V-H+ -ATPase translocation during blood alkalosis in dogfish gills: interaction with carbonic anhydrase and involvement in the postfeeding alkaline tide.

We investigated the involvement of carbonic anhydrase (CA) in mediating V-H(+)-ATPase translocation into the basolateral membrane in gills of alkalotic Squalus acanthias. Immunolabeling revealed that CA is localized in the same cells as V-H(+)-ATPase. Blood plasma from dogfish injected with acetazolamide [30 mg/kg at time (t) = 0 and 6 h] and infused with NaHCO(3) for 12 h (1,000 microeq.kg(-1).h(-1)) had significantly higher plasma HCO(3)(-) concentration than fish that were infused with NaHCO(3) alone (28.72 +/- 0.41 vs. 6.57 +/- 2.47 mmol/l, n = 3), whereas blood pH was similar in both trea

Mechanisms of corneal tissue cross-linking in response to treatment with topical riboflavin and long-wavelength ultraviolet radiation (UVA).

PURPOSE: Treatment of de-epithelialized human corneas with riboflavin (RF) + long-wavelength ultraviolet light (UVA; RFUVA) increases corneal stroma tensile strength significantly. RFUVA treatment retards the progression of keratoconus, perhaps by cross-linking of collagen molecules, but exact molecular mechanisms remain unknown. Research described here tested possible chemical mechanisms of cross-linking. METHODS: Corneas of rabbits and spiny dogfish sharks were de-epithelialized mechanically, subjected to various chemical pretreatments, exposed to RFUVA, and then subjected to destructive te

Built for speed: strain in the cartilaginous vertebral columns of sharks.

In most bony fishes vertebral column strain during locomotion is almost exclusively in the intervertebral joints, and when these joints move there is the potential to store and release strain energy. Since cartilaginous fishes have poorly mineralized vertebral centra, we tested whether the vertebral bodies undergo substantial strain and thus may be sites of energy storage during locomotion. We measured axial strains of the intervertebral joints and vertebrae in vivo and ex vivo to characterize the dynamic behavior of the vertebral column. We used sonomicrometry to directly measure in vivo and

Integrated study of sediments and fish from Croatian rivers: potentially toxic elements and human health concerns.

Aquatic ecosystems are highly vulnerable to contamination by potentially toxic elements (PTEs), with sediments acting as long-term sinks and fish serving as bioindicators of environmental pollution. This study evaluated PTE concentrations in river sediments and three Squalius chub species (S. cephalus, S. squalus, S. svallize) collected from Croatian rivers that belong to the Adriatic and Danube basins. Chemical analyses were performed using inductively coupled plasma-optical emission spectrometry (ICP-OES) and inductively coupled plasma-mass spectrometry (ICP-MS) following acid microwave-assi

Transgenerational enhancement of low-protein feed utilization in Nile tilapia (Oreochromis niloticus) via parental injection with fragmented shark DNA.

The protein fraction is the most expensive component in aquafeed. This study aimed to determine if injecting fragmented shark (Squalus acanthias) DNA into Nile tilapia (Oreochromis niloticus) broodstock could enhance the offspring's ability to utilize low-protein diets. Shark genomic DNA was used as a donor source to investigate if its high metabolic efficiency and growth traits could biologically complement and improve feed utilization in tilapia fed low-protein diets. A 2 × 3 factorial design was used, where offspring from genetically modified (GM) and wild-type (WT) broodstock were fed diet

Differential effects of bicarbonate on severe hypoxia- and hypercapnia-induced cardiac malfunctions in diverse fish species.

We tested in six fish species [Pacific lamprey (Lampetra richardsoni), Pacific spiny dogfish (Squalus suckleyi), Asian swamp eel (Monopterus albus), white sturgeon (Acipenser transmontanus), zebrafish (Danio rerio), and starry flounder (Platichthys stellatus)] the hypothesis that elevated extracellular [HCO3-] protects spontaneous heart rate and cardiac force development from the known impairments that severe hypoxia and hypercapnic acidosis can induce. Hearts were exposed in vitro to either severe hypoxia (~ 3% of air saturation), or severe hypercapnic acidosis (either 7.5% CO2 or 15% CO2), w

Effects of ouabain and furosemide on transepithelial electrical parameters of the isolated shark ciliary epithelium.

Sections of the ciliary epithelium of adult sharks (Squalus acanthias) were mounted in Ussing-type chambers (area 0.2 cm2) for measurements of transepithelial potential difference (PD), short circuit current (SCC) and calculation of transepithelial resistance (R). In 15 preparations PD was aqueous side negative (-0.51 +/- 0.12 mV; SCC 18.3 +/- 2.5 microA cm-2; R 30.7 +/- 3.1 Ohm cm2). However, in 15 other preparations incubated in identical Ringer's solution PD was aqueous side positive (0.53 +/- 0.09 mV; SCC -19.6 +/- 2.3 microA cm-2; R 27.9 +/- 2.8 Ohm cm2). 10(-5) M ouabain or 10(-4) M furo

Differences in microthrix form between stomach and spiral intestine stages of the cestode, Trilocularia: interspecific variation or development?

