Matched Fruit-Larva Metabolomics Identifies Host-Associated Metabolic Signatures After Multi-Generational Laboratory Acclimation in Zeugodacus tau (Tephritidae: Diptera).

Zeugodacus tau (Walker) is a polyphagous invasive tephritid capable of cross-family host shifts from ancestral Cucurbitaceae crops to phylogenetically divergent commercial fruits, causing severe horticultural economic losses. As an obligate fruit-boring pest, Z. tau largely relies on larval metabolic plasticity to achieve successful colonization of novel hosts during host shifts. However, few matched fruit-larva metabolomic studies integrate host chemistry to larval physiology, hindering sustainable pest control development. We performed untargeted LC-MS metabolomics on matched fruit and third

RNAi-mediated knockdown of vitellogenin genes disrupts ovary development and alters ovarian bacterial composition in Zeugodacus tau (Diptera: Tephritidae).

Zeugodacus tau Walker (Diptera: Tephritidae) is an invasive pest of global significance that inflicts substantial losses to the fruit and vegetable production sectors annually. Despite the availability of the Z. tau genome in public database, functional characterization of key genes remained limited. Vitellogenin (Vg) functions as a key precursor of yolk protein and exerts a pivotal part in the insect reproductive process. Here, we identified 3 Vg genes (Vg1, Vg2, and Vg3) based on genetic data from Z. tau. Analysis of amino acid sequences demonstrated that all ZtVgs contain conserved serine r

Olive Fruit Fly, Bactrocera oleae (Diptera: Tephritidae), Attraction to Volatile Compounds Produced by Host and Insect-Associated Yeast Strains.

The olive fruit fly, Bactrocera oleae (Rossi), is one of the most damaging insect pests of olives worldwide, requiring the use of insecticides for fruit protection in many orchards. Olive fruit flies are attracted to volatile composunds, including a female-produced pheromone, and host-plant and bacterial volatiles. Preliminary laboratory bioassays were conducted for olive fruit fly attraction to over 130 yeast strains from among 400 that were isolated from B. oleae adults and larvae or other insects, infested olives, and potential feeding sites. Kuraishia capsulata, Scheffersomyces ergatensis,

A newly identified Y chromosome gene obp-Y is required for sperm storage in female Zeugodacus tau (Diptera: Tephritidae).

In the organisms with XX/XY sex chromosomes, Y chromosome is unique to males and plays an important role in male reproductive development. The study of Y chromosome genes will contribute to the development of pest genetic prevention and control technology. In this study, we identified 9 Y chromosome genes in Zeugodacus tau (Diptera: Tephritidae), including gene 16222. Protein structure analysis showed that 16222 was highly similar to odorant binding protein, and thus gene 16222 was named obp-Y. Obp-Y knockout (KO) significantly reduced hatching rate of offspring. Sperm detection results showed

Oviposition behaviour and biochemical response of an insect pest, Zeugodacus cucurbitae (Coquillett) (Diptera: Tephritidae) to plant phenolic compound phloroglucinol.

Phenolic compounds are the secondary metabolites (SMs) present in plants carrying different bioactivities. In the present study, we explored the influence of a phenolic compound namely phloroglucinol on oviposition behaviour and different biochemical entities of an insect pest Zeugodacus cucurbitae (Coquillett) (Diptera: Tephritidae) using artificial diet. Phloroglucinol (IUPAC name: benzene-1,3,5-triol) affected the activity of antioxidant and detoxifying enzymes viz. superoxide dismutases (SOD), catalase (CAT), ascorbate peroxidases (APOX). dehydroascorbate reductase (DHAR), peroxidases (POX

Using the rDNA Internal Transcribed Spacer 1 to Identify the Invasive Pest Rhagoletis cerasi (Diptera: Tephritidae) in North America.

The cherry-infesting fruit fly Rhagoletis cerasi Loew is a significant commercial pest in Europe that has recently invaded North America. To date, it has been trapped only in Canada and northwestern counties of New York. It has the potential to spread further and threaten production and movement of cherry commodities. Timely diagnosis of the pest will facilitate surveys and quick response to new detections. Adult morphology of the pest is distinct from other flies in North America. However, when flies are significantly damaged on traps or the immature life stages are found in fruits, molecular

Determining the Sterilization Doses under Hypoxia for the Novel Black Pupae Genetic Sexing Strain of Anastrepha fraterculus (Diptera, Tephritidae).

A common strategy used to maintain sterile fly quality without sacrificing sterility is to irradiate the insects under an oxygen-reduced atmosphere. So far, sterilizing doses for the South American fruit fly Anastrepha fraterculus have only been determined under normoxia. Our study reports for the first time the dose-sterility response under hypoxia for two different A. fraterculus strains. The pupae were derived from a bisexual strain (a Brazilian-1 population) and a recently developed genetic sexing strain (GSS-89). Two hours prior to irradiation, pupae were transferred to sealed glass bottl

Structural Differences in the Digestive Tract Between Females and Males Could Modulate Regurgitation Behavior in Anastrepha ludens (Diptera: Tephritidae).

