Mitochondrial genomes of two subspecies of Citellophilus tesquorum: Insights into the molecular evolution and phylogenetic analysis within Siphonaptera.

BACKGROUND: Citellophilus tesquorum is a major flea vector in natural plague foci across the Palaearctic region of China, representing a significant threat to veterinary and public health. Precise identification and sound taxonomic delimitation of its subspecies are critical for effective vector surveillance and evolutionary research. METHODS: We first performed morphological identification to guarantee specimen authenticity and accuracy, then sequenced and annotated the complete mitochondrial genomes of C. t. dzetysuensis and C. t. mongolicus using the Illumina NovaSeq platform. Comparative

[Ecology of flea groups of the species conformis (Siphonaptera: Pulicidae: Xenopsylla) of the fauna of Russia and adjacent countries (review)].

Within the boundaries of the former USSR, the northern part of the conformis group distribution is located. It spreads over the arid regions of the Trans-Cacucasus, Pricaspijckaja lowland, Kazakhstan and Middle Asia. In this area 10 species and subspecies occur. They are mainly parasites of gerbillins. Unlike many other Siphonaptera the conformis fleas, when in the host's home, do not concentrate in the nest but inhabit the passages of burrow and food chambers throughout the year. On this reason the preimaginal development and existence of the adults take place not at the temperature of the ha

Mutation in the pla gene of Yersinia pestis alters the course of the plague bacillus-flea (Siphonaptera: Ceratophyllidae) interaction.

Yersinia pestis possesses a unique gene (pla) encoding coagulase and fibrinolysin which is implicated in the transmission of plague by fleas. This gene is encoded on the highly conserved but poorly characterized 'pesticin' plasmid pKYP1. The role of the pKYP1-encoded gene, pla, in plague transmission was addressed by feeding fleas on blood containing avirulent Y. pestis strain EV76-6 and three derivatives of this strain (K10-2, K10-3, and K10-5) carrying Tn801 insertions in pKYP1. One of these mutant strains, K10-5, contains an insertion within the pla gene that eliminates both coagulase and f

Detection of Rickettsia asembonensis in Fleas (Siphonaptera: Pulicidae, Ceratophyllidae) Collected in Five Counties in Georgia, United States.

We conducted a molecular survey of Rickettsia in fleas collected from opossums, road-killed and live-trapped in peridomestic and rural settings, state parks, and from pet cats and dogs in Georgia, United States during 1992-2014. The cat flea, Ctenocephalides felis (Bouché) was the predominant species collected from cats and among the archival specimens from opossums found in peridomestic settings. Polygenis gwyni (Fox) was more prevalent on opossums and a single cotton rat trapped in sylvatic settings. Trapped animals were infested infrequently with the squirrel flea, Orchopeas howardi (Baker)

Bartonella and Rickettsia from fleas (Siphonaptera: Ceratophyllidae) of prairie dogs (Cynomys spp.) from the western United States.

Fleas of prairie dogs have been implicated in the transmission of Bartonella spp. We used PCR to test DNA extracts from 47 fleas of prairie dogs from 6 states. We amplified DNA from 5 unique genotypes of Bartonella spp. and 1 Rickettsia sp. from 12 fleas collected in North Dakota, Oklahoma, Texas, and Wyoming. Sequences from the Bartonella spp. were similar, but not identical, to those from prairie dogs and their fleas in Colorado.

[Three new species of the genus Medwayella Traub, 1972 (Insecta: Siphonaptera: Pygiopsyllidae) from Sabah (eastern Malaysia, Borneo)].

Medwayella traubiana n. sp., M. pfeifferi n. sp. and M. sabahae n. sp. (Pygiopsyllidae) are described from Sabah (north of Borneo island), the first two on Tupaia tana (Scandentia), the last on Sundasciurus lowii (Rodentia). Sex male is only identified, because these fleas have been collected in sympatry, or even in syntopy. Their determination is based on segment IX and aedeagus. If M. traubiana and M. pfeifferi are related to some known species, M. sabahae is clearly distinct from other Medwayella.

Factors influencing sperm transfer and insemination in cat fleas (Siphonaptera: Pulicidae) fed on an artificial membrane system.

