The surface of gold nanoparticles (AuNPs) can be conjugated with a wide range of highly functional biomolecules. A common pitfall when utilizing AuNPs is their tendency to aggregate, especially when their surface is functionalized with ligands of low molecular weight (no steric repulsion) or ligands of neutral charge (no electrostatic repulsion). For biomedical applications, AuNPs that are colloidally stable are desirable because they have a high surface area and thus reactivity, resist sedimentation, and exhibit uniform optical properties. Here, we engineer the surface of AuNPs so that they r

Cystinuria is an autosomal recessive disorder characterized by excessive urinary excretion of cystine, resulting in recurrent cystine kidney stones, often presenting in childhood. Current treatment options for cystinuria include dietary and/or fluid measures and potassium citrate to reduce cystine excretion and/or increase solubility. Tiopronin and D-penicillamine are used in refractory cases to bind cystine in urine, albeit with serious side effects. A recent study revealed efficacy of nutritional supplement α-lipoic acid (ALA) treatment in preventing kidney stones in a mouse model of cystinu

Glucose transporter type1 (GLUT-1) deficiency may be rare, but it is a preventable cause of severe learning difficulties; and therefore there is an urgency in making an early diagnosis. Suspicions must be roused when intractable seizures occur in infancy. These may be associated with acquired microcephaly and developmental delay. The finding of low glucose sugar levels in the cerebrospinal fluid, but not in the blood will identify the condition. The gene encoding the GLUT-1 protein is located on the short arm of chromosome 1, and inheritance is by a dominant trait. Patients with this syndrome

α-lipoic acid (LA) is an essential cofactor for mitochondrial dehydrogenases and is required for cell growth, metabolic fuel production, and antioxidant defense. In vitro, LA binds copper (Cu) with high affinity and as an endogenous membrane permeable metabolite could be advantageous in mitigating the consequences of Cu overload in human diseases. We tested this hypothesis in 3T3-L1 preadipocytes with inactivated Cu transporter Atp7a; these cells accumulate Cu and show morphologic changes and mitochondria impairment. Treatment with LA corrected the morphology of Atp7a-/- cells similar to the C

rac-a-Lipoic acid (CAS 62-46-4, thioctic acid) is used in human therapy besides the parenteral route also orally in gastric juice soluble galenic formulations in patients suffering from diabetic polyneuropathy which also involves the gastrointestinal tract (GI) tract in about 20% of the diabetic population. In those patients the most common manifestation of the disease due to small intestine dysfunction is diarrhoea as a consequence of which malabsorption of orally administered drugs may result. Senile osteoporosis (SOP) creates a hostile peri-implant microenvironment characterized by impaired osteogenesis, excessive bone resorption, and persistent oxidative stress, leading to poor osseointegration. Here, we developed a dual-functional titanium surface (Ti-Ta@RS) co-modified with a thioctic acid derivative and risedronate sodium to simultaneously regulate oxidative stress and osteoclast activity. Ti-Ta@RS improved the physicochemical properties of the titanium surface and significantly enhanced BMSC viability, migration, spreading, and osteogenic differentiation under senescence-assoc

Amanita phalloides-type mushroom poisoning is well recognized as causing acute liver injury and often death. Less is known, however, of whether maternal Amanita poisoning is associated with fetal damage or not. In August 1991 four members of a family were hospitalized with food intoxication caused by Amanita phalloides and Amanita verna. One of them died from hepatic and renal failure. The survivors included a 26-year-old woman in the 23rd week of pregnancy. Her clinical symptoms and blood chemistry data (lowest prothrombin activity 23%) indicated intoxication of medium severity. The managemen

Supramolecular adhesives enable on-demand detachment due to their dynamic nature. However, such behavior usually at the cost of limited adhesive toughness. To promote the intrinsic compromise between adhesive toughness and switchability, we propose a soft-confinement-regulated nanorod crystallization strategy to construct a supramolecular elastomer (PTA-PCL). The amphiphilic and soft poly (thioctic acid) (PTA) matrix forms a dynamically confined interface through hydrogen bonding, directing the oriented self-assembly of hydrophobic polycaprolactone glycol (PCL-OH) into nanorod crystals with an

Elastomer-based electronics have the potential to maintain both electrochemical conductivity and mechanical properties simultaneously, paving the way for the development of stretchable sensors. However, creating sensors that are extremely stretchable, wearable, and highly sensitive remains a significant challenge. In this work, we present a novel fully bio-based multifunctional polymer (FPL/PTA) designed using thioctic acid (TA) and formaldehyde-protected lignin (FPL). The FPL serves as both a cross-linking agent and a free radical quenching agent. The FPL/PTA features an adaptive polymer netw

