Global gene expression profiles of the cyanobacterium Synechocystis sp. strain PCC 6803 in response to irradiation with UV-B and white light.

We developed a transcript profiling methodology to elucidate expression patterns of the cyanobacterium Synechocystis sp. strain PCC 6803 and used the technology to investigate changes in gene expression caused by irradiation with either intermediate-wavelength UV light (UV-B) or high-intensity white light. Several families of transcripts were altered by UV-B treatment, including mRNAs specifying proteins involved in light harvesting, photosynthesis, photoprotection, and the heat shock response. In addition, UV-B light induced the stringent response in Synechocystis, as indicated by the repress

Apparent redistribution of electron flow between redox cofactors branches in photosystem I from the menB variants of cyanobacterium Synechocystis sp. PCC 6803.

Interruption of the menB gene in cyanobacterium Synechocystis sp. PCC 6803 results in replacement of the native phylloquinones at the A1 sites of photosystem I (PS I) by plastoquinone-9 (PQ) and a slowdown in the rate of forward electron transfer (ET) from plastosemiquinone to 4Fe4S clusters by a factor of ~ 1000 compared with the wild type (WT) (Semenov et al. 2000). We used PS I samples from two menB variants containing either loosely bound PQ (menB-PQL) or its tightly bound analogue (menB-PQT). Flash-induced optical changes were used to characterize both menB-PQL and menB-PQT samples. Photo

The cAMP receptor protein, SyCRP1 acts as a transcriptional repressor of CO2-concentrating mechanism genes at high inorganic carbon levels in Synechocystis PCC 6803.

Cyanobacteria utilize a CO2-concentrating mechanism (CCM) to enhance photosynthetic efficiency by accumulating CO2 around RuBisCO, a process crucial for adapting to fluctuating environmental CO2 levels. While the upregulation of CCM genes under low inorganic carbon (Ci) conditions is known, the precise Ci sensing and regulatory mechanisms governing CCM gene expression remain incompletely understood. We show that a membrane-bound SyCRP1 senses high Ci levels through cAMP binding to its low- and high-affinity sites. We demonstrate its interaction with membrane lipids and liposomes, and a subsequ

Exposure of Synechocystis 6803 cells to series of single turnover flashes increases the psbA transcript level by activating transcription and down-regulating psbA mRNA degradation.

Exposure of Synechocystis sp. PCC 6803 cells to series of single turnover flashes increases specifically the level of psbA and psbD2 messages, encoding the D1 and D2 proteins of photosystem II, as compared to light exposed cells. This increase is due to maintenance the transcription rate as high as in growth light and to the down-regulation of transcript degradation as in darkness. Inhibition of the plastoquinone pool reduction by DCMU or its oxidation by DBMIB does not diminish the transcription of the psbA gene under growth conditions. However, the degradation rate of psbA transcript, as wel

Expression of the psbA gene during photoinhibition and recovery in Synechocystis PCC 6714: inhibition and damage of transcriptional and translational machinery prevent the restoration of photosystem II activity.

The D1 reaction center protein of the photosystem II complex is very sensitive to light. It is continuously being damaged, degraded and resynthesized. Under high light, photosystem II inactivation is observed. This is because the rate of D1 damage is faster than that of its replacement. This process can be reversed if exposure to high light is not too long. In this work we study the changes that occur in the transcriptional and translational machinery that could lead to irreversible photoinhibition in Synechocystis PCC 6714. In the first minutes of photoinhibition, high light induced an accumu

Exploring media optimization and extraction methods to enhance poly-β-hydroxybutyrate (PHB) yield in Synechocystis pevaleikii: Scale-up from shake flask to 10 L photobioreactor.

The surge in demand for sustainable alternatives to petroleum-based plastics has redirected attention towards microbial biopolymers, such as Poly-β-hydroxybutyrate (PHB). This study identified the cyanobacterium Synechocystis pevaleikii as an unexplored potential organism for PHB production. Cultivation was performed in BG-11 media with varying exogenous acetate concentrations (0.08-2.46 g/L). The highest chlorophyll-a content (9.12 ± 0.44 μg/mL), biomass yield (600 ± 17 mg/L), and PHB accumulation (3.01 % w/w) were achieved with 1.64 g/L acetate. Effects of inactivation of the cyAbrB2 transcription factor together with glycogen synthesis on cellular metabolism and free fatty acid production in the cyanobacterium Synechocystis sp. PCC 6803.

