Trimethyltin (TMT) produces cytological damage to several limbic-forebrain structures which is accompanied by behavioral changes including hyperactivity and aggressiveness. The purpose of this investigation was to determine whether the problem-solving capacity of the rat is affected by TMT treatment. Either 7.0 mg/kg of TMT ([CH3]3SnCl) or 0.9% saline was injected by gavage into the gastric lumen of each rat. The treated and non-treated animals were divided into two groups of nine each and tested subsequently on a series of Hebb-Williams maze problems. In comparison to the controls, the tin-tr

The liberation of fatty acids, above all arachidonic acid, in human blood cells is involved in numerous health problems or physiological mechanisms. The activity of cellular phospholipases leads to lipid metabolites such as eicosanoids, platelet activating factor, diacylglycerol, and inositolphosphates that are capable of mediating such pathological symptoms. The results presented here demonstrate that organic heavy metal compounds induce arachidonic acid liberation or its rearrangement within the lipid classes of HL-60 cells before a loss in viability can be detected. Four of the compounds te

The time course of tin distribution in homogenates and subcellular fractions of rat brain was determined following the acute administration of trimethyl tin (TMT) and triethyl tin (TET) to the rat. Exposure to TMT resulted in lower concentrations but greater persistence of tin in subcellular fractions compared to exposure to TET. A delayed accumulation of tin in the mitochondrial fraction was observed following the administration of TMT but not TET. Analysis of total protein and mitochondrial markers did not reveal differences between the compositions of mitochondrial fractions prepared from c

Gastrointestinal symptoms are common health problems found during aging and neurodegenerative diseases. Trimethyltin-induced rat is known as an animal model of hippocampal degeneration with no data on enteric neurodegeneration. This study aimed to investigate the effect of trimethyltin (TMT) induction on the gastrointestinal tract. A 28-day animal study with male Sprague-Dawley rats (3 months old, 150-200 g) given a single TMT injection (8 mg/kg body weight, intraperitoneal) was conducted. The number of neurons in the colonic myenteric plexus was measured using stereological estimation. Histol

Trimethyltin chloride (TMT) is an organotin-based contaminant present in the water environment that poses a great threat to aquatic organisms and humans. The liver is the detoxification organ of the body and TMT exposure accumulates in the liver. Tea polyphenol (TP) is a natural antioxidant extracted from tea leaves and has been widely used as a food and feed additive. To investigate the mechanism of toxicity caused by TMT exposure on grass carp hepatocytes (L8824 cells) and the mitigating effect of TP, we established a hepatocyte model of TMT toxicity and/or TP treatment. L8824 cells were tre

BACKGROUND: Trimethyltin (TMT) acts as a potent neurotoxic compound especially for the hippocampus. The effects of valproic acid (VPA) on TMT-induced learning and memory deficits were investigated.
METHODS: The rats were divided into: (1) control, (2) TMT, (3) TMT-VPA 1, (4) TMT-VPA 5, (5) TMT-VPA 10. TMT was injected as a single dose (12 mg/kg, ip) in groups 2-5. The animals of groups 3-5 were treated by 1, 5, and 10 mg/kg of VPA for 2 weeks. Learning and memory deficits were assessed by Morris water maze (MWM) and passive avoidance (PA) tests. The markers of oxidative stress mainly malondia

Effects of tributyltin chloride (TBT) and other organotin compounds on mitogen-activated protein kinases (MAPKs) were examined in CCRF-CEM human T lymphoblastoid cells. In response to the incubation with 0.25-2 microM TBT for 1 h, the levels of the phosphorylated form of extracellular signal-regulated protein kinase (ERK), c-Jun NH(2)-terminal kinase (JNK), and p38 MAPK increased in a dose-dependent manner. The phosphorylation was observed after 15 min and lasted for 4 h following exposure to 1 microM TBT, while the cell viability was not lowered significantly within 6 h. On the other hand, no

The environmental contaminant, trimethyltin (TMT), produces a profound elevation in tone intensity necessary to inhibit the acoustic startle reflex in laboratory animals which recovers over a prolonged period except at very high frequencies. The recovery that is observed does not begin until 3 to 5 weeks after a single acute administration depending upon dosage. As opposed to the very temporary threshold shifts by the salicylates and loop diuretics or the permanent and progressive ototoxicity resulting from aminoglycoside antibiotics the time course for recovery of acoustic startle reflex inhi

Trimethyltin chloride (TMT), an organotin compound with potent neurotoxicity, is widely used as a heat stabilizer for plastics. However, the precise pathogenic mechanism of TMT remains incompletely elucidated, and there persists a dearth of sensitive detection methodologies for early diagnosis of TMT. In this study, Sprague-Dawley rats were treated with 10 mg/kg TMT to simulate acute exposure in humans. Micro-positron emission tomography/computed tomography (Micro-PET/CT) and molecular imaging quantitative analysis tools were employed to calculate the uptake rate of 18 F-2-fluoro-D-deoxy-gluco

