Resting-state fMRI reveals enhanced functional connectivity in spatial navigation networks after transcranial direct current stimulation.

A number of studies have established that transcranial direct current stimulation (tDCS) modulates cortical excitability. We previously demonstrated polarity dependent changes in parietal lobe blood oxygen level dependent (BOLD) fMRI in a group of young adults during a spatial navigation task [15]. Here we used resting state functional connectivity (rsFC) to examine whether analogous changes were also evident during the resting state. Participants were randomized to either a parietal-anodal, frontal-cathodal (P+F-) or the opposite montage (P-F+) and received 20min of tDCS (2mA) before undergoi

Complementary Roles of the Hippocampus and the Dorsomedial Striatum during Spatial and Sequence-Based Navigation Behavior.

We investigated the neural bases of navigation based on spatial or sequential egocentric representation during the completion of the starmaze, a complex goal-directed navigation task. In this maze, mice had to swim along a path composed of three choice points to find a hidden platform. As reported previously, this task can be solved by using two hippocampal-dependent strategies encoded in parallel i) the allocentric strategy requiring encoding of the contextual information, and ii) the sequential egocentric strategy requiring temporal encoding of a sequence of successive body movements associa

Theta and gamma rhythmic coding through two spike output modes in the hippocampus during spatial navigation.

Hippocampal CA1 neurons generate single spikes and stereotyped bursts of spikes. However, it is unclear how individual neurons dynamically switch between these output modes and whether these two spiking outputs relay distinct information. We performed extracellular recordings in spatially navigating rats and cellular voltage imaging and optogenetics in awake mice. We found that spike bursts are preferentially linked to cellular and network theta rhythms (3-12 Hz) and encode an animal's position via theta phase precession, particularly as animals are entering a place field. In contrast, single

Spatial Navigation and Its Association With Biomarkers and Future Dementia in Memory Clinic Patients Without Dementia.

BACKGROUND AND OBJECTIVES: Impaired spatial navigation is considered an early sign in many neurodegenerative diseases. We aimed to determine whether spatial navigation was associated with future dementia in patients with subjective cognitive decline (SCD) or mild cognitive impairment (MCI) and to explore associations between spatial navigation and biomarkers of Alzheimer disease (AD) and neurodegeneration. METHODS: This study included memory clinic patients without dementia in the longitudinal BioFINDER cohort. The Floor Maze Test (FMT) was used to assess spatial navigation at baseline. Conve

Future trends in brain aging research: Visuo-cognitive functions at stake during mobility and spatial navigation.

Aging leads to a complex pattern of structural and functional changes, gradually affecting sensorimotor, perceptual, and cognitive processes. These multiscale changes can hinder older adults' interaction with their environment, progressively reducing their autonomy in performing tasks relevant to everyday life. Autonomy loss can further be aggravated by the onset and progression of neurodegenerative disorders (e.g., age-related macular degeneration at the sensory input level; and Alzheimer's disease at the cognitive level). In this context, spatial cognition offers a representative case of hig

Adult learning deficits after neonatal exposure to D-methamphetamine: selective effects on spatial navigation and memory.

The effects of neonatal d-methamphetamine (MA) treatment on cued and spatial learning and memory were investigated. MA was administered to neonatal rats on postnatal days 11-20. All groups received four subcutaneous injections per day. Group MA40-4 received 40 mg. kg(-1). d(-1) of MA in four divided doses (10 mg/kg per injection). Group MA40-2 received 40 mg. kg(-1). d(-1) of MA in two divided (20 mg/kg/injection) and saline for the other two injections per day. Controls received saline for four injections per day. As adults, both MA groups showed no differences in swimming ability in a straig

Navigating cell maps by deep learning integration of single-cell and spatially resolved transcriptomics.

Single-cell RNA sequencing (scRNA-seq) enables genome-wide gene expression profiling at single-cell resolution but loses the spatial context essential for interpreting cell identity and tissue organization. In contrast, spatially resolved transcriptomics (SRT) preserves spatial information but typically lacks single-cell resolution or complete transcriptome coverage. To obtain a more comprehensive view of heterogeneous spatial domains and cellular gene expression, we present Cell2Map, an unsupervised deep learning method that integrates scRNA-seq and SRT data from the same tissue region. Insensitivity of Place Cells to the Value of Spatial Goals in a Two-Choice Flexible Navigation Task.

Hippocampal place cells show position-specific activity thought to reflect a self-localization signal. Several reports also point to some form of goal encoding by place cells. We investigated this by asking whether they also encode the value of spatial goals, which is crucial information for optimizing goal-directed navigation. We used a continuous place navigation task in which male rats navigate to one of two (freely chosen) unmarked locations and wait, triggering the release of reward, which is then located and consumed elsewhere. This allows sampling of place fields and dissociates spatial

Galaxy single-cell & spatial omics community update: Navigating new frontiers in 2025.