Deletion of the cyAbrB2 (Sll0822) transcription factor in Synechocystis sp. PCC 6803 causes aberrant accumulation of glycogen. We previously tried to redirect the excess carbon stored as glycogen in the cyabrB2-disrupted (∆ cyabrB2) mutant by knockout of the glgC (slr1176) gene encoding glucose-1-phosphate adenylyltransferase. However, complete knockout could not be attained, suggesting that accumulation of glycogen is essential for the Δ cyabrB2 mutant. In this study, we introduced the cyabrB2 gene fused to the copper-inducible petE promoter into the ∆ cyabrB2 mutant. After complete knockout

Influence of the unusual covalent adduct on the kinetics and formation of radical intermediates in synechocystis catalase peroxidase: a stopped-flow and EPR characterization of the MET275, TYR249, and ARG439 variants.

Catalase-peroxidases (KatGs) are heme peroxidases with a catalatic activity comparable to monofunctional catalases. They contain an unusual covalent distal side adduct with the side chains of Trp(122), Tyr(249), and Met(275) (Synechocysis KatG numbering). The known crystal structures suggest that Tyr(249) and Met(275) could be within hydrogen-bonding distance to Arg(439). To investigate the role of this peculiar adduct, the variants Y249F, M275I, R439A, and R439N were investigated by electronic absorption, steady-state and transient-state kinetic techniques and EPR spectroscopy combined with d

Co-expression analysis, proteomic and metabolomic study on the impact of a Deg/HtrA protease triple mutant in Synechocystis sp. PCC 6803 exposed to temperature and high light stress.

Members of the DegP/HtrA protease family are widespread in nature and play an important role in proteolysis of misfolded and damaged proteins. The cyanobacterium Synechocystis sp. PCC 6803 contains three Deg proteases, HhoA (Sll1679), HhoB (Sll1427) and HtrA (Slr1204). Using the proteomic or metabolomic approach we investigated a triple deletion mutant (Δdeg) exposed to light or temperature stress. To cope with the stress conditions the triple mutant reduces its energy metabolism and stress-related proteins are induced to protect the cells. Additionally the co-expression of the genes encoding

Chemicals Affecting Cyanobacterial Poly(3-hydroxybutyrate) Accumulation: 2-Phenylethanol Treatment Combined with Nitrogen Deprivation Synergistically Enhanced Poly(3-hydroxybutyrate) Storage in Synechocystis sp. PCC6803 and Anabaena sp. TISTR8076.

Various photoautotrophic cyanobacteria increase the accumulation of bioplastic poly(3-hydroxybutyrate) (PHB) under nitrogen deprivation (-N) for energy storage. Several metabolic engineering enhanced cyanobacterial PHB accumulation, but these strategies are not applicable in non-gene-transformable strains. Alternatively, stimulating PHB levels by chemical exposure is desirable because it might be applied to various cyanobacterial strains. However, the study of such chemicals is still limited. Here, 19 compounds previously reported to affect bacterial cellular processes were evaluated for their

The dspA gene product of the cyanobacterium Synechocystis sp. strain PCC 6803 influences sensitivity to chemically different growth inhibitors and has amino acid similarity to histidine protein kinases.

We have cloned and sequenced a gene of the cyanobacterium Synechocystis sp. strain PCC 6803 named dspA (encoding drug sensory protein A; DspA), mutations in which result in cross-resistance to the herbicides difunon and diuron, as well as to the calmodulin antagonists chlorpromazine and trifluoperazine. The dspA gene encodes a polypeptide of 663 amino acids with a predicted molecular mass of 74.5 kDa. The molecular nature of two mutations in the dspA gene leading to the cross-resistance has been determined. Targeted mutagenesis of the dspA gene was performed using a kanamycin-resistance gene c

Purification and characterization of a hydroperoxidase from the cyanobacterium Synechocystis PCC 6803: identification of its gene by peptide mass mapping using matrix assisted laser desorption ionization time-of-flight mass spectrometry.

A cytosolic catalase-peroxidase from the cyanobacterium Synechocystis PCC 6803 was purified to homogeneity by a six-step purification procedure. It is a homodimeric enzyme with a subunit molecular mass of 85 kDa. The isoelectric point of the protein is at pH 5.5; Michaelis constant, turnover number, and catalytic efficiency of the catalase activity for H2O2 were measured to be 4.8 mM, 3450 s-1, and 7.2 x 10(5) M-1 s-1, respectively. Preparation and spectroscopy of the pyridine ferrohemochrome identified an iron protoporphyrin IX as the prosthetic group. The enzyme was shown to exhibit both cat

Spectroscopic evidence from site-directed mutants of Synechocystis PCC6803 in favor of a close interaction between histidine 189 and redox-active tyrosine 160, both of polypeptide D2 of the photosystem II reaction center.

