The antipsychotic drug fluphenazine was obtained in a dry powder form and was screened with respect to 482 strains of bacteria, which included 170 Gram-positive and 326 Gram-negative strains. Nutrient agar plates containing increasing concentrations of fluphenazine (0-200 micrograms/ml) were used for the determination of the minimum inhibitory concentration (MIC) which was demonstrated by inoculating a loopful of an overnight peptone water culture of the organism on nutrient agar plates and determining the MIC against a control. Fluphenazine was detected to possess pronounced action against bo

Photobacterium damselae subsp. damselae (PDD), a halophilic, Gram-negative bacterium of the family Vibrionaceae, infects a wide range of aquatic organisms. PDD isolates originating from fish have been characterized in detail, but much less is known about those infecting invertebrates. Currently, only 3 complete PDD genomes isolated from Kuruma shrimp Penaeus (Marsupenaeus) japonicus from Japan are publicly available, severely limiting comparative studies of host adaptation. To address this, we present complete, plasmid-resolved genome assemblies of 4 novel PDD strains (TUMSAT-PD1, -PD2, -PD3,

Fish contamination has been extensively investigated along the Saudi coasts, but studies pertaining to bacterial pathogens are scarce. We conducted qualitative assessment and molecular identification of culture-dependent bacteria in 13 fish species from three coastal sites and a local fish market in Jeddah, Saudi Arabia. Bacterial counts of gills, skin, gut and muscle were examined on agar plates of Macconkey's (Mac), Eosin Methylene Blue (EMB) and Thiosulfate Citrate Bile Salts (TCBS) culture media. Bacterial counts significantly differed between species, sources and feeding habits of examine

The emergence of antimicrobial resistance in environmental bacteria threatens therapeutic efficacy in clinical settings. Shewanella algae, historically regarded as a marine saprophyte, is increasingly recognized as an emerging opportunistic pathogen. In this study, we analyzed 86 S. algae isolates from Spain (19 clinical and 67 environmental) and integrated them with 178 publicly available genomes to explore antimicrobial susceptibility patterns and genomic diversity. Penicillins and fosfomycin consistently showed poor activity, whereas piperacillin/tazobactam, third- and fourth-generation cep

Dog bites are a source of zoonotic infections to humans, such as pasteurellosis and meningitis. Zoonotic bacterial identification and their antibiotic susceptibility assessment are key towards the successful management of such infections. This study isolated and identified zoonotic bacterial species from the oral cavities of indigenous dogs and also determined their antibiotic susceptibility profile. Oral swab samples collected from 54 indigenous dogs (domestic [36] and stray [18]) were cultured on agar media, and then on selective-differential media. The colony morphology and conventional bio

Enterobacterial common antigen (ECA) was isolated from a number of selected species (including Salmonella montevideo, Shigella sonnei and Plesiomonas shigelloides) using the extraction method described by Männel and Mayer [Eur. J. Biochem. 86, 361-370 (1978)]. ECA of all these species behaved identically in enzyme-linked immunosorption assay (ELISA) and in its inhibition using monoclonal anti-ECA antibodies. Immunoblotting showed a ladder-like pattern of at least 20 bands for all preparations tested. ECA modified at its lipid moiety (e.g. by phospholipases A2 and D or by mild acid hydrolysis)

The burden of antimicrobial resistant bacteria and resistance genes exists in diverse niches, where they can be disseminated to other ecosystems, including the food chain. This study was conducted to investigate the distribution of antibiotic resistant and extended spectrum beta-lactamase (ESBL) producing Enterobacteriaceae in pit latrines in urban and suburban communities in Lagos State, Nigeria. Using standard microbiological methods, bacteria strains were isolated from twenty sludge samples obtained from three sites. Isolates were molecularly identified by 16S rRNA gene amplicon sequencing

Ascites bacterial burden is associated with poor clinical outcomes in patients with end-stage liver disease. However, the impact of ascitic microbial composition on clinical course was still not clear. In this study, the ascitic microbiota composition of 100 cirrhotic patients with culture-negative and non-neutrocytic ascites were researched with 16S rRNA pyrosequencing and enterotype-like cluster analysis. Results: By characterizing the ascitic microbial composition, two distinct microbial clusters were observed, Cluster 1 (86 patients) and Cluster 2 (14 patients). Cluster 1 showed lower micr

