OBJECTIVE: The lateral periaqueductal gray (LPAG), which mainly contains glutamatergic neurons, plays an important role in social responses, pain, and offensive and defensive behaviors. Currently, the whole-brain monosynaptic inputs to LPAG glutamatergic neurons are unknown. This study aims to explore the structural framework of the underlying neural mechanisms of LPAG glutamatergic neurons.
METHODS: This study used retrograde tracing systems based on the rabies virus, Cre-LoxP technology, and immunofluorescence analysis.
RESULTS: We found that 59 nuclei projected monosynaptic inputs to the

The retrograde transneuronal viral tracing method was used to study the CNS nuclei that innervate the parasympathetic preganglionic neurons controlling the submandibular gland in the rat. A genetically engineered beta-galactosidase expressing Bartha strain of pseudorabies virus (PRV) was injected into the submandibular gland of rats. After 4 days, PRV infected tissues were reacted with the Bluo-Gal substrate (halogenated indolyl-beta-D-galactoside) and labeled cell bodies were identified throughout the brain. In the medulla oblongata, cell body labeling was seen in the superior salivatory nucl

Post-mortem neuropathological examination of five cases of Niemann-Pick disease type C revealed neurofibrillary tangles in many parts of the brain. Tangles were a consistent finding in the hippocampus, hypothalamus, substantia innominata, midbrain pons and medulla. Other regions of the brain in which tangles were present included neocortex, basal ganglia, thalamus, cerebellar cortex in one case, and dentate nucleus in another. The tangles were argyrophilic, fluoresced under ultraviolet light when stained with thioflavin S, and reacted strongly with antibody to tau protein. Some of the tangles

The memory disturbance of senile dementia of Alzheimer type has been thought to associate with marked degeneration and loss of cholinergic neurons of the basal forebrain (nucleus basalis of Meynert, NBM). Electrical or chemical destruction of the NBM causes memory deficits in rats. After unilateral lesioning of the NBM in adult rats with excitotoxic amino acid, kainic acid, basal forebrain cells of fetal rats were transplanted through a microsyringe needle, the tip of which was transcortically inserted to the subarachnoid space. Eight weeks after the transplantation, passive avoidance response

Choline acetyltransferase and acetylcholinesterase activities as well as serotonin and imipramine binding were determined in the hypothalamus, nucleus basalis of Meynert, and frontal and temporal poles of subjects with Pick's disease. Choline acetyltransferase activity was decreased in the hypothalamus and nucleus basalis of Meynert, and acetylcholinesterase activity was decreased in the nucleus basalis of Meynert only. Serotonin binding was decreased in all sites but the nucleus basalis of Meynert, and imipramine binding was altered only in the frontal pole. Comparison with previous reports o

Immunohistochemically identified cholinergic neurons in the basal nucleus of Meynert in a brain with SDAT have been compared with those in two age- and sex-matched normal brains. The numbers of such cells at carefully matched levels are not significantly lower, but the cells in SDAT are substantially smaller than in the normal basal nucleus. The persistence of shrunken cholinergic neurons in the basal nucleus in the diseased brain is similar to that seen in an experimental study of retrograde cellular degeneration in the nucleus following damage of the cortex.

The efferent projections of the anterior and posterodorsal part of the medial nucleus (MePD) in the mouse were studied by means of anterograde axonal tracing using biotinylated dextran amine. The MePD axons ran mainly via the stria terminalis and to a lesser extent via the ventral amygdalofugal pathway. The projections to the forebrain were broadly distributed and varied from very strong to scant. The most significant connections were destined to the bed nucleus of the stria terminalis in which all parts of the medial division were innervated by MePD neurons. Moderate projections reached the l

The afferent connections of the parvocellular reticular formation were systematically investigated in the rat with the aid of retrograde and anterograde horseradish peroxidase tracer techniques. The results indicate that the parvocellular reticular formation receives its main input from several territories of the cerebral cortex (namely the first motor, primary somatosensory and granular insular areas), districts of the reticular formation (including its contralateral counterpart, the intermediate reticular nucleus, the nucleus of Probst's bundle, the dorsal paragigantocellular nucleus, the al

N-methyl-D-aspartate (NMDA) receptors have been implicated in learning and memory. Many findings show that NMDA receptor antagonists impair memory. Few studies, however, have investigated the role of NMDA receptor agonists in mnemonic function. The present study examined the effects of nucleus basalis magnocellularis (nbm) injections of NMDA on memory. Rats were trained in a two-component double Y-maze task consisting of a spatial discrimination and a delayed alternation. Rats (n = 7) were surgically implanted with bilateral cannulae in the nbm prior to maze training. Once trained, animals rec

The basal forebrain complex, which includes the nucleus basalis magnocellularis (NBM), provides widespread cholinergic and gamma-aminobutyric acid-containing projections throughout the brain, including the insular and pyriform cortices. A number of studies have implicated the cholinergic neurons in the mediation of learning and memory processes. However, the role of basal forebrain activity in information retrieval mechanisms is less known. The aim of the present study is to evaluate the effects of reversible inactivation of the NBM by tetrodotoxin (TTX, a voltage-sensitive sodium channel bloc

