Ursodeoxycholic acid against TGEV infection via the JAK-STAT1 signaling pathway.

Transmissible gastroenteritis virus (TGEV) causes transmissible gastroenteritis (TGE), leading to severe diarrhea, vomiting, and dehydration in pigs. Mortality rates among newborn piglets are particularly alarming, causing significant economic losses to the livestock industry. In the absence of effective antiviral drugs, developing novel antiviral agents is crucial. Ursodeoxycholic acid (UDCA), as a hydrophilic endogenous bile acid, plays a key role in regulating bile acid metabolism and treating hepatobiliary diseases. Studies indicate that UDCA exhibits specific antiviral effects against the

[Norovirus (infants/children)].

Human norovirus (NoV) is an important cause of the epidemic acute gastroenteritis in children and adults. This review mentions on the required knowledge to understand morphology, genetics, transmission, pathogenesis, and control. The development of molecular diagnostic techniques is clarified the epidemiological impact that is now recognized not only as the major cause of foodborne gastroenteritis outbreaks but also as a cause of sporadic gastroenteritis. It was recognized that the combination pattern to an intestinal epithelium of NoV was prescribed in organization, blood type antigen (histob

Survival of surrogate coronaviruses in water.

The emergence of a previously unknown coronavirus infection, Severe Acute Respiratory Syndrome (SARS), demonstrated that fecally contaminated liquid droplets are a potential vehicle for the spread of a respiratory virus to large numbers of people. To assess potential risks from this pathway, there is a need for surrogates for SARS coronavirus to provide representative data on viral survival in contaminated water. This study evaluated survival of two surrogate coronaviruses, transmissible gastroenteritis (TGEV) and mouse hepatitis (MHV). These viruses remained infectious in water and sewage for

Activation of the NLRP1 Inflammasome and Its Role in Transmissible Gastroenteritis Coronavirus Infection.

The inflammasome pathway is a critical early response mechanism of the host that detects pathogens, initiates the production of inflammatory cytokines, and recruits effector cells to the infection site. Nonetheless, the mechanism of inflammasome activation in coronavirus infection and its biological functions in host defense remain unclear. Transmissible gastroenteritis virus (TGEV), a member of the genus Alphacoronavirus, is a significant pathogen that mainly infects piglets and causes intestinal inflammation and inflammatory cell infiltration. Here, we investigated the mechanism of inflammas

Isolation of porcine respiratory coronavirus from pigs affected with porcine reproductive and respiratory syndrome.

Four cytopathogenic viruses were isolated in CPK cells derived from porcine kidneys from tonsils and lungs of 3 of 15 pigs affected with porcine reproductive and respiratory syndrome virus. Physicochemically and morphologically, the isolates were similar to a coronavirus. The isolates were not distinguished from transmissible gastroenteritis virus (TGEV) by a neutralization test using polyclonal antibodies, but differentiated from TGEV by monoclonal antibodies capable of discriminating between TGEV and porcine respiratory coronavirus (PRCV), indicating that the isolates were PRCV. In a serolog

Experimental infection of Macaca nemestrina with a Toronto Norwalk-like virus of epidemic viral gastroenteritis.

Norwalk virus (NV) and Norwalk-like viruses (NLVs) are common etiologic agents of viral gastroenteritis. Viral gastroenteritis is a common disease that is highly transmissible, spreading rapidly through families, institutions, and communities. Because methods for in vitro cultivation of Norwalk etiologic agents are not available, information regarding this syndrome has come largely from studies in human volunteers. Sequential passaging of an NLV through an immunoincompetent newborn pigtail macaque (Macaca nemestrina) may allow for the adaptation of a human NLV to a primate host, thus providing

DYRK1A is a multifunctional host factor that regulates coronavirus replication in a kinase-independent manner.

Coronaviruses (CoVs) pose a major threat to human and animal health worldwide, which complete viral replication by hijacking host factors. Identifying host factors essential for the viral life cycle can deepen our understanding of the mechanisms of virus-host interactions. Based on our previous genome-wide CRISPR screen of α-CoV transmissible gastroenteritis virus (TGEV), we identified the host factor dual-specificity tyrosine phosphorylation-regulated kinase 1A (DYRK1A), but not DYRK1B, as a critical factor in TGEV replication. Rescue assays and kinase inhibitor experiments revealed that the

PPP2R5B regulates ANPEP expression and TGEV entry via dephosphorylation of HSF1 at Ser304/Ser308.

Transmissible gastroenteritis virus (TGEV) is an enteric coronavirus responsible for severe gastrointestinal disease in swine; however, the host determinants and mechanisms underlying viral entry remain poorly defined. This study systematically examines the role of the serine/threonine protein phosphatase 2A (PP2A) regulatory subunit B'β (PPP2R5B) during TGEV infection. Genetic ablation of PPP2R5B in PK-15 cells significantly reduced TGEV infectivity. Stage-specific analyses revealed that loss of PPP2R5B selectively impaired viral entry, while viral replication and release were unaffected. Tra

Tight junction protein claudin-1 is a novel internalization factor for swine enteric coronaviruses infection.

UNLABELLED: Swine enteric coronaviruses (SeCoVs), including transmissible gastroenteritis virus (TGEV), porcine epidemic diarrhea virus (PEDV), and porcine deltacoronavirus (PDCoV), have been reported to use aminopeptidase N (APN) as a cellular receptor. However, APN alone cannot effectively explain the infection of both APN-positive and APN-negative enterocytes by PEDV and TGEV, nor the wide host range of PDCoV, suggesting the involvement of other host factors. In this study, we demonstrate that TGEV infection in piglets upregulates claudin-1 expression not only in infected cells but also in

Infectious stability of animal gastrointestinal coronaviruses in fasted-state simulated gastric fluid.