Sperm transfer through the epididymis, a prerequisite for insemination of cat fleas, Ctenocephalides felis (Bouché) was stimulated by exposure of unfed male fleas to juvenile hormone III residues for 3 d at 25 degrees C or exposure of unfed fleas to 37 degrees C for 6 d. Sperm transfer was completed at least three times faster in unfed males held at 37 degrees C than in those held at 25 degrees C. Although percentage sperm transfer in fleas fed water or 0.15 M saline at 37 degrees C was not significantly increased over that of unfed fleas, a significantly greater percentage of blood-fed males

The Trick of the Hedgehog: Case Report and Short Review About Archaeopsylla erinacei (Siphonaptera: Pulicidae) in Human Health.

Fleas are ectoparasites of various animals, including Homo sapiens Linnaeus, 1758 (Primates: Hominidae). Among the species relevant to the human health field, either due to their dermatopathological potential or because of their role as vectors of microorganisms responsible for infectious diseases, such as plague or murine typhus, are the human flea, oriental rat flea, closely related cat and dog fleas, and chigoe flea. However, other species can accidentally infest humans. We have herein reported two unusual cases of humans infested and bitten by Archaeopsylla erinacei, the hedgehog flea. Thi

Use of DNA hybridizations probes for detection of the plague bacillus (Yersinia pestis) in fleas (Siphonaptera: Pulicidae and Ceratophyllidae).

The detection of active plaque in nature relies primarily on demonstration of the etiologic agent of the disease. Yersinia pestis, in the flea vectors and susceptible mammalian hosts. A live animal assay is currently used for identification of a Y. pestis virulence antigen that is not expressed in the flea. We have found that DNA hybridization probes specific for Y. pestis, used in very simple sample preparation schemes, allow detection of Y. pestis in three species of fleas as well as tissues of experimentally infected mice at minimum concentrations of 1 x 10(6) bacilli/ml. We detected Y. pes

Factors affecting the seasonal abundance of ground squirrel and wood rat fleas (Siphonaptera) in San Diego County, California.

Abiotic and biotic factors affecting the seasonal abundance of 3 species of sylvatic fleas on their hosts were examined at 7 sites in San Diego County, California. Indices for Oropsylla (Diamanus) montana (Baker) were usually highest on Spermophilus beecheyi nudipes (Huey) when < 18.4 degrees C (October-December), whereas those for Hoplopsyllus anomalus (Baker) were highest when > 18.4 degrees C (July-September). O. montana was affected most by ambient temperature (inversely for the coastal site [134 m] and directly for most mountain sites [> 1,183 m]). O. montana was affected most (directly)

Early-phase transmission of Yersinia pestis by unblocked Xenopsylla cheopis (Siphonaptera: Pulicidae) is as efficient as transmission by blocked fleas.

For almost a century, the oriental rat flea, Xenopsylla cheopis (Rothschild) (Siphonaptera: Pulicidae), was thought to be the most efficient vector of the plague bacterium Yersinia pestis (Yersin). Approximately 2 wk after consuming an infectious bloodmeal, a blockage often forms in the flea's proventriculus, which forces the flea to increase its biting frequency and consequently increases the likelihood of transmission. However, if fleas remain blocked and continue to feed, they usually die within 5 d of blocking, resulting in a short infectious window. Despite observations of X. cheopis tran

[Description of Tunga bonneti n. sp. from Chile (Siphonaptera: Tungidae) with notes on specificity, chorology, dermecos and phenology].

Smit (1968) misreported Tunga libis from Chile. Here we describe this flea as a new species. We analyzed free life females, male and embedded females. Moreover, we include data about its natural history and biology. These data will allow a better understanding about other Tunga fleas, and particularly for collecting males, which remain unknown to date.

Anatomy of Tunga trimamillata Pampiglione et al., 2002 (Insecta, Siphonaptera, Tungidae) and developmental phases of the gravid female.

This paper deals with some internal anatomical features observed in histological sections and freshly dissected mounts of Tunga trimamillata, a Siphonaptera recently discovered in Andean regions of Ecuador from several mammals, including man. It was possible to study in males and also non-gravid and gravid females, the location and anatomy of several organs not previously described for this species: the testes, epididymis, ganglia, Malpighian tubules, eyes, rectal ampulla with one of its pads and structures which could be interpreted as midgut diverticula, whose presence has not been recorded

Parasite island syndromes in the context of nidicolous ectoparasites: Fleas (Insecta: Siphonaptera) in wild passerine birds from Azores Archipelago.