Despite the existence of many preclinical studies, scientific evidence is lacking on the clinical use of alpha-lipoic acid (ALA) for central nervous system disorders. Therefore, we aimed at revising the literature concerning the use of ALA for the treatment of psychiatric and neurological conditions and to point out what is missing for the introduction of this antioxidant to this purpose. For this systematic review we performed a search using PubMed and SCOPUS databases with the following keywords: "alpha-Lipoic Acid AND central nervous system OR psychiatric disorders OR neurological disorders

Protein-based hydrogels have been constrained by modest mechanics and limited responsiveness. In this study, a light-triggered, disulfide-cross-linked hydrogel based on poly(γ-glutamic acid) (γ-PGA) was established by integrating thiolated γ-PGA and α-lipoic acid (LA)-grafted γ-PGA under 405 nm irradiation. Dynamic disulfide chemistry enabled rapid gelation at a critical concentration of ≥7%, with complete gelation at a polymer concentration of 8%, a storage modulus of approximately 100 Pa, and gelation time below 60 s at 0.25% LAP. The hydrogel adhered to diverse substrates, underwent redox-t

Strong and durable adhesion of bioadhesives on wet/underwater tissues is still challenging because of the hindrance of the hydration layer and swelling of the adhesive. Here a water-induced self-hardening bioadhesive (p(LA-ABO)) composed of poly(lipoic acid) (PolyLA) capped with 4-allyl-1,2-benzenediol (ABO), is prepared through a solvent evaporation-induced self-polymerization method. During underwater curing, the water-induced aggregation of the hydrophobic PolyLA chain of the bioadhesive leads to a soft-to-hard transition, therefore enhancing its cohesion and wet/underwater tissue adhesion.

Insulin autoimmune syndrome (IAS) is a rare cause of spontaneous hypoglycemia characterized by the presence of insulin autoantibodies. Alpha-lipoic acid (ALA), a widely used nutraceutical, has been increasingly recognized as a potential trigger for IAS. We report two cases of ALA-induced IAS in Indian patients and review existing literature to highlight its clinical presentation, diagnostic challenges, and management strategies. In the first case, rapid-onset severe hypoglycemia mandated the use of oral steroids with gradual tapering and stoppage over a duration of few months, while in the sec

R-α-lipoic acid (RLA) and dihydrolipoic acid (DHLA), a reduced form of RLA, are potent endogenous antioxidants that can reduce oxidative damage. Despite their numerous nutraceutical potentials, clinical applications of RLA are still limited due to its poor solubility and stability problems. This study aimed to develop an RLA-loaded liposome (LIP/RLA) for the improvement of nutraceutical properties. LIP/RLA was developed by a typical solvent injection method. Uniform liposomes of LIP/RLA were observed by transmission electron microscopy, and the mean particle size was calculated to be ∼150 nm f

Ischemia-reperfusion injury is related to kidney dysfunction due to bilateral lower limb ischemia. This kidney injury may lead to acute kidney failure and mortality. Alpha-Lipoic Acid, a known antioxidant, can ameliorate kidney dysfunction and histopathology related to several etiologies. Ischemia was performed in adult male rats by bilateral femoral artery occlusion, then ischemia-reperfusion was done for 1 day and 7 days. Lipoic acid was administered to rats that had undergone ischemia-reperfusion for 7 days. The renal cortices of the kidneys of the tested groups were processed for light and

After intravenous supplementation of an unintentionally high dose of the antioxidant alpha-lipoic acid (ALA), a 53-year-old female complained of myalgia, chills and nausea, and showed signs of haemorrhagic diathesis. The laboratory findings were excessive hyperferritinemia, leukoerythroblastosis, severe thrombocytopenia, elevated liver enzymes and impaired coagulation. The toxicological tests resulted in an ALA serum concentration of 10 280 µg/L. The peripheral blood film of the patient showed some neutrophil dysplasia with unusual small dark-blue stained round cytoplasmic inclusions resemblin

UNLABELLED: Mycoplasma hyopneumoniae (M. hyopneumoniae) is the causative agent of pandemic pneumonia among pigs, namely, swine enzootic pneumonia. Although M. hyopneumoniae was first identified in 1965, little is known regarding its metabolic pathways, which might play a pivotal role during disease pathogenesis. Lipoate is an essential cofactor for enzymes important for central metabolism. However, the lipoate metabolism pathway in M. hyopneumoniae is definitely unclear. Here, we identified a novel gene, lpl, encoding a lipoate protein ligase in the genome of M. hyopneumoniae (Mhp-Lpl). This g
