DNAH10 interacts with UCHL3-PACRG complex to coordinate sperm head and flagella development during spermiogenesis.

Asthenoteratozoospermia is a common underlying cause of male infertility, with dynein dysfunction playing an important role in the aetiology of the condition. Dysfunction in certain dynein proteins has been implicated in asthenoteratozoospermia, while others exclusively induce asthenozoospermia in the absence of overt morphological abnormalities in the sperm. Dynein axonemal heavy chain 10 (DNAH10), an inner dynein arm heavy chain protein, was identified as being associated with asthenoteratozoospermia in our previous studies. However, the mechanism through which DNAH10 contributes to this con

Pregnancies after ICSI using sperm with abnormal head-tail junction from two brothers: case report.

We report ICSI pregnancies in two couples with a history of long standing primary infertility in which the sperm of the male partner were either acephalic or had abnormal head-midpiece attachments. The two couples, in which the men are brothers, underwent ICSI. Sperm were analysed by transmission electron microscopy and immunocytochemistry with an anti-MPM2 monoclonal antibody. The first couple underwent two ICSI cycles, each consisting of the injection of two mature oocytes and the transfer of two embryos. A successful pregnancy occurred after the second transfer and led to the birth to a hea

Deep-Learning-Based Profiling of Mouse Sperm Head and Neck Morphology Reveals Coordinated Structural Variation.

BACKGROUND: Mammalian sperm head morphology varies across species and dictates fertilization capability. Rodent sperm have asymmetric, hook-shaped heads, complicating analysis. Despite widespread biomedical use, current tools poorly characterize these complex variations. OBJECTIVES: This study aimed to establish a robust framework for the precise, noninvasive, and quantitative analysis of mouse sperm morphology. MATERIALS AND METHODS: We developed ADAM-net (Anomaly-aware Deep-learning Architecture for Morphology), a machine-learning system for mouse sperm head and neck structures. Integratin

Non-canonical RNA polyadenylation polymerase FAM46C is essential for fastening sperm head and flagellum in mice†.

Family with sequence similarity 46, member C (FAM46C) is a highly conserved non-canonical RNA polyadenylation polymerase that is abundantly expressed in human and mouse testes and is frequently mutated in patients with multiple myeloma. However, its physiological role remains largely unknown. In this study, we found that FAM46C is specifically localized to the manchette of spermatids in mouse testes, a transient microtubule-based structure mainly involved in nuclear shaping and intra-flagellar protein traffic. Gene knockout of FAM46C in mice resulted in male sterility, characterized by the pro

Effects of cold shock and phospholipase A2 on intact boar spermatozoa and sperm head plasma membranes.

Head plasma membranes (HPM) isolated from cryopreserved boar spermatozoa show an excessive fluidization, which might be involved in the loss of fertility. The current study assessed the ability of cold shock (5 degrees C) and phospholipase A2 (PA2) to duplicate these effects on membrane structure and to affect 45Ca2+ uptake and gross morphological characteristics of whole, fresh boar-sperm. The HPM from cold-shocked sperm showed a significantly greater rate of fluidization over time than did HPM from control sperm. Addition of PA2 (bee or snake venom, 0.1 or 10.0 ng/ml) to HPM from control spe

Disruption in CYLC1 leads to acrosome detachment, sperm head deformity, and male in/subfertility in humans and mice.

The perinuclear theca (PT) is a dense cytoplasmic web encapsulating the sperm nucleus. The physiological roles of PT in sperm biology and the clinical relevance of variants of PT proteins to male infertility are still largely unknown. We reveal that cylicin-1, a major constituent of the PT, is vital for male fertility in both mice and humans. Loss of cylicin-1 in mice leads to a high incidence of malformed sperm heads with acrosome detachment from the nucleus. Cylicin-1 interacts with itself, several other PT proteins, the inner acrosomal membrane (IAM) protein SPACA1, and the nuclear envelope

Sperm with fibrous sheath dysplasia and anomalies in head-neck junction: focus on centriole and centrin 1.

Spermatozoa with a rare combination of two monomorphic sperm defects, dysplasia of the fibrous sheath (DFS) and alterations in head-mid-piece junction were analysed. The main focus was to explore the status of the centriole, a key organisation during fertilisation, using the centrin 1, a calcium-binding protein linked to this structure. The sperm quality was examined by light, scanning and transmission electron microscopy (SEM, TEM); immunocytochemistry was performed for tubulin, A-kinase anchor protein 4 (AKAP4) and centrin 1. Spermatozoa showed DFS defect associated with anomalies in head-ta

Long-term segmentation-free assessment of head-flagellum movement and intracellular calcium in swimming human sperm.

Human spermatozoa are the archetype of long-term self-organizing transport in nature and are critical for reproductive success. They utilize coordinated head and flagellar movements to swim long distances within the female reproductive tract in order to find and fertilize the egg. However, to date, long-term analysis of the sperm head-flagellar movements, or indeed those of other flagellated microorganisms, remains elusive due to limitations in microscopy and flagellar-tracking techniques. Here, we present a novel methodology based on local orientation and isotropy of bio-images to obtain long

A CCNA1 Missense Variant Associated With Chromatid Non-Disjunction in Abnormal-Headed Sperm and Male Infertility.

