BCG vaccination for bovine tuberculosis; conclusions from the Jerusalem One Health workshop.

The global burden of bovine tuberculosis (bTB) remains poorly characterized, with spill-over impacts on multiple species. The "One Health" concept is especially relevant given the bidirectional risk of cattle infecting humans with Mycobacterium bovis and humans infecting cattle with Mycobacterium tuberculosis. "Test and cull" is the traditional bTB control method, but the strategy may not be economically feasible or culturally acceptable where cattle are highly prized or their killing is a religious taboo; it is also less effective when there are wildlife reservoirs of infection. Vaccination w

Oral delivery of bovine tuberculosis vaccine to free-ranging white-tailed deer.

INTRODUCTION: Free-ranging white-tailed deer (Odocoileus virginianus) are a self-sustaining reservoir for bovine tuberculosis (bTB) in northeastern lower Michigan, (United States) continually putting the area's cattle industry at risk. Liberal recreational deer harvest, baiting bans, and mitigation measures on farms have reduced but not eliminated bTB in deer nor have they eliminated transmission to cattle. With apparent prevalence in deer being low (1-2%) but constant, vaccination could be an additional tool to aid in addressing the problem and merits investigation. Mycobacterium tuberculosis

Biosecurity practices of bovine tuberculosis affected French cattle farms between 2016 and 2023.

In recent years, interest in biosecurity practices on livestock farms has expanded considerably, with improvements noted in pig and poultry farms but limited progress in cattle farms. In France, data regarding biosecurity practices in cattle farms remains scarce. Bovine tuberculosis (bTB) is one of the main animal health challenges. Each case leads to mandatory epidemiological investigation, led by state services. Biosecurity practices are also described. This creates a valuable data source to describe biosecurity practices in bTB-affected farms. This study aimed to describe biosecurity practi

Bovine Tuberculosis Epidemiology in Cameroon, Central Africa, Based on the Interferon-Gamma Assay.

Despite sub-Saharan Africa (SSA) accounting for ~20% of the global cattle population, prevalence estimates and related risk factors of bovine tuberculosis (bTB) are still poorly described. The increased sensitivity of the IFN-γ assay and its practical benefits suggest the test could be useful to investigate bTB epidemiology in SSA. This study used a population-based sample to estimate bTB prevalence, identify risk factors and estimate the effective reproductive rate in Cameroonian cattle populations. A cross-sectional study was conducted in the North West Region (NWR) and the Vina Division (VI

Bovine tuberculosis in eastern Ethiopia: prevalence, risk factors and its public health importance.

BACKGROUND: Bovine tuberculosis is among the primary zoonotic disease caused by Mycobacterium bovis which has significant impact on the health of livestock and human. It has been significantly a cause for great economic loss in animal production. METHODOLOGY: A cross-sectional study was conducted from December 2014 to June 2016 on 315 cattle in selected areas of eastern Ethiopia, aiming to estimate the occurrence of bovine tuberculosis using comparative intradermal tuberculin skin test and assess cattle owners' awareness on its public health implication. Random sampling method was applied in

Disseminated bovine tuberculosis in a wild red fox (Vulpes vulpes) in southern Spain.

A wild-caught, adult red fox (Vulpes vulpes) from Doñana National Park (southern Spain), in very poor condition, died during anesthesia. At necropsy, the submandibular, retropharyngeal, mediastinal, axillary, mesenteric, and popliteal lymph nodes were enlarged, and the right submandibular and mesenteric lymph nodes had hard, white-colored concretions (calcifications). Multiple white pinpoint foci were observed in the lungs, and abscesses were present in the left dorsal lung lobes. No lesions were seen in tonsils, liver, or spleen. On histopathology multiple tuberculous granulomas, with Ziehl-N

A novel live recombinant mycobacterial vaccine against bovine tuberculosis more potent than BCG.

