The role of taurine on skeletal muscle cell differentiation.

Taurine abundantly contained in the skeletal muscle has been considered as one of essential factors for the differentiation and growth of skeletal muscles. The previous studies in the taurine transporter knockout mice showed that deficiency of taurine content in the skeletal muscle caused incomplete muscular developments, morphological abnormalities, and exercise abilities. In fetal and neonatal periods, taurine must be an essential amino acid due to no biosynthesis capacity, and therefore, taurine should be endogenously supplied through placenta and maternal milk. In general cell culture cond

Effect of taurine on alcoholic liver disease in rats.

To investigate the effect of taurine on alcoholic liver disease in rats, male Wistar rats were administered alcohol intragastrically for 3 months. The effect of beta-alanine-mediated taurine depletion and taurine administration on the development of alcoholic liver disease was examined. It was found that taurine administration produced lower levels of aspartate aminotransferase and alkaline aminotransferase than that of the untreated group. In addition, the levels of hepatic total protein, glutathione and superoxide dismutase were higher in the taurine treated groups than those in the untreate

Interaction between the actions of taurine and angiotensin II.

The amino acid, taurine, is an important nutrient found in very high concentration in excitable tissue. Cellular depletion of taurine has been linked to developmental defects, retinal damage, immunodeficiency, impaired cellular growth and the development of a cardiomyopathy. These findings have encouraged the use of taurine in infant formula, nutritional supplements and energy promoting drinks. Nonetheless, the use of taurine as a drug to treat specific diseases has been limited. One disease that responds favorably to taurine therapy is congestive heart failure. In this review, we discuss thre

[Taurine--recent views and importance for artificial feeding].

Taurine is an important age- and compartment-specific part of the pool of free amino acids in all human tissues and fluids examined. Besides an active taurine transport system, the mechanisms to assure compartment-specific taurine-homeostasis in humans is presumably due to a compartment-specific taurine synthesis. By the identity of cysteine-sulfinic acid - and glutamate-oxalate transaminase as well as of cysteine-sulfinic acid - and glutamic acid decarboxylase taurine biosynthesis via cysteine-sulfinic acid shows direct metabolic relationship between biochemical pathway of cysteine-sulfinic a

Correlation of platelet taurine levels with thyroid function.

Changes in neurotransmitter or neuromodular activity and levels have been thought to be responsible for some of the behavioral characteristics observed in patients with thyroid disease. To investigate these relationships, taurine, a proposed inhibitory neurotransmitter or modulator, was measured in platelets, which may serve as biochemical model for synaptosomes. Blood of euthyroid, hypothyroid and hyperthyroid patients was collected. Thyroxine was determined by RIA. Platelets were separated using differential centrifugation techniques. The taurine in platelets was separated by ion exchange ch

Taurine protects against bilirubin-induced neurotoxicity in vitro.

Kernicterus is a bilirubin-induced encephalopathy in newborn. Its spectrum ranges from subtle extrapyramidal to acute encephalopathy and chronic posticteric sequelae. Current treatment of this serious problem is far from optimal. Taurine has been documented to have protective effects on neuronal cells against ischemia in vivo and in vitro. This study used primary neuronal culture to investigate the toxicological effects of unconjugated bilirubin (UCB) and the protection of taurine against UCB-mediated neuron damage. Dose-dependent reduction of cell viability was found. Changes in neurite outgr

Cadmium-induced neurological disorders: prophylactic role of taurine.

The present study was conducted to investigate whether the conditionally essential amino acid taurine could play any protective role against the potent neurotoxin cadmium (Cd)-induced oxidative impairment in mice brain. Cd administration in the form of CdCl(2 )(at a dose of 4 mg kg(-1) body weight for 3 days, orally) increased the intracellular accumulation of metallic Cd, reactive oxygen species and super oxide radicals. The toxin also augmented the extent of lipid peroxidation, protein carbonylation and the levels of glutathione disulfide. Activities of the antioxidant enzymes and the levels

Tritiated taurine handling by isolated rat pancreatic islets.

A gating of volume-sensitive anion channels may participate in the depolarization of the plasma membrane caused by high concentrations of D-glucose in insulin-producing B-cells of the endocrine pancreas. The efflux of tritiated taurine from prelabeled cells is currently used to assess changes in the activity of such channels. The handling of [1,2-3H]taurine by isolated rat pancreatic islets was therefore investigated. The net uptake of [1,2-3H]taurine was found to represent a concentration-, time-, and temperature-dependent process. It was progressively increased in the range of D-glucose conc

Natural polyphenol-taurine-based nanoparticles for oral ulcer therapy.

Oral ulcers are the most common disease of the oral mucosa, and the pathogenesis is caused by a local overload of reactive oxygen species (ROS) that precipitates tissue breakdown, which, in turn, leads to the formation of ulcers. Here, we report a one-step oxidative copolymerization strategy to synthesize pH/ROS dual-responsive protocatechualdehyde taurine nanoparticles (PAT NPs) by cross-linking protocatechualdehyde and taurine (Tau). These PAT NPs combine excellent biocompatibility and multiple functions, such as efficient scavenging of free radicals, neutralization of lipopolysaccharide, pr

Combined metformin and Taurine attenuate age-related bone loss.