The mucosal surfaces of aquatic animals serve as a functional barrier between the host and the aquatic environment, hosting diverse microbial communities that play pivotal roles in host health. In this study, amplicon libraries of the 16S rRNA gene were constructed to investigate the compositional differences between the microbial communities presented in four mucosal surfaces (gills, skin, gut, and ovary) of spawning female olive flounder. To elucidate the unique roles of commensal microbes in external and internal organs, we used PICRUSt2 and ALDEx2, respectively, to predict metabolic functi

Photobacterium damselae subsp. piscicida, the causative agent of fish pasteurellosis, produces a siderophore which is distinct from that produced by P. damselae subsp. damselae. Using suppression subtractive hybridization, a subsp. piscicida-specific DNA region of 35 kb was identified in strain DI21, and 11 genes were defined: dahP, araC1, araC2, frpA, irp8, irp2, irp1, irp3, irp4, irp9 and irp5. The sequence of the predicted proteins encoded by these genes showed significant similarity with the proteins responsible for the synthesis and transport of the siderophore yersiniabactin, encoded wit

Antibiotics, as typical emerging contaminants posing significant ecological risks, are frequently detected in global coastal environments. Their biotransformation in marine environments depends on the stress responses of functional microbial communities, while the underlying response mechanisms remain unclear. Through integrated metagenomic and metabolomic approaches, this study systematically investigated the structural and functional responses of marine microbial communities during the co-metabolic degradation of sulfamethoxazole (SMX), a representative antibiotic. A marine microbial consort

Northern populations of Arctic char (Salvelinus alpinus) can be anadromous, migrating annually from the ocean to freshwater lakes and rivers in order to escape sub-zero temperatures. Such seasonal behavior demands that these fish and their associated microbiomes adapt to changes in salinity, temperature, and other environmental challenges. We characterized the microbial community composition of anadromous S. alpinus, netted by Inuit fishermen at freshwater and seawater fishing sites in the high Arctic, both under ice and in open water. Bacterial profiles were generated by DNA extraction and hi

Vibrio parahaemolyticus belongs to the halophilic genus of Vibrionaceae family that inhabits coastal and marine environments and is a major food-borne pathogen. In the Gulf Cooperation Council (GCC) countries and Saudi Arabia in particular, there is a lack of information regarding the detection of pandemic clone or serovariants of V. parahaemolyticus pandemic clones. Here, 400 seawater samples were collected and examined for the presence of V. parahaemolyticus from 10 locations along the coast of Eastern Province in Saudi Arabia. The recovered isolates were serotyped, and studied for antimicro

Aquacultivated sea cucumbers often suffer from SKin Ulceration Diseases (SKUDs). SKUDs have been observed in six holothuroid species from nine countries. All SKUDs present a similar symptom-the skin ulceration-and can be induced by bacteria, viruses, or abiotic factors. We here provide an update on SKUDs in holothuroids and analyse the case of the SKUD observed in Holothuria scabra in Madagascar. Field observations revealed a seasonality of the disease (i.e. wintertime maximum peak). Morphological analyses of integument ulcers showed that sea cucumbers react by forming a collagen fibre plug. Vibrio mimicus, the species most similar to V. cholerae, is a microbe present in the natural environmental and sometimes causes diarrhea and internal infections in humans. It shows similar phenotypes to V. cholerae but differs in some biochemical characteristics. The molecular mechanisms underlying the differences in biochemical metabolism between V. mimicus and V. cholerae are currently unclear. Several V. mimicus isolates have been found that carry cholera toxin genes (ctxAB) and cause cholera-like diarrhea in humans. Here, the genome of the V. mimicus isolate SX-4, which carries an intact C

Vibrio vulnificus is a Gram-negative, motile, curved bacillus of the family Vibrionaceae that is a rare cause of gastroenteritis, septicemia, and wound infections in humans. V. vulnificus is halophilic, flourishes in warm temperatures, and is part of the bacterial flora of the marine environment. The location of our health care setting, on the Gulf of Mexico, has given us the opportunity to observe a wide variety of clinical presentations of infections caused by this organism. In the first case, a 27-year-old man struck by lightning while windsurfing was found pulseless in the water and was re

BACKGROUND/OBJECTIVES: Currently, readily available data on antimicrobial resistance in bacteria colonizing farmed Atlantic salmon (Salmo salar L.) are limited. Recent data indicate that the gastrointestinal tract mucosa of adult Atlantic salmon farmed in Tasmania (Australia) is consistently colonized by Aliivibrio species. Aliivibrio and other Vibrionaceae may contribute to gut dysbiosis through overgrowth. We investigated Aliivibrio isolates and other Aliivibrio species to link antimicrobial resistance (AMR) phenotypes with the presence of antimicrobial resistance genes (ARGs).
METHODS: We
