Hyperprolactinemia reduces vasoactive intestinal peptide in the anterior pituitary glands of rats.

The concentration of vasoactive intestinal polypeptide (VIP) in the anterior pituitary gland of female rats was significantly reduced by pituitary implants, prolactin-secreting tumours or dopamine blockade-induced hyperprolactinemia. Oestradiol implants increased plasma prolactin (PRL) to 12 times and increased pituitary VIP to 3.5 times that of controls after one week. The dopamine agonist bromocriptine significantly reduced pituitary VIP at doses within the clinically used range. These results provide evidence for a suppressive effect of plasma PRL on pituitary VIP, which may be effected by

Substance P and vasoactive intestinal peptide in bullous and inflammatory skin disease.

Immunoreactivity (IR) of substance P (SP) and Vasoactive Intestinal Peptide (VIP) were determined by radioimmunoassay in serum of 56 patients with inflammatory skin diseases, in blister fluid of 40 patients with spontaneous blisters and 31 subjects with induced skin blisters. Serum concentrations of SP-IR and VIP-IR were mostly low or non-detectable. Spontaneous blisters contained high amounts of SP-IR, particularly in bullous pemphigoid and in some inflammatory dermatoses, while VIP-IR levels usually were low. Suction blisters from inflamed but not from normal skin often contained SP-IR but m

Vasoactive intestinal peptide advances chondrogenesis and modulates pathogenic mediators in human osteoarthritis.

Current therapies for osteoarthritis (OA) focus on symptom management, rather than halting disease progression. Vasoactive intestinal peptide (VIP) has shown promising effects in musculoskeletal diseases, preserving joint integrity and modulating inflammation. This study investigates the potential of VIP to promote chondrogenic differentiation of human bone marrow mesenchymal stem cells (BM-hMSC) and to modulate inflammatory and cartilage extracellular matrix (ECM)-degrading mediators in human osteoarthritis articular chondrocytes (OA-hAC). BM-hMSC from healthy donors were cultured in 3D pelle

Vasoactive intestinal peptide (VIP) localization in the epididymis of two vertebrate species.

VIP and its receptors (VPACRs) are largely investigated in vertebrate testis, as well as their functions in the control of spermatogenesis and steroidogenesis. By contrast, a few data are available about the presence and role of VIP in the epididymis. The aim of the present paper was to investigate the localization of VIP and its receptors in the epididymis of two vertebrates: Podarcis sicula, a seasonal reproducer, and Rattus rattus, a continuous reproducer. By immunohystochemical investigation, we demonstrated for the first time that VIP and its receptors are widely represented in the epidid

Parasympathetic vasoactive intestinal peptide (VIP): a likely contributor to clozapine-induced sialorrhoea.

OBJECTIVE: The parasympathetic transmitter vasoactive intestinal peptide (VIP) increases salivary gland blood flow and evokes protein secretion and, in some species, such as rats, a small fluid secretion. It interacts synergistically with muscarinics for protein and fluid output. Human salivary acini are supplied with VIP-containing nerves. We hypothesise that VIP and clozapine, acting together, evoke a volume of saliva greater than the sum of those induced by each drug given separately. It was further considered whether, in the current test situation, circulatory events influenced the magnitu

Therapeutic potential of vasoactive intestinal peptide and its receptors in neurological disorders.

Vasoactive intestinal peptide (VIP) is a basic 28 amino acid peptide that binds to a member of the class II family of G protein-coupled receptors (GPCRs). It is widely expressed throughout the body and plays an important role in numerous biological functions. VIP acts via three different GPCRs: VPAC1, VPAC2, and PAC1, which have been identified in various tissues, including brain, lung, kidney, gastrointestinal tract, tongue, and also on immunocompetent cells such as macrophages and lymphocytes. There is mounting evidence that VIP expression and signaling is altered in numerous neurological di

Brain delivery of vasoactive intestinal peptide (VIP) following nasal administration to rats.

