Changes in expression of FSH and LH receptors in the ovine main immune organs during early pregnancy.

Follicle-stimulating hormone (FSH) and luteinizing hormone (LH) are mainly involved in follicle development and ovulation, but FSH receptor (FSHR) and LH receptor (LHR) are also expressed in the immune system. Nevertheless, it is not clear if gestation affects the expression of the FSHR and LHR in the maternal main immune organs (thymus, lymph node, spleen, and liver). In this study, these organs were sampled from the ewes at the estrous cycle, and during early pregnancy, and mRNA and protein expression of FSHR and LHR were analyzed. The results showed that early pregnancy downregulated mRNA a

Do thymic-neuroendocrine interactions play a role in the antihypertensive effect of neonatal thymectomy in Lyon hypertensive rats?

Previous studies showed that neonatal thymectomy prevented the spontaneous increase in blood pressure in genetically hypertensive rats (LH) of the Lyon strain, leaving untouched that of their normotensive controls (LN). As the thymus is connected to the neuroendocrine system through secretion of hormonal factors, we investigated the possible role played by these factors in hypertension of LH rats. To that end we studied, in sham-operated and neonatally thymectomized LH rats, the blood pressure effects of thymostimulin, a partially purified thymus extract and examined whether changes in major n

Functional anatomy of the thymic microenvironment.

This paper presents a review of our current understanding of the nature of the thymic microenvironment, after briefly considering the major role of the gland. The epithelial cells and their products are of fundamental importance, and other cells of the macrophage series are implicated in most functional events. The embryological origin of the epithelium is still not clear, although disease conditions would suggest a single origin. Immigration and emigration of thymocytes is considered, and also the passage of antigens into the gland. The events within the thymus are under the control of the CN

Ectopic pituitary transplants restore immunocompetence in Ames dwarf mice.

This work was undertaken to study the effects of prolactin on immune function in Ames dwarf mice. For that purpose, adult Ames dwarf mice were implanted with pituitaries from normal mice under the kidney capsule. Ectopic pituitary transplants produced the expected increase in plasma prolactin levels in male and female dwarf mice as compared to sham-operated dwarf or untreated normal mice. Body weight was significantly increased in pituitary-grafted dwarf mice of both sexes, but did not reach the values found in normal (non-dwarf) animals. Pituitary transplants induced an increase in thymus wei

Biological activity of hormonally active and non-active androgen derivatives.

Androgen derivatives appeared to have different biological activities in vivo and in vitro. Testosterone-17-isobutyrate given in three doses of 50 or 200 microgram increased significantly the weight of seminal vesicles and reduced thymus weight in castrated males, whereas testosterone-17-hemisuccinate, testosterone-D-beta-glucoside and testosterone-3-(O-carboxymethyl)-oxime had no such effects. Similarly, ten 1-mg doses of testosterone-17-isobutyrate, unlike testosterone-17-hemisuccinate, resulted in a marked reduction of thymus weight in non-castrated males and in a significant inhibitory eff

Thymoma and autoimmunity.

The thymus is a central lymphatic organ that is responsible for many immunological functions, including the production of mature, functional T cells and the induction of self-tolerance. Benign or malignant tumors may originate from the thymus gland, with thymoma being the most common and accounting for 50% of anterior mediastinal tumors. Malignancies linked to thymoma include the loss of self-tolerance and the presence of autoimmunity. In this review, we compiled the current scientific evidence detailing the various interactions between thymoma and autoimmune diseases, including myasthenia gra

Toll-like receptors 2 and 4, and acute phase cytokine gene expression in dexamethasone and growth hormone treated dairy calves.

Cattle are exposed to growth hormone stimulants and to stressors that cause cortisol release. Both of these hormones affect immune responses which may reduce disease resistance. Toll-like receptors are the pattern recognition molecules of pathogens that are on immune cells. They then orchestrate the induction of the appropriate acute phase cytokines of the early innate response. The objective of this study was to determine changes in toll-like receptors and acute phase cytokines following treatment with a synthetic glucocorticoid (dexamethasone) and growth hormone (GH). Twenty-eight calves wer

Thymopoietin to thymopentin: experimental studies.

Thymopoietin is a polypeptide hormone of the thymus consisting of 49 amino acids. The pentapeptide thymopentin (TP-5) Arg-Lys-Asp-Val-Tyr, corresponding to amino acids 32-36 of thymopoietin, appears to represent the active site of thymopoietin in that it has all the biological activities of the native hormone. Thymopoietin is secreted by epithelial cells of the thymus and is pleiotropic in action, affecting neuromuscular transmission, induction of early T cell differentiation and immune regulation. The immuno-regulatory actions of thymopentin on peripheral T cells are mediated by intracellular

[Age-related effects in thymocytes differentiation: role of pineal gland peptide factors].

The peculiarities of the differentiation of the thymocytes in adult and old CBA/Ca mice were studied with a help of the monoclonal antibodies to different lymphoid antigens of mice. In the thymus of old mice, the portions of the CD4-CD8-, CD44+ and CD44+Thy-1(-)-cells have been shown to be increased. The percentage of the CD4+CD8(+)-thymocytes did not change, whereas the portion of CD25+, PNA+, CD3+, CD3+CD8+, CD4+CD8- and CD4-CD8(+)-cells diminished. The ratios of different subpopulations of thymocytes, characterizing different degree of their maturation are changing with age towards the prev

Recent studies on the role of the thymus in early stages of lymphopoiesis and immune differentiation.

