Interacting short-term regulatory mechanisms enable the conversion of light energy to chemical energy in photosynthesis.

Photosynthesis is a complex sequence of physical, electrochemical, biochemical, and physiological processes that convert light energy and carbon dioxide into sugars. These sugars then provide the energy and carbon backbone for all metabolic pathways involved in plant growth and development. However, if light energy is not managed effectively within the thylakoid membrane, it can destroy the photosynthetic apparatus in an oxygenic environment generated by photosynthesis itself. Effective photoprotection requires a variety of partially overlapping regulatory mechanisms that control energy and el

Analysis of the Metabolic and Structural Changes in Ulmus pumila 'Zhonghua Jinye' Leaf Under Shade Stress.

Ulmus pumila L. 'Zhonghua Jinye' is a plant variety with colourful leaves that is widely used in landscaping. In our study, the leaves of U. pumila 'Zhonghua Jinye' fade and turn green under light (LT, 45%), moderate (MD, 70%) and high (HG, 95%) shading treatment, reducing its ornamental value. However, the mechanism underlying this adaptation to shade is poorly understood. The objective of this study was to elucidate the mechanism of physiological, cellular microstructural and transcriptional changes involved in leaf regreening in U. pumila 'Zhonghua Jinye'. Our results showed that the pigmen

Chlorophyll deficiency in the maize elongated mesocotyl2 mutant is caused by a defective heme oxygenase and delaying grana stacking.

BACKGROUND: Etiolated seedlings initiate grana stacking and chlorophyll biosynthesis in parallel with the first exposure to light, during which phytochromes play an important role. Functional phytochromes are biosynthesized separately for two components. One phytochrome is biosynthesized for apoprotein and the other is biosynthesized for the chromophore that includes heme oxygenase (HO). METHODOLOGY/PRINCIPAL FINDING: We isolated a ho1 homolog by map-based cloning of a maize elongated mesocotyl2 (elm2) mutant. cDNA sequencing of the ho1 homolog in elm2 revealed a 31 bp deletion. De-etiolation

Nitrogen deficiency in Arabidopsis affects galactolipid composition and gene expression and results in accumulation of fatty acid phytyl esters.

Nitrogen is an essential nutrient for plants because it represents a major constituent of numerous cellular compounds, including proteins, amino acids, nucleic acids and lipids. While N deprivation is known to have severe consequences for primary carbon metabolism, the effect on chloroplast lipid metabolism has not been analysed in higher plants. Nitrogen limitation in Arabidopsis led to a decrease in the chloroplast galactolipid monogalactosyldiacylglycerol (MGDG) and a concomitant increase in digalactosyldiacylglycerol (DGDG), which correlated with an elevated expression of the DGDG synthase

Specific light-harvesting complexes mediate grana stacking and prevent energy spillover between photosystems in plant chloroplasts.

Land plant chloroplasts are characterized by a prominent morphological feature: the stacking of thylakoid membranes into grana, ie defining a continuous membranous structure surrounded by the stroma and folded into appressed grana and non-appressed domains, enclosing the thylakoid lumen. Photosystem II and its antenna system are concentrated, whereas Photosystem I and ATPase are located in the grana end membranes and, in the stroma, membranes connecting grana. Thylakoid stacking promotes the physical separation of Photosystems I and II, thereby ensuring a balanced flow of excitation energy bet

Anticandidal Efficacy of Oral Probiont Limosilactobacillus fermentum Against Dental Caries Pathogens in Children, Tamil Nadu, India.

Functional proteomics of membrane proteins is an important tool for the understanding of protein networks in biological membranes but structural studies on this part of the proteome are limited. In this study we undertook such an approach to analyse photosynthetic thylakoid membranes isolated from wild-type and mutant strains of Chlamydomonas reinhardtii. Thylakoid membrane proteins were separated by high-resolution two-dimensional gel electrophoresis (2-DE) and analysed by immuno-blotting and mass spectrometry for the presence of membrane-spanning proteins. Our data show that light-harvesting

Photo-Empowered Macrophage-Based Drug Delivery System Overcomes Motility Suppression and Significantly Enhances Deep Tumor Drug Delivery.

