INTRODUCTION: Infection-induced orchitis, a leading cause of acquired male infertility affecting 8%-12% of couples globally, is driven by unresolved inflammatory responses following bacterial infection.
METHODS: We employed uropathogenic Escherichia coli (UPEC)- and lipopolysaccharide (LPS)-induced orchitis models to define the mechanisms underlying testicular inflammation. We interrogated the cellular sources of CXCL9/CXCL10 and assessed macrophage-driven inflammatory cell recruitment and spermatogenic disruption. Mechanistic studies were focused on lysine lactylation, STAT1 protein stabilit

Systemic lupus erythematosus (SLE) is a chronic autoimmune disease with diverse manifestations, including rash, arthritis, and nephritis. Although autoantibodies are a key feature of SLE, their levels often poorly reflect disease severity, suggesting the involvement of additional contributing factors. The tripartite motif-containing protein (TRIM) family, which regulates immunity, may play a role in SLE. Although TRIM21 is a known autoantigen, the roles of other TRIM family members remain unclear. The present study examined TRIM expression in SLE and evaluated the potential of TRIM proteins as

OBJECTIVE: Autoantibodies reactive with Ro52 (tripartite motif-containing protein 21 [TRIM21]) are detected in 70% of patients with primary Sjögren's syndrome (SS). TRIM21 belongs to a 34-member C-IV family of TRIM proteins. Although autoantibodies against other TRIM proteins within the C-IV family have been detected in the sera of patients with primary SS, their clinical relevance remains unclear. This study was undertaken to investigate the frequency of anti-TRIM38 in patients with primary SS and evaluate its association with various clinical measures of the disease.
METHODS: Serum samples

Heart failure (HF) persists as the primary cause of death among patients recovering from acute myocardial infarction (AMI). Protein ubiquitination has been implicated as a key modulator of HF pathogenesis, yet the role of ubiquitination in the Aldh2 rs671 mutant - the most common single-nucleotide variant in human populations - remains poorly understood. We discovered TRIM21 as a previously unrecognized E3 ubiquitin ligase for the ALDH2 rs671 mutant and elucidated its mechanistic involvement in HF progression. Using Aldh2 BM chimeric mice to model AMI, we observed that WT mice transplanted wit

BACKGROUND: Japanese encephalitis virus (JEV) infection leads to Japanese encephalitis (JE) in humans. JEV is transmitted through mosquitoes and maintained in a zoonotic cycle. This cycle involves pigs as the major reservoir, water birds as carriers and mosquitoes as vectors. JEV invasion into the central nervous system (CNS) may occur via antipodal transport of virions or through the vascular endothelial cells. Microglial cells get activated in response to pathogenic insults. JEV infection induces the innate immune response and triggers the production of type I interferons. The signaling path

Mitophagy is a major quality control pathway that removes unwanted or dysfunctional mitochondria and plays an essential role in vascular health. Here we show that MCM8 expression is significantly decreased in children with Kawasaki disease (KD) who developed coronary artery aneurysms. Mechanistically, we discovered that nitric oxide signaling promotes TRIM21-mediated MCM8 ubiquitination, which disrupts its interaction with MCM9 and promotes its cytosolic export. In the cytosol, MCM8 relocates to the mitochondria pore-forming proteins and promotes their ubiquitination by TRIM21. In addition, MC

Extracellular vesicles (EVs) have gained prominence as advanced drug delivery systems due to their inherent merits. Recent advancements highlight their utility in transporting therapeutic proteins with significant progress in targeted therapies. To optimize EV efficacy, engineering strategies focus on enhancing cargo encapsulation efficiency. The robust interaction between immunoglobulin G Fc fragments and the cytoplasmic receptor TRIM21 offers a novel framework for stable cargo loading. This study identifies Ras-related GTP-binding protein D (RRAGD) as a critical regulator of lysosomal biogen

BACKGROUND: Interstitial cystitis/bladder pain syndrome (IC/BPS) has become a pressing clinical issue due to its unclear etiology and severe, persistent pelvic pain. Despite extensive research, the pathogenesis of IC/BPS remains unresolved, and current treatments primarily target symptom relief rather than addressing underlying disease mechanisms. This study aimed to investigate the effects of nuclear factor erythroid 2-related factor 2 (NRF2) on IC/BPS and the potential molecular mechanisms.
METHODS: Bladder mucosal biopsies from IC/BPS patients were subjected to RT-qPCR and immunoblotting t

Lung adenocarcinoma (LUAD) remains a leading cause of cancer-related mortality. Integrative multi-omics and machine-learning analyses across independent cohorts identified CYYR1 as a frequently downregulated transmembrane protein in LUAD, with low expression associated with poor prognosis and promoter hypermethylation. CYYR1 restoration suppressed LUAD cell proliferation, migration, invasion, tumor growth, and metastasis. Mechanistically, CYYR1 facilitated TRIM21-dependent ubiquitination and proteasomal degradation of HMMR, thereby reducing CDC20-associated oncogenic signaling. Biochemical and

