Neuronal ceroid lipofuscinoses (NCLs) are a group of neurodegenerative diseases predominantly in childhood that are characterized by psychomotor deterioration, epilepsy, and early death of patients. The NCLs analyzed in the present study are caused by defects of the specific enzymes, CLN1 (palmitoyl protein thioesterase 1; PPT1), CLN2 (tripeptidyl peptidase 1; TPP1), and CLN10 (cathepsin D). In this study, a 2-oxoglutarate-dependent dioxygenase (2OGD), AtS8H, was identified that hydroxylates umbelliferone (UMB) to daphnetin , enabling the design of an artificial biosynthetic pathway. As part of our effort to develop potential tyrosinase inhibitors, we have conjugated the well-known tyrosinase inhibitor kojic acid (KA) with several phenolic natural products such as umbelliferone, sesamol, thymol, carvacrol, eugenol, isoeugenol, vanillin, isovanillin, and apocynin that some reports have shown their activity on tyrosinase enzyme. Anionic charges, cytochemically identified as lipid phosphate groups, cover the outer membrane surface of Spiroplasma floricola. They induce a negative membrane surface potential which affects the distribution of ions, including protons. Accordingly, the pH at the interface differs from the bulk pH. By using the fluorescent lipoid pH indicator 4-heptadecyl-7-hydroxycoumarin, the pH at the membrane surface was determined. From the difference of the bulk and the interfacial pH the membrane surface potential of S. floricola was calculated to be phi = -118 mV.
