Severe cases of Buruli ulcer (infection with Mycobacterium ulcerans) in common ringtail possums in Victoria adversely affect animal welfare.

Buruli ulcer is a chronic ulcerative disease of the skin and subcutaneous tissues caused by infection with Mycobacterium ulcerans. Although Australian possums are known to be susceptible to Buruli ulcer, many aspects of the disease in possums, including welfare impacts, remain largely unreported. Severe clinical Buruli ulcer was identified in four common ringtail possums (Pseudocheirus peregrinus) from Melbourne, Victoria. All four possums were euthanased due to the presence of deep ulcerative lesions on paws, with extensive tissue necrosis that exposed bones and tendons in three cases. Histol

Experimental infection of ringtail possums (Pseudocheirus peregrinus) with Mycobacterium ulcerans, the agent of Buruli ulcer.

Buruli ulcer (BU) is a necrotizing disease of skin and soft tissue caused by the bacterium Mycobacterium ulcerans (MU). In Australia, where the disease is emerging in new geographic areas and human case numbers are increasing, native possum species act as reservoir hosts. To better understand the life history of MU in one of its natural hosts, we conducted intra-dermal challenge of six wild caught, MU-naïve common ringtail possums (Pseudocheirus peregrinus). All six animals developed BU disease consistent with that observed in naturally infected ringtail possums. Time to ulceration varied betw

Sustained protection against tuberculosis conferred to a wildlife host by single dose oral vaccination.

BACKGROUND: Vaccination of wildlife against bovine tuberculosis (TB) is being considered by several countries to reduce the transmission of Mycobacterium bovis infection to livestock. In New Zealand, where introduced brushtail possums (Trichosurus vulpecula) are the major wildlife hosts, we have previously shown that repeat applications of a lipid-encapsulated oral bacille Calmette-Guerin (BCG) vaccine reduce the incidence of naturally acquired TB in wild possums. Here we extend this conceptual demonstration to an operational level, assessing long-term protection against TB conferred to free-l

Mycobacterium ulcerans in Possum Feces before Emergence in Humans, Australia.

We describe emergence of Buruli ulcer in urban Geelong, Victoria, Australia, and examine timing and proximity of human cases to detection of Mycobacterium ulcerans DNA in possum feces. M. ulcerans-positive feces preceded human cases by up to 39 months, constituting an early warning of impending risk for Buruli ulcer.

Serological evidence of Coxiella burnetii exposure in native marsupials and introduced animals in Queensland, Australia.

The state of Queensland has the highest incidence of Q fever in Australia. In recent years, there has been an increase in human cases where no contacts with the typical reservoir animals or occupations were reported. The aim of this study was to determine the seroprevalence of Coxiella burnetii in Australian native animals and introduced animals in northern and southeastern Queensland. Australian native marsupials sampled included the brushtail possum (Trichosurus vulpecula) and common northern bandicoot (Isoodon macrourus). Introduced species sampled included dingoes (Canis lupus dingo), cats

Karyotype relationships between four distantly related marsupials revealed by reciprocal chromosome painting.

Marsupial karyotypes have shown extensive conservation even between distantly related groups with a high diversity of life forms and reproductive biology. Banding analysis has been the main test for assessing their homologies and chromosome rearrangements. More recently, cross-species reciprocal chromosome painting has been developed and applied to several mammalian species and has shown homologies and rearrangements not revealed by banding analysis. Karyotype relationships between four marsupial species, Sminthopsis crassicaudata, Potorous tridactylus, Trichosurus vulpecula and Macropus eugen

Experimental infections of brush-tailed possums, common wombats and water rats with Leptospira interrogans serovars balcanica and hardjo.

Of 12 brush-tailed possums (Trichosurus vulpecula) inoculated with Leptospira interrogans serovar balcanica 11 developed migroagglutination (MA) antibody to jardjo antigen by 14 days postincubation (PI). Leptospiruria was observed in 2 possums 117 to 145 days PI. Of 6 possums inoculated with serovar hardjo 4 developed low short-lived titres by day 18 PI. Two of 3 wombats (Vombatus ursinus) inoculated with balcanica had high MA titres (greater than or equal to 1:128) by day 16 PI and leptospiruria occurred by day 16. One wombat inoculated with hardjo developed a low MA titre. Low transitory MA

Leptospirosis in brushtail possums: is Leptospira interrogans serovar balcanica environmentally transmitted?

