Species and strain-specific phenotypes shape performance and output of a five-species yeast consortium.

Spontaneous wine alcoholic fermentation is initiated by complex yeast communities, which shape the fermentation process and fermentation outcome. While this yeast ecosystem has been described extensively, the biotic factors impacting population dynamics, and the relative ecological importance of the taxonomic levels of species and of strains in defining these parameters remain poorly resolved. Here, we established a consortium of five yeast species that are commonly identified as major contributors to spontaneous wine fermentations: Saccharomyces cerevisiae, Lachancea thermotolerans, Hansenias

Defining species-specific immunodominant B cell epitopes for molecular serology of Chlamydia species.

Urgently needed species-specific enzyme-linked immunosorbent assays (ELISAs) for the detection of antibodies against Chlamydia spp. have been elusive due to high cross-reactivity of chlamydial antigens. To identify Chlamydia species-specific B cell epitopes for such assays, we ranked the potential epitopes of immunodominant chlamydial proteins that are polymorphic among all Chlamydia species. High-scoring peptides were synthesized with N-terminal biotin, followed by a serine-glycine-serine-glycine spacer, immobilized onto streptavidin-coated microtiter plates, and tested with mono-specific mou

Comparative Proteomics of Extracellular Vesicles of Two Species of Mucorales Reveals Species-Specific Cargo.

Rhizopus arrhizus is one of the most common etiological agents of mucormycosis worldwide, whereas Mucor irregularis has been reported with a relatively high frequency in China, exhibiting clear regional characteristics. However, the molecular basis underlying their differential host responses remains incompletely understood. Fungal extracellular vesicles (EVs) play diverse biological roles across fungal species and have emerged as important carriers of bioactive cargos that modulate host-pathogen interactions. In this study, we compared features of EVs derived from R. arrhizus and M. irregular

Green fluorescent protein as a novel species-specific marker in enteric dual-species biofilms.

Green fluorescent protein (GFP) was used as a tool to examine the interactions between pairs of bacterial species and their effects on subsequent biofilm development over 24 h. A plasmid encoding GFP from Aequorea victoria was transformed into strains of Enterobacter agglomerans and Escherichia coli ATCC 11229. The development of dual-species biofilms, containing one fluorescent and one non-fluorescent partner, was examined using viable counts. UV illumination of plates enabled both species to be identified in a mixture. The spatial distribution of each species was examined by UV microscopy, s

Polymorphism at the dnaK locus of Brucella species and identification of a Brucella melitensis species-specific marker.

The dnaK gene and surrounding sequences from reference strains of the six Brucella species were amplified by the polymerase chain reaction (PCR) with primers chosen according to the published sequence of the B. ovis dnaK gene and studied for polymorphism with nine restriction endonucleases. The restriction patterns were identical for all species with all restriction endonucleases tested except for B. melitensis strain 16M that showed a different pattern with EcoRV, consistent with the presence of a single site instead of two for the other Brucella species. The absence of the second EcoRV site

Sialic acid specificity of erythrocyte binding Antigen-140 orthologues in human- and ape-infective Laverania species.

Plasmodium falciparum is the only human-infective species within the Laverania subgenus. In order for Plasmodium falciparum merozoites to invade erythrocytes, parasite ligands interact with a number of erythrocyte surface proteins, several of which are modified with sialic acids. These sialic acids differ between humans, which can synthesise only Neu5Ac sialic acids, and apes which express an enzyme absent in humans that converts Neu5Ac to Neu5Gc, resulting in their surface sialoglycoproteins predominantly being modified with Neu5Gc. This difference between apes and humans has been previously

Melanoma antigens in pediatric medulloblastoma contribute to tumor heterogeneity and species-specificity of group 3 tumors.

Medulloblastoma (MB) is the most malignant childhood brain cancer. Group 3 MB (G3 MB) subtype accounts for about 25% of MB and is associated with the worst outcomes. Herein, we report that more than half of G3 MB tumors express melanoma antigens (MAGEs), which are potential prognostic and therapeutic markers. MAGEs are cancer-testis antigens, aberrantly expressed in several adult cancers, and associated with poorer prognosis and therapy resistance; however, their role in pediatric cancers is mostly unknown. This study aimed to determine whether MAGEs are activated and important in pediatric MB

Nutrient niche specificity for glycosaminoglycans is reflected in polysaccharide utilization locus architecture of gut Bacteroides species.

INTRODUCTION: Glycosaminoglycans (GAGs) present in the mucosal layer can be used as nutrients by certain intestinal bacteria, particularly members of the Bacteroides. GAG abundances are altered in some diseases such as inflammatory bowel diseases, which may affect microbial composition and activity, and it is therefore important to understand GAG utilization by members of the gut microbiota. METHODS: We used growth assays, transcriptomics, and comparative genomics to evaluate chondroitin sulfate (CS) and hyaluronan (HA) degradation ability by multiple gut Bacteroides species. RESULTS AND DIS

Species specificity, surface exposure, protein expression, immunogenicity, and participation in biofilm formation of Porphyromonas gingivalis HmuY.

