Patient-specific effects on sonication heating efficiency during magnetic resonance-guided focused ultrasound thalamotomy.

PURPOSE: During magnetic resonance-guided focused ultrasound (MRgFUS) thalamotomy for refractory tremor, high temperatures must be achieved and sustained for tissue necrosis. We assessed the impact of both patient-specific as well as procedure-related factors on the efficiency of acoustic energy transfer, or heating efficiency (HE). METHODS: Retrospective analysis of 92 consecutive patients (857 sonications) with essential tremor or tremor-dominant Parkinson's disease treated at a single institution. Temperature elevations at the target were measured for each sonication with MR thermometry. H

Simple and rapid method for isolation and quantitation of polyhydroxyalkanoate by SDS-sonication treatment.

We developed a new method for isolation and quantitation of polyhydroxyalkanoate (PHA) from culture broth. In this method, the cells were sonicated in sodium dodecyl sulfate (SDS) solution and centrifuged to recover PHA. The recovered PHA was rinsed with deionized water and ethanol, and then weighed after drying. Hazardous chemicals such as chloroform, methanol, and sulfuric acid were not used, and no expensive analytical instruments were needed. We applied this method to Cupriavidus necator culture broths that included various amounts of poly(3-hydroxybutyrate) (PHB) or poly(3-hydroxybutyrate

Molecular Diagnosis of Orthopedic-Device-Related Infection Directly from Sonication Fluid by Metagenomic Sequencing.

Culture of multiple periprosthetic tissue samples is the current gold standard for microbiological diagnosis of prosthetic joint infections (PJI). Additional diagnostic information may be obtained through culture of sonication fluid from explants. However, current techniques can have relatively low sensitivity, with prior antimicrobial therapy and infection by fastidious organisms influencing results. We assessed if metagenomic sequencing of total DNA extracts obtained direct from sonication fluid can provide an alternative rapid and sensitive tool for diagnosis of PJI. We compared metagenomic

Effect of sonication on B cell development and immunomodulatory functions of Bursa of Fabricius.

A study was initiated with the objective of evaluating the effects of sonication treatment on important quality parameters of extract of Bursa of Fabricius. Sonication of extract was done (frequency 20 kHz and various amplitude levels) at 0 °C for 10 min, 30 min, 50 min, respectively. As results, the yield of bursa peptides significantly increased (p<0.05). Then we found sonicated bursa extract promoted the content of bursin and the CFU pre-B formation, exerted immunomodulatory function on antigen-specific immune responses in C57/BL6 mice immunized with inactivated Japanese encephalitis b viru

Sonication dissociates the synaptic cleft and allows purification of postsynaptic densities with associated postsynaptic membrane.

In the synaptic junction, pre-and post compartments are anchored to each other through trans-synaptic bridges spanning the synaptic cleft. Here we demonstrate that mild mechanical disturbance through sonication dissociates the synaptic cleft, and releases PSDs adjoined to the postsynaptic membrane, but devoid of presynaptic elements. It is the first time, to our knowledge, that dissection of the synaptic cleft has been achieved without the use of chemical/enzymatic treatments. This observation suggests that some of the protein-protein interactions involved in the anchoring of pre- and postsyna

Sonication versus the conventional method for evaluation of the dental microbiome: a prospective pilot study.

OBJECTIVES: To investigate sonication as a new tool in microbiological probing of dental infections. METHODS: Comparison of a standard probing method: intraoperative swab, with sonication, and vortex of the removed tooth, was performed on 20 carious destructed teeth. Illumina high throughput sequencing of the 16S-rRNA-gene was used for assessing the microbial composition. Antibiotic susceptibility has been assigned based on known resistances of each detected species. Probing procedures were compared using Bland-Altmann-Test, and antibiotic susceptibility using the Friedmann-Test and alpha-adj

Ibuprofen elimination from water and wastewater using sonication/ultraviolet/hydrogen peroxide/zeolite-titanate photocatalyst system.

The present investigation was designed to remove ibuprofen from aqueous solutions and wastewater by activating hydrogen peroxide using ultrasonication (US)/ultraviolet (UV) radiation/zeolite-titanium. The physical-chemical properties of the photocatalyst were determined using BET, FTIR, XRD, FESEM, and EDX-mapping techniques. The titanium oxide crystal size and the catalyst BET were determined to be 4.97 nm and 39.88 m2/g, respectively. Tests were performed in a reactor (with a volume of 316 mL) located in an ultrasonic bath to intensify reactions. The synergistic impact of the components of t

Implant sonication versus intraoperative tissue sample cultures for Periprosthetic Joint Infection (PJI) of Shoulder Arthroplasty.

Introduction Periprosthetic joint infection (PJI) is the most problematic complications after shoulder arthroplasty. Many diagnostic tools have been identified to find infection, such as hystopatologic examination of tissue sections or cultures of intraoperative tissue. Implant sonication fluid culture showed good results in order to enhance diagnostic accuracy, but literature results are still controversial. Aim of our study is to compare the results of sonication with intraoperative tissue sample cultures. Patients and Methods From February 2016 to January 2018 we performed 102 revisions of

Mechanisms of Chain Scission under High Strain Rate Sonication in PS-PIB-PS Triblock Copolymers.