Trimethyltin chloride (TMT) is a highly toxic organotin pollutant frequently found in aquatic environments, posing a significant threat to the ecological system. The kidney plays a vital role in the body's detoxification processes, and TMT present in the environment tends to accumulate in the kidneys. However, it remained unclear whether exposure to different doses of TMT could induce pyroptosis and immune dysfunction in grass carp kidney cells (CIK cells). For this purpose, after assessing the half-maximal inhibitory concentration (IC50) of TMT on CIK cells, we established a model for exposur

Neuroinflammation is one of the important mechanisms of trimethyltin chloride (TMT) central neurotoxicity. Artemisinin (ARS) is a well-known antimalarial drug that also has significant anti-inflammatory effects. Prokineticin 2 (PK2) is a small molecule secreted protein that is widely expressed in the nervous system and plays a key role in the development of neuroinflammation. However, it remains unclear whether ARS can ameliorate neuroinflammation caused by TMT and whether PK2/PKRs signaling pathway plays a part in it. In this research, male Balb/c mice were administered TMT (2.8 mg/kg, i.p.)

N-Succinimidyl 5-(trialkylstannyl)-3-pyridinecarboxylates (alkyl = Me, Bu) have been prepared and used as a precursor to label N-succinimidyl 5-[131I]iodo-3-pyridinecarboxylate (SIPC). SIPC was obtained in greater than 80% yield from either the methyl or butyl precursor with N-chlorosuccinimide and heating at 60-65 degrees C. Significantly lower yields were observed with tert-butyl hydroperoxide. After a 30-min incubation with [131I]SIPC at pH 8.5, goat IgG, an intact monoclonal antibody (MAb), and a MAb F(ab')2 fragment were labeled in 60-65% yield. Specific binding of the MAb and MAb fragmen

Trimethyltin (TMT) is a neurotoxicant known to alter auditory function. The present study was designed to compare TMT-induced auditory dysfunction using behavioral, electrophysiological, and anatomical techniques. Adult male Long-Evans hooded rats (n = 9-12/group) were acutely exposed to saline, 3, 5, or 7 mg/kg TMT. Auditory thresholds were determined 11 weeks postdosing for 5- and 40-kHz tones using reflex modification of the auditory startle response (ASR). Brainstem auditory evoked response (BAER) thresholds were determined for 5-, 40-, and 80-kHz tonal stimuli 9 weeks postdosing. Cochlear

The averaged evoked potential technique was used to determine the sequence and relative effects of a single dose of trimethyltin (TMT) on the neurophysiologic functioning of two limbic system pathways, selected for study because they are known to show TMT-induced pathologic changes. Adult female rats were implanted with bipolar electrodes in the olfactory or prepyriform cortex (PPC), dentate gyrus (DG) and distal CA3 subfield of the hippocampus. Evoked potentials were elicited in the DG by stimulation of the PPC, and in CA3 by stimulation of the mossy fiber system which originates in dentate g

Trimethyltin, its tetra-, di-, and monomethyl analogs, inorganic tin (Sn II and Sn IV), triethyltin, tripropyltin, tributyltin, and triphenyltin were tested for their ability in inhibiting the uptake of gamma-[3H]aminobutyric acid (GABA) into mouse forebrain synaptosomes in vitro. All organotins containing three carbon-tin bonds were potent inhibitors of [3H]GABA uptake with IC50 values ranging from 10(-4) to 10(-6) M. Various thiol and sulfur compounds, particularly sodium sulfide, were capable of antagonizing the inhibitory effect of triphenyltin and, to a minor extent, of other organotins.

Trimethyltin chloride (TMT) is widespread in the environment and is harmful to both humans and animals. In order to investigate the toxicity mechanism of TMT exposure on chicken liver, We established an in vivo experimental model by giving chickens oral administration of different concentrations of TMT dilution solution and vitro experiments of treating leghorn male hepatoma (LMH) cells for 12 h. The results showed that Albumin (ALB), total protein (TP) and alanine aminotransferase (ALT) in the blood of TMT-treated chickens, as well as ALT and aspartate aminotransferase (AST) in the liver, wer

Inorganic and organic tin compounds present in aqueous ecosystems have diverse effects on the behavior of living organisms. With the aim of identifying possible correlates of these actions, we studied the effects of both types of Sn2+. The effects of SnCl2 and Sn(CH3)2 on acetylcholine-activate currents were studied on identified neurons of the mollusk Lymnaea stagnalis L. using a two-microelectrode membrane potential clamping technique and by intracellular dialysis with potential and ion concentration clamping. Experiments were performed on single neurons after isolation and on whole ganglion

Aquaculture organisms may accumulate metals to induce health risks. Compared with the focus on total contents, chemical-specific risk assessment makes reasonable but is rare. Herein, we elucidated occurrence of twelve metal compounds in shrimp and fish (edible muscle, one of major metal-containing and generally targeted organs), water, sediment, and feedstuff from two aquaculture ponds in Zhejiang Province (one of the major aquatic production and consumption areas). We detected Cd(II) (0.6 -71.4 μg kg-1 in 100 % prawn but 63 % fish), methylmercury (MeHg, 0.5 -7.1 μg kg-1 in 100 % fish but 61 %