Island syndrome, previously established for isolation process of insular vertebrates' populations, have been adapted to insular parasites communities, termed parasite island syndromes. In this work, were studied for the first time the insular syndromes for nidicolous ectoparasites of the bird species, Turdus merula, Sylvia atricapilla, Fringilla coelebs and Erithacus rubecula from Azores and the mainland Portugal. Flea species were only recorded on Azorean birds, namely Dasypsyllus gallinulae and Ctenocephalides felis felis, known as not host-specific parasites. In the absence of shared flea s

Influence of Bloodmeal Host on Blood Feeding, Egg Production, and Offspring Sex Ratio of Ctenocephalides felis felis (Siphonaptera: Pulicidae).

The cat flea, Ctenocephalides felis felis (Bouché), feeds on different host species, causing annoyance or transmitting disease agents. In this study, the influence of the host of the cat flea on blood feeding, egg production, and sex ratio of the offspring was investigated. Two strains of C. felis were domesticated on either rats or mice for >10 yr in the laboratory, and in this study, these fleas were placed in the following groups and fed on rats or mice continuously: Group A (rat-domesticated C. felis with rats as host); Group B (rat-domesticated C. felis with mice as host); Group C (mouse-

Effect of Yersinia pestis infection on temperature preference and movement of the Oriental rat flea (Xenopsylla cheopis) (Siphonaptera: Pulicidae).

Previous laboratory studies have shown that inoculation of bacterial endotoxin into the hemocoel of some arthropods, or natural infection by a number of pathogens, causes them to seek out a higher ambient temperature. This phenomenon has been called behavioral fever. Yersinia pestis is an endotoxin-producing bacterium that relies on infection of fleas for transmission. Behavioral fever in fleas might enhance the transmission of plague if infected fleas were induced to seek out a warm-bodied host after the death of an infected host. Our study indicates that in thermal gradient Y. pestis infecte

Thermal niche partitioning and phenology of Nearctic and Palearctic flea (Siphonaptera) communities on rodents (Mammalia: Rodentia) from five ecoregions.

Seasonality of fleas (Siphonaptera) may be due to species competition, prompting the idea that flea species partition temperature, along with correlated variables such as moisture (thermal-niche partitioning hypothesis). I compared the fleas of five rodent-flea communities described from the literature for thermal-niche optima by fitting non-linear LRF (Lobry-Rosso-Flandrois) curves to examine whether flea species in a community show distinct, partitioned thermal niches. LRF curves estimate physiological parameters of temperature minimum, optimum, maximum, and maximum abundance, and facilitate

Cost-Effective PCR-Based Identification of Tunga penetrans (Siphonaptera) Larvae Extracted from Soil Samples Containing PCR Inhibitor-Rich Material.

Tungiasis is a neglected tropical disease caused by skin-penetrating female Tunga penetrans fleas. Although tungiasis causes severe health problems, its ecology is poorly understood and morphological descriptions of the larvae are unavailable. To identify T. penetrans immature stages and sites where they develop, diagnostic PCRs are required. However, flea larvae feed on soil organic matter rich in PCR inhibitors. Here, three DNA preparation methods, including a soil DNA kit that removes inhibitors, a simple ammonium acetate precipitation approach (AmAcet) and a crude lysate of larvae (CL), we

Reevaluation of the Role of Blocked Oropsylla hirsuta Prairie Dog Fleas (Siphonaptera: Ceratophyllidae) in Yersinia pestis (Enterobacterales: Enterobacteriaceae) Transmission.

Prairie dogs in the western United States experience periodic epizootics of plague, caused by the flea-borne bacterial pathogen Yersinia pestis. An early study indicated that Oropsylla hirsuta (Baker), often the most abundant prairie dog flea vector of plague, seldom transmits Y. pestis by the classic blocked flea mechanism. More recently, an alternative early-phase mode of transmission has been proposed as the driving force behind prairie dog epizootics. In this study, using the same flea infection protocol used previously to evaluate early-phase transmission, we assessed the vector competenc
