Brepocitinib, a selective TYK2/JAK1 inhibitor, mitigates neutrophilic inflammation and glucocorticoid receptor-β expression in COPD.

Chronic obstructive pulmonary disease (COPD) is a progressive respiratory disorder characterized by neutrophil-dominant, corticosteroid-refractory airway inflammation involving the IL-23/IL-17A axis. IL-23 primarily activates the Janus kinase/signal transducer and activator of transcription (JAK-STAT) pathway through TYK2 and JAK2, whereas IL-17A and other pro-inflammatory cytokines can activate JAK1. However, the contribution of these JAK-dependent pathways to neutrophil-driven inflammation in COPD remains incompletely understood. In this study, we investigated how the IL-23/IL-17A axis modul

Successful Immune Reconstitution in a Patient with a TYK2 Deficiency after Allogeneic Stem Cell Transplantation from Unrelated Donors.

A boy with primary immunodeficiency, caused by a tyrosine kinase 2 (TYK2) mutation, presented with immune defects and a lifelong history of severe infections. Our aim was to determine whether allogeneic hematopoietic stem cell transplantation (HSCT) could restore the patient's immune defenses and reduce susceptibility to infection. In the absence of a suitable HLA-matched blood relative to act as a donor, the patient received an allogeneic HSCT from unrelated donors. The patient's clinical data were analyzed in the Children's Hospital of Chongqing Medical University (Chongqing, China) before t

Influence of TYK2 in systemic sclerosis susceptibility: a new locus in the IL-12 pathway.

OBJECTIVES: TYK2 is a common genetic risk factor for several autoimmune diseases. This gene encodes a protein kinase involved in interleukin 12 (IL-12) pathway, which is a well-known player in the pathogenesis of systemic sclerosis (SSc). Therefore, we aimed to assess the possible role of this locus in SSc. METHODS: This study comprised a total of 7103 patients with SSc and 12 220 healthy controls of European ancestry from Spain, USA, Germany, the Netherlands, Italy and the UK. Four TYK2 single-nucleotide polymorphisms (V362F (rs2304256), P1104A (rs34536443), I684S (rs12720356) and A928V (rs3

Activation of Tyk2 and Stat3 is required for the apoptotic actions of interferon-beta in primary pro-B cells.

The growth-inhibitory effects of type 1 interferons (IFNs) (IFNalpha/beta) are complex, and the role of apoptosis in their antigrowth effects is variable and not well understood. We have examined primary murine interleukin-7-dependent bone marrow-derived pro-B cells, where IFNbeta, but not IFNalpha, induces programmed cell death (PCD). IFNbeta-stimulated apoptosis is the same in pro-B cells derived from wild type and Stat1(-/-) mice. However, in pro-B cells from Tyk2(-/-) mice, where there is normal activation of Stat1 and Stat2, IFNbeta-stimulated PCD is not observed. Loss of B cells in lymph

Green Nail in a Patient With Psoriasis Receiving Deucravacitinib.

Green nail (GN) is typically caused by Pseudomonas aeruginosa and commonly occurs in patients with nail damage, nail psoriasis, or frequent exposure to moist environments. Deucravacitinib is an oral tyrosine kinase 2 (Tyk2) inhibitor effective for psoriasis treatment. Herein, we report a case of GN in a 72-year-old man following four months of treatment with deucravacitinib. Given the absence of pre-existing signs indicative of nail psoriasis prior to the initiation of deucravacitinib, deucravacitinib might have contributed to the development of GN. To the best of our knowledge, no previous ca

Deucravacitinib in moderate-to-severe psoriasis.

Psoriasis is a chronic inflammatory disease that affects up to 1 in 20 people worldwide. A patient's quality of life and health can be drastically affected by psoriasis. The number of therapies for patients with moderate to severe psoriasis has steadily grown over the past two decades, with biologic immunotherapies being the primary agents developed. However, new small-molecule oral therapies have lagged in development. Deucravacitinib is an oral small molecule that inhibits the activity of TYK2, a member of the JAK family. Deucravacitinib works by allosterically inhibiting TYK2, increasing th

Identification of a novel benzyloxy-terminated scaffold as a potent and selective TYK2 inhibitor for anti-inflammatory therapy.

In this study, we described three series of N-phenylpyrimidin-2-amine derivatives as selective TYK2 inhibitors. Systematic exploration of the structure-activity relationship through the introduction of an O-linker and the flexible benzyl substituent based on the reported non-selective JAKs inhibitor yt52 led to the discovery of the optimized derivative compound 29i. Compound 29i showed a potency on TYK2 with an IC50 value of 18 nM and exhibited more than >70-fold selectivity over JAK1/2/3 isoforms. Kinase panel screening, WB assays, and human peripheral blood mononuclear cell assays further va

Modulation of JAK/STAT signaling pathway in the ovine thymus and lymph node during early pregnancy.

There is a dynamic accommodation between the maternal immune system and conceptus, including the thymus and lymph nodes, during early pregnancy. Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathway is involved in early placentation. However, it was not fully understood that early pregnancy had effects on JAK/STAT signaling pathway in the thymus and lymph nodes of ewes. In this study, thymuses and lymph nodes of ewes were collected at day 16 of the estrous cycle (N16), and at days 13, 16, and 25 of gestation (G13, G16, and G25). Expressions of mRNA and prot

Janus kinase (JAK) inhibitors in the treatment of inflammatory and neoplastic diseases.

