Phytohormone-dependent plant defense signaling orchestrated by oral bacteria of the herbivore Spodoptera litura.

A vast array of herbivorous arthropods live with symbiotic microorganisms. However, little is known about the nature and functional mechanism of bacterial effects on plant defense responses towards herbivores. We explored the role of microbes present in extracts of oral secretion (OS) isolated from larvae of Spodoptera litura, a generalist herbivore, in phytohormone signaling-dependent defense responses in Arabidopsis thaliana (Arabidopsis). In response to mechanical damage (MD) with application of bacteria-free OS (OS- ) prepared by sterilization or filtration of OS, Arabidopsis leaves exhibi

Immunological responses and gut microbial shift of fall armyworm Spodoptera frugiperda challenged with entomopathogens.

Fall armyworm (FAW), Spodoptera frugiperda is a dangerous lepidopteran pest that threatens maize production globally. Biopesticides such as entomopathogenic fungi (EPF) and Bacillus thuringiensis present safer alternatives to chemical pesticides for FAW management. This study investigated the impact of Metarhizium anisopliae and B. thuringiensis on the mortality, cellular immune response, and microbiome of FAW under different temperature regimes (15 °C, 20 °C, 25 °C, 30 °C, and 35 °C). M. anisopliae isolates ICIPE 7 and ICIPE 78 caused higher FAW larval mortality than B. thuringiensis isolates

Serratia marcescens SM001-induced oxidative stress accelerates Spodoptera frugiperda mortality by promoting midgut apoptosis.

Spodoptera frugiperda (fall armyworm) is a globally distributed and highly destructive migratory pest, while Serratia marcescens is a widespread entomopathogenic bacterium capable of infecting diverse insects. In this study, oral infection with S. marcescens strain SM001 exhibited potent virulence, with an LC50 of 7.40 × 106 CFU/mL at 168 h post-infection (hpi). Logistic regression modeling demonstrated that the bacterium enters an exponential proliferation phase within the hemolymph approximately 48-54 h after infection. Infection significantly upregulated Duox and Nos, induced severe oxidati

Egg Disease in Spodoptera litura (Fabricius) Caused by Aspergillus ochraceus and Its Metabolic Phenotype.

A pathogenic fungus was isolated from unhatched egg masses of Spodoptera litura (S. litura). The pathogen was identified as Aspergillus ochraceus based on morphological characterization and molecular analyses of the internal transcribed spacer (ITS) region and the β-tubulin gene. The isolate exhibited strong pathogenicity toward S. litura eggs, causing 100% mortality. Phenotypic characteristics were further evaluated using the BIOLOG Phenotype MicroArray (PM) system. A total of 950 distinct growth conditions were assessed across PM plates 1-10. The results indicated that A. ochraceus metaboliz

Three stress-responsive peptides modulate immunity and development in the fall armyworm Spodoptera frugiperda.

Insect stress-responsive peptides (SRPs) orchestrate development and immunity, yet their roles in the globally invasive pest Spodoptera frugiperda remain poorly understood. Here, we identify three SRP genes, SfSRP1, SfSRP2, and SfSRP3, that are strongly induced after Beauveria bassiana infection. Each encodes a 115-119 aa peptide with a conserved five-β-strand scaffold and divergent loop regions. RT-qPCR analysis reveals that all three are most abundant in hemocytes and head tissues, with stage-specific peaks at 2nd to 4th instars. Upon challenge with fungal or bacterial pathogens, SfSRP1 and

Trapping Spodoptera frugiperda (Lepidoptera: Noctuidae) Moths in Different Crop Habitats in Togo and Ghana.

The economic impact of the invasion of Spodoptera frugiperda (J.E. Smith, Lepidoptera: Noctuidae) into Africa has so far been limited to maize agriculture but could potentially impact many other crops. Trapping based on pheromone lures provides a cost-effective method for detecting this important pest (commonly known as fall armyworm) and will be essential for large-scale monitoring of populations to determine its geographical distribution and migration behavior as the species equilibrates to its new environment. However, the effective use of pheromone trapping requires optimization for a give

Synthesis, characterization of two matrine derivatives and their cytotoxic effect on Sf9 cell of Spodoptera frugiperda.

