Dynamics of serum rT3 and 3,3'-T2 during rT3 infusion in patients treated for thyrotoxicosis with propylthiouracil or methimazole.

rT3 metabolism in patients treated for thyrotoxicosis with prophylthiouracil (PTU), or methimazole (MMI) was studied by infusion of rT3 and measurements of the increase in serum rT3 and serum 3,3'-diiodothyronine. The results indicate that the high serum rT3 observed during treatment with PTU is not due to an increase in rT3 production, but to a decrease in the metabolic clearance rate of rT3. rT3 infusion was followed by an increase in serum 3,3'-T2 which was similar whether PTU or MMI was given. However, after stopping rT3 infusion there was a more rapid fall serum 3,3'-T2 during MMI treatme

[Reciprocal changes in T3 nd rT3 serum concentrations in digestive diseases resulting in childhood protein-calorie malnutrition (author's transl)].

In 14 children affected by digestive diseases producing protein-calorie malnutrition T3 and rT3, T4, TSH serum levels and TBG and TBPA maximum binding capacity were measured at the moment of the hospitalisation and up to their clinical and biochemical amelioration. No changes in serum T4 and TSH levels and in TBGcap values were detected. TBPAcap was found to be persistently low. At the moment of the hospitalisation serum T3 and rT3 average values were found to be 65 +/- 25 ng/dl and 58 +/- 25 ng/dl respectively. Opposite changes in serum concentrations of the triiodothyronines were observed at

Treatment of neonatal hyperthyroidism due to Graves' disease with sodium ipodate.

We describe the effect of administration of repeated doses of sodium ipodate in a newborn infant with hyperthyroidism due to transient Graves' disease. Pretreatment (day 3) serum T4 and T3 concentrations were 49 micrograms/dl and 590 ng/dl, respectively. With 24 h after the first dose of ipodate, serum T3 fell by 40%, and it subsequently ranged from 209-278 ng/dl throughout the 39-day ipodate treatment period. Serum T4 also decreased after ipodate administration to 69% and 41% of the pretreatment value after 72 h and 7 days of treatment, respectively; values thereafter during treatment ranged

Effects of sodium ipodate and propylthiouracil in athyreotic human subjects, role of triiodothyronine and pituitary thyroxine monodeiodination in thyrotrophin regulation.

To investigate the respective role of triiodothyronine (T3) and thyroxine (T4) in the regulation of TSH secretion, we studied the action of sodium ipodate and propylthiouracil (PTU) in 11 athyreotic patients. The LT4 replacement dose was adjusted to obtain, in each patient, a normal basal TSH level and a normal TSH response to TRH. In the 5 ipodate-treated patients (single 6 g oral dose), the mean serum T3 level fell by 64% below the baseline value and serum rT3 rose 180% above the baseline. The free T4 index (FT4I) did not change whereas the mean serum TSH concentration increased 280% above b

Propylthiouracil (PTU) pharmacology in the rat. II. Effects of PTU on thyroid function.

Using a sensitive and specific RIA for propylthiouracil (PTU), we examined the effects of short term (1 week) and long term (1 month) PTU treatment on thyroid function in the rat, and correlated changes in thyroid function with serum and thyroid PTU levels. After 1 week, dose-dependent decreases in thyroid PBI, serum T4, and serum T3 were observed, with concomitant elevations in the serum rT3 to T4 ratio and serum TSH. Fifty percent suppression of thyroid PBI occurred at a PTU concentration in the drinking water of 0.0005% (ED50), with concomitant serum and thyroid PTU levels of 0.3 micrograms

Hyperthyroxinemia in patients treated with high-dose propranolol.

Six patients with hyperthyroxinemia (five men and one woman) were evaluated for possible hyperthyroidism. All were taking large daily doses of propranolol--480 +/- 155 (+/- SE) mg--for treatment of angina pectoris. The patients had no clinical evidence of hyperthyroidism, although three had small goiters. Further evaluation of the patients revealed elevated serum free thyroxine levels and/or free thyroxine index, low-normal serum triiodothyronine levels, and elevated serum reverse triiodothyronine levels in all six. The thyroid-stimulating hormone response to thyrotropin-releasing hormone was

Neonatal thyroid function and anaesthesia.

Thyroid function in the neonate and its response to hexobarbitone anaesthesia were studied in newborn Wistar rats between 24 h and 42 days old. 14C-labelled mixed with unlabelled hexobarbitone was injected i.p. in anaesthetic concentrations. Serum T3, reverse T3 (rT3) and T4 and serum and whole brain hexobarbitone concentrations were measured on wakening. A decrease in serum T3 and rT3 and an increase in T4 concentrations were observed after hexobarbitone at 21 and 30 days. This appears to be a critical period in the neonatal rat. A significant negative correlation was found between the change

Characteristics of Dyslipidemia in Adults With Resistance to Thyroid Hormone β.

OBJECTIVE: To assess determinants of dyslipidemia (specifically raised serum low-density lipoprotein cholesterol and triglycerides, and reduced high-density lipoprotein cholesterol) in individuals with Resistance to Thyroid Hormone β (RTHβ). METHODS: Multivariable linear regression analyses of cross-sectional fasting serum lipid profiles from 105 genetically-confirmed individuals with RTHβ (mean age 40.0 years, 40% males) were conducted, including candidate variables with plausible independent associations with the individual components of dyslipidemia such as indices of thyroid function, pla

Uptake of reverse T3 in the human choriocarcinoma cell line, JAr.

