Baicalein is a unique flavonoid compound with important pharmacological activities, derived from Scutellaria baicalensis Georgi. Baicalein, as the aglycone of baicalin, is a key form for exerting pharmacological activity in vivo. β-glucuronidases (GUSs) are the enzymes involved in the conversion of baicalin to baicalein. In this study, the content of baicalein in S. baicalensis was significantly increased by 20.44% after treatment with 5% PEG6000. Seven GUSs from the glycoside hydrolase 79 family were identified through comparative transcriptome analysis. Among them, GUS1 and GUS2 were confirm…

A novel polysaccharide SPS01-2 (87.5 kDa) was isolated from the roots of Scutellaria baicalensis Georgi. Monosaccharide composition revealed that SPS01-2 consists of rhamnose, arabinose, galactose, galacturonic acid, and glucuronic acid in ratio of 4.4: 67.1: 22.2: 6.3: 1.2. Further investigations using methylation, NMR, and mass spectrometry indicated that SPS01-2 is classified as a type II arabinogalactan (AG-II) with a minor presence of type I rhamnogalacturonan (RG-I). The core structure alternates between 1,2/1,2,4-α-L-Rhap and 1,4-α-D-GalpA, with branches including 1,3,6-β-D-Galp, 1,3-β-…

INTRODUCTION: Genetic selection in broiler chickens has led to increased muscle mass without comparable respiratory and cardiovascular system development, limiting the birds' capacity to withstand high ambient temperatures and making them vulnerable to heat stress (HS). Early embryonic Thermal Manipulation (TM) has been suggested as an effective and sustainable way to mitigate the adverse effects of HS. This study investigated how these interventions influenced the immune status of broiler chickens exposed to HS.
METHODS: Cobb 500 fertile eggs (n = 600) were incubated according to guidelines.

In adverse environments, fine dust is linked to a variety of health disorders, including cancers, cardiovascular, neurological, renal, reproductive, motor, systemic, and respiratory diseases. Although PM10 is associated with oral inflammation and cancer, there is limited research on biomaterials that prevent damage caused by fine dust. In this study, we evaluated the effects of biomaterials using microRNA profiling, flow cytometry, conventional PCR, immunocytochemistry, Alizarin O staining, and ELISA. Compared to SBE (Scutellaria baicalensis extract), the preventive effectiveness of SBEIEs (SB…

Psoriasis is a chronic and recurrent skin problem that affects 3% of the global population. Nowadays, most medicines may not promise a complete cure for patients with psoriasis because of the development of pharmacoresistance and the side effects of drugs due to the microenvironment impact in the context of skin imbalance. Herein, we attempt to explore the pharmaceutical efficacy of Scutellaria baicalensis (S. baicalensis) in modulating the microenvironment created by macrophages and keratinocytes in psoriasis. The results indicated that treatment of S. baicalensis extract significantly reduce…

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by clinical symptoms of diarrhea and mucopurulent bloody stools, and its incidence is increasing globally. The etiology and pathogenesis of UC remain elusive. Current therapeutic approaches, including anti-inflammatory, immunosuppressive and immunomodulating agents, are often limited in efficacy and frequently associated with adverse drug reactions. Therefore, there is an urgent need to develop safer and more effective treatment strategies to address the limitations of existing therapies. Scutellaria baicalensis Geor…

OBJECTIVE AND RATIONALE: Inflammatory bowel disease, including Crohn's disease and ulcerative colitis, manifests with chronic intestinal inflammation and frequent sequential fibrosis. Current pharmacological therapies may show harmful side effects and are not useful for prevention or resolution of fibrosis. Thus, the use of alternative therapies is emerging as a novel useful approach. Previous results suggest that Scutellaria baicalensis Georgi (SBG) and Boswellia serrata (BS) display anti-inflammatory properties. The aim of this study was to investigate in intestinal epithelial cells and fibr…

The genus Scutellaria (Lamiaceae) is a phytochemically rich and medicinally important group of plants. Scutellaria species have been characterized by biologically active flavonoids such as baicalin, baicalein, and wogonin. In the present study, the essential oils of S. angustifolia, S. baicalensis, S. barbata, and S. lateriflora, cultivated in eastern Oregon, were obtained by means of hydrodistillation and analyzed using gas chromatographic methods. We hypothesize that the essential oils have compositions that may play a role in the traditional uses and biological activities of the genus. The

Nine flavonoids were isolated and identified from a chloroform-soluble fraction of the roots of Scutellaria baicalensis through a bioactivity-guided fractionation using a proprotein convertase subtilisin/kexin type 9 (PCSK9) monitoring assay in HepG2 cells. All structures were established by interpreting the corresponding spectroscopic data and comparing measured values from those in the literature. All compounds were assessed for their ability to inhibit PCSK9 mRNA expression; compounds 1 (3,7,2'-trihydroxy-5-methoxy-flavanone) and 4 (skullcapflavone II) were found to suppress PCSK9 mRNA via …

BACKGROUND: In plants, flavonoids predominantly exist as glycosides, which exhibit high polarity and low lipid solubility, thereby limiting their absorption across the intestinal mucosa. Baicalin (Bi), a glycoside, and its aglycone baicalein (Ba), derived from Scutellaria baicalensis, serve as representative examples of this bioavailability challenge.
PURPOSE: The aim of this study was to develop a green, one-pot strategy integrating ultra-high pressure (UHP)-assisted hydroxypropyl-β-cyclodextrin (HP-β-CD) extraction with endogenous enzyme-mediated in situ biotransformation and HP-β-CD inclus…

Scutellariae Radix (root of Scutellaria baicalensis) has a long history of application in traditional and in modern herbal medications. The major components of Scutellariae Radix are baicalin, baicalein, wogonoside and wogonin. Accumulating evidence demonstrates that Scutellaria has immunomodulatory effects and possesses compelling anticancer potential. Treatment of peripheral blood leukocytes (PBLs) with Scutellaria extract (SBE) enriched in baicalin, reduced viability of PBLs obtained from patients with acute lymphoblastic leukemia (ALL). SBE had no impact on the survival of healthy, control…

Scutellaria baicalensis Georgi (SBG) is recognized as a species with both medicinal and nutritional value, exhibiting extensive pharmacological activities and edible value. SBG from multiple origins and in two distinct specifications is widely available in the market at the present time. A rapid and accurate analytical method was developed by combining machine learning with metabolomics strategies to discriminate between two specifications and four sources of SBG, and to identify their respective differential markers (DMs). The establishment of a qualitative identification model to distinguish…
