Aberrant splicing of a nicotinic acetylcholine receptor alpha 6 subunit is associated with spinosad tolerance in the thrips predator Orius laevigatus.

Susceptibility to insecticides is one of the limiting factors preventing wider adoption of natural enemies to control insect pest populations. Identification and selective breeding of insecticide tolerant strains of commercially used biological control agents (BCAs) is one of the approaches to overcome this constraint. Although a number of beneficial insects have been selected for increased tolerance to insecticides the molecular mechanisms underpinning these shifts in tolerance are not well characterised. Here we investigated the molecular mechanisms of enhanced tolerance of a lab selected st

Evaluation of Alternatives to an Organophosphate Insecticide with Selected Cultural Practices: Effects on Thrips, Frankliniella fusca, and Incidence of Spotted Wilt in Peanut Farmscapes.

Peanut growers use a combination of tactics to manage spotted wilt disease caused by thrips-transmitted Tomato spotted wilt virus (TSWV). They include planting TSWV-resistant cultivars, application of insecticides, and various cultural practices. Two commonly used insecticides against thrips are aldicarb and phorate. Both insecticides exhibit broad-spectrum toxicity. Recent research has led to the identification of potential alternatives to aldicarb and phorate. In this study, along with reduced-risk, alternative insecticides, we evaluated the effect of conventional versus strip tillage; singl

Emergence and molecular characterization of a resistance-breaking tomato spotted wilt virus isolate on bell pepper plants harboring the Tsw resistance gene in Japan.

Tomato spotted wilt virus (TSWV) is distributed all over Japan and globally, seriously damaging infected plants. A resistance-conferring gene, Tsw, that controls the yellow spotted wilt disease in bell pepper (Capsicum annuum) plants caused by TSWV infection in Japan, is now commercially available. In this study, we isolated for the first time in Japan, a resistance-breaking isolate, TSWV-JRB, from bell pepper plants harboring Tsw. The virus overcame the resistance conferred by Tsw in its heterozygous and homozygous configurations. The host range or virulence of TSWV-JRB and one of the non-res

Complementarity between Orius predators improves control of foliar and flower pests.

BACKGROUND: Multispecies natural enemy assemblages may be more successful in suppressing herbivorous pests compared to low-diversity communities, especially when natural enemies complement each other regarding the niches they exploit. Orius predatory bugs are omnivorous biological control agents used in horticulture, and are widely associated with the control of flower thrips. However, species within the Orius genus may differ significantly in biological characteristics, such as size, thermal development requirements, induction of diapause, degree of omnivory, and within-plant distribution. Comparative Proteomics of Resistant and Susceptible Strains of Frankliniella occidentalis to Abamectin.

Western flower thrips, Frankliniella occidentalis (Thysanoptera: Thripidae) is an invasive agricultural pest with developed resistance to abamectin in some strains due to frequent treatment with the pesticide. In this study, we examined differentially expressed proteins (DEPs) between abamectin-resistant (AbaR; under abamectin selective pressure) and susceptible strains (AbaS; without abamectin selective pressure) of F. occidentalis. Proteins were isolated from second instar larvae of both strains and separated via two-dimensional polyacrylamide gel electrophoresis. Nano-flow liquid chromatogr

Three carboxylesterase genes are involved in spinosad resistance in Frankliniella occidentalis (Pergande).

Carboxylesterases (CarEs) are key enzymes in insect detoxification and metabolism, playing a crucial role in resistance development to various insecticides. In this study, CarE activity was significantly higher in spinosad-resistant strains of Frankliniella occidentalis (NIL-R) than in susceptible strains (Ivf03), suggesting a potential role of CarEs in spinosad resistance. RNA-seq and quantitative PCR (qPCR) validation confirmed that four CarE genes were significantly upregulated in NIL-R. RNA interference (RNAi) validated that three of these genes FoB1, FoB1L, and FoFE4 are directly involved

Toxicity of Ethanolic Plant Extracts to Aphis gossypii, Bemisia tabaci, and Frankliniella occidentalis and Selectivity to Coccinella septempunctata and Menochilus sexmaculatus.

The excessive use of synthetic chemical pesticides in agriculture has led to the emergence of resistant insect populations, posing significant threats to both ecosystems and human health. As part of sustainable pest management, Integrated Pest Management (IPM) strategies have increasingly incorporated eco-friendly alternatives, such as plant ethanolic extracts, to reduce chemical pesticide dependency. This study aimed to evaluate the effectiveness of ethanolic extracts from Cannabis sativa L., Citrullus colocynthis L. Schrad., and Laurus nobilis L. in controlling tomato crop pests. Repellency

Residual effect of commonly used insecticides on key predatory mites released for biocontrol in strawberry.

Florida is the second largest producer of strawberries in the United States. However, the production system faces numerous challenges, especially Scirtothrips dorsalis Hood (Thysanoptera: Thripidae) infestations. Management of this pest involves applying insecticides and use of predatory mites, particularly Amblyseius swirskii Athias-Henriot, Neoseiulus cucumeris Oudemans, and Neoseiulus californicus McGregor (Mesostigmata: Phytoseiidae). Strawberry growers in Florida are concerned about the compatibility of the commercial formulations of insecticides used in strawberry pest management with pr

Pesticide-mediated interspecific competition between local and invasive thrips pests.

Competitive interactions between species can be mitigated or even reversed in the presence of anthropogenic influences. The thrips species Frankliniella occidentalis and Thrips tabaci are highly invasive and damaging agricultural pests throughout the world. Where the species co-occur, one species tends to eventually predominate over the other. Avermectin and beta-cypermethrin are commonly used insecticides to manage thrips in China, and laboratory bioassays demonstrated that F. occidentalis is significantly less susceptible than T. tabaci to these insecticides. In laboratory cage trials in whi

Spinosad resistance, esterase isoenzymes and temporal synergism in Frankliniella occidentalis (Pergande) in Australia.

