Spatial memory improvement by levodopa in parkinsonian MPTP-treated monkeys.

RATIONALE: The ameliorative effects of levodopa (L-3,4-dihydroxy-phenylalanine) on the motor impairment in Parkinson's disease patients is well established, but characterization of its effects on the associated cognitive deficits is still incomplete. OBJECTIVE: The present study determined the effect of different doses of levodopa on performance on a test of working memory in MPTP-treated rhesus monkeys, an animal model of Parkinson's disease. METHODS: Four MPTP-treated monkeys and their age-matched controls with the same experimental history as the MPTP-treated monkeys were tested on a spat

Sense of agency strengthens memory for self-caused spatial information.

Why do some experiences remain vivid while others fade? One factor may be the sense of agency (SoA), which shares self-referential mechanisms with episodic memory. We investigated whether SoA over action outcomes modulates item and spatial long-term memory. During intentional encoding, participants' keypresses moved an object either congruently or incongruently with the chosen direction. One hour later, item and spatial memory were tested. Participants reported higher SoA in congruent trials. Congruency and higher SoA ratings were associated with faster item-recognition responses without chang

Behavioral effects of chronic PFHxA exposure on spatial memory function.

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants widely detected in human biological samples. As regulatory restrictions on long-chain PFAS such as PFOA and PFOS have increased, short-chain alternatives, including perfluorohexanoic acid (PFHxA) have been widely adopted due to their lower bioaccumulation potential. However, their potential neurodevelopmental effects remain poorly understood. In this study, we investigated the long-term behavioral and neurobiological consequences of chronic PFHxA exposure beginning during the neurodevelopmental period and cont

Cerebellar Crus II Regulates Recognition and Spatial Memory in Mice.

The cerebellar Crus II is implicated in the late stages of Alzheimer's disease (AD), yet its specific roles in memory regulation and therapeutic potential remain unclear. Using in vivo fiber photometry, we observed robust activation of Crus II neurons in healthy mice during recognition memory tasks. Acute chemogenetic inhibition of Crus II neurons impaired recognition and spatial memory in mice. Polysynaptic circuit tracing revealed that Crus II neurons modulate neural activity in the contralateral prelimbic cortex (PrL) via the Crus II-cerebellar lateral nucleus (LN)-ventromedial thalamus/zon

Theta-phase locking of single neurons during human spatial memory.

Memory processes may rely on complex interactions between single-neuron activity and local field potentials. To better understand such spike-field relationships in humans, we examined human theta-phase locking-neuronal firing at similar theta phases over time-using single-neuron recordings in epilepsy patients performing a spatial memory task. Applying frequency-adaptive theta-phase estimation in a broad 1-10 Hz frequency range, we found that theta-phase locking was widespread in the human medial temporal lobe during memory encoding and retrieval. Time-resolved spectral parameterization and cy

Spatial Binding Impairments in Visual Working Memory following Temporal Lobectomy.

Disorders of the medial temporal lobe (MTL) adversely affect visual working memory (vWM) performance, including feature binding. It is unclear whether these impairments generalize across visual dimensions or are specifically spatial. To address this issue, we compared performance in two tasks of 13 epilepsy patients, who had undergone a temporal lobectomy, and 15 healthy controls. In the vWM task, participants recalled the color of one of two polygons, previously displayed side by side. At recall, a location or shape probe identified the target. In the perceptual task, participants estimated t

Decoding Spatial Memory Retrieval in Cubical Space Using fMRI Signals.

The way spatial memory retrieval is represented in the brain remains unclear to date. Previous studies have displayed a hippocampus-centered navigation network using functional magnetic resonance imaging (fMRI) analysis. There have been some studies on the representation of navigation behavior by signal distribution patterns, but only in the hippocampus and adjacent structures. In this study, we aimed to determine (1) the brain regions that represent information in both intensity and distribution patterns during spatial memory retrieval and (2) whether the patterns of neural responses represen

'Real-life' hippocampal-dependent spatial memory impairments in Huntington's disease.

Hippocampal-dependent spatial memory impairments are seen in Huntington's disease animal models. Similar impairments were recently reported in Huntington's disease participants on analogous spatial memory tasks (e.g., virtual Morris Water Maze), however, these tasks do not translate well to the range of functions involved in day-to-day spatial cognition. In this study we examined 'real-life' hippocampal-dependent spatial memory in Huntington's disease participants. We studied premanifest Huntington's disease (N = 24), early manifest Huntington's disease (N = 14), and matched healthy controls (

Adolescent social defeat decreases spatial working memory performance in adulthood.

BACKGROUND: Adolescent social stress is associated with increased incidence of mental illnesses in adulthood that are characterized by deficits in cognitive focus and flexibility. Such enhanced vulnerability may be due to psychosocial stress-induced disruption of the developing mesocortical dopamine system, which plays a fundamental role in facilitating complex cognitive processes such as spatial working memory. Adolescent rats exposed to repeated social defeat as a model of social stress develop dopaminergic hypofunction in the medial prefrontal cortex as adults. To evaluate a direct link bet

Representation of dynamic spatial configurations in visual short-term memory.

Locations of multiple stationary objects are represented on the basis of their global spatial configuration in visual short-term memory (VSTM). Once objects move individually, they form a global spatial configuration with varying spatial inter-object relations over time. The representation of such dynamic spatial configurations in VSTM was investigated in six experiments. Participants memorized a scene with six moving and/or stationary objects and performed a location change detection task for one object specified during the probing phase. The spatial configuration of the objects was manipulat

Dopaminergic modulation of visual-spatial working memory in Parkinson's disease.