Dissection of the neural circuits of the cerebral cortex is essential for studying mechanisms underlying brain function. Herein, combining a retrograde rabies tracing system with fluorescent micro-optical sectional tomography, we investigated long-range input neurons of corticotropin-releasing hormone containing neurons in the six main cortical areas, including the prefrontal, somatosensory, motor, auditory, and visual cortices. The whole brain distribution of input neurons showed similar patterns to input neurons distributed mainly in the adjacent cortical areas, thalamus, and basal forebrain

The local cerebral metabolic rate for glucose (LCMRgl) was determined by positron emission tomography (PET) using the 18F-fluorodeoxyglucose method in a series of Alzheimer patients and normal controls. The LCMRgl declined in the cerebral cortex with age, but the decrement was significantly greater in the clinically diagnosed Alzheimer's cases. Comparison of PET and psychological data indicated that, as the disease progressed clinically, the reduction in cortical LCMRgl and the number of cortical regions involved also increased. Variable regions of cortex were involved in the early stages but

The cortical somatostatin interneuron population includes several diverse cell types, among them the Martinotti cells. Layer-specific differences in connectivity and function between different subtypes of Martinotti cells are becoming apparent, which require contemporary studies to investigate cortical interneurons in a layer and subtype-specific manner. In this study, we investigate the connectivity of a subtype of Chrna2+ layer 5 Martinotti cells in the primary motor cortex, using a monosynaptic retrograde rabies viral tracer. We found direct input from pyramidal cells and local parvalbumin

Silver staining (Bodian procedure) in the nucleus of the sublenticular substantia innominata (SI), also referred to as the nucleus basalis of Meynert, was evaluated in autopsy material from patients with Alzheimer's disease or senile dementia of the Alzheimer type (age at death: mean 69 years, range 63 to 81 years; time between onset of symptoms and death: mean 5.6 years, range 2.5 to 11.0 years). Although a decrease in the number of neurons and an increase in gliosis were observed in the SI in the Alzheimer dementia cases, classic senile plaques, as well as neurofibrillary tangles and granulo

The population of neurons and the neuronal size in the nucleus basalis of Meynert (nbM) were studied in 20 patients with definite Creutzfeldt-Jakob disease (CJD). When compared with a normal control group, the 20 CJD brains showed a significant loss of neurons and reduction of neuronal size, mainly in the middle level of the nbM and mostly affecting the right side. Since these findings show some parallelism with the amount of cortical damage and given the scarce gliosis and spongiosis found in only six of the 20 CJD brains, we postulate that the involvement of the nbM in CJD is a retrograde ab

In order to substantiate the hypothesis of a cholinergic pathogenesis of neuritic plaques in Alzheimer's disease the relationship between the loss of cholinergic neurons in six subdivisions of the nucleus basalis of Meynert and density of neuritic plaques in five neocortical target areas and hippocampus was studied in five cases with Alzheimer's disease. Distribution of plaques in different cortical areas as well as degeneration pattern of neurons within the subpopulations of the nucleus basalis were markedly different in the cases of Alzheimer's disease. Quantitative evaluation of the number

The basal forebrain cholinergic neurons (BFCNs) in substantia innominata (SI) and nucleus basalis magnocellularis (nBM) provide dense innervation to multiple cortical areas, shaping cognition, attention, and the sleep-wake cycle. However, the afferent connectome controlling the spiking of BFCNs is poorly characterized. Although biased toward the nearby ventral striatum, monosynaptic rabies virus mapping revealed that spiny projection neurons (SPNs) throughout the striatum innervated BFCNs in the SI/nBM. Roughly 1-5% of SPNs were retrogradely labeled, with both D1 dopamine (DA) receptor-express

Given that sleep and pain are bidirectionally related, we investigated the neural circuits underlying pain-induced sleep disturbances using two acute pain models. Activation of nociceptors in acute inflammatory pain (AIP) significantly reduced sleep by 45-50% in the first 6 h, with reduced sleep spindle density for 1-3 h post-AIP. Additionally, an "optogenetic pain (Opto-Pain)" model is implemented to produce acute peripheral pain-induced awakenings that reduced sleep comparable to AIP. Both pain models are used to test the role of wake-promoting neurons in the parabrachial nucleus that expres

The anteromedial area of the bed nuclei of the stria terminalis (BSTam) is the relatively undifferentiated region of the anterior medial (anteromedial) group of the bed nuclei of the stria terminalis (BSTamg), which also includes the more distinct dorsomedial, magnocellular, and ventral nuclei. The overall pattern of axonal projections from the rat BSTam was analyzed with the Phaseolus vulgaris-leucoagglutinin anterograde pathway tracing method. Brain areas receiving relatively moderate to strong inputs from the BSTam fall into five general categories: neuroendocrine system (regions containing
