In vivo third-harmonic generation microscopy study on vitiligo patients.

Melanin is known to provide strong third-harmonic generation (THG) contrast in human skin. With a high concentration in basal cell cytoplasm, THG contrast provided by melanin overshadows other THG sources in human skin studies. For better understanding of the THG signals in keratinocytes without the influence of melanin, an in vivo THG microscopy (THGM) study was first conducted on vitiliginous skin. As a result, the THG-brightness ratio between the melanin-lacking cytoplasm of basal cells and collagen fibers is about 1.106 at the dermal-epidermal junctions of vitiliginous skin, indicating hig

Nonlinear Imaging Allows to Characterize Granulomas and Fibrosis in Mice Tissue From Experimental Schistosomiasis.

Schistosomiasis is a neglected disease caused by parasites of the genus Schistosoma that poses serious public health problems. The liver is one of the organs seriously affected by the disease as it lodges Schistosoma eggs. We show results by nonlinear imaging techniques, Second Harmonic Generation and Two-Photon Excitation Fluorescence, to characterize mice tissues infected with Schistosoma mansoni in an experimental model. We followed the evolution of the disease in liver tissue sections through three stages: 30, 60, and 120 days post-infection-the pre-patent, acute, and early chronic phases,

Large field-of-view metabolic profiling of murine brain tissue following morphine incubation using label-free multiphoton microscopy.

BACKGROUND: Although the effects on neural activation and glucose consumption caused by opiates such as morphine are known, the metabolic machinery underlying opioid use and misuse is not fully explored. Multiphoton microscopy (MPM) techniques have been developed for optical imaging at high spatial resolution. Despite the increased use of MPM for neural imaging, the use of intrinsic optical contrast has seen minimal use in neuroscience. NEW METHOD: We present a label-free, multimodal microscopy technique for metabolic profiling of murine brain tissue following incubation with morphine sulfate

Machine learning-assisted mid-infrared spectrochemical fibrillar collagen imaging in clinical tissues.

SIGNIFICANCE: Label-free multimodal imaging methods that can provide complementary structural and chemical information from the same sample are critical for comprehensive tissue analyses. These methods are specifically needed to study the complex tumor-microenvironment where fibrillar collagen's architectural changes are associated with cancer progression. To address this need, we present a multimodal computational imaging method where mid-infrared spectral imaging (MIRSI) is employed with second harmonic generation (SHG) microscopy to identify fibrillar collagen in biological tissues. AIM: T

Intravital Microscopy in Atherosclerosis Research.

Atherosclerosis is a lipid-driven inflammatory disorder that narrows the arterial lumen and can induce life-threatening complications from coronary artery disease, cerebrovascular disease, and peripheral artery disease. On a mechanistic level, the development of novel cellular-resolution intravital microscopy imaging approaches has recently enabled in vivo studies of underlying biological processes governing disease onset and progress. In particular, multiphoton microscopy has emerged as a promising intravital imaging tool utilizing two-photon-excited fluorescence and second-harmonic generatio

Multiphoton microscopic imaging of human normal and cancerous oesophagus tissue.

In this paper, microstructures of human oesophageal submucosa are evaluated using multiphoton microscopy, based on two-photon excited fluorescence and second harmonic generation. The content and distribution of collagen, elastic fibers and cancer cells in normal and cancerous submucosa layer have been distinctly obtained and briefly discussed. The variation of these components is very relevant to the pathology in oesophagus, especially in early oesophageal cancer. Our results further indicate that the multiphoton microscopy technique has the potential application in vivo in clinical diagnosis

Second harmonic generation for brain imaging: pathology-related studies.

Microscopy of the brain has been facing problems of contrast and thick tissue imaging. Second harmonic generation (SHG) is a non-linear effect of the light interaction with the imaged material, resulting in photon emission at half the wavelength of the absorbed light. SHG microscopy provides an unprecedented opportunity for imaging collagen and other noncentrosymmetric protein fibrils in unstained thick tissue samples and in the live brain via a regular multiphoton setup. This opens a remarkable methodological window for imaging pathological processes of high importance, including brain trauma

Differential biomechanical properties of mouse distal colon and rectum innervated by the splanchnic and pelvic afferents.

