmission route for some pathogenic microorganisms, but there is little information about the risk of Toxoplasma gondii infection. Blood and milk samples were collected in a paired and random fashion from 106 dairy cows and bulk-tank milk samples were also collected from each of the six farms, in southern Brazil. Serum anti-T.gondii antibodies (IgG) were detected by an indirect fluorescent antibody test (IFAT) with a cutoff point of 1:64. Nested PCR targeting the ITS1 was performed on milk samples to detect the Sarcocystidae family, confirmed
