Contrasting distribution of physiological cell types in different regions of the bed nucleus of the stria terminalis.

We characterized the electroresponsive and morphological properties of neurons in the bed nucleus of the stria terminalis (BNST). Previously, Rainnie and colleagues distinguished three cell types in the anterolateral region of BNST (BNST-AL): low-threshold bursting cells (LTB; type II) and regular spiking neurons that display time-dependent (RS; type I) or fast (fIR; type III) inward rectification in the hyperpolarizing direction (Hammack SE, Mania I, Rainnie DG. J Neurophysiol 98: 638-56, 2007). We report that the same neuronal types exist in the anteromedial (AM) and anteroventral (AV) regio

Central and peripheral distribution of corticotropin-releasing factor.

Corticotropin-releasing factor (CRF) is widely distributed in the central nervous system and is associated with nuclei and pathways that control the release of adrenocorticotropin and beta-endorphin from the pituitary, and others that mediate central and peripheral autonomic responses to stress. Major concentrations of CRF immunoreactive neurons have been described in the hypothalamus, parts of the limbic system, several nuclei of the basal forebrain and brain stem, and the cerebral cortex. Recent data on the distribution of CRF immunoreactive perikarya and fibers in the preoptic-septal area,

Cytometrical and immunocytochemical investigation of brain nuclei in dementia.

Morphometrical investigation of brain nuclei in dementia of the Alzheimertyme and in other types of dementia has been done for the last 10 years. The N. Basalis of Meynert (Acetylcholine pathway) has been screened rather extensively by several groups. The L. Coeruleus (Nor-adrenergic pathway) also has been fairly well documented. The N. Raphe Dorsalis however-an important relay nucleus in the serotonergic pathway-was only studied cytometrically by a few groups. Other nuclei were not investigated systematically f.e. N. Accumbens; this nucleus might play an important role however in memory distu

An ultrastructural study of lipodystrophia centrifugalis abdominalis infantilis, with special reference to fibrous long-spacing collagen.

Lipodystrophia centrifugalis abdominalis infantilis (LCAI) in a 2.5-year-old Japanese girl is reported. Clinically she had a curved cutaneous depression with a slightly elevated erythematous border on the left abdomen beyond the left groin. The regional lymph node was palpable. Histologic examination showed a decrease of the fat, an inflammatory infiltrate mainly composed of lymphocytes, and septal fibrosis in the subcutaneous tissue. Immunohistochemical analysis found that infiltrating lymphocytes were mainly positive against CD4. Ultrastructurally nuclei in the adipocytes were crescent shape

Light microscopic immunocytochemical localization of pyruvate dehydrogenase complex in rat brain: topographical distribution and relation to cholinergic and catecholaminergic nuclei.

Pyruvate dehydrogenase complex (PDHC; EC 1.2.4.1, EC 2.3.1.12 and EC 1.6.4.3) includes 3 catalytically active mitochondrial enzymes involved in the formation of cellular energy through the tricarboxylic acid cycle and in the synthesis of ACh. We sought to determine whether immunocytochemically detected PDHC was enriched in neurons of the rat CNS, and, if so, whether the perikarya containing higher levels of PDHC immunoreactivity were differentially distributed with respect to their size or location within nuclear groups containing ACh, catecholamines or other unidentified transmitters. Dendritic reorganisation in the basal forebrain under degenerative conditions and its defects in Alzheimer's disease. III. The basal forebrain compared with other subcortical areas.

The distribution of the reticular neuronal type in the human brain and its involvement in both degeneration and dendritic reorganisation under the conditions of ageing, Korsakoff's disease (KD), Alzheimer's disease (AD), and Parkinson's disease (PD) was comparatively investigated after Golgi impregnation. Reticular neurones are distributed throughout different areas along the brain axis. The cholinergic basal forebrain nuclei, i.e., the basal nucleus of Meynert, the nucleus of the diagonal band, and the medial septal nucleus form the most rostral part of this network of "open nuclei," which is

An autoradiographic study of the efferent connections of the lateral hypothalamic area in the rat.

The efferent connections of the lateral hypothalamic area (LHA) have been analyzed in a series of 30 rat brains with injections of 3H-amino acids into different parts of the area and the surrounding regions. Our findings indicate that all parts of the LHA contribute ascending and descending fibers to the medial forebrain bundle, and also project medially to certain of the adjoining hypothalamic nuclei. All levels of the LHA appear to send some fibers to a continuous group of structures that extends from the medial septal-diagonal band complex rostrally, through the lateral preoptic and lateral

[Changes in neurotransmission systems after the injection of beta-amyloid protein beta (12-28) in the hypothalamus and anterior thalamus of the rat].

INTRODUCTION AND OBJECTIVE: Alzheimer's disease (AD) is a degenerative central nervous system disorder. The histopathologic features include senile plaques, which are composed primarily of insoluble aggregates of amyloid beta-protein (A beta). The purpose of this study was to analyse the effects of A beta on several neurotransmitter/neuromodulator systems as a possible model for the neuropathological changes that occur in AD. MATERIAL AND METHODS: Adult rats (Wistar, 200-300 g) received injections of A beta (12-28) (0.5 microliter-2 microliters, 1 microgram-4 micrograms) into the hypothalamus

D-aspartate localizations imply neuronal and neuroendocrine roles.

