Photochemical internalization with verteporfin-liposomes enhances oxaliplatin retention and efficacy in models of pancreatic cancer.

Resistance to chemotherapy remains a significant challenge for the treatment of pancreatic cancer. In addition to conventional therapeutic strategies, photodynamic therapy (PDT) has emerged as a compelling alternative for pancreatic cancer as it synergizes with various chemotherapeutics such as irinotecan, and oxaliplatin. However, the exact mechanisms by which PDT overcomes oxaliplatin resistance remains elusive. In this study, we explore the hypothesis that PDT improves oxaliplatin efficacies via photochemical internalization, in which lysosome-borne verteporfin/benzoporphyrin derivative (BP

Verteporfin, an inhibitor of nuclear YAP, improved multi-ciliated cell differentiation in the airway epithelium.

BACKGROUND: Impaired multi-ciliated cell (MCC) differentiation, observed in various airway diseases, causes prolonged inflammation and further airway complications due to malfunction of the airway defense mechanism. YAP/TAZ-TEAD signaling mediates multiple mechanisms associated with MCC differentiation. However, YAP reportedly has both negative and positive roles in MCC differentiation, putatively due to its dual functions in the cytoplasm and nucleus. We hypothesize inhibition of nuclear YAP, while preserving cytoplasmic YAP functions, restores MCCs in various airway disorders. Here, we inves

Targeted and Synergic Glioblastoma Treatment: Multifunctional Nanoparticles Delivering Verteporfin as Adjuvant Therapy for Temozolomide Chemotherapy.

Despite advances in cancer therapies, glioblastoma multiforme treatment remains inefficient due to the brain-blood barrier (BBB) inhibitory activity and to the low temozolomide (TMZ) chemotherapeutic selectivity. To improve therapeutic outcomes, in this work we propose two strategies, (i) photodynamic therapy (PDT) as adjuvant treatment and (ii) engineering of multifunctional theranostic/targeted nanoparticles ( m-NPs) that integrate biotin as a targeting moiety with rhodamine-B as a theranostic agent in pluronic P85/F127 copolymers. These smart m-NPs can surmount the BBB and coencapsulate mul

Half-dose verteporfin photodynamic therapy for bullous variant of central serous chorioretinopathy: a case report.

INTRODUCTION: Central serous chorioretinopathy is characterized by serous neurosensory detachment of the macula and it usually resolves spontaneously with good visual prognosis. In some patients, however, the serous retinal detachment might be very extensive and can result in bullous exudative retinal detachment. We evaluated the use of half-dose verteporfin photodynamic therapy for the treatment of bullous retinal detachment in idiopathic central serous chorioretinopathy. CASE PRESENTATION: We report the case of a 51-year-old Chinese man who presented with blurred vision in his right eye and

Contact transcleral ciliary body photodynamic therapy with verteporfin in pigmented rabbits: effect of repeated treatments.

We studied the effect on the intraocular pressure (IOP) and the ciliary body (CB) morphology after four consecutive contact transcleral photodynamic treatments of the ciliary body (CB-PDT) with verteporfin in pigmented rabbits. Twenty-two pigmented rabbits underwent CB-PDT (study group), performed once (six rabbits) or repeated for up to four times (16 rabbits). Six additional rabbits received only laser treatment without photosensitizer administration (control group). CB-PDT was performed in one eye in rabbits of the study group, with the fellow eye serving as internal control. Verteporfin do

Photo-controlled co-delivery of verteporfin and acriflavine via platelets achieves potentiated glioblastoma-targeted photodynamic therapy.

The potential of glioblastoma (GBM) photodynamic therapy (PDT) is limited by inadequate GBM drug delivery, and the development of resistance to PDT as a result of cellular damage response that critically involves the hypoxia-inducible factor-1α (HIF-1α) and yes-associated protein (YAP). Herein, addressing these challenges, we demonstrated a strategy of photo-controlled, targeted co-delivery of verteporfin (Vp), a photosensitizer and YAP inhibitor as well, and acriflavine (Af), a HIF-1α inhibitor via platelets for enhanced GBM PDT. Mouse platelets were separately loaded with Vp (Vp@Plt) and Af

Verteporfin ameliorates fibrotic aspects of Dupuytren's disease nodular fibroblasts irrespective the activation state of the cells.

Dupuytren's disease is a chronic, progressive fibroproliferative condition of the hand fascia which results in digital contraction. So far, treatments do not directly interfere with the (myo)fibroblasts that are responsible for the formation of the collagen-rich cords and its contraction. Here we investigated whether verteporfin (VP) is able to inhibit the activation and subsequent differentiation of DD nodular fibroblasts into myofibroblasts. Fibroblasts were isolated from nodules of 7 Dupuytren patients. Cells are treated (1) for 48 h with 5 ng/ml transforming growth factor β1 (TGF-β1) follo

Verteporfin inhibits the cGAS-STING pathway and improves the tumor microenvironment during cisplatin treatment in hepatocellular carcinoma.

BACKGROUND: Cisplatin (DDP) is a clinical first-line chemotherapy drug for hepatocellular carcinoma (HCC), but treatment is often ineffective due to drug resistance. Yes-associated protein 1 (YAP1) is a critical regulator/factor in HCC tumor progression. Our previous research showed that DDP promoted the expression of YAP1 in mice bearing H22 cell in situ liver tumors, which might be related to the poor therapeutic effect of DDP. METHODS AND RESULTS: DDP could inhibit tumor growth and decrease tumor volume in DEN/TCPOBOP-induced HCC mice, increase the number of CD8+ T cells in the tumor, redu

Combination Treatment of Docetaxel Liposomes and Sonodynamic Therapy With Verteporfin Liposomes in Patients With Cancer: A Report of Three Cases.

Despite advances in medical technology, cancer treatment remains challenging. Our research focused on combining microtubule polymerization inhibitors with sonodynamic therapy (SDT) as a promising approach for cancer. SDT is a minimally invasive modality that uses tumor-selective sonosensitizers to induce the death of malignant and tumor-associated cells within the tumor microenvironment through targeted ultrasound irradiation. In this study, verteporfin was selected because of its dual properties of high ultrasound sensitivity and the ability to inhibit the yes-associated protein. Liposome-enc