BACKGROUND: Age-related bone loss is closely associated with functional decline and exhaustion of bone marrow mesenchymal stem cells (BMSCs). This study aimed to investigate whether combined Metformin and Taurine (Met + Ta) treatment could attenuate aging-associated skeletal deterioration and preserve BMSC function. METHODS: Eighteen-month-old male C57BL/6J mice were treated with Met, Ta, or Met + Ta for 8 consecutive weeks. Bone microarchitecture was evaluated by micro-computed tomography and histological staining. BMSCs were isolated from aged mice and assessed for osteogenic differentiatio

Taurine and Metabolic Disorders: From Mechanisms to Clinical Implications.

BACKGROUND: Taurine (2-aminoethanesulfonic acid) is a conditionally essential, sulfur-containing amino acid that has attracted growing interest for its multifaceted role in metabolic regulation. Unlike proteinogenic amino acids, taurine exerts diverse physiological functions, including bile acid conjugation, osmoregulation, antioxidant defense, and mitochondrial support. Emerging evidence indicates that taurine modulates key metabolic processes implicated in obesity, type 2 diabetes, metabolic dysfunction-associated liver disease, and metabolic syndrome. Mechanistically, taurine influences ins

Taurine from tumour niche drives glycolysis to promote leukaemogenesis.

Signals from the microenvironment are known to be critical for development, stem cell self-renewal and oncogenic progression. Although some niche-driven signals that promote cancer progression have been identified1-5, concerted efforts to map disease-relevant microenvironmental ligands of cancer stem cell receptors have been lacking. Here, we use temporal single-cell RNA-sequencing (scRNA-seq) to identify molecular cues from the bone marrow stromal niche that engage leukaemia stem-enriched cells (LSCs) during oncogenic progression. We integrate these data with our human LSC RNA-seq and in vivo

Molecular basis of human taurine transporter uptake and inhibition.

The taurine transporter, TauT, regulates various taurine-mediated physiological and pathological functions by facilitating taurine uptake in a sodium- and chloride-dependent manner. Dysfunction of TauT is associated with male infertility, retinal health and cancers. Despite extensive research efforts, the intricate structure of TauT, the molecular mechanisms underlying taurine transport, and the inhibition mechanisms involved, all remain elusive. Here, we present eleven cryo-electron microscopy (cryo-EM) structures of TauT. The structures TauT bound to substrate (taurine) and substrate analogu

Taurine reduction in anaerobic respiration of Bilophila wadsworthia RZATAU.

Organosulfonates are important natural and man-made compounds, but until recently (T. J. Lie, T. Pitta, E. R. Leadbetter, W. Godchaux III, and J. R. Leadbetter. Arch. Microbiol. 166:204-210, 1996), they were not believed to be dissimilated under anoxic conditions. We also chose to test whether alkane- and arenesulfonates could serve as electron sinks in respiratory metabolism. We generated 60 anoxic enrichment cultures in mineral salts medium which included several potential electron donors and a single organic sulfonate as an electron sink, and we used material from anaerobic digestors in com

Taurine status and response to intravenous taurine supplementation in adults with short-bowel syndrome undergoing long-term parenteral nutrition: a pilot study.

Taurine deficiency in patients on long-term parenteral nutrition may be involved in cholestasis. We aimed to assess plasma taurine and tauro-conjugated bile acids in adults with short-bowel syndrome and their response to intravenous taurine. Thirty-two adult patients, who had been on taurine-free parenteral nutrition for a mean of 59 (SE 14) months for short-bowel syndrome, were studied retrospectively. In a second study, a subgroup of ten patients with chronic cholestasis received taurine-enriched (6.0 (SE 0.6) mg/kg per d) parenteral nutrition for 55 (SE 13) months. Post-absorptive plasma ta

Low plasma taurine concentration in Newfoundland dogs is associated with low plasma methionine and cyst(e)ine concentrations and low taurine synthesis.

Although taurine is not dietarily essential for dogs, taurine deficiency and dilated cardiomyopathy (DCM) are sporadically reported in large-breed dogs. Taurine status and husbandry were examined in 216 privately owned Newfoundlands, a giant dog breed with high incidence of idiopathic DCM (1.3-2.5%). Plasma taurine concentration was positively correlated (P < 0.01) with plasma cyst(e)ine (r = 0.37) and methionine (r = 0.35) concentrations and was similar across age, sex, neutering status, body weight, and body-condition scores. Plasma taurine concentration was low (< or =40 micromol/L) in 8% o

The Effect of Taurine on the Alkalinity Stress Resistance of Eriocheir sinensis.

High alkalinity, a major stress in saline-alkaline water, often leads to oxidative damage and apoptosis. Taurine has been shown to enhance the tolerance of aquatic animals to environmental stress, but whether it can reduce toxicity of alkalinity on Chinese mitten crab (Eriocheir sinensis) has not been determined. Thus, in this study, we investigated whether taurine could alleviate the deleterious effects of alkalinity stress on E. sinensis. The results revealed that taurine can improve the survival rate of the crab under 35.00 mmol/L alkalinity stress. Meanwhile, taurine inhibited the producti

In vitro effects of Taurine on Taxol-induced mitochondrial dysfunction.

Mitochondria are intricately involved with essential functions in normal and cancer cells and, as such, mitochondria are a natural target for chemotherapeutic drugs. We previously showed that Taxol, even at low doses, adversely effects mitochondrial energetics, function, and intracellular migration patterns. We observed (by multiple optical means) suppression of oxidative phosphorylation (OXPHOS) and decreased adenosine triphosphate (ATP) production, increased reactive oxygen species (ROS) generation (both immediately and integrated over time), increased mitotic fraction, and the release of LD
