Effects of Mechanical Stress Stimulation on Function and Expression Mechanism of Osteoblasts.

Osteoclasts and osteoblasts play a major role in bone tissue homeostasis. The homeostasis and integrity of bone tissue are maintained by ensuring a balance between osteoclastic and osteogenic activities. The remodeling of bone tissue is a continuous ongoing process. Osteoclasts mainly play a role in bone resorption, whereas osteoblasts are mainly involved in bone remodeling processes, such as bone cell formation, mineralization, and secretion. These cell types balance and restrict each other to maintain bone tissue metabolism. Bone tissue is very sensitive to mechanical stress stimulation. Unl

Molecular mechanisms of oxidative stress in asthma.

The lungs are exposed to reactive oxygen species oxygen (ROS) produced as a result of inhalation of oxygen, as well as smoke and other air pollutants. Cell metabolism and the NADPH oxidases (Nox) generate low levels of intracellular ROS that act as signal transduction mediators by inducing oxidative modifications of histones, enzymes and transcription factors. Redox signalling is also regulated by localised production and sensing of ROS in mitochondria, the endoplasmic reticulum (ER) and inside the nucleus. Intracellular ROS are maintained at low levels through the action of a battery of enzym

Stress as a possible mechanism in melanoma progression.

The incidence of melanoma, the most aggressive type of cutaneous malignant tumor, is currently on the rise. Treatment in advanced stages is still unsuccessful compared with other malignant tumors, thus it is important to indentify the key mechanisms responsible for melanoma progression and metastasis. Genetic and molecular components, in particular, that are up- or downregulated in melanoma cells, affect the invasive potential of melanoma. Another possible important cofactor highlighted by recent studies is chronic stress, involving environmental and psychological factors, which can be an impo

Central mechanisms of stress-induced headache.

Stress is the most commonly reported trigger of an episode of chronic tension-type headache (CTTH); however, the causal significance has not been experimentally demonstrated to date. Stress may trigger CTTH through hyperalgesic effects on already sensitized pain pathways in CTTH sufferers. This hypothesis could be partially tested by examining pain sensitivity in an experimental model of stress-induced headache in CTTH sufferers. Such examinations have not been reported to date. We measured pericranial muscle tenderness and pain thresholds at the finger, head and shoulder in 23 CTTH sufferers

Stress and tension-type headache mechanisms.

Stress is widely demonstrated as a contributing factor in tension-type headache (TTH). The mechanisms underlying this remain unclear at present. Recent research indicates the importance of central pain processes in tension-type headache (TTH) pathophysiology. Concurrently, research with animals and healthy humans has begun to elucidate the relationship between stress and pain processing in the central nervous system, including central pain processes putatively dysfunctional in TTH. Combined, these two fields of research present new insights and hypotheses into possible mechanisms by which stre

Stress and the heart. Mechanisms, measurement, and management.

Unrelieved physical or mental stress and repeated episodic stress are ultimately harmful to the cardiovascular system and thus can be life-threatening. In this article, Dr Eliot describes efforts to quantify the psychophysiologic responses to stress and to identify the components of stress and its clinical consequences. He also explains the importance of controlling the real-life episodic fluctuations in blood pressure that occur daily in response to stress.

Stress injuries of the adolescent extensor mechanism.

Stress injury to the extensor mechanism is common, and we expect it to become more so as adolescent athletes train earlier and more vigorously. The clinical and radiographic findings of a number of these stress injuries are described. Our young athletes challenge us to continue studying these stress injuries with the goal of offering them more effective preventative programs. The success of our study will probably lie in the areas of stretching, cross training, and strength programs.

Oxidative stress and antioxidant therapeutic mechanisms.

Oxidative stress is now understood as a disturbance in the cellular redox balance, involving the accumulation of reactive oxygen, nitrogen, and other reactive species beyond the capacity of antioxidant defenses, with effects that range from essential redox signaling to harmful oxidative damage. Reactive oxygen and nitrogen species are generated from both endogenous metabolic processes and exogenous environmental factors. While controlled levels of oxidative stress contribute to cellular signaling and homeostasis, excessive oxidative damage can lead to pathological conditions, including cardiov

Stress and cardiovascular disease: mechanisms and measurement.

Physiological overreactivity to mental stress has been correlated with the subsequent development of cardiovascular diseases. This coronary-prone behavior (referred to as "hot reacting") is distinct from the Type A behavior pattern and is seen in about 1 in 5 apparently healthy individuals. This paper describes a noninvasive system for measuring hemodynamics that can reveal the mechanisms raising the blood pressure of hot reactors or hypertensives. The subject is challenged with a variety of mental and physical stressors while the hemodynamic responses are monitored. Blood pressure elevations

Disruption of YAP biomolecular condensates by mechanical stress drives intervertebral disc vascularization.