Recently, cell-based drug delivery systems emerge as highly promising alternatives in the field of antitumor therapy. These systems are characterized by their excellent biocompatibility, robust drug-loading capacity, and the ability to effectively traverse biological barriers. However, the immunosuppressive tumor microenvironment significantly weakens the motility of immune cells, which is crucial for efficient drug delivery. A novel photo-empowered macrophage-based (PEM) drug delivery system is successfully developed in this study. The system is modified with thylakoids extracted from spinach

Enhanced Relative Electron Transport Rate Contributes to Increased Photosynthetic Capacity in Autotetraploid Pak Choi.

Autopolyploids often show growth advantages over their diploid progenitors because of their increased photosynthetic activity; however, the underlying molecular basis of such mechanism remains elusive. In this study, we aimed to characterize autotetraploid pak choi (Brassica rapa ssp. chinensis) at the physiological, cellular and molecular levels. Autotetraploid pak choi has thicker leaves than its diploid counterparts, with relatively larger intercellular spaces and cell size and greater grana thylakoid height. Photosynthetic data showed that the relative electron transport rate (rETR) was ma

The Arabidopsis TELLURITE RESISTANCE C is required for the functionality of both photosystems by supplying manganese and calcium.

The oxygen-evolving complex (OEC) of photosystem II (PSII) contains a Mn4CaO5 cluster that catalyzes the light-driven oxidation of water. In Arabidopsis thaliana, optimal PSII function requires the thylakoid membrane-localized manganese transporter PHOTOSYNTHESIS AFFECTED MUTANT 71 (PAM71). Here, we show that overexpression of TELLURITE RESISTANCE C (TerC) restores low chloroplast manganese levels, O2 evolution rates, and growth defects in PAM71 knock-out plants. A TerC knock-down allele (amiR-TerC) exhibits OEC-related phenotypic changes similar to those observed in pam71, including altered p

Calcium binding to the subunit c of E. coli ATP-synthase and possible functional implications in energy coupling.

The 8-kDa subunit c of the E. coli F0 ATP-synthase proton channel was tested for Ca++ binding activity using a 45Ca++ ligand blot assay after transferring the protein from SDS-PAGE gels onto polyvinyl difluoride membranes. The purified subunit c binds 45Ca++ strongly with Ca++ binding properties very similar to those of the 8-kDa CF0 subunit III of choloroplast thylakoid membranes. The N-terminal f-Met carbonyl group seems necessary for Ca++ binding capacity, shown by loss of Ca++ binding following removal of the formyl group by mild acid treatment. The dicyclohexylcarbodiimide-reactive Asp-61

High light induced changes in organization, protein profile and function of photosynthetic machinery in Chlamydomonas reinhardtii.

The green alga Chlamydomonas (C.) reinhardtii is used as a model organism to understand the efficiency of photosynthesis along with the organization and protein profile of photosynthetic apparatus under various intensities of high light exposure for 1h. Chlorophyll (Chl) a fluorescence induction, OJIPSMT transient was decreased with increase in light intensity indicating the reduction in photochemical efficiency. Further, circular dichroism studies of isolated thylakoids from high light exposed cells showed considerable change in the pigment-pigment interactions and pigment-proteins interactio

Mg2+ limitation leads to a decrease in chlorophyll, resulting in an unbalanced photosynthetic apparatus in the cyanobacterium Synechocytis sp. PCC6803.

Mg2+, the most abundant divalent cation in living cells, plays a pivotal role in numerous enzymatic reactions and is of particular importance for organisms performing oxygenic photosynthesis. Its significance extends beyond serving as the central ion of the chlorophyll molecule, as it also acts as a counterion during the light reaction to balance the proton gradient across the thylakoid membranes. In this study, we investigated the effects of Mg2+ limitation on the physiology of the well-known model microorganism Synechocystis sp. PCC6803. Our findings reveal that Mg2+ deficiency triggers both

Photosynthetic medicine: Engineering a plant-derived nanofoundry to power mammalian cells.

