The mitochondrial outer membrane AAA ATPase AtOM66 affects cell death and pathogen resistance in Arabidopsis thaliana.

One of the most stress-responsive genes encoding a mitochondrial protein in Arabidopsis (At3g50930) has been annotated as AtBCS1 (cytochrome bc1 synthase 1), but was previously functionally uncharacterised. Here, we show that the protein encoded by At3g50930 is present as a homo-multimeric protein complex on the outer mitochondrial membrane and lacks the BCS1 domain present in yeast and mammalian BCS1 proteins, with the sequence similarity restricted to the AAA ATPase domain. Thus we propose to re-annotate this protein as AtOM66 (Outer Mitochondrial membrane protein of 66 kDa). While transgeni

Synergistic disruption of brain metabolism and inflammatory signaling in adult zebrafish by co-exposure to alternariol monomethyl ether and titanium dioxide nanoparticles at physiologically relevant levels.

The combined health risks of mycotoxins and nanoparticles (NPs) remain poorly understood, particularly at physiologically relevant concentrations. Herein, we investigated the neurotoxic effects of 21-day exposure to alternariol monomethyl ether (AME, 0.2 nM) and titanium dioxide (TiO2) NPs (4.2 μg/mL), alone and in combination, in 5-month-old adult male zebrafish. While individual exposures caused minimal histopathological alterations and modest increase in swimming distance, metabolomic profiling revealed striking synergistic effects, that co-exposure altered 33 brain metabolites (vs. 11 by A

Novel method for detection of glycogen in cells.

Glycogen, a branched polymer of glucose, functions as an energy reserve in many living organisms. Abnormalities in glycogen metabolism, usually excessive accumulation, can be caused genetically, most often through mutation of the enzymes directly involved in synthesis and degradation of the polymer leading to a variety of glycogen storage diseases (GSDs). Microscopic visualization of glycogen deposits in cells and tissues is important for the study of normal glycogen metabolism as well as diagnosis of GSDs. Here, we describe a method for the detection of glycogen using a renewable, recombinant

Pools of cadmium in Chlamydomonas reinhardtii revealed by chemical imaging and XAS spectroscopy.

The green micro-alga Chlamydomonas reinhardtii is commonly used as a model to investigate metallic stress in photosynthetic organisms. The aim of this study was to explore processes implemented by three C. reinhardtii strains to cope with cadmium (Cd), and particularly to evidence Cd sequestration in the cell. For that, we used a combination of subcellular fractionation and chemical imaging (micro X-ray fluorescence (μXRF) and transmission electron microscopy (TEM/X-EDS)) to identify subcellular compartments of Cd accumulation, and X-ray absorption spectroscopy (XAS) to determine chemical Cd s

Cold acclimation reprograms the flavor metabolome of fresh tea leaves to enhance sensory quality: Insights from metabolomics, GC-O, and transcriptomics.

The enhanced sensory quality of early spring tea is empirically associated with pre-harvest cold exposure, yet the chemical and mechanistic basis for this enhancement requires further clarification. In this study, we investigated the metabolic changes in tea leaves under cold acclimation (4/0 °C) compared to mild cooling (15/11 °C) using metabolomics, HS-SPME/GC × GC-Q-TOF-MS, gas chromatography-olfactometry (GC-O), and transcriptomic analyses. Results indicated that cold acclimation induced the accumulation of key non-volatile tastants, notably soluble sugars and γ-aminobutyric acid (GABA), d

Primary and secondary metabolism in the sun-exposed peel and the shaded peel of apple fruit.

The metabolism of carbohydrates, organic acids, amino acids and phenolics was compared between the sun-exposed peel and the shaded peel of apple fruit. Contents of sorbitol and glucose were higher in the sun-exposed peel, whereas those of sucrose and fructose were almost the same in the two peel types. This was related to lower sorbitol dehydrogenase activity and higher activities of sorbitol oxidase, neutral invertase and acid invertase in the sun-exposed peel. The lower starch content in the sun-exposed peel was related to lower sucrose synthase activity early in fruit development. Dark resp

Hepatic ChREBP orchestrates intrahepatic carbohydrate metabolism to limit hepatic glucose 6-phosphate and glycogen accumulation in a mouse model for acute Glycogen Storage Disease type Ib.

OBJECTIVE: Carbohydrate Response Element Binding Protein (ChREBP) is a glucose 6-phosphate (G6P)-sensitive transcription factor that acts as a metabolic switch to maintain intracellular glucose and phosphate homeostasis. Hepatic ChREBP is well-known for its regulatory role in glycolysis, the pentose phosphate pathway, and de novo lipogenesis. The physiological role of ChREBP in hepatic glycogen metabolism and blood glucose regulation has not been assessed in detail, and ChREBP's contribution to carbohydrate flux adaptations in hepatic Glycogen Storage Disease type 1 (GSD I) requires further in

Multiomics profiling of transcriptional and metabolic responses of Bacillus siamensis LSZ23 to corn straw.

