Efficient targeting of conserved cryptic epitopes of infectious agents by single domain antibodies. African trypanosomes as paradigm.

Antigen variation is a successful defense system adopted by several infectious agents to evade the host immune response. The principle of this defense strategy in the African trypanosome paradigm involves a dense packing of variant surface glycoproteins (VSG) exposing only highly variable and immuno-dominant epitopes to the immune system, whereas conserved epitopes become inaccessible for large molecules. Reducing the size of binders that target the conserved, less-immunogenic, cryptic VSG epitopes forms an obvious solution to combat these parasites. This goal was achieved by introducing drome

The breadth of cross sub-type neutralisation activity of a single domain antibody to influenza hemagglutinin can be increased by antibody valency.

The response to the 2009 A(H1N1) influenza pandemic has highlighted the need for additional strategies for intervention which preclude the prior availability of the influenza strain. Here, 18 single domain VHH antibodies against the 2009 A(H1N1) hemagglutinin (HA) have been isolated from a immune alpaca phage displayed library. These antibodies have been grouped as having either (i) non-neutralising, (ii) H1N1 restricted neutralising or (iii) broad cross-subtype neutralising activity. The ability to neutralise different viral subtypes, including highly pathogenic avian influenza (H5N1), correl

Indium-111-Labeled Single-Domain Antibody for In Vivo CXCR4 Imaging Using Single-Photon Emission Computed Tomography.

C-X-C chemokine receptor type 4 (CXCR4) is highly expressed in a range of pathologies, including cancers like multiple myeloma and non-Hodgkin lymphoma, inflammatory diseases such as rheumatoid arthritis, and viral infections like HIV. Currently, the most advanced radiotracer for CXCR4 imaging in clinics is [68Ga]PentixaFor. However, its structure is prone to modifications, complicating the development of a specific CXCR4 fluorine-18-labeled tracer with good pharmacokinetic properties. This study aimed to screen multiple CXCR4-targeting variable domains of heavy-chain-only antibody (VHH or sin

Characterization of single-domain antibodies against Foot and Mouth Disease Virus (FMDV) serotype O from a camelid and imaging of FMDV in baby hamster kidney-21 cells with single-domain antibody-quantum dots probes.

BACKGROUND: Foot-and-mouth disease (FMD) is a highly contagious disease that affects cloven-hoofed animals and causes significant economic losses to husbandry worldwide. The variable domain of heavy-chain antibodies (VHHs or single domain antibodies, sdAbs) are single-domain antigen-binding fragments derived from camelid heavy-chain antibodies. RESULTS: In this work, two sdAbs against FMD virus (FMDV) serotype O were selected from a camelid phage display immune library and expressed in Escherichia coli. The serotype specificity and affinity of the sdAbs were identified through enzyme-linked i

Single-domain antibodies reveal unique borrelicidal epitopes on the Lyme disease vaccine antigen, outer surface protein A (OspA).

Camelid-derived, single-domain antibodies (VHHs) have proven to be extremely powerful tools in defining the antigenic landscape of immunologically heterogeneous surface proteins. In this report, we generated a phage-displayed VHH library directed against the candidate Lyme disease vaccine antigen, outer surface protein A (OspA). Two alpacas were immunized with recombinant OspA serotype 1 from Borrelia burgdorferi sensu stricto strain B31, in combination with the canine vaccine RECOMBITEK Lyme containing lipidated OspA. The phage library was subjected to two rounds of affinity enrichment ("pann

Isolation of Novel Fully Human Single-Domain Antibodies Targeting GUCY2C from a Heavy-Chain-Only Transgenic Mouse Platform.

IntroductionTraditional natural single-domain antibodies(sdAbs) are primarily derived from camelids and sharks. However, the large size and slow reproduction of these species limit their widespread application in antibody development. In this study, we employed a fully human heavy-chain-only antibody mouse ( huNano mouse ) platform and a rapid, efficient preparation process to successfully develop fully human heavy-chain-only antibodies(HcAbs) targeting the tumor antigen guanylyl cyclase C (GC-C). These sdAbs provide high-quality candidate molecules for subsequent GC-C-based CAR-T therapy and

Screening of Single-Domain Antibodies to Adeno-Associated Viruses with Cross-Serotype Specificity and a Wide pH Tolerance.

