Taste bud cells have a limited lifespan and must be continuously replaced along with the papilla epithelium in which they reside. Previous work has shown that expression of leucine-rich G protein-coupled receptor 5 (Lgr5), a Wnt pathway agonist, serves as a marker of adult stem/progenitor cells for taste buds located in posterior tongue (circumvallate and foliate), but not anterior tongue (fungiform), taste papillae. However, the specific location/niche of the Lgr5-expressing cells supporting renewal and their phenotypic properties have not been fully explored. To address this, the genesis and

Tastants reach the tip of taste bud cells through taste pores which are openings in the epithelium. We found Sprr2a is selectively expressed in the upper layer of the epithelium surrounding taste buds in the circumvallate papilla (CV) where the epithelium is organized into taste pores. Sprr2a is a member of a small proline-rich protein family, which is suggested to be involved in the restitution/migration phase of epithelial wound healing. The expression of Sprr2a was restricted to the upper layer and largely segregated with Ptch1 expression that is restricted to the basal side of the epitheli

A hitherto undescribed cell type within the fungiform papillae of the tongue of the toad, Bufo bufo (L.) and the taste-disks of the soft palate of Bombina variegata L. was identified by means of scanning and transmission electron microscopy. This cell type is characterized by an apical protrusion carrying numerous microvilli. Bundles of filaments are present in the cytoplasm of the protrusion. Vesicles with electron lucent content and microtubules are present thoughout the cytoplasm of the cell.

Taste dysfunction is commonly associated with aging and accompanied by morphological and molecular alterations in the tongue epithelium. To investigate the mechanisms underlying this phenomenon, we utilized Klotho-deficient (Kl -/-) mice, a model of accelerated aging. The tongue epithelium of Kl -/- mice showed altered muscle organization and increased clear spaces within the muscle compartment. The minor salivary glands displayed acinar cell atrophy. Notably, compared with WT, Kl -/- mice presented a reduced taste bud diameter and decreased expression of key taste receptors, as well as type I

The epithelium of the mouse tongue and soft palate consists of at least three distinct epithelial cell populations: basal cells, keratinized cells organized into filiform and fungiform papillae, and taste receptor cells present in tight clusters known as taste buds in the fungiform and circumvallate papillae and soft palate. All three cell types develop from the simple epithelium of the embryonic tongue and palate, and are continually replaced in the adult by cell turnover. Previous studies using pulse-chase tritiated thymidine labeling in the adult mouse provided evidence for a high rate of c

The decrease of the dark cell content and increase of the relative content of the intermediate type cells were observed in the foliate papillae taste buds after denervation or application of colchicine to the nerve. These characteristics were compared with those in the intact animals. Similar changes were found on the contralateral side. Upon colchicine application to the nerve they were most pronounced on the homolateral side. The absence of destruction of the nerve fibers under similar experimental conditions suggests that the blockade of axonal transport may be responsible for the changes u

The localization of Mg++ activated adenosine triphosphatase (ATPase) activity in rat fungiform taste buds was demonstrated by electron microscopic histochemistry. Reaction product was found on the cell membrane of the taste cells, but not within their cytoplasm. Especially strong reaction occurred in the taste pore. Besides, enzymes were observed in axon-Schwann cell and axon-type I cell interspace, but not in the contacts of the nerve fiber and type II cell. The taste bud could well be distinguished from the surrounding epithelium by the existence of the reaction product. ATPase activity was

Microstructural LM and SEM studies of fungiform (Fu), vallate (Vp), and foliate papillae (Fo) were conducted on the entire dog tongue. To demonstrate tongue areas with particular sensitivity to taste stimuli, 3D reconstruction technique of serial histoslides of papillae in Amira™ software was used for the first time. Obtained data indicated the presence of caudally bent Fu's, in which the diameter and the number of taste buds (Tbs) per single papilla increased caudally, and amounted to 29. In Fu's distinguished four types of connective tissue cores with variable distribution of Tbs. Paired Vp'

