In spite of the large number of theories advanced to clarify the etiology of Paget's disease, its cause is still being discussed, and no satisfactory conclusion has been reached. The possibility of a viral origin was raised by the discovery of inclusion bodies, detectable by electron microscopy, in the nuclei and cytoplasm of the osteoclasts in the affected bone tissue, in 1974. In fact, the microcylindrical structures described by various authors, and visible only in osteoclasts and only in Paget's disease, if one excepts certain giant-cell bone tumors, have a close analogy, morphologically,

The antigenicity of the 'prion' protein amyloid fibrils was shown to be preserved after glutaraldehyde/OsO4 fixation in uranyl acetate-stained brain tissue blocks from patients with Gerstmann-Sträussler syndrome (GSS) and from mice infected with Creutzfeldt-Jakob disease (CJD). Amyloid plaques were demonstrated by light microscopy in immunogold silver-intensified semithin sections. Under the electron microscope, the amyloid fibrils were labelled in immunogold-reacted ultrathin sections using an antiserum prepared against GSS amyloid plaque cores and mouse amyloid fibrils respectively. The infl

Recent advances have been made in understanding the aetiology, diagnosis and treatment of Paget's disease of bone (osteitis deformans). The role of viruses in the pathogenesis of the disease has been extensively investigated, although no causative link has yet been demonstrated between infective agents and Paget's disease. Recent advances in the diagnosis of the disease have been made with increasing usage of radionucleotide imaging to determine the extent and distribution of the lesions. Treatment options have been extended with the emergence of the biphosphonates as potential main line thera

We investigated paraffin-embedded brain sections from 41 patients with Creutzfeldt-Jakob disease (CJD) and from 9 with Gerstmann-Sträussler syndrome (GSS) using anti-human prion protein (PrP) antisera (anti-GSS kuru plaque cores and anti-PrP synthetic peptide) and anti-beta protein antiserum. The anti-human PrP antiserum reacted with the plaques in CJD and GSS, with or without degenerative neurites (neuritic components). In addition, the anti-beta protein antiserum immunolabeled kuru plaque-like compact plaques in some cases of CJD. Therefore, previous morphological classifications of the plaq

The diagnosis of Jakob-Creutzfeld disease was made in two women (aged 59 and 76 years) whose course was marked by rapidly progressing signs of dementia, myoclonus and rhythmic bi- and tri-phasic steep waves in the EEG. The diagnoses were confirmed neuropathologically at autopsy. Since the disease is transmissible, precautions must be taken if the disease is suspected: wearing of disposable gloves, disinfection of instruments by one-hour autoclaving at 121 degrees C, disinfection of potentially contaminated surfaces with 5% sodium hypochlorite, and avoidance of skin cuts or punctures by instrum

The main directions of research in the field of viral neuroinfections approached during 35 years in the Institute of Virology are briefly outlined. After some considerations on terminology and on the classification of viral encephalitides, mention is made of the studies in the domain of herpes infections, rabies, meningitis, encephalitis and slow virus infections of the central nervous system (subacute sclerosing panencephalitis, multiple sclerosis, amyotrophic lateral sclerosis, Creutzfeldt-Jakob disease, etc.).

Amyloid plaques have been found in the brains of some patients with Creutzfeldt-Jakob disease (CJD) and all patients with Gerstmann-Sträussler syndrome (GSS). We examined paraffin sections from 45 patients with CJD or GSS and from 51 patients with other neurologic diseases, using an antiserum against GSS amyloid plaque cores. The GSS amyloid plaque core antiserum revealed not only birefringent amyloid plaques but also small plaques that cannot be detected by the staining with Congo red dye. Positive immunolabeling was demonstrated in 59% of 34 Japanese patients with CJD, in 100% of 11 patients

The strong familial association of Alzheimer disease (AD), the difficulty in transmitting the disease to animals, the mapping of the amyloid gene to human chromosome 21, and the non-inflammatory neuropathology have all been considered evidence against a viral etiology for this disease. However, unconventional slow viral infections share some of these traits with AD and yet they are caused by retroviruses or suspected viruses. The recent discovery of 2 human retroviruses causing central nervous system pathology similar to spongiform encephalopathies should prompt renewed search for retroviral c

In spite of a history of more than 100 years, the pathoaetiology of multiple sclerosis is still unknown today. Research is based on three working hypotheses, i.e. on an immunopathological disease origin, on the conception that MS, as an infectious disease, is caused by a specific pathogen (slow virus infection) and on the assumption of a disturbance of basal metabolism or utilisation. The present position of the scientific foundation of the working hypotheses is presented in detail and supplemented by the results of our own investigations. Of particular interest are the geomedical studies whic

In baby mice born to mothers--virus carriers (surviving for various periods after intranasal administration of influenza virus), infectious persisting influenza virus in titres of 10(0.5) to 10(2.5) EID50/0.1 ml was found in the blood, lungs, livers, kidneys, spleens, and brains. Not infrequently, such animals developed a severe pathological process accompanied by growth retardation and characterized by progressive involvement of the hypothalamus, immunocompetent organs, endocrine system and always terminating by the death of the animals. Possible mechanisms of the described slow form of influ