With the aim of understanding the mechanisms involved in the regurgitation behavior of tephritid flies, we performed a structural study of the digestive system of the economically important fruit-fly pest, Anastrepha ludens (Loew) using optical, scanning electronic microscopy (SEM) and transmission electron microscopy (TEM), plus a feeding assay. Most structures studied are similar to those previously reported in other adult dipterans, but, importantly, we found sexual differences in some structures that apparently affect regurgitation. We report for the first time sexual differences in the cr

Managing oriental fruit fly (Diptera: Tephritidae), with spinosad-based protein bait sprays and sanitation in papaya orchards in Hawaii.

The efficacy of GF-120 NF Naturalyte Fruit Fly Bait in combination with field sanitation was assessed as a control for female oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae), in papaya (Carica papaya L.) orchards in Hawaii. Three different bait spray regimes were evaluated: every row (high use of the bait), every fifth row (moderate use), and every 10th row (low use). Orchard plots in which no bait was applied served as controls. For five of the seven biweekly periods that followed the first bait spray, trapping data revealed significantly fewer female B. dorsalis captu

Two Chitin Biosynthesis Pathway Genes in Bactrocera dorsalis (Diptera: Tephritidae): Molecular Characteristics, Expression Patterns, and Roles in Larval-Pupal Transition.

Glucose-6-phosphate isomerase (G6PI) and UDP-N-acetylglucosamine pyrophosphorylase (UAP), two key components in the chitin biosynthesis pathway, are critical for insect growth and metamorphosis. In this study, we identified the genes BdG6PI and BdUAP from the oriental fruit fly, Bactrocera dorsalis (Hendel). The open reading frames (ORFs) of BdG6PI (1,491 bp) and BdUAP (1,677 bp) encoded 496 and 558 amino acid residues, respectively. Multiple sequence alignments showed that BdG6PI and BdUAP had high amino acid sequence identity with other insect homologues. Quantitative real-time polymerase ch

Fitness improvement of mass-reared sterile males of Ceratitis capitata (Vienna 8 strain) (Diptera: Tephritidae) after gut enrichment with probiotics.

Successful mass rearing is crucial for sterile insect technique programs. It has been shown that the sterilizing process using gammaradiation results in damage to midgut tissue, cellular organelles, and gut microbiota of flies. This can be responsible for the inferiority of sterile males compared with wild males. A bacteria-enhanced diet could contribute to the improvement of the fly's fitness. We investigated ways of increasing the competitiveness of mass-reared Mediterranean fruit fly, Ceratitis capitata (Wiedemann) sterile males. We tested the hypothesis that the addition of beneficial bact

Presence and positioning of insecticidal strips in Jackson traps affect catch of oriental fruit flies and melon flies (Diptera: Tephritidae).

Many countries operate trapping programs to detect incursions of pestiferous fruit flies (Diptera: Tephritidae). Three male-specific attractants form the cornerstone of these surveillance networks: methyl eugenol for Bactrocera species, cue-lure for Bactrocera and Zeugodacus species, and trimedlure for Ceratitis species. Traditionally, these lures were applied as a liquid to a cotton wick, but polymeric plugs are being used more widely. Regulations require that any toxicant placed in a trap with a lure-bearing plug must be presented separately from the plug, which, most commonly, is held in a

Comparison of in vitro heat and cold tolerances of the new invasive species Bactrocera invadens (Diptera: Tephritidae) with three known tephritids.

Bactrocera invadens Drew, Tsuruta & White (Diptera: Tephritidae) is spreading throughout central Africa attacking a variety of fruit; quarantines are placed on fruit from this region that are considered hosts. The only phytosanitary treatment that is commercially available is an ionizing irradiation treatment for all Tephritidae at 150 Gy. The development of other treatments, such as heat, cold, or fumigation, usually requires testing tens of thousands of insects at a dose that provides efficacy and may take several years. It may be possible to shorten the time required to develop treatments b

Evaluation of clove-chitosan nanohybrids and clove nanoemulsions as bio-agents against the peach fruit fly, Bactrocera zonata (diptera: tephritidae).

Plant Essential oils (EOs)-based eco-friendly biopesticides area substitute for chemically manufactured insecticides in integrated pest management (IPM) programs. Plant essential oils show chemical instability which causes the active ingredients to evaporate and degrade quickly and that is the primary issue for using them as biopesticides in field applications. EOs could be incorporated into controlled-release nano formulations, which are anticipated to be more efficient than the bulk substance. In this research, clove nanoemulsion (CEO-NE) and clove-chitosan nanohybrid (CEO-NE@CNPs) were asse
