Differential regulation of urease activity in Helicobacter hepaticus and Helicobacter pylori.

Helicobacter hepaticus is a pathogen of rodents, which causes diverse enteric and hepatic inflammatory diseases and malignancies. The urease enzyme is an important colonization factor of gastric Helicobacter species like Helicobacter pylori, but little is known about the role and regulation of urease in enterohepatic Helicobacter species. Here it is reported that urease activity of H. hepaticus does not contribute to acid resistance, and that it is nickel-responsive at the post-translational level. H. hepaticus strain ATCC 51449 did not grow or survive at pH 3.0, and supplementation with urea

Mechanisms of acid resistance due to the urease system of Helicobacter pylori.

BACKGROUND & AIMS: Helicobacter pylori, a neutralophile, uses acid neutralization by urease to combat gastric acidity, allowing gastric colonization. Both acute and chronic acid resistance mechanisms are present. Acute mechanisms of acid adaptation could be due to surface urease, increased inner-membrane urea permeability via UreI, or both. Slower mechanisms may involve increased nickel insertion into apoenzyme, posttranscriptional regulation, or increased enzyme synthesis. The aim of this study was to further define regulation of urease under acidic conditions. METHODS: Surface-bound urease

The role of Helicobacter pylori urease in the pathogenesis of gastritis and peptic ulceration.

Helicobacter pylori produces a 550 kDa, multimeric, nickel-containing urease that catalyses the hydrolysis of urea to yield ammonia and carbonic acid. The ure gene cluster, comprised of seven genes, encodes the two structural subunits UreA (26.5 kDa) and UreB (60.3 kDa), and five accessory proteins: UreI, UreE, UreF, UreG and UreH. Accessory proteins are required for nickel ion insertion into the apoenzyme. The native protein consists of six copies each of UreA and UreB; two nickel ions are coordinated into each UreB active site. Urease is found in the cytosol, but may also localize on the sur

Awakenings to the pathogenicity of urease and the requirement for continuous long term therapy.

Urease is an enzyme found in plants and bacteria, but not mammals. It catalyzes the conversion of urea to carbon dioxide and ammonia. Ammonia shortens the life span of cells; and higher concentrations cause tissue necrosis and cytolysis. Twenty percent of total body urea is converted to ammonia by bacterial urease in the colon. Small injections of urease immunize animals by producing antiurease, a gamma globulin, which inactivates urease. Immunization eliminates the colonic conversion of urea to ammonia. Injection of urease produces ammonia intoxication making immunization hazardous. Although

Antibacterial action of the urease inhibitor acetohydroxamic acid on Helicobacter pylori.

The urease inhibitor acetohydroxamic acid (AHA) was assessed for its bacteriostatic and bactericidal effects on Helicobacter pylori. For eight isolates of H pylori, the minimum inhibitory concentration (MIC) was either 200 mg/l or 400 mg/l. Interactions between AHA and antimicrobial drugs used to treat H pylori were also determined. For most isolates AHA reduced the MIC for colloidal bismuth subcitrate (CBS), tetracycline, metronidazole, and amoxicillin. In a few isolates, however, AHA increased the minimum bactericidal concentration (MBC) for these antimicrobial treatments. In vitro AHA is ac

Usefulness of culture, histology, and urease testing in the detection of Campylobacter pylori.

Several tests have been described which detect the presence of Campylobacter pylori (CP) in the human upper gastrointestinal tract. Antral biopsies from 160 endoscopies were evaluated for the presence of CP; four tests readily available to the practicing endoscopist were used: two urease tests [CLOtest and microtiter biopsy urease test (MBUT)], hematoxylin and eosin staining, and culture. Sensitivity and specificity of each was evaluated and found to be near 90%, except for culture, which was less sensitive. The presence of peptic disease on endoscopy, and age greater than 50 yr, did play a sm

[Comparison of urease test (CLOtest) and histology in the diagnosis of Campylobacter pylori].

Campylobacter pylori has been implicated in the pathogenesis of peptic ulcer, non-ulcer dyspepsia and chronic gastritis. C. pylori produces large amounts of urease which has been used as a biochemical support to identify the microorganism. In this study, we described the use of a rapid urease test (CLOtest) to detect C. pylori. In 46 consecutive patients, biopsy of fundus and antrum were obtained for histology and CLOtest. All specimens showed chronic gastritis. In 22 patients (48%) C. pylori was identified either by histology or CLOtest. In 18 patients (39%) CLOtest was positive and in 16 pat

Rapid urease test in the management of Campylobacter pyloridis-associated gastritis.

Campylobacter pyloridis colonization of the stomach may be an etiological factor in gastritis and peptic ulceration. Campylobacter pyloridis produces large amounts of urease, and the presence of this enzyme in gastric mucosa usually indicates infection with the organism. In this paper we describe the use of a rapid urease test (CLOtest) to detect C. pyloridis infection in gastric mucosal biopsies. In 141 consecutive endoscopy cases, antral biopsies were taken for culture and histology, and an extra biopsy was inserted into the CLOtest gel. There were 79 patients infected with C. pyloridis, 78

Use of a urease-antibody conjugate in an enzyme immunoassay for the detection of blastomycosis.

