Structural Characterisation of Non-Deamidated Acidic Variants of Erwinia chrysanthemi L-asparaginase Using Small-Angle X-ray Scattering and Ion-Mobility Mass Spectrometry.

PURPOSE: Erwinia chrysanthemi L-asparaginase (ErA) is an enzyme commonly used in the treatment regimen for Acute Lymphoblastic Leukaemia (ALL). Biopharmaceutical products such as ErA must be monitored for modifications such as deamidation, typically using ion-exchange chromatography (IEX). Analysis of clinical-grade ErA using native IEX resolves a number of enzymatically-active, acidic variants that were poorly characterised. METHODS: ErA IEX variants were isolated and fully characterised using capillary electrophoresis (cIEF), LC-MS and LC-MS/MS of proteolytic digests, and structural techniq

Structure of the bifunctional aminoglycoside-resistance enzyme AAC(6')-Ie-APH(2'')-Ia revealed by crystallographic and small-angle X-ray scattering analysis.

Broad-spectrum resistance to aminoglycoside antibiotics in clinically important Gram-positive staphylococcal and enterococcal pathogens is primarily conferred by the bifunctional enzyme AAC(6')-Ie-APH(2'')-Ia. This enzyme possesses an N-terminal coenzyme A-dependent acetyltransferase domain [AAC(6')-Ie] and a C-terminal GTP-dependent phosphotransferase domain [APH(2'')-Ia], and together they produce resistance to almost all known aminoglycosides in clinical use. Despite considerable effort over the last two or more decades, structural details of AAC(6')-Ie-APH(2'')-Ia have remained elusive. In

Spectroscopic and Small-angle X-ray scattering analysis of binding between Copper(II) -1-allylimidazole complex, a potential anti-tumor agent, and bovine serum albumin.

Interactions between transport proteins and compounds with therapeutic potential are pharmacologically important. In this study, using fluorescence, circular dichroism (CD), and small-angle X-ray Scattering (SAXS), we investigated the interaction between bovine serum albumin (BSA) and a copper(II)-1-allylimidazole complex with potential anti-cancer properties. The results revealed dynamic fluorescence quenching of the model carrier protein BSA by the copper(II) complex. The enthalpy change (ΔH), free energy (ΔG), and entropy change (ΔS) were calculated to be 108 kJ/mol, -16.47 kJ/mol, and 419

A study combining magic-angle spinning NMR and small-angle X-ray scattering on the interaction in the mixture of poly(benzyl methacrylate) and ionic liquid 1-ethyl-3-methylimidazolium bis(trifluoromethanesulfonyl)amide.

A mixture of poly(benzyl methacrylate) (PBnMA) and 1-ethyl-3-methylimidazolium bis(trifluoromethanesulfonyl)amide ([C2mim][NTf2]) exhibits lower-critical-solution-temperature (LCST)-type phase separation. An investigation combining magic-angle spinning NMR spectroscopy and small-angle scattering was performed to gain new insights into the interaction between PBnMA and the ionic liquid. The molecular mobility and the solute-solvent interaction in the system were investigated using 1H high-resolution magic-angle spinning NMR. Applying a magic-angle spinning frequency of 2 kHz allowed identifying

Small Angle X-Ray Scattering Data Analysis and Theoretical Modelling for the Size and Shape Characterization of Drug Delivery Systems Based on Vitamin E TPGS Micelles.

We developed a simple two-dimensional/two-components theoretical model that describes the structure and functionality of a VitE-TPGS system of micelles assuming a hydrophobic inner core and an outer hydrated hydrophilic shell. We then conceptually applied the developed methodology to a simple system of VitE-TPGS micelles unloaded and loaded with an active pharmaceutical ingredient, eltrombopag, to verify if the model could reliably monitor the size change of the micelle upon loading. The fit of laboratory Small Angle X-Ray Scattering data against such model allows us to extract absolute values

Small-angle neutron scattering studies of mineralization on BSA coated citrate capped gold nanoparticles used as a model surface for membrane scaling in RO wastewater desalination.

