Exploring Blastocystis: hidden complexity in a common intestinal microbe.

Blastocystis is an anaerobic stramenopile that colonizes the large intestine of humans and many animals worldwide. Small-subunit rRNA-based typing reveals deep genetic diversity: at least 44 genetic variants, namely, subtypes (STs), are recognized, with ST1-ST4 accounting for most human carriage. Its medical relevance remains contested. Blastocystis carriage is frequently asymptomatic, and while associations with non-specific gastrointestinal symptoms (e.g. diarrhoea, abdominal pain and bloating), irritable bowel syndrome and extraintestinal manifestations such as urticaria have been proposed.

A P5-ATPase, TgFLP12, diverging from plant chloroplast lipid transporters mediates apicoplast fatty export in Toxoplasma.

Toxoplasma gondii, an apicomplexan parasite and agent of the human disease toxoplasmosis, possesses a non-photosynthetic relic plastid, named the apicoplast. Thought to be evolved from a red algal plastid, the apicoplast houses major metabolic pathways, such as heme, isoprenoid and lipid synthesis, crucial for parasite survival, and thus considered attractive drug targets. However, despite similarities with plant chloroplast lipid synthesis pathways, the apicoplast lacks canonical plant/chloroplast lipid transporters and so metabolite import/export is at present, poorly characterised. Here we

From plants to animals: Pythium, Lagenidium, Aphanomyces, and other oomycete pathogens of animals seen through a one-health lens.

Progress in treating oomycete infections of animals has been slow. Much relevant biological and therapeutic knowledge originates from plant pathology and agricultural science, while medical and veterinary literature develops independently. This review integrates these fields to highlight key knowledge gaps and translational opportunities in animal health. Oomycetes are eukaryotic Stramenopiles, distinct from true fungi; their hyphae are diploid and coenocytic, with cellulose and glucan cell walls. They reproduce via motile zoospores and durable oospores, enabling environmental persistence. Alt

Effects of maternal peripartum supplementation with Schizochytrium oil on metabolic status, colostrum quality, and neonatal calf performance and immunity in dairy cows.

The periparturient period in dairy cattle is characterized by significant metabolic stress, often involving a state of negative energy balance, heightened oxidative stress, and systemic inflammation. Supplementation with n-3 PUFA, such as docosahexaenoic acid (DHA), is a strategy employed to mitigate these negative effects due to their recognized anti-inflammatory and immunomodulatory properties. Furthermore, peripartum cow nutrition during this phase may exert a prenatal programming effect on calf immune development, particularly because DHA is efficiently transported across the placenta to t

Dietary Schizochytrium algal powder is an effective strategy to develop docosahexaenoic acid-enriched eggs in Chinese yellow-feathered laying quails.

Although docosahexaenoic acid (DHA) enrichment in eggs has been extensively studied in laying hens, research on DHA-fortified eggs in quails remains limited. This study evaluated the effects of graded levels of Schizochytrium algae powder (SAP) on production performance, egg quality, egg DHA enrichment and fatty acid composition, and related gene expression responsible for DHA absorption and transport in yellow-feathered laying quails. A total of 960 birds were randomly assigned to four dietary groups (0%, 0.4%, 0.8%, or 1.2% SAP) for an eight-week trial, with each group consisting of six repl

Study of the ichthyotoxic microalga Heterosigma akashiwo by transcriptional activation of sublethal marker Hsp70b in Transwell co-culture assays.

Despite the advance of knowledge about the factors and potential mechanisms triggering the ichthyotoxicity in microalgae, these remain unclear or are controversial for several species (e.g. Heterosigma). Neither typical toxicity tests carried out with cell extracts nor direct exposure to harmful species were proved suitable to unravel the mechanism of harm. Ichthyotoxic species show a complex harmful effect on fish, which is mediated through various mechanisms depending on the species. In this work, we present a method to study sub-lethal effects triggered by reactive oxygen species of a popul

Impact of climate change on the toxicity of bisphenol A in Mytilus galloprovincialis and assessment of phycoremediation using Nannochloropsis salina via a multi-biomarker strategy and modeling.

In the current study, the mussels Mytilus galloprovincialis, exposed to four varying temperatures (17, 20, 23, and 26 °C), were contaminated with 50 μg/L of bisphenol A both with and without Nannochloropsis salina. The toxicity evaluation is determined by quantifying various biomarkers related to oxidative stress, neurotoxicity, and cellular damage. The key findings indicate that the toxicity of bisphenol A is heightened by rising temperature. The impact of bisphenol A is most evident at 26 °C, leading to excessive production of reactive oxygen species, depletion of non-enzymatic antioxidants,

Astaxanthin-rich Nannochloropsis oculata mitigates high-fat diet-induced liver dysfunction in zebrafish by modulating lipid metabolism and oxidative stress.

High-fat diets (HFDs) are widely used in aquaculture but may adversely affect fish liver health by disturbing lipid metabolism and promoting oxidative stress. This study investigated the protective effects of Nannochloropsis oculata enriched with astaxanthin on zebrafish (Danio rerio) fed an HFD. All zebrafish (n = 135) were divided randomly into three groups: conventional diet (CD), high-fat diet (HFD), and HFD supplemented with 2% N. oculata (HFD + NO). After a four-week feeding trial, growth performance, hepatosomatic index, liver oxidative stress markers, and expression of immune cytokine

Blastocystis subtypes detected in humans and animals from Colombia.