Impedance mismatch block copolymers, combining rubbery polyisobutylene (PIB) with glassy polystyrene (PS) segments, show promise for the design of high-strain-rate material applications. However, the molecular-level processes of energy absorption and dissipation under high strain rates in the solution state are poorly understood. Using a grafting-from approach, we synthesized a library of well-defined PS-PIB-PS triblock copolymers with and without mechanophores at the block interfaces to evaluate the effects of molecular weight, block composition, and dispersity on chain scission induced by hi

The value of sonication on orthopaedic implants in an everyday clinical setting - an exploratory study.

BACKGROUND: Sonication of removed orthopaedic implants in suspected implant-associated infections (IAI) is widely applied internationally. However, evaluation of the utility of sonication on all implants removed in everyday standard practice is scarce. This exploratory study was performed to evaluate the application of sonication fluid (SF) culture on removed orthopaedic implants, irrespective of the reason for removal. METHODS: Out of 100 removed orthopaedic implants collected between August 2019 and September 2020, 77 implants with availability of concurrent tissue culture samples were incl

Sonication versus Tissue Sampling for Diagnosis of Prosthetic Joint and Other Orthopedic Device-Related Infections.

Current guidelines recommend collection of multiple tissue samples for diagnosis of prosthetic joint infections (PJI). Sonication of explanted devices has been proposed as a potentially simpler alternative; however, reported microbiological yield varies. We evaluated sonication for diagnosis of PJI and other orthopedic device-related infections (DRI) at the Oxford Bone Infection Unit between October 2012 and August 2016. We compared the performance of paired tissue and sonication cultures against a "gold standard" of published clinical and composite clinical and microbiological definitions of

Sonication-assisted synthesis of large, high-quality mercury thiolate single crystals directly from liquid mercury.

The synthetic formation of mercury thiolates has been known for almost 200 years. These compounds are usually formed by a slow reaction of mercury salts with thiolates or disulfides to produce small (up to 1 μm), plate-like crystals of Hg(S-R)(2). Herein we show that such mercury thiolates can be formed directly from liquid mercury via sonication with neat thiols. The process not only produces crystals very rapidly (within seconds) but also leads to the formation of large crystals (up to hundreds of micrometers). The high quality of these crystals enabled their detailed structural characteriza

Subwavelength imaging with a zero-mass sonic meta-atom.

Acoustic metamaterials offer powerful solutions for manipulating sound at subwavelength scales. One important application is super-resolved acoustic imaging, which relies on access to evanescent waves beyond the diffraction limit. Near-field techniques using subwavelength probes can capture these waves, revealing fine object details. Here, we introduce an experimental platform that harnesses airborne extraordinary transmission to couple evanescent acoustic waves into a subwavelength, zero-mass sonic meta-atom probe. By mounting a circular membrane at the tip of an air-filled waveguide with a c

A simple and scalable zebrafish model of Sonic hedgehog medulloblastoma.

Medulloblastoma (MB) is the most common malignant brain tumor in children and is stratified into three major subgroups. The Sonic hedgehog (SHH) subgroup represents ∼30% of all MB cases and has significant survival disparity depending upon TP53 status. Here, we describe a zebrafish model of SHH MB using CRISPR to create mutant ptch1, the primary genetic driver of human SHH MB. In these animals, tumors rapidly arise in the cerebellum and resemble human SHH MB by histology and comparative onco-genomics. Similar to human patients, MB tumors with loss of both ptch1 and tp53 have aggressive tumor h

Safety and Tolerability of Sonic Hedgehog Pathway Inhibitors in Cancer.

The hedgehog pathway, for which sonic hedgehog (Shh) is the most prominent ligand, is highly conserved and is tightly associated with embryonic development in a number of species. This pathway is also tightly associated with the development of several types of cancer, including basal cell carcinoma (BCC) and acute promyelocytic leukemia, among many others. Inactivating mutations in Patched-1 (PTCH1), leading to ligand-independent pathway activation, are frequent in several cancer types, but most prominent in BCC. This has led to the development of several compounds targeting this pathway as a

HDL nanoparticles targeting sonic hedgehog subtype medulloblastoma.

Medulloblastoma is the most common paediatric malignant brain cancer and there is a need for new targeted therapeutic approaches to more effectively treat these malignant tumours, which can be divided into four molecular subtypes. Here, we focus on targeting sonic hedgehog (SHH) subtype medulloblastoma, which accounts for approximately 25% of all cases. The SHH subtype relies upon cholesterol signalling for tumour growth and maintenance of tumour-initiating cancer stem cells (CSCs). To target cholesterol signalling, we employed biomimetic high-density lipoprotein nanoparticles (HDL NPs) which

Sonic hedgehog in the notochord is sufficient for patterning of the intervertebral discs.

The intervertebral discs, located between adjacent vertebrae, are required for stability of the spine and distributing mechanical load throughout the vertebral column. All cell types located in the middle regions of the discs, called nuclei pulposi, are derived from the embryonic notochord. Recently, it was shown that the hedgehog signaling pathway plays an essential role during formation of nuclei pulposi. However, during the time that nuclei pulposi are forming, Shh is expressed in both the notochord and the nearby floor plate. To determine the source of SHH protein sufficient for formation

The role of sonic hedgehog reemergence during gastric cancer.

Sonic Hedgehog (Shh) signaling has been extensively studied for its role in developmental biology and cancer biology. The association between Shh and cancer development in general is well established but the functional role of Shh in the development and progression of gastric cancer specifically is largely unknown. Bone marrow-derived stem cells, specifically mesenchymal stem cells (MSCs) infiltrate and engraft into the gastric mucosa in response to the chronic inflammatory environment of Helicobacter infection. In this review, MSC infiltration and changes in the cytokine and cellular profiles