Animal gastrointestinal coronaviruses such as porcine epidemic diarrhea virus (PEDV) and transmissible gastroenteritis virus (TGEV) can cause oral infection despite being enveloped viruses, implying that they are capable of passing through the highly acidic gastric lumen. This study focused on the low pH acid resistance of coronaviruses, including PEDV, TGEV and equine coronavirus (ECoV) in fasted-state simulated gastric fluid (FaSSGF) at pH 1.6, which simulated gastric fluid. Interestingly, PEDV, TGEV and ECoV retained infectivity in FaSSGF, whereas SARS-CoV-2 was rapidly inactivated. Althoug

Impact of TGEV infection on the pig small intestine.

BACKGROUND: Pig diarrhea causes high mortality and large economic losses in the swine industry. Transmissible gastroenteritis virus (TGEV) causes pig diarrhea, with 100% mortality in piglets less than 2 weeks old. No investigation has yet been made of the small intestine of piglets that survived infection by TGEV. METHODS: In this study, we evaluated the impact of TGEV infection on the small intestine of recovered pigs. RESULTS: Histological analyses showed that TGEV infection led to villi atrophy, and reduced villous height and crypt depth. The number of SIgA positive cells, CD3+T cells, an

All-trans retinoic acid protects piglets from TGEV-induced diarrhea and intestinal epithelial apoptosis by modulating redox status and endoplasmic reticulum stress pathways.

Transmissible gastroenteritis virus (TGEV) is a significant pathogen responsible for diarrhea in piglets, with its pathogenesis intricately associated with apoptosis. As a bioactive vitamin A derivative, all-trans retinoic acid (ATRA) possesses antiviral, antioxidant and anti-apoptotic characteristics. This research aimed to explore whether ATRA could mitigate TGEV infection in piglets through inhibition of intestinal epithelial apoptosis. In a 19-day trial, 32 piglets were randomized into four treatments: non-challenged group (Control), TGEV-challenged group (TGEV), TGEV+ 5 mg/d ATRA group (T

Establishment of a pseudovirus neutralization assay for TGEV.

INTRODUCTION: Transmissible Gastroenteritis Virus (TGEV) is a major pathogen causing swine enteric diseases, necessitating effective control strategies. Vaccination plays a key role, but assessing vaccine efficacy remains challenging due to variations in immune response and existing detection limitations. Current antibody detection methods, such as neutralization assays and ELISA, are often subjective, labor-intensive, and time-consuming, highlighting the need for a more efficient evaluation approach. METHODS AND RESULTS: The TGEV S gene was amplified and inserted into the eukaryotic vector P

An immunohistochemical investigation of porcine epidemic diarrhoea.

A sudden outbreak of epidemic diarrhoea of piglets occurred in Japan, the principal features being watery diarrhoea, dehydration and high mortality in newborn animals. The microscopical lesions were villous atrophy in the small intestine, the villous enterocytes being vacuolated and cuboidal in shape. The villus-crypt ratio was severely reduced, varying from 1:1 to 3:1. Transmission electron microscopy showed numerous coronaviruses within the cytoplasm of enterocytes and among microvilli. Specific antigens of porcine epidemic diarrhoea (PED) virus were detected in the cytoplasm of enterocytes

Production of porcine aminopeptidase N (pAPN) site-specific edited pigs.

Porcine viral diarrhea is an acute and highly contagious enteric disease in pigs which causes huge economic losses in pig industry worldwide. Transmissible gastroenteritis virus (TGEV) is main pathogens responsible for piglets viral diarrhea. Knockout the host cellular surface receptor for TGEV may be an effective way to accelerate the breeding of resistant pigs. In this study, we applied site-specific editing pAPN which is effective in swine testis (ST) cells. Site-specific editing of pAPN reduced TGEV proliferation in ST cells by 96%-99% at different time periods post-infection. Next, the si

Norwalk-like gastroenteritis epidemic in a Toronto hospital.

An epidemic of gastroenteritis in a teaching hospital affected 57 patients and 69 staff over a 26-day period. The index case was a patient admitted with acute abdominal pain and diarrhea two days prior to the outbreak. The epidemic curve indicated person-to-person transmission. The incubation period, duration and types of symptoms were typical of Norwalk gastroenteritis, and Norwalk-like virus particles, serologically different from the prototype Norwalk virus strain, were observed in 17 of 20 fecal specimens examined by immune-electron microscopy.

Efficacy of a transmissible gastroenteritis coronavirus with an altered ORF-3 gene.

Serial passage of virulent transmissible gastroenteritis virus through cell culture reduced its virulence in 3-day-old piglets. Intramuscular inoculation of pregnant gilts with 2 doses of this modified-live virus elicited a level of lactogenic immunity that protected their nursing piglets against a lethal dose of challenge virus. Sequence analysis of a 637-bp fragment of the spike gene containing most of the aminopeptidase receptor and the 4 major antigenic sites from the original and the serially passed viruses were nearly identical. Gel analysis revealed that the fragment from the ORF-3 gene

A phase trial of the oral Lactobacillus casei vaccine polarizes Th2 cell immunity against transmissible gastroenteritis coronavirus infection.

Transmissible gastroenteritis coronavirus (TGEV) is a member of the genus Coronavirus, family Coronaviridae, order Nidovirales. TGEV is an enteropathogenic coronavirus that causes highly fatal acute diarrhoea in newborn pigs. An oral Lactobacillus casei (L. casei) vaccine against anti-transmissible gastroenteritis virus developed in our laboratory was used to study mucosal immune responses. In this L. casei vaccine, repetitive peptides expressed by L. casei (specifically the MDP and tuftsin fusion protein (MT)) were repeated 20 times and the D antigenic site of the TGEV spike (S) protein was r
