Effects of Ursolic Acid on Intestinal Health and Gut Bacteria Antibiotic Resistance in Mice.

Ursolic acid (UA), a natural pentacyclic triterpenoid, has been widely reported to exert anti-oxidant and anti-inflammatory properties. However, the effects of UA on the intestinal homeostasis and gut microbiota were rarely explored. The aim of the present study was to investigate the effects of UA on intestinal health and gut microflora antibiotic-resistance in antibiotic-exposed mice. Kunming mice (n = 80) were randomly allocated into three groups and fed with one of the following diets, respectively: Cont group (n = 20), the basal diet; UA group (n = 20), the basal diet supplemented with 15

Evaluation of Anti-Toxoplasma gondii Effect of Ursolic Acid as a Novel Toxoplasmosis Inhibitor.

This study was carried out to evaluate the anti-parasitic effect of ursolic acid against Toxoplasma gondii (T. gondii) that induces toxoplasmosis, particularly in humans. The anti-parasitic effects of ursolic acid against T. gondii-infected cells and T. gondii were evaluated through different specific assays, including immunofluorescence staining and animal testing. Ursolic acid effectively inhibited the proliferation of T. gondii when compared with sulfadiazine, and consistently induced anti-T. gondii activity/effect. In particular, the formation of parasitophorous vacuole membrane (PVM) in h

Efficacy of ursolic acid against Echinococcus granulosus in vitro and in a murine infection model.

BACKGROUND: Cystic echinococcosis is a global public health problem; however, the drugs (albendazole and mebendazole) currently recommended by WHO for its treatment, have limited efficacy. Therefore, novel drugs are required to provide more choices for the treatment of this disease. METHODS: The anthelmintic effects of ursolic acid (UA) were tested on Echinococcus granulosus protoscoleces, germinal cells and metacestodes in vitro. The in vivo efficacy of UA was investigated in mice following secondary infection with E. granulosus. Furthermore, the corresponding ultrastructural damage induced

The effect of oleanolic and ursolic acids on the hemolytic properties and biofilm formation of Listeria monocytogenes.

Oleanolic acid and ursolic acid are pentacyclic triterpenoids isolated from a variety of medicinal plants, which have antibacterial activity. Listeria monocytogenes is a Gram-positive facultative pathogen, being the causative agent of listeriosis. The present study was carried out to evaluate the in vitro effect of sub-inhibitory concentrations of both triterpene acids on the pathogenicity determinants of L. monocytogenes: their hemolytic activity and biofilm forming ability. Oleanolic and ursolic acids inhibited listeriolysin O activity without influencing toxin secretion. Biofilm formation,

Synthesis, stability and pharmacological evaluation of a novel codrug consisting of lamivudine and ursolic acid.

A novel codrug (LMX) was obtained from lamivudine (LMV) and ursolic acid (UA) coupled with ethyl chloroacetate through an amide and ester linkage. The structure of LMX was confirmed by ¹H NMR, ¹³C NMR, IR and HRMS. Herein, the in vitro non-enzymatic and enzymatic hydrolysis and in vivo pharmacological activities of LMX were studied. The kinetics of hydrolysis of LMX was studied in aqueous solution of pH 1-10, 80% buffered human plasma and in the presence of lipase from Porcine pancreas (EC 3.1.1.3) at 37°C. It is found that LMX hydrolysis rate was significantly faster in lipase with half-life

Impact of ursolic acid on chronic ethanol-induced oxidative stress in the rat heart.

Oxidative stress plays an important role as a mediator of myocardial damage produced by ethanol. This work was designed to investigate the effect of ursolic acid (UA), a reported radical scavenger and antioxidant, on oxidative stress in the heart of chronically ethanol-administered rats. Chronic ethanol administration (7.9 g/kg/day for 60 days) caused tissue damage that was manifested by the elevation of serum lactate dehydrogenase (LDH) and creatine phosphokinase (CPK). It also induced oxidative stress in the heart by increasing the lipid peroxidation process and by decreasing the antioxidant

Apoptotic activity of ursolic acid may correlate with the inhibition of initiation of DNA replication.