A urease-antibody conjugate was used in an enzyme-linked immunoassay (ELISA) for the detection of antibodies in blastomycosis. A Blastomyces dermatitidis immunodiffusion antigen was used as the reference antigen in a solid phase indirect ELISA procedure and the endpoints were determined visually. Urease ELISA results on serum specimens from patients with blastomycosis compared favorably (90%) with results obtained spectrophotometrically by the alkaline phosphatase enzyme system. Specificity was evaluated with assays on sera from patients with histoplasmosis (20% cross reactivity), coccidioidom

Inhibition of urease activity by hydroxamic acid derivatives of amino acids.

Hydroxamic acids have been reported to be potent and specific inhibitors of urease (EC 3.5.1.5) activity of plant and bacterial origin. The present investigation was performed on the inhibitory effect of hydroxamic acid derivatives of naturally occurring amino acids on the urease activity of the Jack Bean and the alimentary tracts of rats. Methionine-hydroxamic acid was the most powerful inhibitor (I50=3.9 X 10(-6) M) among nineteen alpha-aminoacyl hydroxamic acids. Phenylalanine-, serine-, alanine-, glycine-, histidine-, threonine-, leucine-, and arginine-hydroxamic acids followed, in order o

Fur-regulated urease contributes to the environmental adaptation of Yersinia pseudotuberculosis.

Urease converts urea into ammonia and carbon dioxide, providing a nitrogen and carbon source for microbial growth and serving as an important mechanism for human bacterial pathogens to survive in acidic conditions, which can be regulated by many factors. As a global regulator, the ferric uptake regulator (Fur) regulates a series of genes and pathways involved in many different cellular processes and the virulence of the enteric bacterium Yersinia pseudotuberculosis (Yptb). However, whether Fur regulates the urease activity in Yptb was still unknown. In this study, we found that urease is posit

In vitro assessment of Helicobacter pylori ureases inhibition by honey fractions.

BACKGROUND AND AIMS: Pathogenic effects associated with Helicobacter pylori infection include peptic ulcer. Pathogenicity is mediated by the bacteria ureases. The emerging resistance of H. pylori to clarithromycin used in combination with amoxicillin in the treatment of H. pylori infection has necessitated the search for other means of combating the scourge of H. pylori, hence the search for potent H. pylori urease inhibitor. The aim of the study was to evaluate honey fractions obtained from different geographic zones for H. pylori urease inhibitory potentials. METHODS: Chloroform and diethyl

Design, synthesis, and biological evaluation of phosphoramide derivatives as urease inhibitors.

The design, synthesis, and biological evaluation of phosphoramide derivatives as urease inhibitors to reduce the loss of ammonia has been carried out. Forty phosphorus derivatives were synthesized and their inhibitory activities evaluated against that of jack bean urease. In addition, in vivo assays have been carried out. All of the compounds were characterized by IR, (1)H NMR, MS, and elemental microanalysis. In some cases, detailed molecular modeling studies were carried out, and these highlighted the interaction between the enzyme active center and the compounds and also the characteristics

UreA2B2: a second urease system in the gastric pathogen Helicobacter felis.

Urease activity is vital for gastric colonization by Helicobacter species, such as the animal pathogen Helicobacter felis. Here it is demonstrated that H. felis expresses two independent, and distinct urease systems. H. felis isolate CS1 expressed two proteins of 67 and 70 kDa reacting with antibodies to H. pylori urease. The 67-kDa protein was identified as the UreB urease subunit, whereas the N-terminal amino acid sequence of the 70-kDa protein displayed 58% identity with the UreB protein and was tentatively named UreB2. The gene encoding the UreB2 protein was identified and located in a gen

The influence of warming on rapid urease test results: a prospective evaluation.

BACKGROUND: A rapid urease test is the initial test of choice for the diagnosis of Helicobacter pylori at endoscopy. Incubating the CLOtest at 30 degrees to 40 degrees C is recommended, but this decreases simplicity and requires the purchase of additional equipment. We compared results of testing at room temperature versus incubation at 37 degrees C. METHODS: Four biopsy specimens were taken from the same portion of the antrum in 200 patients undergoing upper endoscopy. One was placed in each of two CLOtests and two were sent for histologic examination. One CLOtest was incubated at 37 degrees

Clotest (rapid urease test) in the diagnosis of Helicobacter pylori infection.

Over a period of 3 months, 85 patients who underwent gastroscopy had antral biopsy taken for Clotest, histology and/or culture for Helicobacter pylori (HP). The sensitivity and specificity of Clotest were found to be 77% and 96% respectively with negative predictive value of 63% and positive predictive value of 98%. Ninety-two percent of the positive Clotests were positive within 20 minutes, thus giving rapid result. Therefore, Clotest is a rapid, sensitive and highly specific test for HP infection. A high correlation between HP infection and chronic gastritis was noted and the prevalence of H