Visceral pain is one of the principal complaints of patients with irritable bowel syndrome, and this pain is reliably evoked by mechanical distension and stretch of distal colon and rectum (colorectum). This study focuses on the biomechanics of the colorectum that could play critical roles in mechanical neural encoding. We harvested the distal 30 mm of the colorectum from mice, divided evenly into three 10-mm-long segments (colonic, intermediate and rectal), and conducted biaxial mechanical stretch tests and opening-angle measurements for each tissue segment. In addition, we determined the col

Optical characterization of ocular collagen microstructure and anisotropy in animal and human eyes.

Second-harmonic generation (SHG) microscopy enabled label-free, high-resolution visualization of ocular collagen structures in mouse, rabbit, and human pterygium tissues. Quantitative analyses using fast Fourier transform (FFT), gray-level co-occurrence matrix (GLCM), and polarization-resolved SHG revealed regional variations in collagen alignment, texture anisotropy, and optical polarization dependence. The results demonstrate that SHG imaging can sensitively capture microstructural organization and pathological remodeling within corneal and conjunctival tissues. This study highlights the pot

Three-dimensional investigation and scoring of extracellular matrix remodeling during lung fibrosis using multiphoton microscopy.

The organization of collagen during fibrotic processes is poorly characterized because of the lack of appropriate methodologies. Here we show that multimodal multiphoton microscopy provides novel insights into lung fibrosis. We characterize normal and fibrotic pulmonary tissue in the bleomycin model, and show that second-harmonic generation by fibrillar collagen reveals the micrometer-scale three-dimensional spatial distribution of the fibrosis. We find that combined two-photon excited fluorescence and second-harmonic imaging of unstained lung tissue allows separating the inflammatory and fibr

Enabling 3D refractive index tomography in laser scanning microscopy with a simple tape-on detector.

We introduce quantitative scanned oblique back-illumination microscopy (qsOBM) for laser scanning microscopes (LSMs), enabling quantitative three-dimensional refractive index (RI) tomography in thick scattering samples. By exploiting the reciprocity of light scattering, the established camera-based qOBM reconstruction framework is extended to a scanning implementation. Experimentally, qsOBM is realized with a slide-mounted four-quadrant photodetector requiring only minimal optomechanical modification of a conventional LSM. We demonstrate quantitative RI imaging and multimodal qsOBM/epi-detecte

Quantitative changes in human epithelial cancers and osteogenesis imperfecta disease detected using nonlinear multicontrast microscopy.

We show that combined multimodal nonlinear optical (NLO) microscopies, including two-photon excitation fluorescence, second-harmonic generation (SHG), third harmonic generation, and fluorescence lifetime imaging microscopy (FLIM) can be used to detect morphological and metabolic changes associated with stroma and epithelial transformation during the progression of cancer and osteogenesis imperfecta (OI) disease. NLO microscopes provide complementary information about tissue microstructure, showing distinctive patterns for different types of human breast cancer, mucinous ovarian tumors, and ski

Minoxidil decreases collagen I deposition and tissue-like contraction in clubfoot-derived cells: a way to improve conservative treatment of relapsed clubfoot?

AIM: Clubfoot is a congenital deformity affecting the musculoskeletal system, resulting in contracted and stiff tissue in the medial part of the foot. Minoxidil (MXD) has an inhibitory effect on lysyl hydroxylase, which influences the quality of extracellular matrix crosslinking, and could therefore be used to reduce the stiffness and to improve the flexibility of the tissue. We assessed the in vitro antifibrotic effects of minoxidil on clubfoot-derived cells. METHODS: Cell viability and proliferation were quantified by xCELLigence, MTS, and LIVE/DEAD assays. The amount of collagen I deposite

Ex vivo and in vivo second-harmonic-generation imaging of dermal collagen fiber in skin: comparison of imaging characteristics between mode-locked Cr:forsterite and Ti:sapphire lasers.