The aim of this work was to study in rats the nasal route for the brain delivery of the vasoactive intestinal peptide (VIP) neuropeptide. After evaluating VIP stability in solutions obtained from nasal washes, the effect of formulation parameters (pH 4-9, 0-1% (w/v) lauroylcarnitine (LC), hypo- or isoosmolality) on the brain uptake of intranasally administered VIP (10(-8)M)/125I-VIP (300,000 cpm/ml) was studied, using an in situ perfusion technique. Brain radioactivity distribution was assessed by quantitative autoradiographic analysis. Results were compared to intravenously administered VIP.

Vasoactive intestinal peptide enhances its own expression in sympathetic neurons after injury.

Neurons in the adult rat superior cervical sympathetic ganglion (SCG) dramatically increase their content of vasoactive intestinal peptide (VIP) and its mRNA after axotomy in vivo and after explantation. Because the VIP gene contains a functional cAMP response element, the effects of cAMP-elevating agents on VIP expression were examined. VIP, forskolin, or isoproterenol increased cAMP accumulation in explanted ganglia. Secretin, a peptide chemically related to VIP, or forskolin increased VIP levels above those seen in ganglia cultured in control medium, whereas treatment with VIP or secretin i

Cloning and functional expression of a human neuroendocrine vasoactive intestinal peptide receptor.

Vasoactive intestinal peptide (VIP) is a potent mediator of gastrointestinal, nervous, pulmonary, vascular and immune functions. A cDNA was obtained from human HT29 intestinal epithelial cells and found to encode a 457 amino acid, 52 kDa VIP receptor. Transfection of the cDNA into COS-7 cells and 293 cells resulted in expression of specific saturable binding of VIP with a Kd of 0.8 nM, and induction of increases in intracellular cAMP by VIP with an EC50 of 1 nM. The human VIP receptor is homologous to other G protein-coupled receptors of the secretin-parathyroid hormone receptor family. A 2.8

Nucleotide regulation of vasoactive intestinal peptide binding to bovine thyroid plasma membranes.

The specific binding of vasoactive intestinal peptide (VIP) to bovine thyroid plasma membranes is inhibited by guanine nucleotides. Guanosine 5'-triphosphate (GTP) and the non-hydrolyzable GTP analogs guanosine 5'-beta,gamma-imidotriphosphate (Gpp(NH)p) and guanosine 5'-O-(3-thiotriphosphate) (GTP-gamma-S) inhibited markedly the binding of VIP to its receptors. This inhibition was higher with GTP than with Gpp(NH)p and GTP-gamma-S and was due to an increase of the rate of dissociation of peptide bound to membranes. Other nucleotides did not show any effect.

Expression and localization of VPAC1, the major receptor of vasoactive intestinal peptide along the length of the intestine.

Vasoactive intestinal peptide (VIP) is an endogenous neuropeptide with a broad array of physiological functions in many organs including the intestine. Its actions are mediated via G protein-coupled receptors, and vasoactive intestinal peptide receptor 1 (VPAC1) is the key receptor responsible for majority of VIP's biological activity. The distribution of VPAC1 along the length of the gastrointestinal tract and its subcellular localization in intestinal epithelial cells have not been fully characterized. The current studies were undertaken to determine VPAC1 distribution and localization so th

Vasoactive Intestinal Peptide: A Neuropeptide that Plays an Important Role in Parkinson's Disease.

Parkinson's disease (PD) is primarily characterized by rigidity and tremor, which are pathologically associated with α -synuclein aggregation, especially in dopaminergic neurons in the midbrain. As it is a multi-factorial disease, there are currently no effective treatments but only mitigative therapies. Vasoactive intestinal peptide (VIP), a 28-amino acid neurotransmitter, is widely distributed in both the central and peripheral nervous system with a broad biological effect. Studies have shown that VIP exhibits anti-inflammatory, antioxidant, and anti-apoptotic effects and regulates glial cel

Vasoactive Intestinal Peptide Tumor as the Cause of Persistent Diarrhea: A Diagnostic Challenge.