Bovine serum albumin (BSA) coated on citrate capped gold nanoparticles (BSA-GNPs) was exposed to a simulated wastewater effluent (SSE) in order to study the mineralization and thereby mimic scaling at biofouled membranes of reverse osmosis (RO) wastewater desalination plants. RO is a leading technology of achieving freshwater quality as it has the capability of removing both dissolved inorganic salts and organic contaminants from tertiary wastewater effluents. The aim was to better understand one of the major problems facing this technology which is fouling of the membranes, mainly biofouling

Toward reliable low-density lipoprotein ultrastructure prediction in clinical conditions: A small-angle X-ray scattering study on individuals with normal and high triglyceride serum levels.

Atherosclerosis is the main killer in the west and therefore a major health challenge today. Total serum cholesterol and lipoprotein concentrations, used as clinical markers, fail to predict the majority of cases, especially between the risk scale extremes, due to the high complexity in lipoprotein structure and composition. In particular, low-density lipoprotein (LDL) plays a key role in atherosclerosis development, with LDL size being a parameter considered for determining the risk for cardiovascular diseases. Determining LDL size and structural parameters is challenging to address experimen

An insight into third-phase formation during the extraction of thorium nitrate: evidence for aggregate formation from small-angle neutron scattering and validation by computational studies.

Small-angle neutron scattering (SANS) studies were carried out to compare the aggregation behavior of 1.1 M solutions of tributyl phosphate (TBP) and N,N-dihexyl octanamide (DHOA) dissolved in different deuterated diluents, viz., n-dodecane, chloroform, and benzene, during the extraction of Th(IV) from nitric acid medium. The scattering data was treated using the Baxter sticky spheres model. The third phase formed in the case of DHOA displayed higher aggregation tendency compared to that of TBP. These studies have demonstrated that the nature of the diluents plays an important role in the aggr

Effect of Mutations in the N-Terminal Peptide of the Coat Protein on the Structure of Potato Virus X According to Small-Angle X-Ray Scattering and Molecular Dynamics.

Mutations in the N-terminal peptide (Ser-Thr to Ala-Gly substitution) of the coat protein (CP) of potato virus X (PVX-ST) render its genomic RNA translationally competent, unlike in the wild-type PVX virions. Consequently, RNA within the PVX-ST virions can be translated without additional triggers (such as phosphorylation or interaction with the triple gene block 1 protein), unlike the encapsidated RNA of the wild-type virus. Comprehensive structural analysis using molecular dynamics (MD), small-angle X-ray scattering (SAXS), and tritium planigraphy revealed differences in the virion organizat

Influence of in situ Limited Proteolysis of Potato Virus X on Change in the Structure of Virions According to the Small-Angle X-Ray Scattering Data and Tritium Labeling.

Capsids of the potexvirus family virions are characterized by the presence on the surface of virions of partially disordered N-terminal protein fragments of various lengths. The present study is devoted to studying the effect of in situ removal of the N-terminal domain of coat protein (CP) on structural organization and physicochemical properties of the potato virus X (PVX) virions. The work considers PVX virions containing an intact Ps-form CP, as well as virions including an in situ degraded Pf-form devoid of 19/21 amino acid residues from the N-end (PVXΔN). Dynamic Self-Assembly and Stimuli-Responsive Disassembly of Bioactive-Loaded Cubosomes in Biomimetic Media Traced by Real-Time Small-Angle X-ray Scattering and Cryogenic Transmission Electron Microscopy.

Designing advanced functional materials capable of passing through complex biological environments requires a deep understanding of their dynamic structural behavior in situ. We investigate pH-responsive core-shell cubosomes for oral drug delivery applications. These nanoparticles comprise a lipid-based core of cubic Im3m liquid crystalline structure and are coated with a chitosan-N-arginine/alginate polyelectrolyte shell (PS). The cubosomes encapsulate varying concentrations (0-30% w/w) of Aloe vera-derived acemannan, an immunomodulatory macromolecular drug. Utilizing synchrotron small-angle

Insights into the Solution Structure and Oligomeric State of Fructose-1,6-bisphosphate Aldolase and Pyruvate Kinase from Nakaseomyces glabratus by Small-Angle X‑ray Scattering (SAXS) and AlphaFold Prediction.

Nakaseomyces glabratus is an opportunistic pathogen of humans, causing invasive candidiasis (IC). Among the risk factors that favor IC are various host-specific factors. Although drugs are available to treat candidiasis, their clinical application remains limited. Therefore, it is necessary to identify therapeutic targets that will enable the development of new antifungals. In this regard, our research group has identified enzymes as potential therapeutic targets in N. glabratus, including fructose-1,6-bisphosphate aldolase (Fba1) and pyruvate kinase (Pk). Enzyme activity studies on these two

Dried hybrid imogolite nanotubes as solids with a changeable surface area: an insight into textural properties based on the correlation between nitrogen gas adsorption, immersion calorimetry into water, and small angle X-ray scattering.