Adeno-associated virus (AAV) vectors are the preferred gene delivery tool in gene therapy owing to their safety, long-term gene expression, broad tissue tropism, and low immunogenicity. Affinity ligands that can bind multiple AAV serotypes endure harsh clean-in-place (CIP) conditions and are critical for industrial-scale purification. However, current ligands lack broad serotype recognition and adequate alkaline stability, which limits their reusability in large-scale manufacturing. In this study, we employed a competitive biopanning strategy to isolate a single-domain antibody (VHH) that simu

Single domain antibodies derived from ancient animals as broadly neutralizing agents for SARS-CoV-2 and other coronaviruses.

With severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) as an emergent human virus since December 2019, the world population is susceptible to coronavirus disease 2019 (COVID-19). SARS-CoV-2 has higher transmissibility than the previous coronaviruses, associated by the ribonucleic acid (RNA) virus nature with high mutation rate, caused SARS-CoV-2 variants to arise while circulating worldwide. Neutralizing antibodies are identified as immediate and direct-acting therapeutic against COVID-19. Single-domain antibodies (sdAbs), as small biomolecules with non-complex structure and intrins

A C-type single-domain antibody with protective efficacy against H1N1 via respiratory administration.

Influenza A viruses, particularly H1N1, pose a significant public health threat, highlighting the continued need for novel prophylactics and therapeutics. Monoclonal antibodies are promising, but conventional IgGs face challenges in respiratory delivery due to their large size. This study presents A1-B7, a C-type single-domain antibody (C-sdAb) based on a scaffold derived from a human IgG1 CH2 domain, targeting the hemagglutinin (HA) of H1N1 influenza virus. A1-B7 potently neutralizes multiple H1N1 subtypes in vitro. Both prophylactic intranasal administration and therapeutic aerosol inhalatio

Tuning the Structural Properties of a Single-Domain Antibody Scaffold for Improved Fibroblast Activation Protein Targeting.

Fibroblast activation protein (FAP) is an attractive target for the development of cancer theranostics due to its selective expression on cancer-associated fibroblasts (CAFs). While a number of small-molecule FAP inhibitors (FAPIs) have been developed, few biologics have been investigated as FAP-targeting vectors. Camelid-derived single-domain antibodies, or variable-heavy-heavy domains (VHHs), offer a compelling alternative, combining high affinity with versatile engineering options. In this study, we first identified a novel anti-FAP VHH, F7, from an affinity-matured camelid phage display li

Topical delivery of a human single-domain antibody targeting IL-33 to inhibit mucosal inflammation.

Addressing mucosal inflammatory disorders in the ocular surface or respiratory system remains a formidable challenge owing to the limited penetration of biological therapeutics across epithelial barriers. In this study, we explored the potential of human single-domain antibodies (UdAbs) as topical therapeutics for the targeted modulation of interleukin-33 (IL-33) in two mucosal-associated inflammatory disorders. The anti-IL-33 UdAb A12 demonstrated potent inhibition of the IL-33-mediated signaling pathway, despite not potently blocking the IL-33 receptor interaction. Compared with the anti-IL-

Preparation of divalent camelid single-domain antibody and its application in immunoassays for Salmonella detection in food.

Salmonella-related foodborne infections are commonly caused by the serovars of S. Typhimurium, which can be detected using antibody-based immunoassays. The monovalent variable domain of the camelid heavy chain antibody (VHH) performs excellently in constructing multivalent VHH variants, which generally exhibit higher affinities with antigens and consequently enhance the assay sensitivity. In this study, the divalent variants of VHHs (diVHHs) targeting S. Typhimurium were generated by encoding the monovalent VHH genes assembled in tandem with a flexible linker peptide (G4S)2. Soluble diVHHs wer

Development of an Engineered Single-Domain Antibody for Targeting MET in Non-Small Cell Lung Cancer.