Tetraphyllidean cestodes of the genus Trilocularia have been reported from Squalus acanthias throughout this shark's global distribution, occupying both the stomach and spiral intestine. This study investigated whether variation seen in microthrix form among specimens within this host species is indicative of interspecific variation or is attributable to developmental transformation of a single cestode species. Sequence data for the 2 nuclear genes 28S rDNA (D1-D3) and ITS1, and the mitochondrial gene 16S rDNA, were generated for 22 specimens of Trilocularia; 3 from the stomach and 19 from the

NASIGULPER ITSGOTATAO N. GEN., N. SP. (MONOCOTYLIDAE: CATHARIOTREMATINAE) INFECTING OLFACTORY LAMELLAE OF DEEPWATER GULPER SHARKS, CENTROPHORUS GRANULOSUS (BLOCH AND SCHNEIDER, 1801) (SQUALIFORMES: CENTROPHORIDAE) FROM THE GULF OF MEXICO OUTER CONTINENTAL SHELF.

We herein describe a new species of Cathariotrematinae Bullard, 2021 (Monogenoidea: Monocotylidae Taschenberg, 1879) and propose a new genus to accommodate it. Nasigulper itsgotatao n. gen., n. sp. infects the olfactory lamellae (nose) of the deepwater gulper shark, Centrophorus granulosus (Bloch and Schneider, 1801) (Squaliformes: Centrophoridae) from the north-central Gulf of Mexico outer continental shelf. The new species was assigned to Cathariotrematinae by having a 3-part attachment organ (TAO) and by lacking open loculi that symmetrically encircle a cluster of >2 loculi in the center of

The Rectal Gland of the Shark: The Road to Understanding the Mechanism and Regulation of Transepithelial Chloride Transport.

Pictured, described, and speculated on, for close to 400 years, the function of the rectal gland of elasmobranchs remained unknown. In the late 1950s, Burger discovered that the rectal gland of Squalus acanthias secreted an almost pure solution of sodium chloride, isosmotic with blood, which could be stimulated by volume expansion of the fish. Twenty five years later, Stoff discovered that the secretion of the gland was mediated by adenyl cyclase. Studies since then have shown that vasoactive intestinal peptide (VIP) is the neurotransmitter responsible for activating adenyl cyclase; however, t

Mechanically stripped pigmented and non-pigmented epithelium of the shark ciliary body: morphology and transepithelial electrical properties.

Sections of intact ciliary epithelium and mechanically stripped non-pigmented (NPE) and pigmented (PE) cell layers of adult sharks (Squalus acanthias) were mounted in Ussing-type chambers (area 0.1 cm2). Addition of 10(-5) M forskolin to the aqueous side of intact epithelium significantly increased short-circuit current (Isc) within 15 min and a maximum of approx. 30 microA cm-2 was reached after 45-60 min. Transepithelial potential difference (V) increased from -0.8 mV (aqueous side negative as compared with blood/stromal side) to -1.5 mV, whereas resistance (R) was unchanged (50 omega cm2).

A spermine-coupled cholesterol metabolite from the shark with potent appetite suppressant and antidiabetic properties.

OBJECTIVE: We describe the pharmacological properties of a novel spermine-cholesterol adduct, MSI 1436 (3beta-N-1(spermine)-7alpha, 24R-dihydroxy-5alpha-cholestane 24-sulfate), which causes reversible suppression of food and fluid intake in mammals resulting in profound weight loss, not associated with other signs or symptoms of illness, and which exhibits antidiabetic properties in genetically obese mice. METHODS: Wild-type rodents and strains with genetic obesity were studied. Effects on food and fluid intake, body weight and composition were examined along with pharmacological and toxicolo

Divergent dogfish: morphological correlates of vertebral column bending mechanics in two species of distantly related sharks.

BACKGROUND: Dogfish sharks from two different Orders - Squalus acanthias and Mustelus canis - offer the opportunity to compare whether known differences in vertebral morphology correlate with differences in the dynamic mechanical properties of the vertebral column. Histology of a caudal intervertebral joint in both species, combined with morphometrics of the entire column, provide the morphological information. Dynamic bending tests of ten-vertebrae sections of the vertebral column from the head to the base of the caudal fin are conducted by varying the amplitude of lateral curvature and bendi

Cranial nerves of the coelacanth, Latimeria chalumnae [Osteichthyes: Sarcopterygii: Actinistia], and comparisons with other craniata.

We reconstructed the cranial nerves of a serially sectioned prenatal coelacanth, Latimeria chalumnae. This allowed us to correct several mistakes in the literature and to make broad phylogenetic comparisons with other craniates. The genera surveyed in our phylogenetic analysis were Eptatretus, Myxine, Petromyzon, Lampetra, Chimaera, Hydrolagus, Squalus, Mustelus, Polypterus, Acipenser, Lepisosteus, Amia, Neoceratodus, Protopterus, Lepidosiren, Latimeria and Ambystoma. Cladistic analysis of our data shows that Latimeria shares with Ambystoma two characters of the cranial nerves. Our chief findi

A phase I and pharmacokinetic study of squalamine, a novel antiangiogenic agent, in patients with advanced cancers.

PURPOSE: A Phase I study of squalamine, a novel antiangiogenic agent originally isolated from the dogfish shark Squalus acanthias, was conducted in patients with advanced cancers to: (a) determine the maximum tolerated dose (MTD), dose-limiting toxicity (DLT) and pharmacokinetics of squalamine lactate when given as a 120-h continuous i.v. infusion every two weeks; and (b) to obtain information on prolonged (>120-h) continuous i.v. infusions in patients who have tolerated 120-h infusions. EXPERIMENTAL DESIGN: A rapid dose escalation scheme was used that permitted intrapatient dose escalation.