Ursolic acid (UA), a pentacyclic triterpene acid, has been reported to exhibit anti-tumor activity. In this study, we investigated the pro-apoptotic effect of UA on HepG2 human hepatoblastoma cells. Treatment with UA decreased the viability of HepG2 cells in a concentration- and time-dependent manner. Furthermore, 30 microM of UA induced DNA fragmentation and subdiploid cells and enhanced the release of cytochrome c and the activation of caspase-3. These results suggest that UA induces cell death through apoptosis, which may be mediated by cytochrome c-dependent caspase-3 activation. In additi

Co-Amorphous Ursolic Acid-Quercetin Complex Improves Solubility and In Vivo Expectorant-Antitussive Activity.

This study investigated the formation of a co-amorphous complex between ursolic acid and quercetin as a chemically defined model system for improving the formulation performance of a poorly soluble triterpenoid acid. Co-amorphous systems were prepared by solvent evaporation and physical grinding at different mass ratios. The optimal formulations were identified and evaluated by water-solubility measurement, powder X-ray diffraction (PXRD), Fourier-transform infrared spectroscopy (FTIR), and in vivo expectorant-antitussive assays. The solvent-derived system showed higher water solubility than t

Ursolic acid induces apoptosis and disrupts host-parasite interactions in Theileria annulata-infected cells.

This study explores the anti-proliferative and anti-parasitic properties of ursolic acid (UA) in Theileria annulata-infected bovine (TA) cells. Dose-response analyses determined an IC50 value of approximately 5 μg/mL for UA, demonstrating selective cytotoxicity toward infected cells with minimal impact on healthy cells. UA treatment induced pronounced morphological alterations and apoptosis in TA cells, as evidenced by light microscopy and a time-dependent increase in cell mortality. Notably, UA exhibited consistent efficacy against both buparvaquone (BPQ)-sensitive and BPQ-resistant TA cell l

Ursolic Acid Ameliorated Neuronal Damage by Restoring Microglia-Activated MMP/TIMP Imbalance in vitro.

PURPOSE: The oxygen and glucose deprivation-reoxygenation (OGDR) model is widely used to evaluate ischemic stroke and cerebral ischemia-reperfusion (I/R) injury in vitro. Excessively activated microglia produce pro-inflammatory mediators such as matrix metalloproteinases [MMPs] and their specific inhibitors, tissue inhibitors of metalloproteinases [TIMPs], causing neuronal damage. Ursolic acid (UA) acts as a neuroprotective agent in the rat middle cerebral artery occlusion/reperfusion (MCAO/R) model keeping the MMP/TIMP balance with underlying mechanisms unclear. Our study used OGDR model to d

Ursolic Acid Suppresses Cholesterol Biosynthesis and Exerts Anti-Cancer Effects in Hepatocellular Carcinoma Cells.

Abnormally upregulated cholesterol and lipid metabolism, observed commonly in multiple cancer types, contributes to cancer development and progression through the activation of oncogenic growth signaling pathways. Although accumulating evidence has shown the preventive and therapeutic benefits of cholesterol-lowering drugs for cancer management, the development of cholesterol-lowering drugs is needed for treatment of cancer as well as metabolism-related chronic diseases. Ursolic acid (UA), a natural pentacyclic terpenoid, suppresses cancer growth and metastasis, but the precise underlying mole

Optimizing microwave-assisted extraction of ursolic acid from apple pomace using response surface methodology.

This study investigates the optimization of microwave-assisted extraction (MAE) to recover ursolic acid (UA) from Hanfu apple pomace using response surface methodology (RSM). The effects of three key variables-extraction time, sample-to-solvent ratio, and ethanol concentration-on UA yield were studied. A Box-Behnken design was employed to model and optimize these variables. The results showed that extraction time had the most significant effect on UA yield, followed by ethanol concentration and sample-to-solvent ratio. The optimal conditions for UA extraction were 118.25 s for extraction time,