Though L-amino acids predominate in living organisms, substantial levels of free D-serine and D-aspartate occur in mammals, especially in nervous and endocrine tissues. Using an antibody specific for glutaraldehyde-fixed D-aspartate, we have localized D-aspartate in rat tissues. In the brain we observe discrete neuronal localizations of D-aspartate, especially in the external plexiform layer of the olfactory bulb, hypothalamic supraoptic and paraventricular nuclei, the medial habenula, and certain brainstem nuclei. In rats 3-4 weeks old, we observe D-aspartate in septal nuclei and in a subset

The distribution of melatonin binding sites in neuroendocrine tissues of the ewe.

Specific high-affinity binding of 2-[125I]iodomelatonin (IMEL) was examined in 20-micrometer sections prepared from intact Suffolk ewes killed during late anestrus or the breeding season. The pars tuberalis contained by far the highest concentration of IMEL binding sites of all areas studied. Within the telencephalon, intense labeling was found in the mediolateral septum, the ventrolateral septal and septohypothalamic nuclei, the entorhinal cortex, the subiculum, and the inner and outer molecular layers of the hippocampus adjacent to the dentate gyrus. Melatonin binding in the medial preoptic

Closed-loop stimulation of the medial septum terminates epileptic seizures.

Temporal lobe epilepsy with distributed hippocampal seizure foci is often intractable and its secondary generalization might lead to sudden death. Early termination through spatially extensive hippocampal intervention is not feasible directly, because of the large size and irregular shape of the hippocampus. In contrast, the medial septum is a promising target to govern hippocampal oscillations through its divergent connections to both hippocampi. Combining this 'proxy intervention' concept and precisely timed stimulation, we report here that closed-loop medial septum electrical stimulation ca

Oscillatory activity in the BNST/ALIC and the frontal cortex in OCD: acute effects of DBS.

Deep brain stimulation (DBS) of the bed nucleus of the stria terminalis/anterior limb of the internal capsule (BNST/ALIC) is successfully used for treatment of patients with obsessive-compulsive disorder (OCD). Clinical and experimental studies have suggested that enhanced network synchronization in the theta band is correlated with severity of symptoms. The mechanisms of action of DBS remain unclear in OCD. We here investigate the effect of acute stimulation of the BNCT/ALIC on oscillatory neuronal activity in patients with OCD implanted with DBS electrodes. We recorded the oscillatory activi

Distribution of electric field in patients with obsessive compulsive disorder treated with deep brain stimulation of the bed nucleus of stria terminalis.

BACKGROUND: Deep brain stimulation (DBS) is being investigated as a treatment for therapy-refractory obsessive compulsive disorder (OCD). Many different brain targets are being trialled. Several of these targets such as the ventral striatum (including the nucleus accumbens (NAc)), the ventral capsule, the inferior thalamic peduncle, and the bed nucleus of stria terminalis (BNST)) belong to the same network, are anatomically very close to one another, or even overlap. Data is still missing on how various stimulation parameters in a given target will affect surrounding anatomical areas and impac

Efferent connections of the hippocampus and adjacent regions in the pigeon.

In this investigation, the projections of the dorsomedial (viz. hippocampus) and adjacent dorsolateral cortical mantle (viz. parahippocampus) of the pigeon telencephalon were studied with the use of 3H-amino acid radioautography. The results indicate that the fibers which arise from these cortical areas project to the septum. The projection to this structure is topographically organized in a crude manner along the longitudinal axis of the hippocampal complex. Fibers arising from rostral levels of the dorsomedial hippocampal complex project principally to central portions of the precommissural

What hope for repair of the brain?

The consequences of axonal injuries in the adult nervous system depend on the location of the axons: complete anatomical and functional repair can occur in the peripheral but not the central nervous system. A quantitative electron microscopical comparison of the reactions to axonal injuries in a peripheral site (the superior cervical sympathetic ganglion) and in a central site (the septal nuclei) in adult rats shows that in both cases the denervated postsynaptic sites are reinnervated by new formation of synapses. However, in the ganglion it is the originally severed axons which form the new s

Cerebellar stimulation: effects on septal region, hippocampus, and amygdala of cats and rats.

The influence of the cerebellum on activity of the septal region, hippocampus, and amygdala of cats and rats was determined by obtaining unit recordings from those supratentorial sites with cerebellar stimulation. With stimulation of the postral vermis, fastigial nucleus, and intervening midline folia of the cerebellum, units in the septal region were facilitated, whereas those in the hippocampus were inhibited. Mixed results were obtained in the amygdala, some units being facilitated and others inhibited. Stimulation over the lateral cerebellar hemispheres and dentate nucleus, on the other ha

Stress ensemble in the BNST mediates comorbid anxiety and sleep disruption.

Anxiety and insomnia are highly comorbid; however, the underlying brain-circuit mechanisms remain inadequately explored. Here, we identify a dual-functional ensemble within the bed nucleus of the stria terminalis (BNST) that modulates both anxiety-like behaviors and insomnia through distinct neural circuits. Specifically, a subset of BNST neurons exhibits enhanced activity in response to anxiety-related cues and during wakefulness. Activation of these ensemble neurons exacerbates anxiety-like behaviors and reduces non-rapid eye movement (NREM) sleep in chronic restraint stress (CRS) mice. Conv

When stress becomes wakefulness: Stress-arousal circuitry and the state control of insomnia.

Insomnia disorder (ID) is the most common sleep disorder and among the most prevalent neuropsychiatric conditions, affecting ∼9-20% of adults with substantial burden. Chronic ID is linked to cardiometabolic and psychiatric morbidity, cognitive impairment, and accelerated neurodegeneration. Although behavioral and pharmacologic therapies help, relapse is common, suggesting a pathophysiology not simply of "too little sleep" but a persistent bias toward wakefulness. Current models converge on ID as a disorder of hyperarousal, yet the circuit, cellular, and synaptic mechanisms by which stress prod