The invasion of Spodoptera frugiperda has imposed a serious impact on global food security. Matrine is a botanical pesticide with a broad spectrum of insecticidal activity which was recommended for controlling Spodoptera frugiperda. In order to discover effective insecticide for Spodoptera frugiperda, two matrine derivatives modified with carbon disulfide and nitrogen-containing groups were systhesized. And their inhibition activities on Sf9 cell were evaluated. The structural configuration of compounds were characterized by IR, HPLC, MS, NMR and XRD, with yields of 52% and 65%, respectively.

Effects of cyanogenic plants on fitness in two host strains of the fall armyworm (Spodoptera frugiperda).

The generalist moth, Spodoptera frugiperda (J. E. Smith) consists of two genetic subgroups (host strains) that differ in their distribution among host plant species. The corn strain prefers crop plants such as corn, sorghum, and cotton, while the rice strain is found in small grasses such as Cynodon spp. and rice. Little is known about the physiological factors that drive this host preference. Here, we report a feeding study with natural host plants and an artificial diet containing cyanide. We found that corn, two Cynodon spp. (bermudagrass C. dactylon (L.) Persoon, 'NuMex Sahara', and stargr

Functional conservation of IR75q.2 in the recognition of volatile acids and aldehydes in two Spodoptera species.

Ionotropic receptors (IRs) serve as important chemosensory receptors for the perception of diverse ecological volatiles in insects, but their specific functions in Lepidoptera remain largely unexplored. This study systematically compared the ligand recognition profiles and key binding residues of the highly expressed IR75q.2 receptor in two Spodoptera species. Phylogenetic and conserved motif analyses revealed sequence and structural conservation of the IR75 receptors across five noctuid species. Transcriptome data analysis further showed that IR75q.2 had the highest expression in the antennae

Seasonal Migratory Activity of the Beet Armyworm Spodoptera exigua (Hübner) in the Tropical Area of China.

The beet armyworm Spodoptera exigua (Hübner), a global pest, feeds on and affects a wide range of crops. Its long-distance migration with the East Asian monsoon frequently causes large-scale outbreaks in East and Southeast Asia. This pest mainly breeds in tropical regions in the winter season every year; however, few studies have investigated associations with its population movements in this region. From 2017 to 2023, we monitored its population dynamics in a tropical site, located in Hainan Province of China, using a searchlight trap. Dissection of the ovaries of female S. exigua moths captu

Identification and pharmacological characterization of histamine-gated chloride channels in the fall armyworm, Spodoptera frugiperda.

Histamine-gated chloride channels (HACls) mediate fast inhibitory neurotransmission in invertebrate nervous systems and have important roles in light reception, color processing, temperature preference and light-dark cycle. The fall armyworm, Spodoptera frugiperda is a main destructive pest of grain and row crops. However, the pharmacological characterization of HACls in S. frugiperda remain unknown. In this study, we identified two cDNAs encoding SfHACl1 and SfHACl2 in S. frugiperda. They had similar expression patterns and were most abundantly expressed in the head of larvae and at the egg s

Antiviral Treatment Reveals a Cooperative Pathogenicity of Baculovirus and Iflavirus in Spodoptera exigua, a Lepidopteran Insect.

NPVThe beet armyworm, Spodoptera exigua, is a serious insect pest infesting various vegetable crops. Two infectious insect viruses, baculovirus and iflavirus, are known to induce epizootics in S. exigua populations. Indeed, some laboratory colonies have appeared to be covertly infected by these viruses. Diagnostic PCR tests detected two different viruses: Spodoptera exigua multiple nucleopolyhedrosis virus (SeMNPV) and iflaviruses (SeIfV1 and SeIfV2). Viral extract from dead larvae of S. exigua could infect Sf9 cells and produce occlusion bodies (OBs). Feeding OBs to asymptomatic larvae of S.

