Picocyanobacteria contribute significantly to carbon export but the mechanism by which their biomass is transported to depth is unclear. By analysing16S rRNA gene sequences from sediment trap material and size fractionated water samples from different depths in the Baltic Sea, we show that picocyanobacterial contribution to phytoplankton biomass export was largely proportional (70-90%) to their surface water abundances. However, we also found that both cell aggregation (enrichment in >10 μm fraction) and sedimentation (trap enrichment) varied among Synechococcus ASVs (amplicon sequence variant

Despite being fundamental to multiple biological processes, phosphorus (P) availability in marine environments is often growth-limiting, with generally low surface concentrations. Picocyanobacteria strains encode a putative ABC-type phosphite/phosphate/phosphonate transporter, phnDCE, thought to provide access to an alternative phosphorus pool. This, however, is paradoxical given most picocyanobacterial strains lack known phosphite degradation or carbon-phosphate lyase pathway to utilise alternate phosphorus pools. To understand the function of the PhnDCE transport system and its ecological co

The capacity of photosynthetic microorganisms to fix carbon dioxide into biomass positions them as promising cell factories for sustainable biomanufacturing. However, limitations in screening throughput hinder the identification of enzymes, strains, and growth conditions needed to realize this potential. Here we present a microplate-based high-throughput cultivation system that can be integrated into existing automation infrastructure and supports growth of both prokaryotic and eukaryotic photosynthetic microorganisms. We validate this system by optimizing BG-11 medium compositions for Synecho

A defining feature of circadian rhythms is an internal oscillator that is self-sustaining in constant conditions, but the utility of this free-running property is not clear.1 In cyanobacteria, two alternative timing systems are found: a canonical circadian clock and an hourglass-like system incapable of free-running.2 By swapping genetic elements, we engineer a pathway to convert a circadian clock into an hourglass. We show that the performance of these systems is similar in a balanced light-dark cycle, but the hourglass shows dysregulated transcription in long photoperiod days and fails to pr

Five cyanophyte species (Amorphonostoc punctiforme, Gloeocapsa turgidus, Sphaeronostoc coeruleum, Stratonostoc linckia f. spongiaeforme and Synechococcus cedrorum) were isolated and identified from sandy Egyptian soils. Polysaccharides extracted from these species showed a pronounced antiviral activity against Rabies and Herpes-1 viruses represented by the absence of the characteristic cytopathic effects of these viruses. It was found that 100 μg polysaccharide/ml induced 100% inhibition of the two viruses which, depending on the polysaccharide concentration. Both of Gloeocapsa turgidus and Sy

Cultivation of microalgae or/and cyanobacteria in nutrient-rich wastewaters offers an opportunity for enhancing sustainability of tertiary wastewater treatment processes via resources/energy recovery/production, mitigation of emitted GHGs and provision of added value products. However, maintaining a monoculture in wastewater-media constitutes a significant challenge to be addressed. In this regard, the present work assesses the efficiency of the low-cost wastewater substrate disinfection techniques of filtration, use of NaClO, H2O2 or Fe(VI), as a preliminary treatment stage upstream a cyanoba

A 3.3 kb HindIII restriction-digest DNA fragment was isolated from a Synechocystis sp. strain PCC6803 subgenomic plasmid library which strongly hybridized to a 349 bp fragment of the icfA (ccaA) gene from Synechococcus sp. strain PCC7942. DNA sequence analysis of the fragment revealed three open reading frames (ORFs), two of which potentially coded for pantothenate synthetase (ORF275) and cytidylate kinase (ORF230). The third, ORF274, was 825 bp in length, encoding a deduced polypeptide of 274 aa (Mr, 30747) that bears 55% sequence identity to the Synechococcus icfA (ccaA) translation product,

Strains of the picocyanobacteria genera Synechococcus and Cyanobium occur in cold-temperate zones, from coastal waters to the open ocean, spanning a range of salinities and water depths. To thrive across different salinities, picocyanobacteria strains acclimate internal turgor by expending metabolic energy to accumulate compatible solutes and extrude inorganic ions. We grew five picocyanobacterial strains, derived from a range of source habitat salinities, under a matrix of light and salinity levels. Although chlorophyll is a general proxy for phytoplankton biomass, Chl/cell is itself a key co

Light-driven microbial communities consisting of phototrophs and heterotrophs represent an emerging frontier for biochemicals production from carbon dioxide (CO2). However, the construction of stable and robust light-driven artificial microbial communities remains challenging because the dominant strain wins the competition for nutrient and leads to the instability of subpopulations. Inspired by natural ecosystems, one promising approach to assemble stable consortia is to construct spatial niches partitioning subpopulations-that is, physically separating different microbial members into distin

Sidestreams in wastewater treatment plants can serve as concentrated sources of nutrients (i.e., nitrogen and phosphorus) to support the growth of photosynthetic organisms that ultimately serve as feedstock for production of fuels and chemicals. However, other chemical characteristics of these streams may inhibit growth in unanticipated ways. Here, we evaluated the use of liquid recovered from municipal anaerobic digesters via gravity belt filtration as a nutrient source for growing the cyanobacterium Synechococcus sp. strain PCC 7002. The gravity belt filtrate (GBF) contained high levels of c