The influence of thymus deprivation and thymus restoration was studied: a) on the cycling capacity of colony forming cells (CFU-S) in the bone marrow and b) on the establishment of tolerance in liver radiation chimeras. After neonatal thymectomy a reduction in the number of CFU-S in the bone marrow was observed. This reduction was accompanied by a striking decrease in the proportion of cycling cells in the bone marrow of thymus deprived mice. On the other hand, restoration of thymus function by thymic hormone (THF), by implantation of thymus in semi-impermeable cellophane bags, or by pregnancy

Parathyroid cyst of the thymus.

A case is described of primary hyperparathyroidism in a patient with both a parathyroid cyst within the thymus gland and a concomitant parathyroid adenoma. The parathyroid adenoma contained microcystic areas of degeneration, and it is thought that the parathyroid cyst reflected degenerative change in a pre-existing adenoma. Parathyroid cyst should be considered in the differential diagnosis of cystic lesions within the thymus. Fine needle aspiration and parathyroid hormone assay on cyst fluid may provide a preoperative diagnosis.

Epigenetic modifications in thymic epithelial cells: an evolutionary perspective for thymus atrophy.

BACKGROUND: The thymic microenvironment is mainly comprised of thymic epithelial cells, the cytokines, exosomes, surface molecules, and hormones from the cells, and plays a vital role in the development, differentiation, maturation and homeostasis of T lymphocytes. However, the thymus begins to degenerate as early as the second year of life and continues through aging in human beings, leading to a decreased output of naïve T cells, the limited TCR diversity and an expansion of monoclonal memory T cells in the periphery organs. These alternations will reduce the adaptive immune response to tumo

Comparison of the effects of the growth factor produced by Spirometra mansonoides and growth hormone in diabetic-hypophysectomized rats: lymphoid tissue.

Growth and development of the thymus is dependent on secretions from the anterior pituitary, presumably growth hormone. Diabetes mellitus is known to reduce immunological competence. These studies compare the effects of bovine growth hormone (bGH) and the growth factor produced by plerocercoid larvae of the tapeworm, Spirometra mansonoides, on metabolism of lymphoid tissue, thymus and spleen, in hypophysectomized rats made diabetic with a single intraperitoneal injection of alloxan. Whereas the control diabetic-hypophysectomized rats gradually lost weight throughout the experimental period, bo

Therapeutic application of calf thymus extract (TFX).

The semipurified calf thymus extract, TFX, produced since 1973 by the Polish Pharmaceutical Industry (POLFA) has been found to exert biological activities meeting criteria for thymic hormones. During the last 15 yr TFX has been evaluated independently by several Polish medical centers in over 1000 patients with symptomatically different but pathogenically similar human diseases, including primary immunodeficient states, bone marrow failure, autoimmune disorders, chronic skin diseases, recurrent viral and bacterial infectious diseases and some oncological disorders. Tuberculosis, the Disrupted Immune-Endocrine Response and the Potential Thymic Repercussion As a Contributing Factor to Disease Physiopathology.

Upon the pathogen encounter, the host seeks to ensure an adequate inflammatory reaction to combat infection but at the same time tries to prevent collateral damage, through several regulatory mechanisms, like an endocrine response involving the production of adrenal steroid hormones. Our studies show that active tuberculosis (TB) patients present an immune-endocrine imbalance characterized by an impaired cellular immunity together with increased plasma levels of cortisol, pro-inflammatory cytokines, and decreased amounts of dehydroepiandrosterone. Studies in patients undergoing specific treatm

SART stress effects on lymphocytes in the thymus and spleen of normal, adrenalectomized, and sympathectomized mice.

The effects of stress due to specific alteration of rhythm of temperature (SART) on lymphocyte subpopulations of the thymus and spleen were examined in C3H/He mice. Thymic and splenic involution were evident in SART-stressed mice, and lymphocyte subpopulations were significantly changed. In the thymus, CD4+ CD8+ and CD3- or CD3low lymphocytes were decreased by SART stress more than other lymphocytes, and CD4- and CD8-single-positive lymphocytes and B lymphocytes in the spleen were decreased equally. Immunoregulation via the adrenal glands and sympathetic nervous system was studied in chemicall

[Features of therapy of hematologic diseases in old age. 3. Features in the occurrence and clinical course of aplastic and neoplastic diseases of the blood].

The development and course of blood diseases in the aged often are determined by two factors: The first is the disturbance of the immune system based on a defective interaction between the T- and B-cell system. These are due to the involution of the thymus gland accompanied by reduced incretion of thymus hormones. The second factor consists in a regressive alteration of a "haemopoietic inductive micro-environment (Trentin), which has its morphological substrate in the circulatory system and the stroma of the bone marrow. Both factors lead to atrophic or diminished recovery of the bone marrow.

Gestation Regulates Growth Hormone and Its Receptor Expression in Sheep Immune Organs.

There are multiple adaptations in maternal physiology, hormones, and immunology during pregnancy. Growth hormone (GH) is not only produced by the pituitary but also secreted by extra-pituitary tissues. In this study, 24 ewes were randomly divided into four groups and mated with either adult intact rams (pregnant ewes) or a vasectomized ram (nonpregnant ewes), and maternal thymus, lymph node, spleen, and liver were harvested at day 16 of nongestation and at days 13, 16, and 25 of gestation. The specified primers for GH and GH receptor (GHR) were utilized to analyze mRNA expression of GH and GHR

Effect of thymus on the thyroid and pituitary thyrotropic function.

Thymus inflammation induced in infantile guinea pigs and rabbits by administration of melted paraffin drops into the thymic parenchyma resulted in enhancement of the thyroid functional activity. Thymosin injections provoked an identical effect. In both variants of these experiments the pituitary thyrotropin level remained within the norm. Hence, the pituitary thyrotropic function as in thyrotoxicosis lost the ability of response to enhancement of the thyroid hormone production. This result suggests that the cause of disorganization of the negative feedback between the thyroid and the pituitary
