Subcutaneous adipose tissue radiation attenuation is associated with chemotoxicity and survival in metastatic colorectal cancer.

BACKGROUND: Skeletal muscle mass and radiation attenuation (RA) have been shown to be associated with chemotoxicity and survival in cancer patients. However, little is known about the clinical significance of adipose tissue RA, which has been hypothesized to be associated with tissue inflammation and insulin resistance. We investigated the association between adipose tissue RA, chemotoxicity, and survival in patients with metastatic colorectal cancer (mCRC) by performing a secondary analysis using data from a randomized controlled trial. METHODS: We included 104 mCRC patients treated with sta

T cell-dependent survival of CD20+ and CD20- plasma cells in human secondary lymphoid tissue.

The signals mediating human plasma cell survival in vivo, particularly within secondary lymphoid tissue, are unclear. Human tonsils grafted into immunodeficient mice were therefore used to delineate the mechanisms promoting the survival of plasma cells. Tonsillar plasma cells were maintained within the grafts and the majority were nonproliferating, indicating a long-lived phenotype. A significant depletion of graft plasma cells was observed after anti-CD20 treatment, consistent with the expression of CD20 by most of the cells. Moreover, anti-CD52 treatment caused the complete loss of all graft

Long-term survival of human fetal pancreatic tissue transplanted into an insulin-dependent diabetic patient.

The explants of two human fetal pancreases of 15 weeks gestational age were cultured for 6-7 days before being implanted in a 29-year-old insulin-dependent diabetic woman who had received a renal graft two months previously. One pancreas was placed in the flexor muscles of the forearm whilst the other was implanted in an omental pouch. To reduce the chances of rejection the tissue was cultured in vitro, the donor of the tissue placed in the forearm was DR antigen matched with the recipient and the patient remained on cyclosporin and prednisone therapy. At 3 months a mass developed in the forea

Chronic Exposure to Field-Level Thiamethoxam Impairs Gut Tissue and Reduces Honeybee (Apis cerana) Survival.

Pesticides such as neonicotinoids frequently harm beneficial insect pollinators and affect their survival, social behavior, digestive system, and metabolism. Investigating the mechanisms behind these impairments is crucial for enhancing pesticide risk assessments. Apis cerana, a native honeybee species in Asia, has received limited research attention regarding the toxicological mechanisms of thiamethoxam (TMX) exposure. We exposed newly emerged worker bees of A. cerana to a field-relevant dose of TMX (400 ng/g) under laboratory conditions to examine whether TMX exposure triggers similar or dis

Plasma tissue factor activity in lung cancer patients predicts venous thromboembolism and poor overall survival.

BACKGROUND: Biomarkers to identify lung cancer (LC) patients with high risk of venous thromboembolism (VTE) are needed. OBJECTIVES: To evaluate the usefulness of plasma tissue factor activity (TFA) and D-dimer levels for the prediction of VTE and overall survival in patients with LC. METHODS: In a prospective multicenter observational cohort of consecutive LC patients, TFA and D-dimer levels were measured at diagnosis before any cancer treatment (V1) and between 8 and 12 weeks after diagnosis (V2). RESULTS: Among 302 patients, 38 (12.6%) experienced VTE within the first year after diagnosis

Genomic profiling of tissue and blood predicts survival outcomes in patients with resected pleural mesothelioma.

PURPOSE: Pleural mesothelioma (PM) is an aggressive tumor still considered incurable, in part due to the lack of predictive biomarkers. Little is known about the clinical implications of molecular alterations in resectable PM tissues and blood. Here, we characterized genetic alterations to identify prognostic and predictive biomarkers in patients with resected PM. EXPERIMENTAL DESIGN: Targeted next-generation sequencing was performed in retrospective pleural tumor tissue and paired plasma samples from stage IB-IIIB resected PM. Association between prognosis and presence of specific mutations

Conditional survival of pediatric, adolescent, and young adult soft tissue sarcoma and bone tumor patients.

BACKGROUND: Survival estimates for soft tissue sarcomas (STS) and malignant bone tumors (BT) diagnosed in pediatric, adolescent, and young adult patients are not easily available. We present survival estimates based on a patient having survived a defined period of time (conditional survival). Conditional survival estimates for the short-term were calculated for patients from diagnosis to the first five years after diagnosis and for patients surviving in the long-term (up to 20 years after diagnosis). METHODS: We identified 703 patients who were diagnosed with a STS or BT at age ≤25 years from

Survival and outcomes after lung transplantation for non-scleroderma connective tissue-related interstitial lung disease.