Mammalian eyes receive constant visible light yet lack photosynthesis. Xing et al. engineer an intact thylakoid grana nanoparticle enabling light-driven NADPH production within corneal cells, restoring antioxidant defenses and interrupting inflammatory cycles in dry eye disease. This work positions borrowed photosynthesis as a therapeutic strategy for oxidative, light-exposed tissue pathologies.

Transplanting light-dependent reactions for mammalian eye photosynthesis.

Mammalian eyes are exposed to visible light but cannot perform photosynthesis. Here, we show that introducing a nanoscale, structurally and functionally preserved thylakoid system, LEAF (light-reaction enriched thylakoid NADPH-foundry), into corneal cells enables light-driven bona fide photosynthetic production of NADPH and ATP, similar to plant leaves, which alleviates oxidative stress and inflammation. LEAF acts in two domains. Intracellularly, it integrates with host cells to supply NADPH and ATP via intact photosynthetic electron transport, restoring redox balance. Extracellularly, photosy

Evidence for an association of the early light-inducible protein (ELIP) of pea with photosystem II.

The precursor to the nuclear-coded 17 kDa early light-inducible protein (ELIP) of pea has been transported into isolated intact chloroplasts. The location of the mature protein in the thylakoid membranes was investigated after using cleavable crosslinkers such as DSP and SAND in conjunction with immunofractionation methods and by application of mild detergent fractionation. We show that ELIP is integrated into the membranes via the unstacked stroma thylakoids. After isolation of protein complexes by solubilization of membranes with Triton X-100 and sucrose density-gradient centrifugation the c

Multilayer gyroid cubic membrane organization in green alga Zygnema.

Biological cubic membranes (CM), which are fluid membranes draped onto the 3D periodic parallel surface geometries with cubic symmetry, have been observed within subcellular organelles, including mitochondria, endoplasmic reticulum, and thylakoids. CM transition tends to occur under various stress conditions; however, multilayer CM organizations often appear associated with light stress conditions. This report is about the characterization of a projected gyroid CM in a transmission electron microscopy study of the chloroplast membranes within green alga Zygnema (LB923) whose lamellar form of t

The multiple roles of light-harvesting chlorophyll a/b-protein complexes define structure and optimize function of Arabidopsis chloroplasts: a study using two chlorophyll b-less mutants.

The multiple roles of light-harvesting chlorophyll a/b-protein complexes in the structure and function of Arabidopsis chloroplasts were investigated using two chlorophyll b-less mutants grown under metal halide lamps with a significant far-red component. In ch1-3, all six light-harvesting proteins of photosystem (PS) II were greatly decreased; in ch1-3lhcb5, Lhcb5 was completely absent while the other five proteins were further decreased. The thylakoids of ch1-3 were less negatively-charged than the wild type, and those of ch1-3lhcb5 were even less so. Despite the expected weaker electrostatic

Structural and functional alterations of cyanobacterial phycobilisomes induced by high-light stress.

Exposure of cyanobacterial or red algal cells to high light has been proposed to lead to excitonic decoupling of the phycobilisome antennae (PBSs) from the reaction centers. Here we show that excitonic decoupling of PBSs of Synechocystis sp. PCC 6803 is induced by strong light at wavelengths that excite either phycobilin or chlorophyll pigments. We further show that decoupling is generally followed by disassembly of the antenna complexes and/or their detachment from the thylakoid membrane. Based on a previously proposed mechanism, we suggest that local heat transients generated in the PBSs by

Photosynthetic and ultrastructural responses of the chlorophyte Lobosphaera to the stress caused by a high exogenic phosphate concentration.

Biotechnology of microalgae holds promise for sustainable using of phosphorus, a finite non-renewable resource. Responses of the green microalga Lobosphaera sp. IPPAS C-2047 to elevated inorganic phosphate (Pi) concentrations were studied. Polyphosphate (PolyP) accumulation and ultrastructural rearrangements were followed in Lobosphaera using light and electron microscopy and linked to the responses of the photosynthetic apparatus probed with chlorophyll fluorescence. High tolerance of Lobosphaera to ≤ 50 g L-1 Pi was accompanied by a retention of photosynthetic activity and specific induction