Angiogenesis represents a key pathological basis for intervertebral disc degeneration (IDD) and discogenic low back pain (LBP), yet its underlying mechanisms remain elusive. Here, we show that mechanical stress induces disc vascularization by activating Hippo signaling and inducing Yes-associated protein (YAP) degradation in nucleus pulposus (NP) cells. This finding was corroborated in NP-specific Yap conditional knockout mice, which exhibited enhanced disc vascularization. Mechanistically, YAP undergoes liquid-liquid phase separation to form biomolecular condensates that recruit and sequester

Mechanical stress may initiate osteoderm development in the American alligator (Alligator mississippiensis).

Osteoderms are bones that form in the dermis of several species, but their development and function are not well understood. Comparing cellular and molecular events that initiate and drive pathologies similarly characterized by ossification of soft tissues (heterotopic ossification) may provide insight into how osteoderms develop. It is thought that the origin of some heterotopic ossification lesions is mediated through mechanotransduction, or the conversion of mechanical forces on the extracellular matrix (ECM) to biochemical signals that initiate bone formation. Discrete osteoderm developmen

Role of mechanical stress and neutrophils in the pathogenesis of plaque erosion.

Mechanical stress is a well-recognized driver of plaque rupture. Likewise, investigating the role of mechanical forces in plaque erosion has recently begun to provide some important insights, yet the knowledge is by far less advanced. The most significant example is that of shear stress, which has early been proposed as a possible driver for focal endothelial death and denudation. Recent findings using optical coherence tomography, computational sciences and mechanical models show that plaque erosion occurs most likely around atheromatous plaque throats with specific stress pattern. In paralle

Optical coherence tomography for the investigation of skin adaptation to mechanical stress.

BACKGROUND: Skin breakdown due to limb-socket interface stress is a significant problem for lower limb prosthesis users. While it is known that skin can adapt to stress to become more resistant to breakdown, little is understood about skin adaptation and few methods exist to noninvasively investigate it. In this study, we present novel, noninvasive imaging methods using Optical Coherence Tomography (OCT) to assess key features of the cutaneous microvasculature that may be involved in skin adaptation. MATERIALS AND METHODS: Eight able-bodied participants wore a modified below-knee prosthetic s

LRP5, Bone Density, and Mechanical Stress: A Case Report and Literature Review.

The Wnt-β-catenin pathway receptor, low-density lipoprotein receptor-related protein 5 (LRP5), is a known regulator of bone mineral density. It has been hypothesized that specific human polymorphisms in LRP5 impact bone density, in part, by altering the anabolic response of bone to mechanical loading. Although experiments in animal models support this hypothesis, there is limited evidence that LRP5 polymorphisms can alter the anabolic response of bone to mechanical loading in humans. Herein, we report a young male who harbors a rare LRP5 missense mutation (A745V) and who provides potential pro

Mechanical loading mitigates osteoarthritis symptoms by regulating endoplasmic reticulum stress and autophagy.

Osteoarthritis (OA) is a disease characterized by cartilage damage and abnormal remodeling of subchondral bone. Our previous study showed that in the early stage of OA, knee loading exerts protective effects by suppressing osteoclastogenesis through Wnt signaling, but little is known about loading effects at the late OA stage. Endoplasmic reticulum (ER) stress and autophagy are known to be involved in the late OA stage. We determined the effects of mechanical loading on ER stress and autophagy in OA mice. One hundred seventy-four mice were used for a surgery-induced OA model. In the first set

Mechanical stress influences the viability and morphology of human parametrial ligament fibroblasts.

The present study aimed to investigate damage to human parametrial ligament fibroblasts by detecting cell proliferation, cytoskeletal structure, cellular alterations and senescence. Uterosacral and cardinal ligaments were obtained from 10 patients with cervical intraepithelial neoplasia grade II‑III, who had received total vaginal hysterectomies, and fibroblasts were derived from this tissue. Fibroblasts were stretched using a four‑point bending system with a force of 0 (control), 1,333 µ strain (1 mm) or 5,333 µ strain (4 mm) for 4 h. The present study revealed that mechanical force significa

The mechanical stress-strain properties of single electrospun collagen type I nanofibers.

Knowledge of the mechanical properties of electrospun fibers is important for their successful application in tissue engineering, material composites, filtration and drug delivery. In particular, electrospun collagen has great potential for biomedical applications due to its biocompatibility and promotion of cell growth and adhesion. Using a combined atomic force microscopy (AFM)/optical microscopy technique, the single fiber mechanical properties of dry, electrospun collagen type I were determined. The fibers were electrospun from a 80 mg ml(-1) collagen solution in 1,1,1,3,3,3-hexafluro-2-pr