Mycobacterium bovis infection of cattle and other domesticated animals exacts a significant economic toll in both economically developing and industrialized countries. Vaccination of herds and/or wild animals that share their grazing land and serve as reservoirs of infection has been proposed as a strategy to combat bovine tuberculosis. However, the only currently available vaccine, M. bovis Bacille Calmette-Guerin (BCG), is not highly efficacious. Here we show that a live recombinant vaccine, rBCG30, which expresses large amounts of the Mycobacterium tuberculosis 30 kDa major secretory protei

Recombinant proteins expressed in Mycolicibacterium smegmatis enhance antibody-based detection of bovine tuberculosis.

Bovine tuberculosis (bTB), caused by Mycobacterium bovis, is a zoonosis that threatens public health and causes substantial economic losses in livestock. The suboptimal Escherichia coli-expressed recombinant proteins limit the diagnostic performance of current bTB serological tests. To overcome this limitation, we evaluated Mycolicibacterium smegmatis as an expression host capable of producing recombinant antigens with post-translational modifications comparable to those of M. bovis. Ten antigen candidates were individually expressed in E. coli using the pET-26b( +) vector and in M. smegmatis

A bovine pulmosphere model and multiomics reveal early host response signature in tuberculosis.

Early interactions between tubercle bacilli and lung cells are critical in tuberculosis (TB) pathogenesis. Conventional two-dimensional cell cultures fail to replicate the multicellular complexity of lungs. We introduce a three-dimensional pulmosphere model for Mycobacterium tuberculosis infection in bovine systems, demonstrating through comprehensive transcriptome and proteome analyses that these multicellular spheroids closely mimic lung cell diversity, interactions, and extracellular matrix (ECM) composition. Cell viability, hypoxia, and reactive oxygen species assessments over three weeks

Bovine ownership and reduced pulmonary tuberculosis risk in Nepal: A case-control study.

This case-control study sought to confirm and investigate in more depth protective associations previously found of bovine (cattle and water buffalo) ownership with reduced risk of both pulmonary tuberculosis (PTB) and latent tuberculosis infection (LTBI) in humans. The study recruited male and female PTB cases from a diagnostic centre and a frequency-matched community-based control group in Kaski District, Nepal. Controls were tested for LTBI status and a separate nested case-control study was conducted based on LTBI status. Data were collected on participant household animal ownership. Using

Diagnosis of human bovine tuberculosis aided by PET/CT scanning and EBUS-TBNA.

Human bovine tuberculosis is a rare zoonotic infection in developed countries which has been achieved predominantly by effective eradication programmes in cattle. The principal modes of transmission are consumption of unpasteurised dairy products and close contact with infected cattle. The clinical and radiological presentation is indistinguishable from tuberculosis caused by Mycobacterium tuberculosis The diagnosis should be considered in individuals with relevant risk factors who present with intra/extrathoracic pathology. We describe and discuss a case of bovine tuberculosis with a synchron

Bovine tuberculosis in working foxhounds: lessons learned from a complex public health investigation.

In 2017, Public Health England South East Health Protection Team (HPT) were involved in the management of an outbreak of Mycobacterium bovis (the causative agent of bovine tuberculosis) in a pack of working foxhounds. This paper summarises the actions taken by the team in managing the public health aspects of the outbreak, and lessons learned to improve the management of future potential outbreaks. A literature search was conducted to identify relevant publications on M. bovis. Clinical notes from the Public Health England (PHE) health protection database were reviewed and key points extracted

Low-dose BCG vaccination protects free-ranging cattle against naturally-acquired bovine tuberculosis.

Vaccination of cattle with Mycobacterium bovis BCG has been shown to protect against infection with virulent strains of M. bovis, and against resultant bovine tuberculosis (TB). Here we report on a large-scale trial in New Zealand where free-ranging cattle were vaccinated with 3 x 105 BCG via injection, a lower dose than any previously trialed in cattle against exposure to a natural force of M. bovis infection. In a multi-year enrolment study involving >800 animals, three cohorts of 1-2 year old cattle were randomised to receive vaccine or to serve as non-vaccinated controls. Cattle were slaug

Milk yield and reproductive performance of Holstein cows testing positive for bovine tuberculosis.