Powdered samples of three imogolite materials with an inner surface covered with Si-CH3 groups (hybrid IMO-CH3) were prepared by ambient drying at 323 K, freeze drying, and spray drying. Reliable estimates of the intra-tube (SA), inter-tube (SB), and inter-bundle (SC) specific surface areas of these samples were inferred from a model-guided correlation between the results of measurements by small angle X-ray scattering (SAXS), nitrogen gas adsorption, and immersion calorimetry into water. Since the SAXS studies indicated no significant deformations of IMO-CH3 nanotubes upon drying, further stu

Strain-Induced Density Fluctuations in Glassy Polymers.

Deformation of glassy polymers induces plastic flow as a result of mechanical instability, ultimately leading to cavitation and fracture of the liquid. Thus, understanding the origin of such mechanical instability is crucial. Here, we demonstrate that strain deformation of polymeric glass is accompanied by marked shear-thinning behavior and leads to enhanced density fluctuations. These results are consistent with Furukawa and Tanaka's theory, which predicts that strain induces the self-amplification of the density fluctuations. Thus, we conclude that the enhancement results from the coupling b

Complex supramolecular fiber formed by coordination-induced self-assembly of benzene-1,3,5-tricarboxamide (BTA).

HYPOTHESIS: In the quest for large but well-controlled supramolecular structures, the discotic benzene-1,3,5-tricarboxamide (BTA) has received quite some attention, because it can form hydrogen-bonded stacks that can be regarded as supramolecular polymers of which the single BTA molecule is the monomer. In this report, we consider a more complex BTA-based supramolecular polymer, namely one that is built up from supramolecular 'monomers'. EXPERIMENTS: We design a tris-ligand L3 consisting of a BTA core carrying three dipicolinic acid (DPA) groups. L3 itself is too small to form polymers, but i

Comparison of macro-, micro- and nanomechanical properties of clinically-relevant UHMWPE formulations.

We characterized a set of eleven clinically relevant formulations of UHMWPE for total joint replacements. Although their molecular and supermolecular structure were quite similar as evidenced by IR, DSC and SAXS measurements, there were slight differences in their crystallinity (DSC crystallinity ranging from 52 to 61%), which were connected with processing conditions, such as the total radiation dose, thermal treatment and/or addition of biocompatible stabilizers. Mechanical properties were assessed at all length scales, using macroscale compression testing, non-instrumented and instrumented

Supramolecular arrangements in human amyloid tissues using SAXS.

Amyloid diseases are characterized by the accumulation of misfolded protein aggregates in human tissues, pose significant challenges for both diagnosis and treatment. Protein aggregations known as amyloids are linked to several neurodegenerative conditions including Alzheimer's disease, Parkinson's disease, and systemic amyloidosis. The key goal of this research is to employ Small-Angle X-ray Scattering (SAXS) to examine the supramolecular structures of amyloid aggregates in human tissues. We present the structural analysis of amyloid using SAXS, which is employed directly to analyze thin tiss

Exploring the influence of mesoporosity in hard carbon-templated hierarchical SAPO-5 for ethanol dehydration.

Microporous solid acid catalysts offer a vast amount of control over chemical processes. However, their coveted smaller pores also have several drawbacks, including a limited substrate scope, faster deactivation, and pore blockage. As such, there are significant advantages to introducing mesopores alongside the microporous framework, to create hierarchically porous frameworks. This work explores the influence of adapting our microporous synthetic procedure for silicoaluminophosphate (SAPO-5) to include different shaped carbon nanotemplates. The differing size of the mesopores formed is explore

Alginate-ionic liquid injectable hydrogels supporting protein crystallization.

Protein crystallization is essential for determining 3D structures, yet innovative media are needed to enhance crystallization efficiency and optimize crystallography experiments. Injectable hydrogels offer a dynamic, tunable environment for protein crystallization while enabling continuous sample delivery for serial crystallography. In this study, we combined two emerging media and developed novel injectable alginate-ionic liquid hydrogels to support protein crystallization. Our results demonstrate that the ionic liquid ethylammonium nitrate promoted the formation of stable, continuous inject