BACKGROUND: Patients with non-scleroderma connective tissue-related lung disease (NS-CTLD), including rheumatoid arthritis, idiopathic inflammatory myopathies, Sjögren syndrome, mixed connective tissue disease, and systemic lupus erythematosus, may be at risk for worse outcomes after lung transplantation because of immune dysregulation or extrapulmonary manifestations of their underlying disease. We compared survival, acute and chronic rejection, and extrapulmonary organ dysfunction after transplantation in patients with NS-CTLD and idiopathic pulmonary fibrosis (IPF). METHODS: This was a ret

Optimized cell survival and seeding efficiency for craniofacial tissue engineering using clinical stem cell therapy.

Traumatic injuries involving the face are very common, yet the clinical management of the resulting craniofacial deficiencies is challenging. These injuries are commonly associated with missing teeth, for which replacement is compromised due to inadequate jawbone support. Using cell therapy, we report the upper jaw reconstruction of a patient who lost teeth and 75% of the supporting jawbone following injury. A mixed population of bone marrow-derived autologous stem and progenitor cells was seeded onto β-tricalcium phosphate (β-TCP), which served as a scaffold to deliver cells directly to the d

Oxygen diffusion through collagen scaffolds at defined densities: implications for cell survival in tissue models.

For the success of any biomaterial for tissue engineering, its mechanical properties and ability to support nutrient diffusion will be critical. Collagen scaffolds are ideal candidates, due to their ability to immerse cells in a biomimetic nanofibrous matrix. We have established O(2) diffusion coefficients through native, dense collagen scaffolds at two tissue-like densities, with and without photo-chemical crosslinking, by adapting an optical fibre-based system for real-time core O(2) monitoring deep within collagen constructs. Using a Fick's law model, we then derived O(2) diffusion coeffici

Long-term composite tissue allograft survival in a porcine model with cyclosporine/mycophenolate mofetil therapy.

BACKGROUND: Low-dose cyclosporine (CsA)/mycophenolate mofetil (MMF) therapy has significantly reduced the frequency of rejection and drug-induced side effects in rat hindlimb allograft recipients. With an eye toward direct clinical application, we developed a large-animal extremity composite tissue allograft model to assess the antirejection efficacy and systemic toxicity of combination CsA/MMF treatment. METHODS: Radial forelimb osteomyocutaneous flap transplants were performed between size-matched, outbred pigs assigned to one of two groups: 5 control pigs received no immunosuppression, and

Cardiac valvular disease in chronic inflammatory disorders of connective tissue. Factors influencing survival after surgery.

Three cases of cardiac valvular disease complicating chronic inflammatory disorders of connective tissue are described. Each underwent surgical valve replacement. The outcome in the first case, a patient with aortitis complicating Reiter's syndrome and in good health otherwise, has been excellent. The other two patients, one with rheumatoid arthritis and vasculitis, the other with systemic lupus erythematosus and nephritis, died after operation as the result of their systemic disease. The importance of systemic features in assessing this type of patient is emphasized.

[Effects of thymus tissue transplantation on the survival of mice exposed to a lethal X-Ray dose.].

During the radiation exposure of the body, processes similar to those characteristic of the natural aging of mammals occur. This phenomenon is most often referred to as accelerated aging. By transplanting the thymus tissue, it is possible to achieve a normalization of food and water consumption, a stabilization of weight gain, and a significant increase in the life expectancy of experimental animals, even after their lethal irradiation.

Strategies to Enhance Implantation and Survival of Stem Cells After Their Injection in Ischemic Neural Tissue.

High post-transplantation cell mortality is the main limitation of various approaches that are aimed at improving regeneration of injured neural tissue by an injection of neural stem cells (NSCs) and mesenchymal stromal cells (MStroCs) in and/or around the lesion. Therefore, it is of paramount importance to identify efficient ways to increase cell transplant viability. We have previously proposed the "evolutionary stem cell paradigm," which explains the association between stem cell anaerobic/microaerophilic metabolic set-up and stem cell self-renewal and inhibition of differentiation. Effects of creatine treatment on the survival of dopaminergic neurons in cultured fetal ventral mesencephalic tissue.

Parkinson's disease is a disabling neurodegenerative disorder of unknown etiology characterized by a predominant and progressive loss of dopaminergic neurons in the substantia nigra. Recent findings suggest that impaired energy metabolism plays an important role in the pathogenesis of this disorder. The endogenously occurring guanidino compound creatine is a substrate for mitochondrial and cytosolic creatine kinases. Creatine supplementation improves the function of the creatine kinase/phosphocreatine system by increasing cellular creatine and phosphocreatine levels and the rate of ATP resynth