The objective of this study was to determine if high milk-yielding Holstein cows testing positive for bovine tuberculosis (bTB) are affected in their reproductive performance and milk yield. For this purpose, 1044 healthy cows and 105 bTB reactor cows were used. Tuberculosis reactor cows were from four large commercial dairy operations from the same region which were transferred from their barns to an isolated dairy facility. Cows free from this disease were placed in the same barn as the bTB reactor cows but in an isolated division and served as control animals. The analysis of variance with

Evaluation of BCG vaccine and Mycobacterium bovis culture filtrate proteins against bovine tuberculosis.

The objective of this study was to evaluate the degree of protection against TB of three immunogens in calves experimentally challenged with a pathogenic Mycobacterium bovis strain by identification of lesions during necropsy. Twenty-four calves free of TB were distributed into four groups: group 1 was inoculated with M. bovis CFPE, group 2 with CFPE/IFN-gamma, group 3 with M. bovis BCG, and group 4 remained as control. After 6 months all animals were challenged with an M. bovis field strain, and 6 months later the animals were euthanized and the presence of lesions was evaluated. Several degr

An experimental intratonsilar infection model for bovine tuberculosis in African buffaloes, Syncerus caffer.

An infection model for Mycobacterium bovis in African buffaloes, Syncerus caffer, was developed, using the intratonsilar route of inoculation. Two groups of 11 buffaloes each, aged approximately 18 months, were infected with either 3.2 x 10(2) cfu (low dose) or 3 x 10(4) cfu (high dose) of M. bovis strain isolated from a buffalo. A control group of six buffaloes received saline via the same route. The infection status was monitored in vivo using the comparative intradermal tuberculin test, and in vitro by the modified interferon-gamma assay. All buffaloes were euthanazed 22 weeks post infectio

Unseen Threats to Meat Safety: Exposing the Hidden Epidemic to Bovine Tuberculosis in Slaughterhouses.

Bovine tuberculosis (BTB) is a zoonotic illness of significant public health concern. This research aims to investigate the prevalence of BTB and its risk factors in Upper Egypt governorates. A total of 600 cattle (200 from each of New Valley, Qena, and Aswan) were tested using a single intradermal cervical tuberculin test (SICTT), and the positive animals were slaughtered at the central abattoirs of each governorate. The tissues of the affected carcasses were inspected by Ziehl-Neelsen (ZN) staining and culturing, and confirmation of results was achieved through ELISA and PCR. The findings re

Comparison of qPCR methods for the diagnosis of bovine tuberculosis from granulomas collected at slaughterhouses.

To improve the direct diagnosis of bovine tuberculosis (bTB) in lesions from carcasses condemned at slaughterhouses - the primary method for identifying infected farms within the Surveillance System - two qPCR assays were evaluated against the gold standard: culture and identification of Mycobacterium bovis. A total of 167 lesion samples were collected by inspection services in Mato Grosso and Santa Catarina. Samples were homogenized and analyzed using culture and qPCR targeting the IS1081 sequence (for the M. tuberculosis complex) and the RD4 region (specific to M. bovis). Both qPCR assays de

Epidemiological situation of bovine and bubaline tuberculosis in the state of Pará, Amazon region of Brazil.

INTRODUCTION: Bovine tuberculosis is one of the primary infectious diseases affecting cattle. Although several countries have managed to eradicate this zoonosis it remains endemic and uncontrolled across many countries in Africa, Asia, Latin America, and the Middle East. Brazil launched its national control and eradication program in 2001, and since then, epidemiological studies have been carried out to define optimal control strategies and to enable the management of the process in each region. METHODS: This study covered the state of Pará, which was divided into three regions, in each of wh