BACKGROUND: Macrozoospermia is a rare form of teratozoospermia characterized by tetraploids, large-headed spermatozoa with multiple flagella, usually caused by bi-allelic AURKC mutations. The etiology of atypical phenotypes with a lower proportion of large headed spermatozoa and single flagella however often remains unresolved. OBJECTIVE: To investigate the genetic cause of severe sperm-head abnormalities with moderate macrozoospermia without multiflagellated spermatozoa in a patient with repeated ICSI failure. An infertile male with three failed ICSI attempts underwent semen analysis, reveal

Heads or tails? Structural events and molecular mechanisms that promote mammalian sperm acrosomal exocytosis and motility.

Sperm structure has evolved to be very compact and compartmentalized to enable the motor (the flagellum) to transport the nuclear cargo (the head) to the egg. Furthermore, sperm do not exhibit progressive motility and are not capable of undergoing acrosomal exocytosis immediately following their release into the lumen of the seminiferous tubules, the site of spermatogenesis in the testis. These cells require maturation in the epididymis and female reproductive tract before they become competent for fertilization. Here we review aspects of the structural and molecular mechanisms that promote fo

Reproductive outcomes in patients with male infertility because of Klinefelter's syndrome, Kartagener's syndrome, round-head sperm, dysplasia fibrous sheath, and 'stump' tail sperm: an updated literature review.

PURPOSE OF REVIEW: To describe the reproductive outcomes of a heterogeneous group of male infertility conditions causing severe alterations in the sperm parameters (counts, motility, and morphology) because of chromosomal, genetic, or still unknown causes. Source of sperm, fertilization, pregnancy, live birth, and miscarriage rates of patients with Klinefelter's syndrome, Kartagener's syndrome, round-head sperm, dysplasia of the fibrous sheath (DFS), and stump-tail sperm were reviewed. RECENT FINDINGS: There are differences in the outcome according to the conditions and the source of sperm (e

Perinuclear theca protein FNDC8 interacts with CCIN and ACTL7A to ensure proper sperm head shaping during spermiogenesis.

Precise shaping of the sperm head is essential for successful fertilization, and defects in this process are frequently associated with severe forms of male infertility. Sperm head morphology is coordinated by several testis-specific structures, including the apical ectoplasmic specialization (ES), manchette, and perinuclear theca (PT)-a dense cytoskeletal layer that envelops the nucleus during spermiogenesis. However, the molecular components and physiological functions of the PT remain incompletely understood. This study identified FNDC8 as a testis-enriched protein primarily localized to th

SUN5 interacts with nuclear membrane LaminB1 and cytoskeletal GTPase Septin12 mediating the sperm head-and-tail junction.

Acephalic spermatozoa syndrome (ASS) is a severe teratospermia with decaudated, decapitated, and malformed sperm, resulting in male infertility. Nuclear envelope protein SUN5 localizes to the junction between the sperm head and tail. Mutations in the SUN5 gene have been identified most frequently (33-47%) in ASS cases, and its molecular mechanism of action is yet to be explored. In the present study, we generated Sun5 knockout mice, which presented the phenotype of ASS. Nuclear membrane protein LaminB1 and cytoskeletal GTPases Septin12 and Septin2 were identified as potential partners for inte

Protamine labeling with Reactive Black-5 and its molecularly designed derivatives visualize vacuoles in human sperm head.

To date, numerous studies have examined the etiological relationship between vacuoles and DNA fragmentation in human sperm, with vacuoles typically assessed using differential interference contrast microscopy. To explore the optimal dye for visualizing vacuoles, we collected 12 commercially available reactive dyes and compared their staining performance. Labeling protamines with 9.0 μmol/L Reactive Black-5 (RB5) stained the nuclear body bluish and visualized vacuoles as unstained spots. Next, we synthesized 30 molecularly designed RB5 derivatives to identify an optimal molecular structure for

Development of rat oocytes following intracytoplasmic injection of sperm heads isolated from testicular and epididymal spermatozoa.

The possibility of obtaining normal development of rat oocytes following intracytoplasmic injection of rat sperm heads, obtained by sonicating spermatozoa from testes and epididymides, was evaluated. Irrespective of the source of spermatozoa, sperm heads were successfully injected into approximately 45% of oocytes used; after 9-12h of culture, approximately 55% of injected oocytes still had normal morphology. Of the oocytes injected with testicular sperm heads 45% were activated, with a female pronucleus and a second polar body, but significantly more oocytes (approximately 68%) injected with

Effect of ethanol and sperm head reducing agents on the preimplantation development of bovine embryos generated by Piezo-ICSI.

Intracytoplasmic sperm injection (ICSI) is a widely used assisted reproduction technique, but in cattle it faces major challenges due to inefficient oocyte activation after sperm microinjection. This study investigated different oocyte activation strategies and assessed the potential role of reducing agents glutathione (GSH), cysteamine (Cys) and dithiobutylamine (DTBA) to improve sperm head decondensation and embryo development following Piezo-ICSI. Haploid parthenogenetic activation using different ethanol concentrations (1%, 3%, 7% and 10%) failed to yield blastocysts, while diploid activat