Single-cell omics, named Method of the Year three times, have revolutionized biological research by enabling the high-resolution exploration of cellular heterogeneity and molecular processes. Initially centered on transcriptomics, this rapidly evolving field now ranges from multiomics to spatial analysis, with expanding customization options. The ubiquity of such analyses and the lack of a unified pipeline necessitate the development of scalable, flexible, and integrated tools and workflows. The Galaxy platform has responded to these technological advancements, extending its repertoire of free

Pure representational neglect and navigational deficits in a case with preserved visuo-spatial working memory.

We describe a patient who, after right hemisphere damage, showed severe, persistent, pure representational neglect but no evidence of perceptual neglect and no deficits in spatial working memory when evaluated with a traditional clinical test (Corsi Block Tapping test). This finding provides evidence against a full explanation of representational neglect within the context of visuo-spatial working memory. Indeed, this patient showed a peculiar deficit in navigational tasks requiring re-orientation in a novel environment by means of his mental representation of the environment. Since no represe

Altering sensory cues for spatial navigation does not impose a dual-task effect on gait and balance.

Walking is an attentionally demanding process that draws from a limited pool of attentional resources. Dual-task assessments, where individuals perform a cognitive task while walking, often reveal changes in gait and balance due to competing attentional demands. As cognitive task difficulty increases, the attentional resources necessary to complete the task also increase, leading to greater interference with gait and balance. However, these interactions are typically examined using contrived lab-based tasks, leaving it unclear how the cognitive processes engaged during real-world movement impa

Dopamine differentially modulates medial temporal lobe activity and behavior during spatial navigation in young and older adults.

Aging is associated with changes in spatial navigation behavior. In addition to an overall performance decline, older adults tend to rely more on proximal location cue information than on environmental boundary information during spatial navigation compared to young adults. The fact that older adults are more susceptible to errors during spatial navigation might be partly attributed to deficient dopaminergic modulation of hippocampal and striatal functioning. Hence, elevating dopamine levels might differentially modulate spatial navigation and memory performance in young and older adults. In t

Dissociating spatial strategies in animal research: Critical methodological review with focus on egocentric navigation and the hippocampus.

One major challenge in animal research on spatial learning and memory pertains to designing methods to dissociate spatial strategies (allocentric vs. egocentric). This is crucial for understanding the underlying cognitive processes and neural circuits that are recruited in navigational tasks. Taking the egocentric reference frames as a starting point, this review argues that in many extensively used spatial paradigms, multiple spatial reference frames are often available to the animals but remain unaccounted for. We discuss the implications this has for the inferences that can be made and prop

Tetrahydroaminoacridine and D-cycloserine fail to alleviate the water maze spatial navigation defect induced by hippocampal inactivation.

The present study examined the efficacy of single and combined treatment with an anticholinesterase, tetrahydroaminoacridine (i.p.), and a glycine-B site partial agonist, D-cycloserine (i.p.; a positive allosteric modulator of NMDA receptors), in alleviating the deficit in water maze spatial navigation induced by electrolytic lesion of the medial septum or lidocaine infusion into the dorsal hippocampi. In medial septum-lesioned rats, a combination of tetrahydroaminoacridine 3 mg kg(-1) and D-cycloserine 10 mg kg(-1) facilitated acquisition of the water maze test more effectively than either of

Route efficiency in spatial navigation as an early indicator of cognitive decline in amnestic mild cognitive impairment.

INTRODUCTION: The hippocampus is one of the brain regions most affected by neurodegeneration in Alzheimer's disease (AD). This structure and its neural circuits are critically involved in spatial learning and memory. Poor spatial navigation performance in virtual environments such as the Virtual Morris Water Navigation Task (VMWNT) may precede other clinical findings in amnestic mild cognitive impairment (aMCI), a pre-dementia stage of AD. To explore this idea, we correlated aMCI and cognitively healthy control (HC) performance in the VMWNT with Montreal Cognitive Assessment (MoCA) and Memory

Developmental exposure to legacy environmental contaminants, medial temporal lobe volumes and spatial navigation memory in late adolescents.

Exposure to lead, mercury, and polychlorinated biphenyls (PCBs) has been causally linked to spatial memory deficits and hippocampal changes in animal models. The Inuit community in Northern Canada is exposed to higher concentrations of these contaminants compared to the general population. This study aimed to 1) investigate associations between prenatal and current contaminant exposures and medial temporal brain volumes in Inuit late adolescents; 2) examine the relationship between these brain structures and spatial memory; and 3) assess the mediating role of brain structures in the associatio

Network QoS Impact on Spatial Perception through Sensory Substitution in Navigation Systems for Blind and Visually Impaired People.

A navigation system for individuals suffering from blindness or visual impairment provides information useful to reach a destination. Although there are different approaches, traditional designs are evolving into distributed systems with low-cost, front-end devices. These devices act as a medium between the user and the environment, encoding the information gathered on the surroundings according to theories on human perceptual and cognitive processes. Ultimately, they are rooted in sensorimotor coupling. The present work searches for temporal constraints due to such human-machine interfaces, w