Concerted impacts of antiherbivore defenses and opportunistic Serratia pathogens on the fall armyworm (Spodoptera frugiperda).

Insects frequently confront different microbial assemblages. Bacteria inhabiting an insect gut are often commensal, but some can become pathogenic when the insect is compromised from different stressors. Herbivores are often confronted by various forms of plant resistance, but how defenses generate opportunistic microbial infections from residents in the gut are not well understood. In this study, we evaluated the pathogenic tendencies of Serratia isolated from the digestive system of healthy fall armyworm larvae (Spodoptera frugiperda) and how it interfaces with plant defenses. We initially s

Knockout of a P-glycoprotein gene increases susceptibility to abamectin and emamectin benzoate in Spodoptera exigua.

P-glycoprotein [P-gp or the ATP-binding cassette transporter B1 (ABCB1)] is an important participant in multidrug resistance of cancer cells, yet the precise function of this arthropod transporter is unknown. The aim of this study was to determine the importance of P-gp for susceptibility to insecticides in the beet armyworm (Spodoptera exigua) using clustered regularly interspaced short palindromic repeats/CRISPR-associated 9 (CRISPR/Cas9) gene-editing technology. We cloned an open reading frame (ORF) encoding the S. exigua P-gp protein (SeP-gp) predicted to display structural characteristics

The calyx fluid of Microplitis rufiventris parasitoid and growth of its host Spodoptera littoralis larvae.

The morphology of the female reproductive system of pupal and adult stages of Microplitis rufiventris and the ultrastructure of the ovaries are described and illustrated. Two morphologically distinct types of particles of nuclear origin, i.e., polydnavirus (PDV) and a virus-like filamentous particle (VLFP) were detected in the ovarian calyx fluid of the female wasp. It is likely that these particles are injected into the host during oviposition. PDV initiated replication in the calyx of mid-aged pupae and in pharate adults and were present throughout adult life. VLFP were only seen in the caly

Constitutive overexpression of ATP-binding cassette transporters contributes to emamectin benzoate resistance in Spodoptera frugiperda.

Emamectin benzoate is a key insecticide for controlling Spodoptera frugiperda, a highly destructive agricultural pest. However, the emergence of resistance frequently results in control failures. Monitoring insecticide resistance in S. frugiperda populations and elucidating the underlying genetic mechanisms are essential for developing informed pest management decisions and effective resistance management strategies. This study demonstrates that six field populations of S. frugiperda from China have developed moderate resistance to emamectin benzoate, with resistance ratios (RR) ranging from 1

Targeting DOPA decarboxylase disrupts immune-metabolic coordination and renders Spodoptera frugiperda susceptible to bacterial infection.

DOPA decarboxylase (DDC) is a key enzyme that connects melanin production and innate immunity in insects, yet its regulatory network in major lepidopteran pests remains poorly understood. We combined CRISPR/Cas9 gene editing with multi-omics analysis to explore DDC's role in immune responses in the invasive pest Spodoptera frugiperda. Knocking out DDC caused complete albinism, severe developmental delays, pronounced developmental defects, and increased susceptibility to bacterial challenge. Following the E. coli challenge, DDC-mutant larvae showed substantially lower final survival than infect

The genetic adaptations of fall armyworm Spodoptera frugiperda facilitated its rapid global dispersal and invasion.

The fall armyworm (Spodoptera frugiperda) is a lepidopteran insect pest that causes huge economic losses. This notorious insect pest has rapidly spread over the world in the past few years. However, the mechanisms of rapid dispersal are not well understood. Here, we report a chromosome-level assembled genome of the fall armyworm, named the ZJ-version, using PacBio and Hi-C technology. The sequenced individual was a female collected from the Zhejiang province of China and had high heterozygosity. The assembled genome size of ZJ-version was 486 Mb, containing 361 contigs with an N50 of 1.13 Mb.
