Binding affinity screening and MAPK/NF-κB mechanisms of antioxidant and anti-inflammatory phenolics compounds from Gynura procumbens stem extract (GPSE).

Gynura procumbens is a traditional medicinal plant widely used in Southeast to east Asia for its various therapeutic properties, but its stems are a byproduct that is less commonly utilized than its leaves. In this study, GPSE was fractionated using hexane, chloroform, ethyl acetate, butanol, and water, and HPLC-MS/MS analysis identified 11 phenolic compounds, including scopolin, caffeic acid, scopoletin, and 3,5-dicaffeoylquinic acid. Subsequently, we screened candidate phenolic compounds with antioxidant and anti-inflammatory activities using DPPH and COX-2 HPLC binding affinity assays. Afte

Inhibition of luminol-dependent chemiluminescence of alveolar macrophages by possible etiological agents of byssinosis.

Aqueous extracts of cotton bracts inhibited luminol-dependent chemiluminescence of phagocytosing alveolar macrophages. The inhibitory compounds were partitioned into organic solvents. Chromotographic analysis indicated that these compounds were either lacinilene C or its 7-methyl ether and scopoletin. The fact that the inhibitory activity of these compounds was always less than that of the total extract suggests synergistic action. These compounds may function as etiological agents of byssinosis.

Production of Two Isomers of Sphaeralcic Acid in Hairy Roots from Sphaeralcea angustifolia.

The Sphaeralcea angustifolia plant is used as an anti-inflammatory and gastrointestinal protector in Mexican traditional medicine. The immunomodulatory and anti-inflammatory effects have been attributed to scopoletin (1), tomentin (2), and sphaeralcic acid (3) isolated from cells in suspension cultures and identified in the aerial tissues of the wild plant. The hairy roots from S. angustifolia established by infecting internodes with Agrobacterium rhizogenes were explored to produce active compounds based on biosynthetic stability and their capacity to produce new compounds. Hydrogen peroxide is not released following reaction of cyanide with several catalytically important derivatives of cytochrome c oxidase.

We have looked for the production of hydrogen peroxide following reaction of oxidized cytochrome c oxidase and two oxy derivatives (compounds P and F) with cyanide. In each case the final product was the cyanide adduct of cytochrome c oxidase. In no case release of hydrogen peroxide was detected, as gauged by the scopoletin plus horse radish peroxidase assay. The simplest conclusion is that none of these forms of the enzyme contains intact hydrogen peroxide.

Reduction of anoxia-induced bioenergetic disturbance in astrocytes by methanol fruit extract of Tetrapleura tetraptera and in silico evaluation of the effect of its antioxidative constituents on excitotoxicity.

Oxidative stress and excitotoxicity are some of the pathophysiological abnormalities in hypoxia-induced brain injury. This study evaluated the intrinsic antioxidant property of methanol fruit extract of Tetrapleura tetraptera (TT), traditionally used for managing brain diseases such as cerebral infarction in West Africa, and its ability to protect primary astrocytes from anoxia-induced cell death. The effect of the phytochemicals present in TT on excitotoxicity was assessed in silico, through docking with human glutamate synthetase (hGS). Chromatographic and spectrophotometric analyses of TT w

BTH and BABA induce resistance in pea against rust (Uromyces pisi) involving differential phytoalexin accumulation.

Systemic acquired resistance elicitors, BTH and BABA, reduce rust penetration in pea through phytoalexins pathway but differing in their mode of action. It has been previously shown that rust (Uromyces pisi) infection can be reduced in pea (Pisum sativum) by exogenous applications of systemic acquired resistance elicitors such as BTH and BABA. This protection is known to be related with the induction of the phenolic pathway but the particular metabolites involved have not been determined yet. In this work, we tackled the changes induced in phytoalexin content by BTH and BABA treatments in the

Cytokinins mediate resistance against Pseudomonas syringae in tobacco through increased antimicrobial phytoalexin synthesis independent of salicylic acid signaling.

Cytokinins are phytohormones that are involved in various regulatory processes throughout plant development, but they are also produced by pathogens and known to modulate plant immunity. A novel transgenic approach enabling autoregulated cytokinin synthesis in response to pathogen infection showed that cytokinins mediate enhanced resistance against the virulent hemibiotrophic pathogen Pseudomonas syringae pv tabaci. This was confirmed by two additional independent transgenic approaches to increase endogenous cytokinin production and by exogenous supply of adenine- and phenylurea-derived cytoki

Inhibition of hepatitis C virus replication by chalepin and pseudane IX isolated from Ruta angustifolia leaves.

Hepatitis C virus (HCV) infection is highly prevalent among global populations, with an estimated number of infected patients being 170 million. Approximately 70-80% of patients acutely infected with HCV will progress to chronic liver disease, such as liver cirrhosis and hepatocellular carcinoma, which is a substantial cause of morbidity and mortality worldwide. New therapies for HCV infection have been developed, however, the therapeutic efficacies still need to be improved. Medicinal plants are promising sources for antivirals against HCV. A variety of plants have been tested and proven to b

Computational and Biophysical Approaches to Identify Cell Wall-Associated Modulators in Salmonella enterica serovar Typhi.

An increase in the number of antibiotic-resistant bacterial pathogens, in recent times, has posed a great challenge for treating the affected patients. This has paved the way for the development and design of antibiotics against the previously less explored newer targets. Among these, peptidoglycan (PG) biosynthesis serves as a promising target for the design and development of novel drugs. The peptidoglycan cell wall synthesis in bacteria is essential for its viability. The enzyme class, Mur ligases, plays a key role in PG biosynthesis. Therefore, compounds with the ability to inhibit these e