In New Zealand, the biological control of introduced brushtail possums (Trichosurus vulpecula) may be the only affordable option for achieving a significant long term reduction in pest numbers on a national scale. Leptospira interrogans serovar balcanica is among the potential biocontrol agents and vectors currently being investigated for this purpose. As the transmission pathways of L. interrogans serovar balcanica between possums are poorly understood, the objective of the study was to determine whether infection could result from exposure to contaminated environments. Buruli ulcer surveillance in south-eastern Australian possums: Infection status, lesion mapping and internal distribution of Mycobacterium ulcerans.

Buruli ulcer (BU) is a neglected tropical disease of skin and subcutaneous tissues caused by Mycobacterium ulcerans. BU-endemic areas are highly focal, and M. ulcerans transmission dynamics vary by setting. In Victoria, Australia, BU is an endemic vector-borne zoonosis, with mosquitoes and native possums implicated in transmission, and humans incidental hosts. Despite the importance of possums as wildlife reservoirs of M. ulcerans, knowledge of BU in these animals is limited. Opportunistic necropsy-based and active trap-and-release surveillance studies were conducted across Melbourne and Geelo

Tawny frogmouths and brushtail possums as sentinels for Angiostrongylus cantonensis, the rat lungworm.

The objective of this study was to determine the prevalence of angiostrongylosis in tawny frogmouths (Podargus strigoides) and brushtail possums (Trichosurus vulpecula) with signs of neurological disease, and to describe the clinicopathological features of angiostrongylosis in both species. Tawny frogmouths and brushtail possums with signs of neurological disease were sampled from the Sydney metropolitan area between October 1998 and June 2010. Samples from 100 tawny frogmouths and 31 brushtail possums from the Australian Registry of Wildlife Health (ARWH), the Wildlife Assistance and Informat

Heritable transgenesis of Parastrongyloides trichosuri: a nematode parasite of mammals.

Germline transformation of a parasitic nematode of mammals has proven to be an elusive goal. We report here the heritable germline transformation of Parastrongyloides trichosuri, a nematode parasite whose natural hosts are Australian possums of the genus Trichosurus. This parasite can undergo multiple free-living life cycles and these replicative cycles can be maintained indefinitely in the laboratory. Transformation was achieved by microinjection of DNA into the ovary syncytium of either free-living or parasitic adult females. By selecting for the transgenic progeny of successive free-living

Efferent connections of the main olfactory bulb in the opossum (Monodelphis domestica): a characterization of the olfactory entorhinal cortex in a marsupial.

Olfactory projections have been investigated for decades, but few reports using modern, sensitive neural tracers are available. In marsupials, only lesion-degeneration studies exist and they are restricted to the genera Didelphis and Trichosurus. Some of the territories described as olfactory-recipient such as the upper portion of the rhinal fissure and the vomeronasal amygdala are, however, controversial. Also, the characterization of the olfactory portion of the entorhinal cortex is far from clear in acallosal mammals. The present report investigates, using biotinylated dextran-amine, the ol

Digesta retention and fibre digestion in brushtail possums, ringtail possums and rabbits.

1. Mean retention times (MRTs) of fluid (marked with Co-EDTA), fine particles (mordanted with Yb) and large particles (mordanted with Cr) were measured in brushtail possums (Trichosurus vulpecula), ringtail possums (Pseudocheirus peregrinus) and laboratory rabbits fed semipurified diets. 2. In brushtail possums there were no significant differences in MRT among the three digesta markers. 3. In ringtail possums MRTs of the fluid and fine particle markers were approximately twice that of the large particle marker, indicative of selective retention of both fluid and fine particles in the caecum.

Bio-sensor system discriminating between the biliary and pancreatic ductal systems.

