Toltrazuril and sulphonamide treatment against naturally Isospora suis infected suckling piglets: is there an actual profit?

A study was carried out to assess the efficacy and the economic profit of prophylactic treatment against Isopsora suis with toltrazuril or with a sulfamethazine/trimethoprim combination in piglets from an intensive pig farm. Thirty-one litters were included in study. Eight litters were treated once with toltrazuril (20 mg/kg b.w.) at 3 days of age (Toltra group); 8 litters were treated with 2 ml/animal of a [corrected] sulphonamide combination (sodium sulfamethazine 250 [DOSAGE ERROR CORRECTED] mg and trimethoprim 50 [DOSAGE ERROR CORRECTED] mg/kg b.w.) for 3 consecutive days starting at 3 day

Screening methods for the detection of antibiotic residues in slaughter animals: comparison of the European Union Four-Plate Test, the Nouws Antibiotic Test and the Premi®Test (applied to muscle and kidney).

Microbial growth inhibition tests are widely used as the primary screening approach for the detection of antibiotic residues in slaughter animals. In this study we evaluated and compared the performance of the European Union Four-Plate Test (EU4pt), the Nouws Antibiotic Test (NAT), and a commercial ampoule test, the PremiTest (applied to both muscle and kidney), by parallel analysis of 735 slaughter animals. The EU4pt only showed significant inhibition with two muscle samples containing 305 µg kg(-1) doxycycline and 648 µg kg(-1) tulathromycin, while an maximum residue limit (MRL) violation of

Acetaminophen is an inhibitor of hepatic N-acetyltransferase 2 in vitro and in vivo.

Slow acetylators of the polymorphic N-acetyltransferase 2 (NAT2, EC 2.3.1.5) suffer more often from side-effects of NAT-substrates than fast acetylators. Since concomitant administration of drugs may inhibit NAT2, we studied the influence of acetaminophen on NAT2 in human hepatic cytosol in vitro and in healthy individuals. In-vitro acetylation was assessed in liver homogenate of one fast and one slow acetylator using sulfamethazine as a test substrate. Acetaminophen competitively inhibited sulfamethazine acetylation in fast and slow acetylator liver samples with Ki values of acetaminophen of

Naturally occurring biopterin as a photosensitizer for enhanced photodegradation of sulfonamides in aquatic environments.

The natural photosensitizer Biopterin (Bip) can significantly accelerate the photodegradation of sulfamethazine (SMZ). Under simulated sunlight, 5 μM Bip achieved a 3-h degradation rate of 69.06 ± 2.50% for SMZ, while direct photolysis was less than 3%. The reaction rate increases linearly with the concentration of Bip, with the optimal pH ≈ 8. HCO3- and Cl- enhanced SMZ degradation, likely through the generation of reactive secondary species and ion-mediated processes. Laser flash photolysis and quenching experiments confirmed that the triplet excited state (3Bip∗) of Bip is the main active i

Acetylator status of kwashiorkor children in Ibadan (south-west Nigeria).

Acetylator status was determined in 25 kwashiorkor children, aged between 8 months and 3 years and in 25 age-matched control group of healthy children after a single oral dose of sulphamethazine (40 mg/kg body weight) and by measuring the acetylated sulphamethazine in blood samples, collected 6 h after the administration of sulphamethazine. The percentage of slow acetylators among kwashiorkor children was 40% while among the control group of children it was 48%. The difference between the two groups was not significant. Therefore, it is probable that the slow acetylator status of the Nigerian

Methemoglobinemia after ingestion of Chinese herbal medicine in a 9-day-old infant.

Traditional Chinese Medicine has long been popular among Chinese people, but it is always difficult for physicians to ascertain the nature and use of different ingredients involved. We report case of a 9-day-old infant who ingested home-made herbal medicine and developed cyanosis with methemoglobinemia. He responded to a single dose of methylene blue therapy. The suspected causative agent identified in the herbal medicine was sulfamethazine. This is the first reported case of methemoglobinemia related to the use of Chinese herbal medicine in the newborn period.

Fate of antibiotics and antibiotic resistance genes in a full-scale restaurant food waste treatment plant: Implications of the roles beyond heavy metals and mobile genetic elements.

Is our food safe and free of the crisis of antibiotics and antibiotic resistance (AR)? And will the derived food waste (FW) impose AR risk to the environment after biological treatment? This study used restaurant FW leachates flowing through a 200 tons-waste/day biological treatment plant as a window to investigate the fate of antibiotics and antibiotic-resistance genes (ARGs) during the acceptance and treatment of FW. Sulfonamides (sulfamethazine, sulfamethoxazole) and quinolones (ciprofloxacin, enrofloxacin, ofloxacin) were detected during FW treatment, while tetracyclines, macrolides and ch

Molecular and Functional Characterization of N-Acetyltransferases NAT1 and NAT2 in Cynomolgus Macaque.