Mouse esophagus is lined with a stratified epithelium, which is maintained by the constant renewal of unipotent progenitors. In this study, we profiled mouse esophagus by single-cell RNA sequencing and found taste buds specifically in the cervical segment of the esophagus. These taste buds have the same cellular composition as the ones from the tongue but express fewer taste receptor types. State-of-the-art transcriptional regulatory network analysis allowed the identification of specific transcription factors associated to the differentiation of immature progenitors into the three different t

Mutations in polycystin proteins PKD1 and TRPP2 lead to autosomal dominant polycystic kidney disease. These two proteins form a receptor-ion channel complex on primary cilia. PKD1 undergoes an autoproteolysis at the N terminal G-protein-coupled receptor proteolytic site (GPS), which is essential for the function of PKD1. Whether GPS cleavage happens in other PKD proteins and its functional consequence has remained elusive. Here we studied the GPS cleavage of PKD1L3, a protein that associates with TRPP3 in taste cells and may play a role in sour taste. Our results show that PKD1L3 also undergoe

This study aimed to achieve a better understanding of taste receptor cell development relative to endothelin receptor B (ETB) in circumvallate papillae (CVP). ETB localization was assessed by immunohistochemistry during tongue development of the mouse. Co-localization of ETB with taste receptor type III cell marker, Synaptosomal-Associated Protein 25 kDa (SNAP25), was evident in both the developing and adult CVP. ETB was strongly localized in the stromal core region. As development progressed, ETB became localized in the CVP mesenchyme and partially in the epithelium. ETB and SNAP25 co-localiz

Taste buds in the European catfish Silurus glanis were examined with electron microscopic lectin histochemistry. For detection of carbohydrate residues in sensory cells and adjacent epithelial cells, gold-, ferritin- and biotin-labeled lectins were used. A post-embedding procedure carried out on tissue sections embedded in LR-White was applied to differentiate between the sensory cells: The lectins from Helix pomatia (HPA) and Triticum vulgare (WGA) bound to N-acetyl-galactosamine and to N-acetylglucosamine residues occurring especially in vesicles of dark sensory cells. This indicates a secre

The effects of aging and zinc deficiencies on taste were studied. The soft palate of zinc-deficient old rats, prepared by feeding with a zinc-deficient diet, were studied by a light microscope and a scanning electron microscope, and comparative examination was made on the morphological changes in soft palate papillae and taste buds of young normal rats and control old rats. In the case of young normal rats, papillae similar to lingual fungiform papillae were observed on the soft palate; one taste bud per one papilla was observed. The papillae were concentrated in the central area of the soft p

Two lipid sensors, CD36 and GPR120, are crucial for the orosensory detection of fat taste and for mediating fat preference. However, the mechanism by which endogenous lipid (FAHFA) binds to CD36 to initiate intracellular signaling remains unexplained. Hence, the primary objective of this study is to investigate the binding mechanism of FAHFA to CD36 and its role in isolated mouse taste bud cells (mTBCs). The Schrodinger platform was used to assess the molecular dynamics of protein and ligand interactions, and an in vitro experiment was used to validate the findings. Based on the docking score

Previous studies on the cat's tongue focused on the anatomy of the tongue and on the lingual papillae micromorphology. According to new challenges in studies on gustatory papillae, we adopted a modern computer-aided method of 3D reconstruction of gustatory papillae from serial 2D cross-sections in the description of the taste bud system. The study in short-hair cats aimed for the first time in carnivores to analyze fungiform (Fu papillae) and vallate papillae (Vp papillae) with spatial visualization of connective tissue cores (CTCs) and the arrangement and exact number of taste buds (Tbs). Type III IP3 receptor (IP3R3) is one of the common critical calcium-signaling molecules for sweet, umami, and bitter signal transduction in taste cells, and the total IP3R3-expressing cell population represents all cells mediating these taste modalities in the taste buds. Although gustducin, a taste cell-specific G-protein, is also involved in sweet, umami, and bitter signal transduction, the expression of gustducin is restricted to different subsets of IP3R3-expressing cells by location in the tongue. Based on the expression patterns of gustducin and taste receptors in the tongue, the functio