Although chronic diarrhea is frequent, some of its causes are rare, namely, neuroendocrine tumors (NETs). Due to their rarity and non-specific symptoms, such as diarrhea, they are often underdiagnosed. An 80-year-old woman presented to the emergency department due to emesis and watery diarrhea. Blood tests showed acute kidney injury, hypokalemia, and metabolic acidosis. An abdominal computed tomography revealed a 51 mm pancreatic lesion. An endoscopic ultrasound-guided biopsy raised the hypothesis of a NET. The patient refused surgery and was lost to follow-up. At the eighth hospitalization, 1

Prospect of vasoactive intestinal peptide therapy for COPD/PAH and asthma: a review.

There is mounting evidence that pulmonary arterial hypertension (PAH), asthma and chronic obstructive pulmonary disease (COPD) share important pathological features, including inflammation, smooth muscle contraction and remodeling. No existing drug provides the combined potential advantages of reducing vascular- and bronchial-constriction, and anti-inflammation. Vasoactive intestinal peptide (VIP) is widely expressed throughout the cardiopulmonary system and exerts a variety of biological actions, including potent vascular and airway dilatory actions, potent anti-inflammatory actions, improvin

Regulation of vasoactive intestinal peptide and substance P in the human pyloric sphincter.

BACKGROUND/AIMS: Brain-gut hormones (i.e., neuropeptides) such as vasoactive intestinal peptide (VIP) and substance P (SP) have been shown to exist in the enteric nervous system (ENS) of the pyloric sphincter (PS) in studies of immunohistochemistry. To clarify the role of neuropeptides in the ENS of the normal human PS, we investigated the enteric nerve responses to VIP and SP in normal human PS specimens in vitro. METHODOLOGY: Normal human PS specimens derived from 45 patients with early gastric cancer (35 men and 10 women aged from 44 to 65 years, average 57.4 years) were used. A total of 1

Vasoactive intestinal peptide as a new drug for treatment of primary pulmonary hypertension.

Primary pulmonary hypertension is a fatal disease causing progressive right heart failure within 3 years after diagnosis. We describe a new concept for treatment of the disease using vasoactive intestinal peptide, a neuropeptide primarily functioning as a neurotransmitter that acts as a potent systemic and pulmonary vasodilator. Our rationale is based on the finding of a deficiency of the peptide in serum and lung tissue of patients with primary pulmonary hypertension, as evidenced by radioimmunoassay and immunohistochemistry. The relevance of this finding is underlined by an upregulation of c

Immunohistochemical localization of vasoactive intestinal peptide (VIP) in the circumventricular organs of the rat.

The indirect peroxidase-antiperoxidase immunohistochemical technique was used to investigate the possible presence of vasoactive intestinal peptide (VIP) in the circumventricular organs of the rat. Considerable numbers of VIP-immunoreactive fibers were seen in the pineal gland. A moderate amount of VIP-immunoreactive fibers was present in the median eminence, the posterior lobe of the pituitary and the area postrema, but only few fibers were found in the organum vasculosum laminae terminalis. No immunoreactivity was observed in the subfornical organ or the subcommissural organ. The circumventr

Vasoactive intestinal peptide in canine portosystemic shunt in the absence of portal hypertension.

BACKGROUND: The congenital portosystemic shunt (PSS) is a common vascular anomaly in dogs. Vasoactive intestinal peptide (VIP) is produced in various organs (including the small intestine, large intestine, and pancreas), leading to abdominal vasodilation, increased blood flow, increased pancreatic blood flow, and promotion of pancreatic endocrine and exocrine secretions. However, there have been no reports on the concentration of VIP in the portal and peripheral veins in canine PSS. AIM: The aim of this pilot study was to evaluate whether dogs with PSS have a different VIP concentration in th