The Janus kinase (JAK) family of non-receptor protein-tyrosine kinases consists of JAK1, JAK2, JAK3, and TYK2 (tyrosine kinase-2). Each of these proteins contains a JAK homology pseudokinase (JH2) domain that regulates the adjacent protein kinase domain (JH1). JAK1/2 and TYK2 are ubiquitously expressed whereas JAK3 is found predominantly in hematopoietic cells. The Janus kinase family is regulated by numerous cytokines including interleukins, interferons, and hormones such as erythropoietin, thrombopoietin, and growth hormone. Ligand binding to cytokine and hormone receptors leads to the activ

Clinical genomic profiling identifies TYK2 mutation and overexpression in patients with neurofibromatosis type 1-associated malignant peripheral nerve sheath tumors.

BACKGROUND: Malignant peripheral nerve sheath tumors (MPNSTs) are aggressive sarcomas that arise at an estimated frequency of 8% to 13% in individuals with neurofibromatosis type 1 (NF1). Compared with their sporadic counterparts, NF1-associated MPNSTs (NF1-MPNSTs) develop in young adults, frequently recur (approximately 50% of cases), and carry a dismal prognosis. As such, most individuals affected with NF1-MPNSTs die within 5 years of diagnosis, despite surgical resection combined with radiotherapy and chemotherapy. METHODS: Clinical genomic profiling was performed using 1000 ng of DNA from

Non-receptor tyrosine kinase 2 reaches its lowest expression levels in human breast cancer during regional nodal metastasis.

Almost half of breast Ductal Carcinoma in situ are likely to remain non threatening in situ lesions with no invasion to the surrounding stroma and no metastases. The majority of focal disruptions in myoepithelial (ME) cell layers indicative of invasion onset were found to be overlying epithelial cell clusters with no or substantially reduced estrogen receptor α (ERα) expression. Here we report the down-regulation of tyrosine kinase-2 (TYK2) and up-regulation of strumpellin expression, among other proteins in ERα(-) cells located at disrupted ME layers compared to adjacent ERα(+) cells overlyin

The rs2304256 Polymorphism in TYK2 Gene Is Associated with Protection for Type 1 Diabetes Mellitus.

BACKGROUND: Tyrosine kinase 2 (TYK2) is a candidate gene for type 1 diabetes mellitus (T1DM) since it plays an important role in regulating apoptotic and pro-inflammatory pathways in pancreatic β-cells through modulation of the type I interferon signaling pathway. The rs2304256 single nucleotide polymorphism (SNP) in TYK2 gene has been associated with protection for different autoimmune diseases. However, to date, only two studies have evaluated the association between this SNP and T1DM, with discordant results. This study thus aimed to investigate the association between the TYK2 rs2304256 SN

Janus kinases in immune cell signaling.

The Janus family kinases (Jaks), Jak1, Jak2, Jak3, and Tyk2, form one subgroup of the non-receptor protein tyrosine kinases. They are involved in cell growth, survival, development, and differentiation of a variety of cells but are critically important for immune cells and hematopoietic cells. Data from experimental mice and clinical observations have unraveled multiple signaling events mediated by Jaks in innate and adaptive immunity. Deficiency of Jak3 or Tyk2 results in defined clinical disorders, which are also evident in mouse models. A striking phenotype associated with inactivating Jak3

JAK/STAT signaling pathway in the spleen and liver during early pregnancy in ewes.

Pregnancy induces adaptations in the maternal immune system, including the liver and spleen, in a gestation-dependent manner. The Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathway participates in immune tolerance at the maternal-fetal interface. Nevertheless, whether pregnancy affects the JAK/STAT signaling pathway in the liver and spleen of ewes has remained unclear. In this study, samples of ovine liver and spleen were collected at day 16 of the estrous cycle and at days 13, 16, 25, and 70 of gestation (n = 6 for each group). The expression of JAK/STA

Clearance of Cutaneous Lichen Planus with Deucravacitinib.

Lichen planus (LP) is an idiopathic, immune-mediated inflammatory disorder commonly characterized by pruritic, violaceous papules and plaques that often involve the extremities. Although a variety of topical and systemic therapies are used, no targeted treatments are currently approved by the US Food and Drug Administration for this indication, and management of refractory disease remains challenging. Tyrosine kinase 2 (TYK2) inhibitors selectively modulate several cytokine signaling pathways, including interleukin (IL)-12, IL-23/IL-17, and type I interferon signaling, which are implicated in

Pharmacological inhibition of tyrosine protein-kinase 2 reduces islet inflammation and delays type 1 diabetes onset in mice.

BACKGROUND: Tyrosine protein-kinase 2 (TYK2) mediates inflammatory signalling through multiple cytokines, including interferon-α (IFNα), interleukin (IL)-12, and IL-23. TYK2 missense mutations protect against type 1 diabetes (T1D), and inhibition of TYK2 shows promise in other autoimmune conditions. METHODS: We evaluated the effects of specific TYK2 inhibitors (TYK2is) in pre-clinical models of T1D, including human β cells, cadaveric islets, iPSC-derived islets, and mouse models. FINDINGS: In vitro studies showed that TYK2is prevented IFNα-induced β cell HLA class I up-regulation, endoplasmi

Progress on the Pharmacological Targeting of Janus Pseudokinases.

The Janus kinases (JAKs) are key components of the JAK-STAT signaling pathway and are involved in myriad physiological processes. Though they are the molecular targets of many FDA-approved drugs, these drugs manifest adverse effects due in part to their inhibition of the requisite JAK kinase activity. However, the JAKs uniquely possess an integrated pseudokinase domain (JH2) that regulates the adjacent kinase domain (JH1). The therapeutic targeting of JH2 domains has been less thoroughly explored and may present an avenue to modulate the JAKs without the adverse effects associated with targeti
