Cardiac telocytes. From basic science to cardiac diseases. II. Acute myocardial infarction.

INTRODUCTION: The purpose of this study was to evaluate the scientific evidence regarding a potential role of telocytes in myocardial infarction. MATERIALS AND METHODS: To this purpose, we performed a systematic review of relevant scientific literature, indexed in PubMed, Web of Science, and Scopus. RESULTS AND DISCUSSIONS: We found six articles containing relevant studies aimed at liking myocardial infarction and telocytes. The studies that were analysed in this review failed to show, beyond a reasonable doubt, that telocytes do actually have significant roles in myocardial regeneration aft

Podoplanin (D2-40) is a reliable marker of urinary bladder myofibroblasts (telocytes).

Podoplanin, D2-40, has been described in a variety of normal and neoplastic tissues. It is often used for highlighting lymphatics. We evaluated the expression of podoplanin in α-smooth muscle actinpositive myofibroblasts producing the suburothelial layer in tunica propria of the urinary bladder that have some similar features with telocytes. Our results showed that these cells demonstrate distinct D2-40 immunoreactivity from telocytes occurring in the renal pelvis and ureter. We observed positive reaction not only in bioptic specimens from women with interstitial cystitis, but also in a contro

Telocytes accompanying cardiomyocyte in primary culture: two- and three-dimensional culture environment.

Recently, the presence of telocytes was demonstrated in human and mammalian tissues and organs (digestive and extra-digestive organs, genitourinary organs, heart, placenta, lungs, pleura, striated muscle). Noteworthy, telocytes seem to play a significant role in the normal function and regeneration of myocardium. By cultures of telocytes in two- and three-dimensional environment we aimed to study the typical morphological features as well as functionality of telocytes, which will provide important support to understand their in vivo roles. Neonatal rat cardiomyocytes were isolated and cultured

Telocytes in pleura: two- and three-dimensional imaging by transmission electron microscopy.

Information about the ultrastructure of connective (interstitial) cells supporting the pleural mesothelium is scarce. Our aim has been to examine whether telocytes (TCs) are present in pleura, as in epicardium and mesentery. TCs are a distinct type of cell, characterized by specific prolongations named telopodes (Tp). We have used transmission electron microscopy (TEM) and electron tomography (ET) to determine whether ultrastructural diagnostic criteria accepted for TCs are fulfilled by any of the cell subpopulations existing in the sub-mesothelial layer in mouse and human pleura. TCs have bee

Analysis of the spatial-temporal distribution and functional morphology of telocytes in goat testes.

Telocytes (TCs), a distinct interstitial cell population, are increasingly being reported in the male reproductive system. This study aimed to confirm TC presence in goat testes, to characterize their immunophenotype and age-related differences. Testes from one-, two-, and twelve-month-old goats were examined by using transmission electron microscopy (TEM) and double immunofluorescence (DIF) staining. TEM revealed that TCs were widely distributed among peritubular myoid cells (PMC) in testes from 12-month-old goats and exhibited reduced cell bodies and cytoplasm compared to TCs in juvenile goa

Telocytes Twenty Years on: A Critical Reappraisal of Identity, Function, and Pathological Relevance.

Telocytes are stromal cells defined by extremely long, moniliform prolongations termed telopodes and have been described in most mammalian organs since their formal designation in 2010. During the past two decades, the field has expanded from ultrastructural organ-mapping to hypotheses concerning stem-cell niche regulation, extracellular-vesicle-mediated communication, fibrosis, inflammation, and cancer-associated stromal remodelling. This expansion, however, has also generated methodological heterogeneity, with frequent reliance on non-specific markers such as CD34, PDGFRα, vimentin, and c-ki

First report of telocytes in human ovarian stroma: an ultrastructural and immunohistochemical study.

Telocytes (TCs) are interstitial cells characterized by long, thin cytoplasmic extensions called telopodes. Although their presence has been established in various organs, data on their existence and organization within the human ovary remain limited. This study aimed to identify and describe telocytes in the ovarian stroma using immunohistochemical, immunofluorescence, and ultrastructural methods. Ovarian tissues from women aged 24-65 years who underwent total hysterectomy and bilateral salpingo-oophorectomy for nonmalignant uterine diseases were analyzed. TCs were identified by CD34 co-expre

Recently discovered interstitial cells "telocytes" as players in the pathogenesis of uterine leiomyomas.

Uterine telocytes are interstitial cells characterized by a very long cytoplasmic prolongations, which form a 3D network, functionally integrating a wide variety of different cells. Leiomyomas (uterine fibroids) are benign tumors, which pose a huge threat concerning various health problems in women affected by this condition. The exact cause of leiomyomas development is, however, still largely unknown. Therefore, in an attempt to clarify their etiology, we performed an immunohistochemical characterization of telocytes in leiomyomas as well as in normal myometrium. Tissue samples of intramural

Telocytes are reduced during fibrotic remodelling of the colonic wall in ulcerative colitis.

Ulcerative colitis (UC) is characterized by chronic relapsing intestinal inflammation finally leading to extensive tissue fibrosis and resulting in a stiff colon unable to carry out peristalsis or to resorb fluids. Telocytes, a peculiar type of stromal cells, have been recently identified in the human gastrointestinal tract. Several roles have been proposed for telocytes, including mechanical support, intercellular signalling and modulation of intestinal motility. The aim of the present work was to investigate the presence and distribution of telocytes in colonic specimens from UC patients com

Mast Cells, Pancreatic Stellate Cells, and Telocytes in Chronic Pancreatitis: Ultrastructural Study.