Immunotherapy has shed light on clinical oncology for several decades. However, it still suffers from heterogeneous therapeutic outcomes and potential immune-related adverse events (irAEs). Inspired by a natural phenomenon that microalgae can absorb and accumulate selenium (Se), a well-known healthcare element, we engineered a Se-enriched cyanobacterium Synechococcus Sp (Se-Ssp) for advanced immunotherapy. Simply by culturing in the presence of Na2SeO3, Se nanoparticles could be bio-fabricated within Synechococcus Sp, underscoring a facile preparation process with promising translational poten

This study investigates whether a single initial cyanobacterial microbiome, originating from a mixed community, retains its capacity for polyhydroxybutyrate (PHB) production under non-sterile long-term operation when subjected to cultivation conditions that promote different dominant cyanobacterial populations. Distinct selective pressures were applied in two photobioreactors: one subjected to high pH (>10) and nutrient limitation, which favored the dominance of Synechococcus sp., and one operating under controlled pH (8.0-8.25) and sufficient nutrient supplementation, which promoted the enric

Polyphosphate (polyP) is a conserved inorganic polymer traditionally viewed as a stress-induced phosphate and energy reserve. In cyanobacteria, however, polyP granules are constitutively present and are frequently observed in close proximity to carboxysomes, the bacterial microcompartments that mediate CO2 fixation. Here, we show that polyP functions as a spatially organized regulator of the photosynthetic cytoplasm in Synechococcus elongatus. PolyP granules localize to the nucleoid and are periodically arranged along the cell axis, independent of the McdAB carboxysome positioning system. Desp

Toxic and non-toxic cyanobacterial strains from Anabaena, Aphanizomenon, Calothrix, Cylindrospermum, Nostoc, Microcystis, Planktothrix (Oscillatoria agardhii), Oscillatoria and Synechococcus genera were examined by RFLP of PCR-amplified 16S rRNA genes and 16S rRNA gene sequencing. With both methods, high 16S rRNA gene similarity was found among planktic, anatoxin-a-producing Anabaena and non-toxic Aphanizomenon, microcystin-producing and non-toxic Microcystis, and microcystin-producing and non-toxic Planktothrix strains of different geographical origins. Understanding how tropical cyclones affect phytoplankton communities is important for studies on ecological variability. Most studies assessing the post-storm phytoplankton response rely on satellite observations of chlorophyll a concentration, which inform on the ocean surface conditions and the whole phytoplankton community. In this work, we assess the potential of the Massachusetts Institute of Technology marine ecosystem model to account for the response of individual phytoplankton functional types (PFTs, coccolithophores, diatoms, diazotrophs, mixotrophic dinoflagellates, picoeukaryotes,

Microbial consortia combining phototrophic and heterotrophic partners offer a sustainable and cost-effective strategy for producing polyhydroxyalkanoates, which have attracted considerable attention as promising alternatives to petroleum-derived plastics. Here, a synthetic cyanobacteria-heterotroph consortium composed of Synechococcus elongatus and Priestia aryabhattai was employed to produce polyhydroxybutyrate (PHB) directly from CO2. Transcriptomic analysis of the heterotrophic partner revealed the sucrose-to-PHB biosynthesis pathway and key gene cluster. Optimal coculture conditions were e

UNLABELLED: Cyanobacteria are photosynthetic organisms that play important roles in carbon cycling and are promising bioproduction chassis. Here, we isolate two novel cyanobacteria with 4.6Mbp genomes, UTEX 3221 and UTEX 3222, from a unique marine environment with naturally elevated CO₂. We describe complete genome sequences for both isolates and, focusing on UTEX 3222 due to its planktonic growth in liquid, characterize biotechnologically relevant growth and biomass characteristics. UTEX 3222 outpaces other fast-growing model strains on a solid medium. It can double every 2.35 hours in a liqu

Quantification of cyanobacterial CO2 fixation rates is vital to determining their potential as industrial strains in a circular bioeconomy. Currently, however, CO2 fixation rates are most often determined through indirect and/or low-resolution methods, resulting in an incomplete picture of both dynamic behaviors and total carbon fixation potential. To address this, we developed the "Automated Carbon and CO2 Experimental Sampling System" (ACCESS); a low-cost system for in situ off-gas analysis that supports the automated acquisition of high-resolution volumetric CO2 uptake rates from multiple c

The ever-increasing number of available microbial genomes and metagenomes provides new opportunities to investigate the links between niche partitioning and genome evolution in the ocean, especially for the abundant and ubiquitous marine picocyanobacteria Prochlorococcus and Synechococcus. Here, by combining metagenome analyses of the Tara Oceans dataset with comparative genomics, including phyletic patterns and genomic context of individual genes from 256 reference genomes, we show that picocyanobacterial communities thriving in different niches possess distinct gene repertoires. We also iden

Direct conversion of carbon dioxide to valuable compounds is a desirable way to reduce the environmental burden and switch from fossil to renewable fuels. Cyanobacteria are photosynthetic bacteria that perform oxygenic photosynthesis and are able to produce valuable compounds from carbon dioxide in the air. Synechocystis and Synechococcus species, model unicellular cyanobacteria, can produce succinate and lactate, which are commodity chemicals used to generate bioplastics. Several cyanobacteria are also able to produce polyhydroxybutyrate, a biodegradable polyester that accumulates under nitro