BACKGROUND: Olfactomedin-like protein 3 (OLFML3) is a secreted glycoprotein that is primarily deemed to be associated with embryonic development, angiogenesis, and tumorigenesis. Recent studies have highlighted the function of OLFML3 as an important regulatory protein in viral and bacterial infections. This study explores the role of OLFML3 in type I interferon (IFN-I) signaling in RNA virus infection and related mechanism of action.
METHODS: RT-qPCR was used to determine the effects of OLFML3 on IFN-I production in the RNA virus infection. Olfml3 -/- mice was used to evaluate the effects of

BACKGROUND: Hepatitis B virus (HBV) infection is a major health problem with hundreds of millions of people still chronically infected worldwide. Although it is known that cytokines can inhibit HBV replication in infected hepatocytes, much is still unknown about the underlying mechanisms or mediators.
METHODS: In this study, we systematically analyzed tumor necrosis factor-alpha (TNF-α) and interferon-gamma (IFN-γ) induced proteins by proteomic analysis and identified interferon-inducible protein 35 (IFI35) as a novel host restriction factor for HBV replication.
RESULTS: Overexpression of IF

Antiretroviral therapy (ART) strategies against HIV-1 have been successful in suppressing HIV-1 replication and preventing disease progression. However, drug-resistance development, side-effects and life-long adherence of current drugs have driven the development of novel inhibitors against HIV-1. Inhibition of capsid production by targeting the highly conserved capsid protein (CA) has shown to be promising in blocking viral replication. Here, we developed a capsid-targeting biological composed of a single-domain Llama VHH that binds CA, fused to human IgG1 Fc to activate TRIM21-mediated degra

Rationale: Cardiac hypertrophy is an important pathological basis for heart failure. Most physiological activities of cardiomyocytes are regulated by proteins and their post-translational modification. Deubiquitinating enzymes (DUBs) are involved in protein stability maintenance and closely related to myocardial hypertrophy. In this study, we aimed to clarify the regulatory role of a DUB, ubiquitin-specific peptidase 28 (USP28), in cardiac hypertrophy and explore the molecular mechanism behind. Methods: Transcriptome and single-cell mRNA sequencing was used to demonstrate the association of US

Microtubule-associated protein 1A/1B-light chain 3 (LC3)-associated phagocytosis (LAP) plays a critical role in host defense against invading pathogens, including Listeria monocytogenes (Listeria. monocytogenes), Salmonella typhimurium (S. typhimurium), and Francisella novicida (F. novicida). However, the precise regulatory mechanisms controlling LAP remain poorly understood. Here, we identify enkurin domain-containing protein 1 (ENKD1) as a key negative regulator of LAP during infection with these pathogens. Macrophages infected with L. monocytogenes (10403S), S. typhimurium (ATCC14028), or F

RNA-dependent RNA polymerases (RdRps) play a key role in the life cycle of RNA viruses and impact their immunobiology. The arenavirus lymphocytic choriomeningitis virus (LCMV) strain Clone 13 provides a benchmark model for studying chronic infection. A major genetic determinant for its ability to persist maps to a single amino acid exchange in the viral L protein, which exhibits RdRp activity, yet its functional consequences remain elusive. To unravel the L protein interactions with the host proteome, we engineered infectious L protein-tagged LCMV virions by reverse genetics. A subsequent mass

OBJECTIVE: Multisystem inflammatory syndrome in children (MIS-C) is a rare hyperinflammatory syndrome that follows SARS-CoV-2 infection. Prior plasma proteomic analysis from a 2020 cohort of patients with MIS-C at our center revealed a profile characterized by thrombotic microangiopathy (TMA), macrophage activation syndrome (MAS)-associated proteins, and dysregulated interferon-γ (IFNγ) responses. However, a limitation of that study was that samples were often acquired after treatment. The objective of this study was to identify plasma proteomic signatures that uniquely define MIS-C versus oth

Bortezomib resistance in multiple myeloma (MM) is a significant clinical challenge that limits the long-term effectiveness. Currently, there is a lack of reliable biomarkers to predict bortezomib resistance. Previous studies reported that several proteins regulate bortezomib resistance through targeting ubiquitin-proteasome pathways, including heat shock protein family A member 9 (HSPA9), dickkopf Wnt signaling pathway inhibitor 1 (DKK1), proteasome 26S subunit non-ATPase 14 (PSMD14), and tripartite motif containing 21 (TRIM21). In our study, we aimed to analyze the expression of these protein

The mechanics of the cellular microenvironment continuously modulates cell functions such as growth, survival, apoptosis, differentiation and morphogenesis via cytoskeletal remodelling and actomyosin contractility1-3. Although all of these processes consume energy4,5, it is unknown whether and how cells adapt their metabolic activity to variable mechanical cues. Here we report that the transfer of human bronchial epithelial cells from stiff to soft substrates causes a downregulation of glycolysis via proteasomal degradation of the rate-limiting metabolic enzyme phosphofructokinase (PFK).