Visual-spatial working memory (WM) impairment is frequently associated with the early stage of Parkinson's disease (PD). The aim of this study was to evaluate the performance of a group of PD patients in visual-spatial and visual-object WM tasks and to investigate the effect of administering the dopaminergic agonist apomorphine (experiment 1) or the dopamine precursor L-dopa (experiment 2) on the performance of tests assessing these functions. To study WM processes, the PD patients and age-matched normal controls were given an n-back task paradigm. In both experiments, the PD patients were sub

Spatial memory deficits in patients with chronic bilateral vestibular failure.

The role of the vestibular system for navigation and spatial memory has been demonstrated in animals but not in humans. Vestibular signals are necessary for location-specific "place cell" activity in the hippocampus which provides a putative neural substrate for the spatial representation involved in navigation. To investigate the spatial memory in patients with bilateral vestibular failure due to NF2 with bilateral neurectomy, a virtual variant (on a PC) of the Morris water task adapted to humans was used. Significant spatial learning and memory deficits were shown in 12 patients as compared

BDNF protects against spatial memory deficits following neonatal hypoxia-ischemia.

Hypoxic-ischemic (H-I) brain injury in the human perinatal period often leads to significant long-term neurobehavioral dysfunction in the cognitive and sensory-motor domains. Using a neonatal H-I injury model (unilateral carotid ligation followed by hypoxia) in postnatal day seven rats, previous studies have shown that neurotrophins, such as brain-derived neurotrophic factor (BDNF), can be protective against neural tissue loss. The present study explored potential relationships between neural protective and behavioral protective strategies in this neonatal H-I model by determining if neonatal

Rightward exogenous attentional shifts impair perceptual memory of spatial locations in patients with left unilateral spatial neglect.

Spatial remapping implies the updating and maintaining of the spatial position of objects in successive visual images across time, despite their displacement on the retina due to eye movements. In the parietal cortex, the representation of spatial locations appears to be partly centered on gaze direction, and thus modulated by current eye-gaze position. It has been suggested that short-term memory for spatial locations across delays might be impaired in right brain-injured patients with left spatial neglect, but more so after rightward than leftward gaze shifts - an asymmetry attributed to a l

Memory on ice: spatial memory is retained over a 9-month hibernation period in Columbian ground squirrels.

Memory allows animals to utilize ecologically relevant information, yet whether it endures through prolonged metabolic shutdown is poorly understood. We tested spatial memory retention in a long-term, free-living hibernator, the Columbian ground squirrel (Urocitellus columbianus), using a field-based spatial maze over two consecutive active seasons. Ten individuals were tested before and after a 9-month hibernation period and compared with eight naive individuals. Maze-solving performances-time to exit, path sinuosity and navigational errors-were analysed over 10 consecutive trials using Bayes

Attenuating corticosterone levels on the day of memory assessment prevents chronic stress-induced impairments in spatial memory.

This study investigated whether chronic stress-induced spatial memory deficits were caused by changes in the hypothalamic-pituitary-adrenal axis, such as corticosterone (CORT) elevations on the day of memory assessment, rather than the consequence of structural changes in the hippocampus. Male Sprague-Dawley rats were restrained for 6 h/day/21 days, and spatial memory was assessed on the Y-maze on day 22. Ninety minutes before training, rats received a subcutaneous injection of vehicle or metyrapone, a CORT synthesis inhibitor, and then spatial memory was determined 4-h later. The highest dose

Normal spatial memory following postseizure treatment with ketamine: selective damage attenuates memory deficits in brain-damaged rodents.

Within 30 min after the initiation of status epilepticus (SE) by lithium and pilocarpine, rats were injected with either acepromazine or ketamine. Compared to the rats that had received the acepromazine, the group that had received the ketamine displayed more accurate spatial memory. Their scores did not differ significantly from normal (non-seized) controls. Although the ketamine treatment did not significantly change the amount of neuronal loss within about 100 Paxinos and Watson structures, it was neuroprotective for several structures within the thalamus and portions of the temporal and pa

Aberrant increased GABAergic neuronal activity in postrhinal cortex impairs spatial memory.

The postrhinal cortex (POR) in rodents is homologous with the primate parahippocampal cortex (PHC), and has been shown to play a crucial role in spatial and object recognition memory functions. Our previous study identified that POR glutamatergic neurons are specifically regulate spatial memory retrieval. However, the function of POR γ-aminobutyric acidergic (GABAergic) neurons in spatial and object recognition memory remains to be elucidated. In this study, we first confirmed that the POR contains plenty of GABAergic neurons. Then we determined that hyperactivity of POR GABAergic neurons impa

Comparison of Different Exercise Modalities on Neuronal Plasticity and Spatial Memory.

PURPOSE: The relationship between exercise and brain function has been studied extensively; however, whether different exercise modalities promote hippocampal neurogenesis and cognitive function through shared or distinct biological pathways remains unclear. This study examined the effects of endurance training (ET), resistance training (RT), and high-intensity interval training (HIIT) on hippocampal neurogenesis, spatial memory, and neurotrophic factors. METHODS: Forty 3-month-old male C57BL/6J mice were randomized to control (SED), ET, RT, or HIIT groups. Following the training intervention

Pharmacological stimulation of hippocampal GPR35 impairs spatial navigation, learning, and memory.

GPR35 is an orphan receptor expressed in astrocytes and neurons of the central nervous system (CNS), including the hippocampus; however, its role in cognitive functions is largely unknown. This study administered intra-hippocampal injections of GPR35 compound-10 (GPR35C10; a GPR35-selective agonist), ML194 (a GPR35-selective antagonist), and the combination of both to elucidate their pharmacological actions in the Barnes maze. Compared to the vehicle (10 % DMSO) and ML194 groups, animals treated with GPR35C10 showed a higher latency and longer distance to solve the maze (p < 0.05) (an effect p