BACKGROUND: Porphyromonas gingivalis is a major etiological agent of chronic periodontitis. The aim of this study was to examine the species specificity, surface exposure, protein expression, immunogenicity, and participation in biofilm formation of the P. gingivalis heme-binding protein HmuY. RESULTS: HmuY is a unique protein of P. gingivalis since only low amino-acid sequence homology has been found to proteins encoded in other species. It is exposed on the cell surface and highly abundant in the outer membrane of the cell, in outer-membrane vesicles, and is released into culture medium in

Developmental appearance, species and tissue specificity of mouse 23-kDa, a retinal calcium-binding protein (recoverin).

A 23-kDa, soluble, calcium-binding photoreceptor-specific protein (23-kDa) has been shown to be identical to recoverin and the cancer-associated retinopathy protein. Recoverin has been reported to activate guanylate cyclase to increase the amount of cyclic GMP and thereby reopen cation channels within the photoreceptor cells. In this study, the 23-kDa protein was purified from bovine retinas and monospecific antibodies against it were generated in rabbits. Western analysis demonstrated 23-kDa in retinas from human, monkey, bovine, dog, rabbit, rat, mouse, frog, chameleon and iguana although it

Specificity Evaluation of a Polyprotein-Based ELISA Designed for the Detection of Paratuberculosis in Multiple Species.

Paratuberculosis (PTB) is a disease affecting ruminant animals. The etiological agent, Mycobacterium avium subspecies PTB (Map), is a short, Gram-positive, acid-fast bacillus. Due to the nature of PTB, diagnosis often occurs at advanced stages of the disease, following the onset of clinical symptoms and prior dissemination of the agent. The specificity of current humoral diagnostic techniques, such as ELISA, is limited, mainly due to the presence of various species of mycobacteria in the environment and other closely related pathogenic mycobacteria that can interfere with the accurate diagnosi

Sequencing of transcriptomes from two Miscanthus species reveals functional specificity in rhizomes, and clarifies evolutionary relationships.

BACKGROUND: Miscanthus is a promising biomass crop for temperate regions. Despite the increasing interest in this plant, limited sequence information has constrained research into its biology, physiology, and breeding. The whole genome transcriptomes of M. sinensis and M. sacchariflorus presented in this study may provide good resources to understand functional compositions of two important Miscanthus genomes and their evolutionary relationships. RESULTS: For M. sinensis, a total of 457,891 and 512,950 expressed sequence tags (ESTs) were produced from leaf and rhizome tissues, respectively, w

Identification of residues in the ectromelia virus gamma interferon-binding protein involved in expanded species specificity.

Gamma interferon (IFN-gamma) production is important in the host response to, and recovery from, infection with Ectromelia virus (ECTV) and Vaccinia virus (VACV). The orthopoxviruses have evolved several mechanisms to subvert the IFN-gamma response. IFN-gamma-binding protein (IFN-gammaBP) is a virally encoded homologue of the host IFN-gamma receptor that blocks the effects of IFN-gamma in the infected host. Unlike the cellular receptors, whose ligand specificity is restricted to their own species, the orthopoxvirus IFN-gammaBPs bind IFN-gamma from several species. The reason for this relaxed s

Two known and one new species of Proctoeces from Australian teleosts: Variable host-specificity for closely related species identified through multi-locus molecular data.

Species of Proctoeces Odhner, 1911 (Trematoda: Fellodistomidae) have been reported from a wide range of marine animals globally. Members of the genus tend to lack strongly distinguishing morphological features for diagnosis, making identification difficult and the true number of species in the genus contentious. Combined morphological and molecular analyses were used to characterise three species of Proctoeces from Moreton Bay and the southern Great Barrier Reef. Data for two ribosomal regions and one mitochondrial region were generated for specimens collected from Australia. Three unique 18S-

Coral-associated denitrification is seasonally variable and species-specific.

Nitrogen (N) plays a critical role in coral growth, but maintaining an N-limited state is essential for coral-algal symbiosis stability. Coral-associated denitrifiers may help regulate excess N, though denitrification in corals remains poorly quantified. To better understand species-specific and environmental effects on the denitrification pathway, we investigated year-long denitrification dynamics in four Red Sea corals, using acetylene inhibition assays alongside physiological and environmental measurements. All species exhibited measurable denitrification activity, ranging from 0 - 0.8 nmol

Structural origins of species-specific differences in TRPV2 activation.

Transient receptor potential vanilloid 2 (TRPV2) is a broadly expressed ion channel implicated in diverse physiological and pathological processes. Despite strong conservation, human TRPV2 (hTRPV2) displays markedly reduced sensitivity to stimuli such as 2-aminoethoxydiphenyl borate (2-APB) and heat compared to rodent orthologs. Here we combine electrophysiology and cryo electron microscopy to define the basis of this species-dependent divergence. The structure of hTRPV2 is remarkably different at the voltage sensor-like domain (VSLD) compared to rodent channels and functional analyses show a
