Intraerythrocytic protozoan species of the genera Theileria and Babesia are known to infect both wild and domestic animals, and both are transmitted by hard-ticks of the family Ixodidae. The prevalences of hemoprotozoa and ectoparasites in 15 free-living Mazama gouazoubira, two captive M. gouazoubira and four captive Blastocerus dichotomus from the State of Minas Gerais, Brazil, have been determined through the examination of blood smears and the use of nested polymerase chain reaction (nPCR). The cervid population was inspected for the presence of ticks and any specimens encountered were iden

Babesia canis and Babesia gibsoni have, until recently, been considered the only piroplasms that parasitise dogs. However, recent reports indicate that "small" Babesia infections in Spanish dogs are surprisingly frequent and molecular phylogenetic analysis indicates that the infecting agent is closely related to Babesia microti. Because the 18SrDNA sequence was not completely identical to that of B. microti, the new name "Theileria annae" was assigned to the canine agent. No information is available regarding the possible vector of the new piroplasm, T. annae. As part of an effort to identify

A steer was infected with Theileria parva parva Kilae stabilate; nymphal Rhipicephalus appendiculatus ticks were applied to its ears so that they completed repletion when the steer had a high piroplasm parasitaemia. The engorged nymphs were subsequently incubated at 28 degrees C for 26-29 days to complete moulting, when the adult ticks were divided into two groups; one was incubated at 18 degrees C for 20 days and the other at 18 degrees C for 14 days and then at 37 degrees C for 6 days. Groups of ticks incubated at 37 and 18 degrees C were triturated and each resultant supernatant fluid inocu

Hyalomma anatolicum anatolicum ticks were collected around cattle pens at two locations close to Khartoum. They were assessed for theileria infection by four methods. Salivary glands were stained whole with methyl green pyronin and examined for parasite masses. Adult ticks were partially fed on rabbits, ground up in medium, and the suspensions were examined in Giemsa stained smears and by inoculation into bovine lymphocyte cultures. Ticks were fully fed on calves which were monitored for developing theileriosis. H a anatolicum were found infected with parasite masses similar to those seen in e

Theileria parva, a protozoan parasite, is the causative agent of East Coast fever (ECF), an economically important disease of cattle in sub-Saharan Africa. The Muguga cocktail vaccine which comprises 3 T. parva strains, namely Muguga, Kiambu 5 and Serengeti transformed, is used for immunization of cattle to control ECF. However, the relative contributions of these T. parva strains to vaccine efficacy are not fully understood. This study compared the in vitro infectivity of the strains at varying concentrations of 2.75, 84.5, and 169 infected acini/ml using peripheral blood mononuclear cells is

Bovine tropical theileriosis, caused by Theileria annulata, is a significant economic burden in endemic regions. The limitations of current control measures necessitate the development of new therapeutic agents. This study evaluated the anti-Theileria potential and host cell safety of rutin, andrographolide, dodecyltrimethylammonium bromide (DTAB), and fusidic acid. Antiparasitic efficacy was assessed using a resazurin-based growth inhibition assay on T. annulata-infected bovine lymphocytes. Cytotoxicity and haemolytic activity were evaluated using healthy bovine peripheral blood mononuclear c

The aim of this non-blinded, randomised, controlled study was to determine the efficacy of toltrazuril for the prevention of oriental theileriosis in calves. Thirty calves were infected with Theileria orientalis Ikeda genotype through the application and retention of infected female bush ticks (Haemaphysalis longicornis). 15 Group 1 calves were treated with a single, oral dose of toltrazuril at the dose registered in Australia for coccidiosis (15 mg/kg), concurrently with tick infection, and 15 Group 2 (control group) calves were untreated. There were no significant differences (P > 0.05) in m

Tropical theileriosis is a significant tick-borne disease affecting cattle. For decades an empirical live attenuated vaccine has been the primary method of controlling disease. The vaccine is produced through prolonged culture of Theileria annulata schizont-transformed macrophages, but how loss of virulence occurs remains unclear. Notably attenuated (vaccine) macrophages display dampened dissemination potential compared with their original, virulent counterparts. In addition, parasite schizonts in attenuated macrophages have significantly lost their ability to differentiate into merozoites. Th

The mRNA vaccine platform should help research on East Coast fever subunit vaccines, but it remains unexplored. A theoretical strength of this platform, namely its capacity to prime cytotoxic T lymphocyte responses, is appealing as these, rather than antibodies, are the major mediators of immunity induced by a live parasite-based vaccine. Here, I highlight knowledge on functionally relevant bovine adaptive cellular and antibody immune responses to Theileria parva and antigens targeted by them that could help to assess this vaccine platform, and in the design of a broad-spectrum subunit vaccine