The uptake and efflux of reverse triiodothyronine (rT3) in JAr cells were investigated. Uptake of 125I-rT3 was time dependent and reversible with a saturable component of around 70 per cent of total uptake after 30 min of incubation. Efflux was not saturable. Kinetic analysis of the initial specific uptake rates revealed an uptake process with a Michaelis constant of 3.04+/-0.53 microM (mean+/-SEM, n=15) and a corresponding maximum velocity of 9.65+/-2.49 pmol/min/mg protein (n=15). Uptake of rT3 was stereospecific, but not specific for rT3, as unlabelled L stereoisomers of thyroid hormone ana

Thyroid function and thyroid size in normal pregnant women living in an iodine replete area.

OBJECTIVE: The interpretation of the changes in thyroid hormone concentrations during normal pregnancy is a matter of debate involving, in some geographical regions, enhanced thyroid activity in early pregnancy and a hypothyroid state in the third trimester. A clinically detectable increase in thyroid size has been found in areas of mild iodine deficiency ('goitre of pregnancy'), but not in iodine replete areas. DESIGN: A prospective study. We have studied thyroid size and function in normal pregnant women living in an iodine replete area. PATIENTS: Healthy women before and during a normal p

Thyrotoxicosis without elevated serum triiodothyronine levels during diabetic ketoacidosis.

Thyrotoxicosis with a normal serum triiodothyronine (T3) concentration has been described with a variety of acute and chronic illnesses occurring in association with thyrotoxicosis. We describe the first case to our knowledge of thyroxine (T4) toxicosis in a 16-year-old boy with diabetic ketoacidosis. Although the clinical manifestations of hyperthyroidism were mild, thyromegaly and persistent tachycardia suggested thyrotoxicosis. Serum T4 levels were elevated; however, the serum T3 level was normal. Measurement of reverse T3 (rT3) initially revealed an elevated level that decreased over sever

The influence of endogenous cortisol on the peripheral conversion of thyroxine in patients with acute myocardial infarction.

A study was performed to elucidate whether endogenous cortisol, as previously suggested, could be responsible for the decreased T3 levels seen in euthyroid patients with acute myocardial infarction. Levels of these hormones as well as levels of T4 and reverse-T3 were monitored in 31 consecutive patients admitted to the Coronary Care Unit with symptoms of precordial pain or with acute arrhythmias. Sixteen of the patients had proven myocardial infarction, the remaining 15 were used as a control group. The results demonstrated that a reduction of T3 levels was seen in the infarction group without

[The effect of age on parameters of thyroid function].

The study was aimed at determination of the effect of age on the function of thyroid gland in all consecutive phases of human life--from the newborn period to the late senility. Blood serum concentrations of total and free thyroxine, total and free triiodothyronine, reverse--triiodothyronine and TSH were determined by radioimmunoassay in 1350 euthyroid persons of age ranging from 3 days to 81 years. It was found that both secretory function of the thyroid and the peripheral metabolism of the hormones secreted undergo the changes parallel to the advancement of age and to the development of the

Hyperthyroxinaemia: abnormal binding of T4 by an inherited albumin variant.

Two euthyroid subjects with high total concentrations of T4 in their sera have been studied (J.D.: T4, 170; T3, 1.90; rT3, 0.54 nmol/l. E.T.: T4, 185; T3, 1.63; rT3, 0.37 nmol/l). Concentrations of all three T4-binding proteins were within normal limits in both cases. However, on reverse-flow electrophoresis an abnormally large amount of thyroxine was found to travel with albumin. The three T4-binding proteins in the sera of both patients were separated from each other by a novel affinity chromatographic method using dye-Sepharose conjugates. Affinity constants for T4 binding to TBG and to pre

Studies on the deiodination of 3,3',5'-T3 (reverse T3) to 3,3'-T2 (diiodothyronine) in rat liver.

Properties of the deiodination reaction of rT3 to 3,3' T2 in rat liver homogenate are reported and compared with T4 to T3 conversion under similar conditions. pH optimum and SH-group dependency of these two reactions are quite different, though both are concerned with 5' deiodination. The most potent activator of the reaction rT3 to T2' is dithiothreitol; the enzyme activity increases almost linearly even at very high concentration of this compound (the same is true for mercaptoethanol). Glutathione and coenzyme A, show only small activating effects. T4 to T3 and rT3 to T2' converting is being

3,3',5'-triiodothyronine, thyroxine, triiodothyronine, and thyrotropin levels in maternal and cord blood sera from endemic goiter regions of Brazil.

T4, T3, rT3, and TSH were measured in cord blood of infants born in mild to severe endemic goiter areas and nonendemic urban areas in South and North America. A significantly high rT3 concentration was found in newborns living in the urban area of South America as compared to a Canadian newborn population. There were no differences among the various thyroid parameters in the groups of newborns studied. Blood from goitrous mothers showed significantly higher concentrations of T3 and TSH than nongoitrous mothers. However, these changes did not affect the concentrations of thyroid hormones, rT3,

Neonatal thyroid function: influence of perinatal factors.

Indices of thyroid function were measured in 229 healthy term neonates at birth and at 5, 10, and 15 days of age. Results were analysed to assess whether maternal diabetes mellitus, toxaemia of pregnancy, intrapartum fetal distress, duration of labour, method of delivery, asphyxia at birth, race, sex, birthweight, birth length, head circumference, or method of feeding influenced any index. Thyroxine, the free thyroxine index, and free thyroxine concentrations at birth correlated with birthweight. Method of delivery influenced mean thyroxine and free thyroxine index values at birth and at age 5