Arylamine N-acetyltransferases (NATs) are drug-metabolizing enzymes essential for the metabolism of endogenous substrates and xenobiotics, and their molecular characteristics have been extensively investigated in humans, but not in cynomolgus macaques, nonhuman primate species important for drug metabolism studies. In this study, cynomolgus NAT1 and NAT2 cDNAs were isolated from livers. NAT1 and NAT2 were characterized by molecular analyses and drug-metabolizing assays. A total of 9 transcript variants were found for cynomolgus NAT1, similar to human NAT1, and contained 1-4 exons with the codi

Leaching of veterinary antibiotics in calcareous Chinese croplands.

Veterinary antibiotics reach the soil environment by manure application, where they accumulate or are prone to vertical translocation. We assumed that a high pH value at slightly calcareous soil properties should retain tetracyclines and fluoroquinolones even at high manure loads, whereas it should facilitate leaching of sulfonamides. Hence, we investigated the transport of manure-added antibiotics in a soil developed from calcareous parent material on a farm in peri-urban Beijing, China. Leachate was collected from zero-tension samplers, installed at 40cm depth under undisturbed soil, which w

Use and environmental occurrence of antibiotics in freestall dairy farms with manured forage fields.

Environmental releases of antibiotics from concentrated animal feeding operations (CAFOs) are of increasing regulatory concern. This study investigates the use and occurrence of antibiotics in dairy CAFOs and their potential transport into first-encountered groundwater. On two dairies we conducted four seasonal sampling campaigns, each across 13 animal production and waste management systems and associated environmental pathways: application to animals, excretion to surfaces, manure collection systems, soils, and shallow groundwater. Concentrations of antibiotics were determined using on line

Possible association between different congenital abnormalities and use of different sulfonamides during pregnancy.

The objective of the study presented here was to check the debated human teratogenic potential of sulfonamide drugs. Five different sulfonamides such as sulfamethazine, sulfathiourea, sulfamethoxypyridazine, sulfamethoxydiazine and the combination of sulfamethazine-sulfathiourea-sulfamethoxypyridazine were differentiated. Cases with congenital abnormalities were compared with their matched controls without congenital abnormalities in the population-based large data set of the Hungarian Case-Control Surveillance of Congenital Abnormalities between 1980 and 1996. Of 38,151 newborn infants withou

Understanding flow patterns from the field - Controlled laboratory experiments on the transport behavior of veterinary antibiotics in the presence of liquid manure.

The main entry path of veterinary antibiotics to the environment is the application of liquid manure on agricultural land. Along with the manure, they can infiltrate into soils and leach into groundwater. As the environmental behavior of veterinary antibiotics is strongly affected by the process of sorption, the comprehensive knowledge regarding their sorption behavior is key to a reliable risk assessment. However, the flow patterns in field experiments are influenced by several factors that can hardly be distinguished, while most of the sorption studies on veterinary antibiotics were designed

Transport of three veterinary antimicrobials from feedlot pens via simulated rainfall runoff.

Veterinary antimicrobials are introduced to wider environments by manure application to agricultural fields or through leaching or runoff from manure storage areas (feedlots, stockpiles, windrows, lagoons). Detected in manure, manure-treated soils, and surface and ground water near intensive cattle feeding operations, there is a concern that environmental contamination by these chemicals may promote the development of antimicrobial resistance in bacteria. Surface runoff and leaching appear to be major transport pathways by which veterinary antimicrobials eventually contaminate surface and grou

Systems-wide analysis of A. fumigatus using kinetic modeling of metabolic pathways to identify putative drug targets.

Aspergillosis is a major causative factor for morbidity in those with impaired immune systems, often caused by Aspergillus fumigatus. The diagnosis and treatment are difficult due to the diversity of individuals and risk factors and still pose a challenge for medical professionals. To understand the pathogenicity of any organism, it is critical to identify the significant metabolic pathways that are involved. Our work focused on developing kinetic models of critical pathways crucial for the survival of A. fumigatus using COPASI. While focusing on the folate biosynthesis, ergosterol biosynthesi

Hazard prioritization and risk characterization of antibiotics in an irrigated Costa Rican region used for intensive crop, livestock and aquaculture farming.

Antibiotics alter the homeostasis of microbial communities and select for antibiotic-resistant bacteria in the wild. Thus, the accumulation of unnaturally high concentration of these substances in the environment due to their use in human activities can be regarded as a neglected form of pollution, especially in countries with agricultural-based economies. Qualitative and quantitative information on antibiotic usage in Costa Rica is scarce, hence the design and enforcement of prevention strategies and corrective measures is difficult. To address this issue, and aiming in the long run to contri

Comparison of supercritical CHF3 and CO2 for extraction of sulfonamides from various food matrices.

Supercritical CHF3 and methanol-modified CHF3 were compared with supercritical CO2 and methanol-modified CO2 for the extraction of sulfonamides from a spiked sand sample. In addition, a fortified nonfat milk powder, fortified egg yolk, and fortified beef liver were studied. The results showed CHF3 has higher solvating power and selectivity for extraction of two of the three sulfonamides than CO2. Extraction efficiencies of sulfamethazine and sulfadimethoxine using pure CHF3 were more than 45% higher from spiked sand samples and over 200% higher from fortified beef liver samples than pure CO2.