Sphincter of Oddi manometry is the "gold standard" to evaluate the ductal anatomy of patients with suspected sphincter of Oddi dysfunction. During such procedure it can be unclear which duct is being investigated without radiographs. Bilirubin and other pigments in bile strongly absorb blue light, whereas pancreatic fluid is clear and does not absorb blue light. The bio-sensor system incorporates fibre-optic technology to measure blue light absorbance in the ducts. Bench tests and animal tests were conducted utilising possums. In bench and animal experiments the bio-sensor correctly discrimina

Skeletal fluorosis in marsupials: A comparison of bone lesions in six species from an Australian industrial site.

In this study we explored the prevalence, type, location and severity of skeletal lesions in six species of Australian marsupial (Macropus giganteus, Notamacropus rufogriseus, Wallabia bicolor, Phascolarctos cinereus, Trichosurus vulpecula and Pseudocheirus peregrinus) from high and low-fluoride environments. Lesions occurred to varying extents in all species, and lesion distribution varied with biomechanical differences in gait and mastication. Bone fluoride levels increased with severity of periosteal hyperostosis. The mean bone fluoride concentration of individuals lacking hyperostosis (acr

Protection of free-living and captive possums against pulmonary challenge with Mycobacterium bovis following oral BCG vaccination.

In New Zealand, possums (Trichosurus vulpecula) are the main wildlife reservoir for bovine tuberculosis (Tb), which they transmit to livestock. This study investigated oral vaccination with lipid-formulated Mycobacterium bovis BCG and subsequent protection against virulent M. bovis challenge in wild-caught possums. Possums were trapped from the field and either hand-vaccinated and released back into the wild, or acclimatised to captive conditions prior to voluntary uptake of flavoured vaccine. Possums were subsequently exposed to pulmonary challenge with virulent M. bovis, administered either

Progress in the development of tuberculosis vaccines for cattle and wildlife.

Vaccination against bovine tuberculosis is likely to become an important disease control strategy in developing countries, which cannot afford a test and slaughter control programme, or in countries which have a wildlife reservoir of Mycobacterium bovis infection. In the past decade, considerable progress has been made in the development and evaluation of tuberculosis vaccines for cattle and for a range of wildlife maintenance hosts including possums, badgers, deer and African buffaloes. Experimental challenge systems have been established for the different target species and the resulting dis

Uptake and persistence of the vertebrate pesticide, sodium monofluoroacetate (compound 1080), in plants of cultural importance.

Field research was undertaken to determine if naturally occurring plants utilised by a Maori (the indigenous people of New Zealand) community for food and medicine would take up the toxin sodium monofluoroacetate (Compound 1080) from baits used to control the brush-tailed possum Trichosurus vulpecula. Single baits were placed at the base of individual plants of two species, pikopiko (Asplenium bulbiferum) and karamuramu (Coprosma robusta). Plants were sampled at various times up to 56 days, and samples analyzed for 1080 content. No 1080 was detected in any of the pikopiko samples, whereas 1080

A sero-epidemiological study of Leptospira interrogans serovar Balcanica in four brush-tailed possum populations in Victoria, Australia.

The microscopic agglutination (MA) test was utilised to study the prevalence of antibodies to Leptospira interrogans serovar hardjo in 4 populations of brush-tailed possums (Trichosurus vulpecula). The overall antibody prevalence varied from 14% to 66%; however, the age distribution of MA test titres was remarkably similar in all 4 populations. Antibody prevalence was similar in both males and females and demonstrable antibodies were limited to sexually mature animals. The greater prevalence of high titres (greater than or equal to 1:128) in the 18- to 24-month age group suggested that primary

Transmission of Leptospira interrogans serovar Balcanica infection among socially housed brushtail possums in New Zealand.

Leptospira interrogans serovar balcanica is a potential vector being investigated for spreading a biological control agent among introduced brushtail possums (Trichosurus vulpecula) in New Zealand. As previous studies have shown that possums are unlikely to contract leptospirosis through a contaminated environment alone, the objective was to determine whether L. interrogans serovar balcanica could be transmitted between sexually mature, socially housed possums. Possums were infected experimentally with L. interrogans serovar balcanica and housed in pairs or groups with uninfected possums for e