Theileriosis is a haemo protozoan disease affecting livestock, causing high mortality and production loss. Although therapeutic and vector control measures are available, selecting Theileria-resistant/tolerant animals is considered an alternate control measure for which the identification of genetic markers is essential. The present study screened dwarf Vechur (Bos indicus) and crossbred (B. taurus × B. indicus) cattle for natural infection to Theileria using peripheral blood smear and clinical and haematology evaluation. Polymorphisms in a previously identified candidate gene were screened in

BACKGROUND: No tick-borne pathogens (TBPs) causing haemolytic anaemia in cattle have been reported, except Theileria orientalis and complete blood count (CBC) profile is the only haematological parameter to determine the severity of regenerative haemolytic anaemia.
OBJECTIVES: To identify the causative agents of TBP-induced haemolytic anaemia and determine haematological parameters that indicate haemolytic anaemia in grazing cattle.
METHODS: Eighty-two Korean indigenous cattle (Hanwoo) were divided into two groups: grazing (n = 67) and indoor (n = 15) groups. CBC and serum biochemistry were

Hyalomma anatolicum is a tick of significant one-health importance due to its role as a vector for various pathogens affecting humans, animals and the environment, such as Theileria annulata, which causes tropical theileriosis in cattle, leading to severe economic losses. When infected with pathogens like T. annulata, the salivary glands of H. anatolicum undergo gene expression changes, secrete modified proteins and activate immune responses, all of which facilitate pathogen survival and transmission by modulating the host immune response and optimizing conditions for pathogen development. Equine piroplasmosis (EP) is a tick-borne infectious disease highly prevalent in tropical and subtropical regions, such as Venezuela. EP affects wild and domestic equids leading to several clinical presentations, from asymptomatic to severely affected animals. In this study, thirty-three (33) sport horses under regular training activities and from endemic regions of north-central Venezuela were submitted to an observational survey, case-control, to describe the presence of clinical signs and natural EP infections. A conventional PCR assay targeting the SSU rRNA gene revealed EP etiologic agent

Theileria parasites commonly infect African wild artiodactyls. In rare roan (Hippotragus equinus) and sable (H. niger) antelopes, Theileria sp. (sable)-associated calf mortalities constrain breeding programs. The pathogenicity of most leukocyte-transforming Theileria spp. originates in their invasion of and multiplication in various mononuclear leukocytes, the transformation of both infected and uninfected leukocytes, and their infiltration of multiple organs. Understanding the pathogenesis of theileriosis can be improved by the use of immunohistochemistry (IHC) to identify the localization of

Pathogens transmitted by ticks are an emerging problem worldwide, this study aimed to diagnose the causal agents of infection in dogs presenting suspected hemoparasitoses. Fifty-eight dogs with clinical signs such as depression, hemorrhagic diathesis and fever were evaluated regarding clinical presentation, hemogram, blood smears and serological tests, using the indirect immunofluorescence method for the agents Babesia vogeli and Ehrlichia canis and conventional PCR for Babesia spp. (gene 18S rRNA), Rangelia vitalii (gene 18S rRNA) and Ehrlichia spp. (gene dsb). Five (8.6%) of the 58 dogs were

East Coast Fever is a tick-transmitted disease in cattle caused by Theileria parva protozoan parasites. Quantification of the clinical disease can be done by determining a number of variables, derived from parasitological, haematological and rectal temperature measurements as described by Rowlands et al. (2000). From a total of 13 parameters a single ECF-score is calculated that allows categorization of infected cattle in five different classes that correlate with the severity of clinical signs. This score is complicated not only by the fact that it requires estimation of 13 parameters but als

Blood and serum samples were taken from 481 horses, from a stud farm or a racecourse, and tested by microscopic examination of blood smears and cELISA for Theileria equi (T. equi) and Babesia caballi (B. caballi) infections. At the time of sampling, animals were also examined for tick infestations and clinical disease, which were not observed in any of the sampled horses. During the microscopic examination of thin blood smears, parasites were detected in the three horses from the racecourse. Overall seroprevalence of infection was detected as 18.50% (89 of 481 horses) by cELISA, with T. equi b

During 1973-1990 cattle immunization, in Iran, was induced by two strains within an interval of one month: first the milder and then the mild strain. Although this method of vaccination rendered satisfactory results in the field, yet production, maintenance in deep-freezers and transportation in liquid nitrogen particularly to remote areas of the country proved to be uneconomical and time consuming. Therefore, in order to reduce cost and save time, a new method involving only one local and live attenuated vaccine strain was sought. Reports received from different ecological areas of the countr