The reaction center of photosystem II of oxygenic photosynthesis contains two redox-active tyrosines called Z and D, each of which can act as an electron donor to the oxidized primary electron donor, P680+. These tyrosines are located in homologous positions on the third transmembrane alpha-helix of each of the two homologous polypeptides, D1 and D2, that comprise the reaction center. Tyrosine D of polypeptide D2 has been proposed, upon oxidation, to give up its phenolic proton to a nearby basic amino acid residue, forming a neutral radical. Modeling studies have pointed to His190 (spinach num

A Cyanobacterial Screening Platform for Rubisco Mutant Variants.

Rubisco is the main entry point of inorganic carbon into the biosphere and a central player in the global carbon system. The relatively low specific activity and tendency to accept O2 as a substrate have made Rubisco an attractive but challenging target for enzyme engineering. We have developed an enzyme engineering and screening platform for Rubisco using the model cyanobacterium Synechocystis sp. PCC 6803. Starting with the Form II Rubisco from Gallionella, we first show that the enzyme can replace the native Form I Rubisco in Synechocystis and that growth rates become sensitive to CO2 and O

Synthetic consortia towards photosynthetically derived acetate for heterotrophic production of 1-butanol.

Synthetic consortia is an emerging field in biotechnology, offering advantages such as division of labor between the individual members and reduced risk of contamination. Consortia combining phototrophic and heterotrophic bacteria are attractive, as phototrophs can utilize CO₂ and light energy to sustain growth. Several studies have explored such systems where sucrose is most commonly used as the provided carbon source. An alternative is acetate, a smaller two-carbon molecule produced as a by-product by many microorganisms, including the model cyanobacterium Synechocystis PCC 6803 (thereafter

dCas12a-mediated CRISPR interference for multiplex gene repression in cyanobacteria for enhanced isobutanol and 3-methyl-1-butanol production.

BACKGROUND: Cyanobacteria of the genera Synechocystis and Synechococcus have emerged as promising platforms for metabolic engineering endeavors aimed at converting carbon dioxide into valuable fuels and chemicals, thus addressing the pressing energy demand and mitigating global climate change. Notably, Synechocystis sp. strain PCC 6803 (Synechocystis) has been engineered to produce isobutanol (IB) and 3-methyl-1-butanol (3M1B) via heterologous expression of α-ketoisovalerate decarboxylase (Kivd). Despite these advances, the achieved IB/3M1B titers remain low. CRISPR interference (CRISPRi), an

Two cyanobacterial response regulators with diguanylate cyclase activity, Rre2 and Rre8, participate in biofilm formation.

Phototrophic bacteria face diurnal variations of environmental conditions such as light and osmolarity that affect their carbon metabolism and ability to generate organic compounds. The model cyanobacterium, Synechocystis sp. PCC 6803 forms a biofilm when it encounters extreme conditions like high salt stress, but the molecular mechanisms involved in perception of environmental changes that lead to biofilm formation are unknown. Here, we studied two two-component regulatory systems (TCSs) that contain diguanylate cyclases (DGCs), which produce the second messenger c-di-GMP, as potential compon

Adaptation strategies of cyanobacteria under trace tetracycline exposure: A comparative study of toxigenic and non-toxigenic strains.

Trace antibiotics exert unclear ecological impacts on cyanobacteria, especially the differential responses between toxigenic and non-toxigenic strains. To address this, we established a paired-strain comparative framework exposing toxigenic Microcystis aeruginosa and non-toxigenic Synechocystis sp. to environmentally relevant tetracycline (0.2, 2, 20 μg/L) for 14 days. Results showed that TC induced hormetic growth in both strains, with final cell density increases of 2.47-9.79% (M. aeruginosa) and 4.98-8.59% (Synechocystis sp.) relative to controls, but exhibited divergent adaptation strategi

Recycling nutrients from greenhouse effluent with a stress-resistant cyanobacterial strain.

Hydroponic agriculture is being increasingly utilized for its high productivity. Current wastewater treatment methods lead to loss of phosphate and nitrate, so a future circular economy calls for better methods to treat soilless systems' effluents, allowing reuse of nutrients. Cyanobacteria can use wastewaters as cultivation media and nutrient sources, atmospheric or anthropogenic CO2 as a carbon source and artificial or sunlight for energy, while producing biomass and valuable products. This proof-of-concept study tests, at a laboratory scale, if the cyanobacterium Synechocystis sp. PCC 6803