Spinosad has been widely used in Australia to control western flower thrips Frankliniella occidentalis (Pergande) but spinosad usefulness is now compromised by resistance. Here we studied a highly spinosad resistant strain of F. occidentalis to explore if esterases had a role in spinosad resistance. Enhanced esterase activity in pressured spinosad-resistant F. occidentalis was confirmed via PAGE electrophoresis and estimated to be approximately three times higher than that in a susceptible strain. Spinosad-esterase inhibition data in the resistant strain, showed a concentration effect with sig

Perception of solar UVB radiation by phytophagous insects: behavioral responses and ecosystem implications.

Most of our present knowledge about the impacts of solar UVB radiation on terrestrial ecosystems comes from studies with plants. Recently, the effects of UVB on the growth and survival of consumer species have begun to receive attention, but very little is known about UVB impacts on animal behavior. Here we report that manipulations of the flux of solar UVB received by field-grown soybean crops had large and consistent effects on the density of the thrips (Caliothrips phaseoli, Thysanoptera: Thripidae) populations that invaded the canopies, as well as on the amount of leaf damage caused by the

Comparative mitogenomics of six Thrips species reveals conserved architecture, repeat variation, and nucleotide composition bias.

Thrips are widely distributed insects with economic importance, but their evolutionary dynamics of the mitochondrial genome remain poorly understood. This study presented a computational mitogenomic analysis of six Thrips species obtained from a public repository. An analysis was performed to investigate patterns of genomic architecture and evolutionary variations. Mitochondrial genomes were analyzed using bioinformatic approaches, including annotation, syntenic comparison, gene content analysis, and SSR identification. Codon usage bias was measured using RSCU and CAI metrics, while skewness i

CRISPR/Cas9 knock-out of nAChR α6 confers resistance to spinosyns in Frankliniella occidentalis and is associated with a higher fitness cost than target-site mutation G275E.

Thrips are major agricultural pests globally and spinosyn insecticides like spinosad and spinetoram are commonly used for their control. However, numerous cases of resistance have emerged, often linked to mutations in the nicotinic acetylcholine receptor (nAChR) α6 subunit, the main molecular target of spinosyns. In this study, toxicological data for spinosad and spinetoram were obtained from a susceptible strain of Frankliniella occidentalis, as well as two field-collected resistant strains carrying the G275E resistance mutation. Notably, a new candidate resistance mutation never reported bef

Methyl hexenoate and linalool emitted by hot peppers repel western flower thrips (Frankliniella occidentalis) and attract their predator (Orius laevigatus).

A plant emits diverse volatile compounds and communicates with other organisms at different trophic levels. Hot pepper, Capsicum annuum, leaves produced different compositions of 41 organic compounds upon an infestation by the western flower thrips, Frankliniella occidentalis. These compounds induced a non-preference behavior to the thrips while they attracted a natural enemy, Orius laevigatus. Among the differentially emitted compounds, Z-3-methyl hexenoate (MeH) and linalool repelled the herbivore but attracted the predator. MeH or linalool biosynthesis was suppressed by virus-induced gene s

Nanoscale 3D anatomy of the western flower thrips reproductive system revealed by volume electron microscopy: extreme miniaturization and evolutionary adaptations for pest fecundity†.

Frankliniella occidentalis (western flower thrips) is a globally destructive pest causing major crop losses via oviposition, feeding, and plant virus transmission. Previous work has focused on its reproductive physiology and behavior, but comprehensive 3D structural knowledge remains limited. We used volume electron microscopy to achieve nanoscale 3D visualization of the reproductive systems in both sexes, providing the first high-resolution, in situ reconstruction, and establishing a new anatomical framework for insect reproductive physiology. Males exhibit extreme miniaturization, with repro

Binding characteristics and physicochemical basis of C-minus odorant-binding protein 8 in western flower thrips, Frankliniella occidentalis, with various host vegetable volatiles.

Western flower thrips (WFT), Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae), is a globally invasive polyphagous pest that relies on olfaction to sense host plants. Odorant-binding proteins (OBPs) in insect sensillar lymph mediate interactions with plant volatiles. However, the physicochemical mechanisms by which OBPs in F. occidentalis bind to host-plant volatiles, the key amino acid residues involved in these interactions and the structural characteristics of the preferred ligands remain unclear. Here, we cloned a novel OBP, FoccOBP8, which belongs to the C-minus OBP subfamil

Incorporation of the 15N-labeled simulated arthropod rain in the soil food web.

Direct trophic links between aboveground and belowground animal communities are rarely considered in food web models. Most invertebrate animals inhabiting aboveground space eventually become prey of soil predators and scavengers forming a gravity-driven spatial subsidy to detrital food webs, but its importance remains unquantified. We used laboratory-grown 15N-labeled Collembola to trace the incorporation of arthropod rain into soil food webs. Live or euthanized Collembola were supplemented once to field mesocosms in the amount equivalent to the mean daily input of the arthropod rain (19 mg d.

Mutation (G275E) of nAChR subunit Foα6 associated with spinetoram resistance in Australian western flower thrips, Frankliniella occidentalis (Pergande).

Western flower thrips, Frankliniella occidentalis is an economically important agricultural pest. It causes damage by feeding and oviposition or indirectly by plant virus transmission. Australian F. occidentalis are resistant to many insecticides including spinosad and the related chemical spinetoram. Spinetoram resistance to F. occidentalis has been recently reported in three different Australian States, however, mechanisms conferring that resistance have not been investigated. To identify the mechanisms underlying resistance to spinetoram in F. occidentalis, we sequenced the genomic region o