Agricultural straw is an abundant yet underutilised lignocellulosic resource whose microbial valorisation requires cellulolytic strains that sustain enzyme production under substrate-imposed stress. Although Bacillus siamensis is valued for plant growth promotion and biocontrol, its lignocellulose-degrading capacity and regulation remain largely unexplored. B. siamensis LSZ23, isolated from humic soil in Inner Mongolia, China, produced a balanced cellulolytic system (CMCase 0.321, β-glucosidase 0.275, avicelase 0.240 and FPase 0.209 U/mL in 24 h LB culture) and reduced sterilised corn straw dr

Prevention and repair of circulatory shock and cerebral ischemia/injury by various agents in experimental heatstroke.

The current report summarized animal models of heatstroke experimentation that advance our current knowledge of therapeutic effects on cerebrovascular dysfunction, hypercoagulable state and/or systemic inflammation with various agents in the setting of heatstroke. This was a narrative review of selected published primary basic literature from MEDLINE for 1973-2006. It was found that rodents shared with humans almost the same heatstroke reactions such as hyperpyrexia, hypotension, hyperventilation, pulmonary edema, hepatic and renal failure, hypercoagulable state, metabolic acidosis, systemic i

Exogenous Eugenol Alleviates Salt Stress in Tobacco Seedlings by Regulating the Antioxidant System and Hormone Signaling.

Salt stress seriously affects crop growth, leading to a decline in crop quality and yield. Application of exogenous substances to improve the salt tolerance of crops and promote their growth under salt stress has become a widespread and effective means. Eugenol is a small molecule of plant origin with medicinal properties such as antibacterial, antiviral, and antioxidant properties. In this study, tobacco seedlings were placed in Hoagland's solution containing NaCl in the presence or absence of eugenol, and physiological indices related to stress tolerance were measured along with transcriptom

Cerebrovascular dysfunction is an attractive target for therapy in heat stroke.

1. The aim of the present review is to summarize clinical observations and results of animal models that advance the knowledge of the attenuation of cerebrovascular dysfunction in the setting of heat stroke. It is a narrative review of selected published literature from Medline over the period 1959-2005. 2. All heat-stressed rodents, even under general anaesthesia, have hyperthermia, systemic inflammation, hypercoagulable state, arterial hypotension and tissue ischaemia and injury in multiple organs. These findings demonstrate that rodent heat stroke models can nearly mirror the full spectrum

Novel resuscitation strategy for pulmonary contusion after severe chest trauma.

BACKGROUND: Adenosine A2a receptor stimulation can increase coronary perfusion and also reduce leukocyte-mediated inflammatory responses in some conditions. Hextend is a novel colloid solution that may have antioxidant properties. All these actions might be beneficial after severe chest trauma, but have never been investigated. To fill these gaps, this study evaluated the therapeutic potential of a novel adenosine A2a agonist during fluid resuscitation from severe chest trauma with either standard-of-care crystalloid or Hextend. METHODS: Anesthetized, ventilated swine received unilateral, blu

Biochemical characterization of Paracoccidioides brasiliensis α-1,3-glucanase Agn1p, and its functionality by heterologous Expression in Schizosaccharomyces pombe.

α-1,3-Glucan is present as the outermost layer of the cell wall in the pathogenic yeastlike (Y) form of Paracoccidioides brasiliensis. Based on experimental evidence, this polysaccharide has been proposed as a fungal virulence factor. To degrade α-1,3-glucan and allow remodeling of the cell wall, α-1,3-glucanase is required. Therefore, the study of this enzyme, its encoding gene, and regulatory mechanisms, might be of interest to understand the morphogenesis and virulence process in this fungus. A single gene, orthologous to other fungal α-1,3-glucanase genes, was identified in the Paracoccidi

Evaluation of aphid resistance on different rose cultivars and transcriptome analysis in response to aphid infestation.

BACKGROUND: The rose is one of the most important ornamental flowers in the world for its aesthetic beauty but can be attacked by many pests such as aphids. Aphid infestation causes tremendous damage on plant tissues leading to harmed petals and leaves. Rose cultivars express different levels of resistance to aphid infestation yet the information remains unclear. Not only that, studies about the transcriptional analysis on defending mechanisms against aphids in rose are limited so far. RESULTS: In this study, the aphid resistance of 20 rose cultivars was evaluated, and they could be sorted in