Pancreatic inflammation and subsequent fibrosis drive serious disease complications. However, the pathogenesis of this process and the mechanisms underlying excessive extracellular matrix (ECM) deposition remain poorly understood. Our aim was to study intercellular interactions and ultrastructural changes in mast cells, pancreatic stellate cells, and telocytes, as well as in the extracellular matrix in various degrees of pancreatic fibrosis. Histological, immunohistochemical, and electron microscopic (EM) studies were performed on surgical materials from 17 patients. Mapping of fibrosis fields

Sex steroid hormone receptors of telocytes - potential key role in leiomyoma development.

BACKGROUND: Uterine leiomyoma is the most widespread benign tumor affecting women of childbearing age. There are still gaps in the understanding of its pathogenesis. Telocytes are unique cells found in more than 50 different locations inside the human body. The functional relationship between cells could clarify the pathogenesis of leiomyomata. Examination of membrane receptors on telocytes could explain their role in fibrosis, oxidative stress, and myometrial contractility. AIM: This research was conducted to assess the density of telocytes in terms of their putative role in leiomyoma format

Telocytes promote VEGF expression and alleviate ventilator-induced lung injury in mice.

Mechanical ventilation (MV) is an important procedure for the treatment of patients with acute lung injury or acute respiratory distress syndrome in a clinical setting; however, MV can lead to severe complications, including ventilator-induced lung injury (VILI). Telocytes (TCs) can promote tissue repair following injury in the heart, kidneys, and other organs. The aim of this study was to investigate the role of TCs in VILI in mice and the associated mechanisms. By using in vivo studies in mice and in vitro studies in cells, we demonstrated that an airway injection of TCs can reduce the pulmo

Isolated human uterine telocytes: immunocytochemistry and electrophysiology of T-type calcium channels.

Recently, telocytes (TCs) were described as a new cell type in the interstitial space of many organs, including myometrium. TCs are cells with very long, distinctive extensions named telopodes (Tps). It is suggested that TCs play a major role in intercellular signaling, as well as in morphogenesis, especially in morphogenetic bioelectrical signaling. However, TC plasma membrane is yet unexplored regarding the presence and activity of ion channels and pumps. Here, we used a combination of in vitro immunofluorescence and patch-clamp technique to characterize T-type calcium channels in TCs. Myome

Telocytes damage in endometriosis-affected rat oviduct and potential impact on fertility.

Women with endometriosis (EMs) have unexplained infertility. The recently identified telocytes (TCs) might participate in the maintenance of structural and functional integrity of oviduct tissue, but so far the involvement of TCs in EMs-affected oviduct tissue and potential impact on fertility capacity remain unknown. By an integrated technique of haematoxylin and eosin staining, in situ immunohistochemistry and double-labelled immunofluorescence staining and electron microscopy approach, TCs were studied in the autotransplantation Sprague-Dawley rat model of EMs-affected oviduct tissue and in

Telocytes link epithelial nutrient sensing with amplification of the ILC2-tuft cell circuit.

UNLABELLED: Group 2 innate lymphocytes (ILC2s) are prevalent in small intestine but engagement of type 2 immunity during basal processes are incompletely described. Thymic stromal lymphopoietin (TSLP), a cytokine implicated in ILC2 activation, was constitutively expressed in villus telocytes and crypt-associated trophocytes, specialized fibroblasts that sustain epithelial identity. Feeding increased TSLP and induced ILC2 type 2 cytokines that were attenuated by deletion of TSLP in PDGFRα + stromal cells or TSLP receptor on ILC2s. Mouse and human telocytes expressed receptors for glucagon-like

The telocytes relationship with satellite cells: Extracellular vesicles mediate the myotendinous junction remodeling.

The peripheral nerve injury (PNI) affects the morphology of the whole locomotor apparatus, which can reach the myotendinous junction (MTJ) interface. In the injury condition, the skeletal muscle satellite cells (SC) are triggered, activated, and proliferated to repair their structure, and in the MTJ, the telocytes (TC) are associated to support the interface with the need for remodeling; in that way, these cells can be associated with SC. The study aimed to describe the SC and TC relationship after PNI at the MTJ. Sixteen adult Wistar rats were divided into Control Group (C, n = 8) and PNI Gro

The Function of Efhd1 + Telocytes in the Synovial Lymphatic System and Inflammatory-Erosive Arthritis.

Resting collecting lymphatic vessels (cLVs) sense edema in distal joints and initiate contractions via unknown mechanisms. Rheumatoid arthritis (RA) patients have lymphatic drainage deficiencies from affected joints, and defects in the synovial lymphatic system exacerbate inflammatory-erosive arthritis in animal models. To understand this, we generated Efhd1-CreERT2 and Myoc-CreERT2 mice for cell-specific genetic gain and loss of function studies. These mice were crossed with tdTomato reporter (Ai9) mice, and studies showed selective tamoxifen-induced transgene expression in CD31-/CD34+ telocy

Microscopical Identification of Telocytes in the Fundic Stomach of African Catfish (Clarias gariepinus).

Clarias gariepinus catfish fundic stomach telocytes (TCs) were identified using light microscopy (LM), transmission electron microscopy (TEM), scanning electron microscopy (SEM), and immunohistochemistry. For LM, histochemical, hematoxylin, and eosin staining, as well as toluidine blue-stained semithin sections, were employed. These approaches revealed characteristic morphological features that differentiated TCs, which possessed a cell body with a nucleus and a telopode. Specifically, mucosal TCs were identified beneath the epithelium, within the lamina propria, and dispersed throughout the s