The Mesenchymal Epithelial Transition (MET) receptor tyrosine kinase is upregulated or mutated in 5% of non-small-cell lung cancer (NSCLC) patients and overexpressed in multiple other cancers. We sought to develop a novel single-domain camelid antibody with high affinity for MET that could be used to deliver conjugated payloads to MET expressing cancers. From a naïve camelid variable-heavy-heavy (VHH) domain phage display library, we identified a VHH clone termed 1E7 that displayed high affinity for human MET and was cross-reactive with MET across multiple species. When expressed as a bivalent

Generation of HIV-resistant cells with a single-domain antibody: implications for HIV-1 gene therapy.

The cure or functional cure of the "Berlin patient" and "London patient" indicates that infusion of HIV-resistant cells could be a viable treatment strategy. Very recently, we genetically linked a short-peptide fusion inhibitor with a glycosylphosphatidylinositol (GPI) attachment signal, rendering modified cells fully resistant to HIV infection. In this study, GPI-anchored m36.4, a single-domain antibody (nanobody) targeting the coreceptor-binding site of gp120, was constructed with a lentiviral vector. We verified that m36.4 was efficiently expressed on the plasma membrane of transduced TZM-b

Advanced genetic engineering to achieve in vivo targeting of adenovirus utilizing camelid single domain antibody.

For the developing field of gene therapy the successful address of the basic requirement effective gene delivery has remained a critical barrier. In this regard, the "Holy Grail" vector envisioned by the field's pioneers embodied the ability to achieve efficient and specific in vivo gene delivery. Functional linkage of antibody selectivity with viral vector efficiency represented a logical strategy but has been elusive. Here we have addressed this key issue by developing the technical means to pair antibody-based targeting with adenoviral-mediated gene transfer. Our novel method allows efficie

Single oral dose of bovine Fc-fused single-domain antibody enables multi-week serum detectability in neonatal calves.

Neonatal calf diarrhea caused by bovine rotavirus (BRV) remains a major health and economic burden in cattle production, and current oral antibody approaches require frequent dosing because of poor systemic persistence. This study investigated whether a single oral dose of a BRV-specific antibody engineered with an immunoglobulin Fc region-designed to extend circulatory half-life-could be detected in the blood of newborn calves for up to 28 days. The antibody construct showed high-affinity binding to BRV antigen and effectively neutralized viral infection in cell culture. In a controlled calf

Structural basis of influenza A virus neutralization by broadly reactive single-domain antibody G2.3 recognizing glycosylated epitope within hemagglutinin stem domain.

Influenza remains one of the most common and contagious respiratory infections causing around a billion cases of seasonal illness annually and five million cases of severe consequences. A recently reported G2.3 antibody exhibited a potent cross-subtype activity against Group 1 influenza A viruses. Here, we shed light on the structural basis of the broad neutralizing activity of G2.3 by using cryoEM. Structural analysis of the G2.3 complex with H1 hemagglutinin revealed a partly conserved epitope located on the stem domain. The structural data were confirmed by the assessment of binding and neu

Controlled immobilization of single-domain antibodies using cellulose-binding modules for AB5-type toxin detection in paper-based assays.

The rapid and sensitive detection of toxigenic Vibrio cholerae and enterotoxigenic Escherichia coli remains a critical challenge for effective disease screening and environmental monitoring, which is essential for guiding clinical management and outbreak control in regions where diarrheal diseases are endemic. To address this, we developed a single-domain antibody (VHH)-based sandwich-format lateral flow assay for the rapid and sensitive detection of cholera toxin from V. cholerae and heat-labile enterotoxin from enterotoxigenic Escherichia coli in human fecal and environmental water samples.

Rapid isolation and characterization of high-affinity single-domain antibodies for broad-Spectrum detection of SARS-CoV-2 variants.

The rapid evolution of SARS-CoV-2 presents substantial challenges to maintaining diagnostic accuracy. Single-domain antibodies (variable domains of camelid heavy-chain antibodies, VHHs), commonly referred to as nanobodies, are attractive tools for diagnostic applications due to their high specificity, stability, affinity and expression yield. In this study, three high-affinity VHHs that recognize the receptor-binding domain (RBD) of the SARS-CoV-2 BA.2 variant were isolated from a large naïve alpaca VHH phage display library. These nanobodies exhibited nanomolar affinities and high thermal sta