Blastocystis is a common enteric protist colonizing probably more than 1 billion people along with a large variety of non-human hosts. This protist has been linked to symptoms and diseases such as abdominal pain, constipation, diarrhea, flatulence and irritable bowel syndrome (IBS). Remarkable genetic diversity has been observed, leading to the subdivision of the genus into multiple subtypes (ST), some of which are exclusively found in non-human hosts. The aim of this study was to determine the distribution of Blastocystis STs in different Colombian hosts. We obtained fecal samples positive fo

Bioremediation of marine shrimp wastewater using high-light-tolerant Rose Bengal mutants of Nannochloropsis oceanica offers a climate-resilient method for co-producing eicosapentaenoic acid under warming and flue gas CO₂.

This study investigates high-light-tolerant Nannochloropsis oceanica Rose Bengal mutants (RB2 and RB113) for bioremediation of shrimp aquaculture wastewater (SWW) under increased temperature and light, simulating future climate change. Cultivations were performed under 250 μmol photons m-2·s-1 with flue gas CO₂ supply. At 18 °C, RB mutants and wild-type (WT) strain showed similar growth. At 25 °C, representative of warming in subtropical and temperate regions, the RB113 mutant outperformed WT and RB2 in terms of biomass accumulation, photosynthetic efficiency, and non-photochemical quenching.

Hemolytic and cytotoxic activity from cultures of Aureococcus anophagefferens-a causative species of brown tides in the north-western Bohai Sea, China.

Brown tides were first observed in 2009 in the north-western Bohai Sea (Qinhuangdao sea area), China, and blooms have occurred at different scales in late spring every year since then. Although the detrimental effects on marine organisms of the causative phytoplankton species Aureococcus anophagefferens have been extensively studied, the mechanism remains poorly understood. We used erythrocytes and adrenal gland chromaffin tumor cells (PC12) to explore the hemolytic activity and cytotoxicity, respectively, of chloroform and methanol extracts of cultured A. anophagefferens isolated from the nor

Blastocystis, an Enigmatic Parasite.

Blastocystis spp. is the most prevalent human enteric protist, yet its clinical significance remains controversial. Currently recognized as a genetically diverse stramenopile with zoonotic potential, it comprises at least 42 subtypes, of which ST1-ST4 account for most human infections. Despite more than a century of research, its pathogenic role and ecological interactions are still debated. We performed a comprehensive bibliometric analysis of global scientific production on Blastocystis spp. from 1917 to 2025 using the Scopus database, retrieving 3710 publications. To develop greener extraction alternatives for microalgae biomass, ultrasound assisted extraction (UAE) and pressurized liquid extraction (PLE) with different biobased solvents were investigated, demonstrating that both techniques are useful alternatives for algal lipid extraction. Specifically, Nannochloropsis gaditana lipids were extracted by UAE and PLE at different temperatures and extraction times with sustainable solvents like 2-Methyltetrahydrofuran (2-MeTHF) and its mixtures with ethanol and other alcohols. The best oil yields for both PLE and UAE of N. gaditana were achieved with the

Meta-taxonomic analysis of prokaryotic and eukaryotic gut flora in stool samples from visceral leishmaniasis cases and endemic controls in Bihar State India.

Visceral leishmaniasis (VL) caused by Leishmania donovani remains of public health concern in rural India. Those at risk of VL are also at risk of other neglected tropical diseases (NTDs) including soil transmitted helminths. Intestinal helminths are potent regulators of host immune responses sometimes mediated through cross-talk with gut microbiota. We evaluate a meta-taxonomic approach to determine the composition of prokaryotic and eukaryotic gut microflora using amplicon-based sequencing of 16S ribosomal RNA (16S rRNA) and 18S rRNA gene regions. Holed up, but thriving: Impact of multitrophic cryoconite communities on glacier elemental cycles.

Cryoconite holes (water and sediment-filled depressions), found on glacier surfaces worldwide, serve as reservoirs of microbes, carbon, trace elements, and nutrients, transferring these components downstream via glacier hydrological networks. Through targeted amplicon sequencing of carbon and nitrogen cycling genes, coupled with functional inference-based methods, we explore the functional diversity of these mini-ecosystems within Antarctica and the Himalayas. These regions showcase distinct environmental gradients and experience varying rates of environmental change influenced by global clima

A novel protein kinase-like domain in a selenoprotein, widespread in the tree of life.

Selenoproteins serve important functions in many organisms, usually providing essential oxidoreductase enzymatic activity, often for defense against toxic xenobiotic substances. Most eukaryotic genomes possess a small number of these proteins, usually not more than 20. Selenoproteins belong to various structural classes, often related to oxidoreductase function, yet a few of them are completely uncharacterised. Here, the structural and functional prediction for the uncharacterised selenoprotein O (SELO) is presented. Using bioinformatics tools, we predict that SELO protein adopts a three-dimens
