Genetic mapping of fruit texture in American cranberry reveals multi-trait QTL and a complex genetic architecture.

Fruit texture is a key quality trait in cranberry (Vaccinium macrocarpon Ait.), with a direct impact on processing efficiency and overall profitability for the industry. To elucidate the genetic basis of cranberry texture, we conducted the first QTL mapping study in this crop implementing a fruit compression method with 10 mechanical texture traits in two biparental populations, CNJ02 (n=168) and CNJ04 (n=67), over two years. These 10 traits, associated with flesh hardness/stiffness and elasticity, showed similar phenotypic patterns of transgressive segregation and generally strong positive co

Cranberry extract attenuates indomethacin-induced gastriculcer in rats via its potential atioxidant and anti-inflammatory effects.

Indomethacin (IND) belongs to nonsteroidal anti-inflammatory drugs (NSAIDs) prescribed for the treatment of rheumatoid diseases and linked to the development of gastric ulcers in many people. Cranberry is a rich source of polyphenols and flavonoids, which have powerful antioxidant and anti-inflammatory properties. The study aimed to evaluate the activity of cranberry aqueous extract on IND-induced gastric ulcers in albino rats. Twenty adult male rats were sequentially assigned to four groups of five each. The control group consumes distilled water (DW) orally for 15 days. The IND group receive

Characterization of Anti-Inflammatory and Anti-Proliferative Triterpenoids and Phytosterols from Cranberry Pomace.

Vaccinium macrocarpon (American Cranberry) fruit is processed to make juice, supplements, and other edible products, leaving pomace as a sidestream that contains secondary metabolites with potential anti-inflammatory and anti-proliferative activities. Ultrasound-assisted extraction and chromatographic methods were developed to prepare an extract (POM-ACE) and fractions rich in pentacyclic triterpenoids and phytosterols for bioactivity evaluation. These were characterized using UPLC-MS and GC-MS. Anti-inflammatory effects were assessed in a human monocyte (THP-1) model, identifying several frac

Proanthocyanidin-Rich Cranberry Extract Lowers Glycemia in Established Obesity by Delaying Glucose Absorption.

Dietary polyphenols, including proanthocyanidins, have emerged as potential modulators of metabolic health. Evidence supports benefits on glucose and hepatic metabolism in diet-induced obesity. However, reported effects vary widely across polyphenol sources and experimental design, and the key physiological mediators of benefit in established obesity remain incompletely defined. Moreover, ambient temperature, a key determinant of metabolic phenotype that may influence therapeutic responses, is rarely considered. Here we aim to determine the metabolic effects and mechanisms of action of a proan

Cranberry and Sumac Extracts Exhibit Antibacterial and Anti-Adhesive Effects Against Streptococcus pyogenes.

Group A Streptococci (GAS) or Streptococcus pyogenes is responsible for acute bacterial pharyngitis in children as well as adults. Streptococcal pharyngitis is initiated by successful attachment and colonization of the bacteria, followed by the establishment of the biofilm in various environments. In this study, we examined the antibacterial activities of in-house prepared aqueous and ethanolic extracts of 10 Atlantic Canada fruits in the context of minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), time-kill kinetics, and adhesion inhibition properties against S

Phenolic composition and antioxidant potential of grain legume seeds: A review.

Legumes are a good source of bioactive phenolic compounds which play significant roles in many physiological as well as metabolic processes. Phenolic acids, flavonoids and condensed tannins are the primary phenolic compounds that are present in legume seeds. Majority of the phenolic compounds are present in the legume seed coats. The seed coat of legume seeds primarily contains phenolic acids and flavonoids (mainly catechins and procyanidins). Gallic and protocatechuic acids are common in kidney bean and mung bean. Catechins and procyanidins represent almost 70% of total phenolic compounds in

Gas Chromatography/Mass Spectrometry Chemical Profiling of Volatile Compounds from Cranberry Plant Byproducts as Potential Antibacterials, Antifungals, and Antioxidants.

The increasing resistance of microorganisms to currently used antimicrobials requires the urgent development of new effective treatments. Plant-based natural products can be an alternative solution. The aerial plant parts of the cranberry (Vaccinium macrocarpon) present a potential new source of antimicrobial secondary metabolites. Volatile essential oils were extracted from Stevens, Early Black, and Mullica Queen variety plants by steam distillation (SD) and the Clevenger method (CM), and their profiles were characterized by GC-MS. The extracts and two identified constituents, cinnamaldehyde

Cranberry phytochemicals inhibit glycation of human hemoglobin and serum albumin by scavenging reactive carbonyls.

Protein glycation caused by sugars and reactive carbonyls is a contributing factor to diabetic complications, aging, and other chronic diseases. The objective of this study was to investigate the inhibitory effects of cranberry phytochemicals on protein glycation. Cranberries, purified to yield sugar-free phytochemical powder, were fractionated into ethyl acetate and water fractions. Water fraction was further separated into water fraction I, II, and III on a Sephadex LH-20 column. Cranberry phytochemical powder and its fractions significantly inhibited the formation of glycated hemoglobin. Th

Synergistic Inhibition of Candida albicans by Cranberry Proanthocyanidins and Probiotics: Novel Strategies for Vulvovaginal Candidiasis Treatment.

Vulvovaginal candidiasis (VVC) is a common gynecological condition primarily caused by Candida albicans. The excessive use of antifungal drugs has led to increased drug resistance, necessitating the search for alternative therapies. This study investigates the synergistic antifungal effects of cranberry proanthocyanidins (PACs) and probiotics against C. albicans. PACs were prepared at different concentrations (low, medium, high) and tested alone and in combination with multi-strain probiotics, including Lactobacillus rhamnosus and Lactobacillus plantarum. The antifungal activity of their cell-

Proanthocyanidins mitigate bile acid-induced changes in GSTT2 levels in a panel of racially diverse patient-derived primary esophageal cell cultures.

Persistent and symptomatic reflux of gastric and duodenal contents, known as gastroesophageal reflux disease (GERD), is the strongest risk factor for esophageal adenocarcinoma (EAC). Despite similar rates of GERD and other risk factors across racial groups, EAC progression disproportionately impacts Caucasians. We recently reported that elevated tissue levels of the detoxification enzyme GSTT2 in the esophagi of Blacks compared to Caucasians may contribute protection. Herein, we extend our research to investigate whether cranberry proanthocyanidins (C-PAC) mitigate bile acid-induced damage and