Second-harmonic-generation (SHG) microscopy is an interesting new tool for observing dermal collagen fiber in skin. However, conventional SHG microscopy using a mode-locked Ti:sapphire laser suffers from low penetration depth and a slow image acquisition rate caused by scattering and absorption in tissue, making it difficult to use for in vivo applications on human skin. We develop an SHG microscope equipped with a mode-locked Cr:forsterite laser with a long wavelength and compare its imaging characteristics with that of a Ti:sapphire-laser-based SHG microscope for the measurement of dermal co

Label-Free Non-Linear Optics for the Study of Tubulin-Dependent Defects in Central Myelin.

The satisfactory visualization of cytoskeletal components in the brain is challenging. The ubiquitous distribution of the networks of microtubules, microfilaments, and intermediate filaments in all the neural tissues, together with the variability in the outcomes of fluorescent protein fusion strategies and the limited applicability to dynamic studies of antibodies and drugs as chromophore vehicles, make classical optical approaches not as effective as for other proteins. When tubulin needs to be studied, the label-free generation of second harmonics is a very suitable option due to the non-ce

Uterine Vulnerability to Environmental PM2.5: Chronic Wood Smoke Exposure Alters Morphogenesis Before First Pregnancy.

Chronic exposure to fine particulate matter (PM2.5) derived from residential wood combustion is a major environmental health concern in southern Chile and other cold-climate regions. Although PM2.5 has been linked to adverse reproductive outcomes, it remains unclear whether sustained exposure induces pregestational uterine alterations that compromise reproductive competence before the first pregnancy. This study evaluated the effects of chronic wood smoke-derived PM2.5 exposure on uterine morphology and molecular markers in nulliparous rats. A two-generation exposure model was used to assess c

Detecting drug-induced nephrotoxicity using simultaneous label-free autofluorescence multiharmonic microscopy.

Drug-induced nephrotoxicity (DIN) is a major cause of drug development failure, yet many cases are detected only late in clinical trials or after approval. Conventional methods to detect DIN often lack sensitivity and specificity, particularly for early or region-specific kidney injury. Here, we evaluate Simultaneous Label-free Autofluorescence Multiharmonic (SLAM) microscopy as a rapid, label-free imaging approach to quantify kidney microstructural and functional metabolic changes associated with DIN. SLAM simultaneously captures endogenous NAD(P)H and FAD autofluorescence, collagen-derived s

HDAC Inhibition Reverses Preexisting Diastolic Dysfunction and Blocks Covert Extracellular Matrix Remodeling.

BACKGROUND: Diastolic dysfunction (DD) is associated with the development of heart failure and contributes to the pathogenesis of other cardiac maladies, including atrial fibrillation. Inhibition of histone deacetylases (HDACs) has been shown to prevent DD by enhancing myofibril relaxation. We addressed the therapeutic potential of HDAC inhibition in a model of established DD with preserved ejection fraction. METHODS: Four weeks after uninephrectomy and implantation with deoxycorticosterone acetate pellets, when DD was clearly evident, 1 cohort of mice was administered the clinical-stage HDAC

Second Harmonic Generation Imaging as a Virtual Biopsy for Upper Extremity Nerve Injuries: A Cadaver Study.

BACKGROUND: A timely assessment of nerve damage is needed for early clinical decision-making. Second Harmonic Generation (SHG) microscopy visualizes collagen. This study investigated: Can SHG imaging distinguish collagenous substructures in human nerves? Can SHG imaging detect structural damage in human nerves following destructive stretch injury? METHODS: Six human upper extremities were equally divided into 2 groups: A no-injury and a load-to-failure (LTF) group. The median, radial, and ulnar nerves were surgically exposed. Arms were placed on an Instron material testing system. Eight centi
